Objective To explore the correlation between chromosome fragile site in peripheral blood lymphocytes and proto-oncogene expression rate in patients with hepatocellular carcinoma.Methods The peripheral blood lymphocyte chromosome preparation was made routinely.The cells were cultivated with low concentrations of calf serum and folic acid,higher pH medium and G-banding method.The fragile sites in 20 patients with hepatocellular carcinoma(experimental group) and 18 healthy subjects(control group) were exactly located and analyzed.(Results The) fragile sites in experimental group were determined as 44 species mainly distributed on the chromosomes of A,B,C group.The expression rate of the fragite site in experimental group(9.90%) was significantly higher than that in control group(0.67%)(P
2.Effect Evaluation and Mechanism Study of Gastrodin against Migraine of Rat

Objective To research the effects and mechanism of gastrodin against migraine and on single amine neurotransmitter. Methods The model of nitroglycerin migraine was made after rat treated by gastrodin for 5 days, the time points of ear rubescence and the times of scratch head were observed to take as evaluation index of pharmacodynamic action, the concentration of 5-HT, NE and DA were measured in the brain with HPLC. Results Gastrodin could obviously prevent the attack of migraine and increase the concentration of 5-HT, DA and NE in the brain. Conclusions The effects of gastrodin against migraine is related with the concentration of 5-HT, DA and NE in the brain, gastrodin can regulate neurosystematic functions and enhance the secretion and metabolism of neurotransmitters.
3.Evaluation and Analysis of Drug Resistance of Escherichia Coli and Relationship of the Antibacterial Use

Objective:To investigate the variation of the drug resistance of Escherichia coli(Eco)against antibacterial in clinical specimens in the hospital, and to evaluate the simultaneous use of antibacterial, to explore to provide the reference for the reasonable clinical use of antibacterial. Method:Eco was separated from all kinds of clinical specimens of out-patients and in-patients in the hospital from August of 2005 to April of 2006,and ESBLs was detected with Kirby-Bauer (K-B) disk diffusion method. It was also counted that the antibacterial drugs ranked the first 30 in sales per month and their use frequencies in the same period in the hospital. Result:The detectable rate of producing ESBLs Eco was 58.6%. Eco was completely sensitive to imipenem. It had lower resistance rates, only 2% and 4% , against piperacillin/tazobactam and cefoperazone/sulbactam; and had 59% and 55% resistance rates respectively against cefotaxime claforan and ceftazidime pentahydrate in the third generation cephalosporin. The third generation cephalosporin and quinolones kept high in sales and their use frequencies ranked the first three. The use of the third generation cephalosporin with anhydrase inhibitors also had an uptrend. Conclusion:There is a direct relation between the use of a great deal of antibacterial and the imitative drug resistance of Eco. It is necessary to strengthen its clinical management and to raise the level of drug use.
4.Relationship between activity and virulence of secretory acid proteinase and extracellular phosphlipase of Candida albicans

Objective To study the relationship between activity and virulence of secretory acid proteinase and extracellular phosphlipase of different Candida albicans isolated from various specimens.Methods 190 strains of Candida albicans were isolated from various specimens(sputum,blood,thrush,wound,secretion of vaginitis) of patients with Candida albicans infection in hospital.Milk-plate medium and egg-yolk medium were used respectively to test the activities of secretory acid proteinase and extracellular phosphlipase of Candida albicans.The suspension of two forms of Candida albicans(5?106CFU?mL-1) were injected to vein of tail in mice respectively in virulence test.The mortality and mean survival time of mice were observed in 1 month.The virulence was appreciated with the mean survival time of mice.Results All of 190 strains,the positive detectable rates of secretory acid proteinase and extracellular phosphlipase were 83.68% and 85.26%,respectively.Animal experimental results showed the activities of secretory acid proteinase and extracellular phospolipase in hyphal forms were significantly higher than that in spore form(P

Objective: To explore the mechanism of Fujian Tablet in promoting neural regeneration by observing the effect of Fujian Tablet on the expression of slit in MCAO rats at different stages. Methods: 240 SD rats were randomly divided into normal control, sham operation, model and medicine groups, which were randomly divided into five subsets with 12 rats according to the day 3 and week 1, 2, 4, 6 stages. The rat models with middle cerebral artery occlusion were successfully established by the improved Longa EZ. Objective To investigate the effects of 15-deoxy-?12,14-prostaglandin J2(15d-PGJ2) and DDP on the growth of human pulmonary carcinoma PLA-801D cells and the mechanisms of apoptosis.Methods The human pulmonary carcinoma PLA-801D cells were selected and added to each well of 96-well place and cultivated for 24 h.Then the cells were treated with different concentrations of 15d-PGJ2(0,5,10,20,40 and 80 ?g?L-1) or 15d-PGJ2 combined with DDP(3 mg?L) for 24 h.0 ?g?L-1 15d-PGJ2 group was control group.The morphological changes of cells were observed under inverted microscope.Microculture tetrazolium(MTT)dye was applied to detect the proliferation of the human pulmonary carcinoma PLA-801D cells treated with 15d-PGJ2 and DDP.Diphenylamine assay(DPA) was used to evaluate the activation.Flow cytometry assay(FCM) was used to detect the apoptosis proportion and the changes of cell cycle.Results When the human pulmonary carcinoma PLA-801D cells were treated with low-concentration 15d-PGJ2 alone(5,10 and 20 ?g?L-1),no significant difference was observed in the inhibitory rate of cell growth and the apoptotic indexes such as the apoptosis proportion,the percent of DNA fragmentation and the activity of caspase-3 compared with control group(P
7.Establishment of a SD Rat Model with Vasomotor Abnormalities Induced by Dopamine and Nitroglycerin

Objective: To determine the appropriate time of early administration of dopamine (DA) before nitroglycerin (GTN) for establishing rat model with vasomotor abnormalities. Methods: Rats were divided into 5 groups: normal group, DA Group and other 3 groups in accordance with DA at different time points (10, 30, 45 min) plus GTN .Common carotid artery (CCA) blood flow velocity was dynamically measured with pulsed Doppler flow probe to evaluate the influence of DA, GTN on blood flow velocity rate(BFVR). Results:The blood flow velocity rate induced by DA of All medication administration was different from the normal group within 10 minutes(P < 0.05). However, the differences were not statistically significant after 11 min(P > 0.05). The BFVR of 10min interval group had differences with the DA group at 45min, 50min, 55min, 60min, 65min and 70min(P < 0.05 or P < 0.01). CCA BFVR of 45 min interval group was statistically significant different from the DA group at 80, 85, 90, 95 min and 100 min(P < 0.05). No statistically significant differences existed between CCA BFVR of 30min interval group and DA group (P > 0.05). Conclusion: Subcutaneous injection of DA, 45m in after injection of GTN can induce vasomotor abnormalities in rat with phase characters. The model may be useful for vasoactive screening of anti-migraine drugs.
8.Effect of Fujian Tablets on Expression of Microtubule-associated Protein 2 in Cerebral Infarction Rats at Different Time Points

Objective To observe the effect of Fujian Tablets(FT),a prescription with the actions of nourishing liver and kidney,on the expression of microtubule-associated protein 2(MAP-2) in cerebral infarction rats at different time points,and to explore its mechanism on promoting nerve cell differentiation.Methods A total of 240 male Sprague Dawley rats were divided into normal control group,sham-operation group,model group and FT group.Focal cerebral ischemia models were established by occlusion of the middle cerebral artery according to the reformed Longa method in rats,and were subdivided into 3-,7-,14-,28-,and 42-day cerebral infarction subgroups.FT group was given FT in the dose of 9 g?kg-1?d-1,and the other groups were given the same dose of distilled water.Results In the model group,MAP-2 expression in the hippocampus increased and reached a peak on day 3,which were notably different from that in the normal control group(P0.05).In FT group,MAP-2 expression increased on day 3,reached a peak on day 7,remained high level till day 14,and then decreased,the difference being significant compared with the model group(P
9.Safety evaluation of dexmedetomidine sedation in ICU patients undergoing mechanical ventilation

Objective To observe the occurrence of cardiovascular adverse events in patients undergoing mechanical ventilation with dexmedetomidine sedation,and to evaluate its safety in intensive care unit (ICU).Methods A prospective randomized controlled trial was conducted.Adult critical patients undergoing mechanical ventilation over 48 hours admitted to ICU of Zigong First People's Hospital in Sichuan Province were enrolled.The patients were divided into dexmedetomidine group (Dex group) and midazolam group (Mid group) according to the randomise number generated by computer.The patients in both groups were given slow intravenous infusion of 0.05 mg/kg midazolam and 1-2 μg/kg fentanyl to induce anesthesia before tracheal intubation,followed by 0.06 mg· kg-1· h-1 midazolam and 20-50 μg· kg-1 · h-1 fentanyl or 0.1-0.2 μg· kg-1 · h-1 sufentanil for continuous intravenous pumping to maintain analgesia and sedation;on the next day,the patients in Dex group was given dexmedetomidine (with the initial dose of 0.4 μg· kg-1 · h-1,and maintenance dose of 0.1-0.7 μg· kg-1 · h-1),and midazolam was stopped half an hour later;the original sedation and analgesia plan remained unchanged in Mid group.The goal of sedation was to maintain a Richmond agitation-sedation scale (RASS) score of-2 to 1 or a Ramsay sedation score of 3 to 4;patients were given midazolam if obvious agitation occurred,in combination with propofol for sedation if necessary;wakeup test was performed every day.Observation endpoints included patients discharged from ICU,death or mechanical ventilation over 28 days.Occurrence of cardiovascular adverse events during sedation such as hypertension,hypotension,bradycardia,tachycardia and arrhythmia,dose of sedatives and analgesics,duration of mechanical ventilation,length of ICU stay and 28-day mortality were observed in two groups.Results A total of 383 patients were enrolled,with 190 patients in Dex group and 193 in Mid group.There was no statistically significant difference in general data such as gender,age,acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score and predicted mortality between two groups.Doses of midazolam,propofol and fentanyl in Dex group were reduced as compared with those of Mid group,while the dose of sufentanil was similar in two groups.The Ramsay score and RASS score of Dex group were significantly reduced as compared with those of Mid group (3.34± 0.63 vs.3.95 ± 0.86,-1.33 ±0.87 vs.-1.98 ± 1.27,both P < 0.01).Bradycardia was prominent in Dex group,which was observed in more than half of patients at the beginning of the treatment (1-2 hours),but the patients requiring isoprenaline treatment for heart rate lower than 50 bpm were less than that of Mid group (3.7% vs.5.2%,x 2 =0.506,P =0.477).The incidences of hypotension (45.3% vs.68.4%),tachycardia (16.3% vs.33.7%) and arrhythmia (14.7% vs.31.1%) in Dex group were significantly lower than those of Mid group (all P < 0.01),and no other cardiovascular adverse events such as cardiac arrest,sinus arrest,hyperglycemia or hypoglycemia were observed in two groups.There was no statistically significant difference in duration of mechanical ventilation between Dex group and Mid group [days:5 (2,28) vs.4 (2,56),Z =-1.917,P =0.055],but compared with Mid group,the length of ICU stay in Dex group was significantly prolonged [days:9 (2,67) vs.6 (2,57),Z =-4.302,P =0.000],and the 28-day mortality in Dex group was significantly reduced (22.6% vs.44.6%,x2 =20.610,P =0.000).Conclusion Long time dexmedetomidine sedation is safe in critical patients undergoing mechanical ventilation,which can significantly reduce cardiovascular adverse events except bradycardia,and lower the 28-day mortality.
10.Expression of ADM and ET-1 was decreased by Losartan in renal of diabetic rats

Objective To investigate the therapeutic effect of Losartan on ADM,ET-1 and their receptors in the kidney of diabetic rats.Methods Experimental diabetes was induced in uninephrectomized rats by STZ.The animals were divided into three groups: group A(control group),group B(diabetic group) and group C(Losartan-treated group).Immunohistochemistry,Western bloting,in situ hybridyzation and RT-PCR were used to detect the protein and mRNA expressions of ADM,ADMR,ET-1 and EDNR-A.Results Compared with group A,the expressions of ADM,ADMR,ET-1 and EDNR-A in group B significantly increased.After Losartan treatment,those indicators decreased.Conclusion The expressions of ADM,ET-1 and their receptors may be responsible for the renal protective effect of Losartan in diabetic rats.
