1.Study on the mechanism of different concentrations of celecoxib on pancreatic cancer cells resistance

Objective:Study on the inhibitory effect of celecoxib on pancreatic cancer SW1990 cells growth.Methods:The experiment was divided into two parts.(1) SW1990 cells were divided into fluorouracil group (50 μ g/mL),celecoxib group(50 μ mol/L),the combined group (celecoxib 50 μ mol/L+fluorouracil 50 μ g/mL),blank group,after 24 h culture of each group using the MTT assay SW1990 cell growth inhibition rate was detected by flow cytometry cell cycle distribution ratio;(2) were used at different concentrations (12.5,25.0,50.0 and 100.0 μ mol/L) of celecoxib,while using different concentrations above celecoxib were combined with 50 μ g/mL of fluorouracil treated SW1990 cells were treated 24 h,using real-time fluorescence polymerase chain reaction(RT-PCR) to detect differential expression Survivn RNA and detect the cyclooxygenase-2 (COX-2) enzyme-linked immunosorbent assay(ELISA),matrix metalloproteinase-14(MMP-14) expression differences.Results:Celecoxib group,fluorouracil group was significantly higher than the rate of the combination group cytostatic blank group (P<0.05),the inhibition rate of the combination group was significantly higher than the celecoxib group and fluorouracil group (P<0.05),celecoxib cloth inhibition rate was significantly lower than the fluorouracil group(P<0.05);significantly higher than the proportion of cells celecoxib group,fluorouracil group,joint group of G0/G1 phase control group (P<0.05),celecoxib group was significantly higher than the proportion of cells fluorouracil group G0/G1 phase combination group(P<0.05),significantly higher than the proportion of cells fluorouracil group G0/G1 phase celecoxib group (P<0.05);with the increasing concentration of celecoxib,celecoxib group,celecoxib+Survivn RNA fluorouracil group,COX-2 and MMP-14 protein levels were decreased and group differences were between statistically significant(P<0.05),Under the condition of the same concentration of celecoxib and fluorouracil plus celecoxib group RNA survivin,COX-2 and MMP-14 protein expression levels were significantly lower than that of celecoxib group,the difference has statistical significance (P<0.05).Conclusion:Celecoxib on the growth of pancreatic cancer SW1990 cells was significantly inhibited in a dose dependent manner,and its mechanism may be related to reduced Survivn RNA,COX-2 and MMP-14 protein expression.
2.The effect of bFGF on apoptosis of cultured human retinal pigment epithelial cells exposed to visible light

Objective To observe the effect of exogenous basic fibroblast growth factor (bFGF) on apoptosis of cultured human retinal pigment epithelial (RPE) cells exposed to visible light,and determine the role of bFGF, fibroblast growth factor receptor 1 (FGFR1),bcl 2 and caspase 3. Methods (2000? 500) lx cold white light was used. Exogenous bFGF was utilized during culture. Annexin annexin V fluorescein isothiocyanate/propidium iodium (V FITC/PI) labeling,flow cytometry, Immunocytochemical staining, enzyme associated absorb examing and reverse transcriptional polymerase chain reaction (RT PCR) were used to determine the apoptosis, the expression levels of bFGF, FGFR1, bcl 2, as well as the activity of caspase 3. Results No protective effect of bFGF was observed under the concentration 5 ng/ml. 3.Mesh infection after abdominal wall hernia mesh repair:an analysis of 14 cases

Objective To evaluate the causes,prevention and treatment of mesh infection after abdominal wall hernia mesh repair.Methods The clinical data of 14 mesh infections admitted from De-cember 1997 to December 2013 were analyzed retrospectively.There were one case of inguinal hernia with Lichtenstein repair,eleven cases of inguinal hernia with preperitoneal repair,one case of incisional hernia with Bard Composix Mesh and 1 case of parastomal hernia with mesh repair above the abdominal muscle. Based on prothetic materials and infection status,the infection meshes were removed in 4 cases and open dressing change were operated in 10 cases.Results All patients were healed and discharged without peri-operative death.There was no hemorrhage and bladder injury during the procedures.The time of dressing change ranged from 3 weeks to 6 months,with a median of 4 weeks.All patients were followed up for 8 to 64 months.One patient had a recurrence of abdominal incisional hernia.Conclusion There are many fac-tors related to mesh infection after mesh repair and preventing mesh infection is the most important.Once the infection occurs,the management should be individualized.Antibiotic treatment and surgical drainage can be effective in most polypropylene mesh(PPM)infection However,infected expanded polytetrafluoro-ethylene(ePTFE)mesh should be removed completely.
4.The effect of visible light on apoptosis of cultured human retinal pigment epithelium cells

Objective To observe the effect of visible light on apoptosis of cultured human retinal pigment epithelium (RPE) cells. Methods Being the light source,500lx,(2 000?500)lx and (3 400?200)lx cold white light were used. The duration of exposure was 0,6,12 and 24 hours respectively. Apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labelling, Annexin V flunorescein isothiocyanate/Propidium iodium labelling and flow cytometry. Results Apoptosis and necrosis were found in cultured human RPE cells which were exposed to visible light.(1)A significant increase in apoptotic and necrotic percentages was consistent with a higher light intensity.(2)Apoptosis was the main response to shorter (6 h and 12 h) exposure duration,while necrosis was more pronounced correlated to the prolongation of post exposure culture ( P 500 lx) increases the proportion of apoptosis and necrosis of human RPE cells in vitro.The extent is related to exposure intensity and duration. It demonstrates that the lower intensity and the shorter duration of exposure to light are, the more pronounced apoptotic percentages are observed,otherwise necrosis.
5.Influence of Sanchi gel on TIMP-1 and MMP-1 expression in epidural adhesion

Objective To explore the effects of Sanchi gel on MMP-1 and TIMP-1 expression in epidural adhesion after laminectomy. Methods Laminectomy model was set up in SD rats. 72 SD model rats were divided randomly into Saline group, Zhanlp group, Carbopol Gel group and Sanchi Gel group, with 18 rats in each group. On 7, 14 and 21 days after the laminectomy, immunohistochemistry (method of S-P) was adopted to detect the expression of TIMP-1 and MMP-1. Results Different treatments had different influence on TIMP-1 and MMP-1 expression in epidural adhesion. The expression of TIMP-1 increased gradually on 14 dand 21 d after operation, and Sanqi Gel group showed weaker expression than the other groups (P<0.05), but the number of positive cells decreased gradually. As to the expression of MMP-1, there was no difference among each group at the end of the second week (P>0.05). There was a difference between Sanchi Gel group and the other groups at the end of third week (21 d) (P<0.05) after operation. Sanchi Gel group showed higher expression of TIMP-1 than the other groups. The number of positive cells sharply decreased from 14 d to 21 d after operation. Conclusion Sanchi Gel has a significant preventive effect on fibrous scar formation after laminectomy. The possible mechanism of preventing epidural adhesion after laminectomy by Sanchi Gel could be its regulating and controlling the expression of TIMP-1 and MMP-1 in epidural tissue.
6.Study on the levels of cytokines in first-episode depressant patients before and after the treatment with sertraline and venlafaxin

Objective To detect the levels of cytokines in first-episode depressant patients and the influence on the levels of cytokines after the treatment with sertraline and venlafaxin. Methods The levels of IL-2,IL-6,TNF-α and IL-10 were measured in 80 first-episode depressant patients and 40 healthy control subjects. 80first-episode depressant patients were divided into two groups which were treated by sertraline and venlafaxin respectively. The levels of the same items were measured after 6-week treatment with sertraline and venlafaxin. All data were statistically analyzed by SPSS. Results The levels of IL-2,IL-6,TNF-α in 80 first-episode depressant patients were significantly higher than that in control group( IL-2:(6.79 ± 2.89 )pg/ml vs (4.06 ± 2.05 )pg/ml;IL-6:(10.12 ±4.52)pg/ml vs (6.04 ± 1.79) pg/ml;TNF-α: ( 14.81 ±4.38) pg/ml vs ( 10.69±2.54) pg/ml)(P<0.05). The level of IL-6 after treatment by sertraline and venlafaxin was lower than that before treatment.There were no significant difference of the levels of IL-2,TNF-α, IL-10 before and after treatment. No significant difference of the levels of cytokines were observed between the two groups which were treated by sertraline and venlafaxin. Conclusions There are some changes of the levels of cytokines in first-episode depressant patients, which might be corrected by the treatment with sertraline and venlafaxin. Sertraline and venlafaxin might have the same mechanisms to cytokines.
7.Spontaneous regression of a giant basilar artery aneurysm in a young adult after surgical injury:case report and literature review

A giant basilar artery aneurysm of young woman with endocrine disturbance was misdiagnosed as a large pituitary adenoma and treated surgically via a trans-sphenoidal approach was planned.But the neurosurgery was finally aborted because of massive bleeding during the procedure. One year later, a cerebral angiography confirmed this basilar artery aneurysm was obviously regressed and then endovascular coiling was successfully performed. No neurological complication occurred post-procedure and the endocrine dysfunction symptom was obviously improved.
8.A comparison of four dimensional time-resolved with keyhole and three dimensional time-of-flight MR angiography for the evaluation of cerebral aneurysms

Objective To evaluate the accuracy and reliability of 4D time-resolved MRA with keyhole (4D-TRAK) for the detection and characterization of cerebral aneurysms ( CAs),with a comparison of 3D time-of-flight MRA (3D-TOF-MRA).Methods3D-TOF-MRA,4D-TRAK and 3D-DSA were performed sequentially in 52 patients with suspected CAs.4D-TRAK was acquired using a combination of sensitivity encoding (SENSE) and contrast-enhanced (CE) timing robust angiography ( CENTRA ) k-space sampling techniques at a contrast dose of 10 ml at 3 T scanner. Accuracy,sensitivity,specificity of 4D-TRAK and 3D-TOF-MRA were calculated and compared for the detection of CAs on patient-based and aneurysm-based evaluation using 3D-DSA as a reference. Wilcoxon signed rank test were used. Results The overall image quality of 4D-TRAK was appropriate for the diagnostic purpose,but yet not comparable with that of 3D-TOF-MRA.In 52 patients with suspected GAs,58 CAs were confirmed on 3D-DSA finally.Fifty-one (with 2 false-positives and 9 false-negatives) and 58 (with 1 false-positive and 1 false-negative)CAs were visualized on 4D-TRAK and 3D-TOF-MRA,respectively.Accuracy,sensitivity and specificity on patient-based evaluation of 4D-TRAK and 3D-TOF-MRA were 92.31% ( 48/52 ),93.33% ( 42/45 ),85.71 % (6/7) and 98.08% ( 51/52 ),100.00% ( 45/45 ),85.71% ( 6/7 ),respectively,and 74.07%(20/27),75.00% ( 18/24),66.67% (2/3) and 96.30% (26/27),95.83% (26/27),100.00% (3/3)on aneurysm-based evaluation in patients with multiple CAs,respectively.Subgroup analysis revealed that for 19 very small CAs ( maximal diameter <3 mm,measured on 3D-DSA),9 were missed on 4D-TRAK and 1 on 3D-TOF-MRA( Z =- 2.464,P < O.01 ). However,for 39 CAs with maximal diameter more than 3 mm,there was no significantly difference in the diagnostic accuracy (39 on 4D-TRAK vs.39 on 3D-TOFMRA) (Z =0.000,P >0.05).In 4 large CAs with maximal diameter more than 10 mm,4D-TRAK provided a better characterization of morphology than 3D-TOF-MRA.Conclusions 4D-TRAK with a combination of SENSE and CENTRA at 3 T shows potential value in the diagnosis of cerebral aneurysms.However,due to the compromise in spatial resolution and vascular edge artifacts,it does not yet have a diagnostic accuracy of CAs comparable with 3D-TOF-MRA.TRAK imaging can be of great help in patients with large-giant CAs to characterize the morphology of CAs and to diminish the risk of NSF in patients with renal impairment by using a lower-dose contrast.
9.Behavioral and endocrine changes following conflict-related stress

Objective To investigate behavioral and endocrinal changes following the conflict-related stress. Method Male SpragueDawley rats were exposed to stressors using Vogel conflict test. Body weight and behavioral responses in open field test were investigated during the whole procedure. Serum corticosterone level was analyzed using immunoassay. Results Compared with the control, the rats of the 1-week, 2-week and 4-week stressed group showed significantly decreased body weight. There was increasing tendency or significant difference in locomotion between the 1 -week, 2-week and 4-week stressed group and their control. And the hormone levels for the stressed animal were reliably high. Conclusions The increased locomotion, and high corticosterone level and reduced body weight in stress animal,was consistent with the features of anxiety-like disorders. Furthermore, the behavioral and endocrine changes produced by conflict-related stress could maintain a long time. The conflictrelated stress paradigm may be used as psychological stress animal mode in the future.
10.Effects of PE on estrogen receptor expressions and its transcriptional activation on target genes

Aim To investigate the effect of piperazinyl estrone(PE) on estrogen receptor expression and the transcriptional regulation of target genes.Methods Ovariectomized mice were given with PE in different doses (0.5 mg·kg~(-1),1 mg·kg~(-1),2 mg·kg~(-1))and estrone(0.71 mg·kg~(-1)) for 42 days,the protein expressions of Ers(Erαand Erβ)were shown by immunohistochemical method; To study transcriptional regulation of PE, PACT2-hERα and ERE2-TATA-LUC were co-transfected into MCF-7 cells by using Tfx 50 cationic liposome.Results Compared with ovx group, the groups with PE could up-regulate Ers of uteri in a dose-dependent manner,but its effect on Erα subtype was obvious.The classical ER signaling pathways could be activated by PE in co-transfected MCF-7 cells,but activation of PE was feebler than estrone with the same dose.Conclusion PE can up-regulate estrogen receptors in uteri. PE can transactivate ERE reportor gene through Erα and Erβ in MCF-7 cells, but its effect is feeble.
