1.Calculation of the surface density of the RGD-containing peptide on allogenic bone using isotopic tracing technique

Objective To investigate the feasibility of determining the surface density of RGD-containing peptide on allogenic bone by isotopic tracing technique using RGD peptide labelled with 125Ⅰ,and the impact of the input concentration of RGD-containing peptide on the surface density,and to obtain the history between the surface density and the input concentration of RGD-containing peptide.Methods The synthetic RGD-containing peptide was labelled with 125Ⅰ,and the specific radioactivity was calculated.The reactive solutions of RGD-containing peptide with the radioactive 125Ⅰ-RGD as a probe were prepared at the input concentrations of 0.01 mg/ml,0.10 mg/ml,0.50 mg/ml,1.00 mg/ml,2.00 mg/ml,4.00 mg/ml.Using EDC as the cross-linking agent,the reaction was carried out by placing the allogenic bone pieces into the reactive solutions of RGD-containing peptide with different input concentrations.After the reaction,the surface density of RGD-containing peptide grafted onto the allogenic bone pieces was calculated by evaluating the radioactivity and the surface area of the bone pieces.The impact of the input concentration of RGD-containing peptide on the surface density was measured and the curve was ascertained.Results After measuring in the radiodensity γ-counter,the result showed the RGD peptides have been marked with 125Ⅰ successfully.The allogenic bone pieces were radioactive after the reaction,which demonstrated that the RGD-containing peptide had been grafted onto the surface of bone pieces successfully.It was also found that with the increasing of input concentration,the surface density raised.Conclusion The surface density of RGD-containing peptide is related to its input concentration.With the increasing of input concentration,the surface density raises to the saturation value gradually.
2.Study on secure tunnel in the fixation of talar neck fracture based on digital technology

Objective To investigate the establishment of secure tunnel in fixation for the talar neck fracture when screws were fixed from the medial wall of talus.Methods The age of volunteers was limited from 20 to 60 years old,and the height of male volunteers was from 165 to 185 centimeters,while the female volunteers' height was from 155 to 175 centimeters.The body mass index (BMI) was less than 25.The volunteers who were not heavy manual workers or standing working for long time had no history of ankle or foot fractures,and there were no evidence of degeneration changes in ankle joints according to X-ray.At last there were 33 males in this study with an average age of 43.7 (21-59) years and an average height of 176.0 (168-184) centimeters.There were 22 females with an average age of 43.2 (22-60) years and an average height of 165.4 (158-172) centimeters.After the volunteers' ankle joints and feet were scanned by CT,the reconstructed images were stored in CD and the slice thickness was 0.75 millimeters.The SuperImageTM Orthopedics Edition1.1 software was used to display the images and perform three dimensional reconstruction.The height of talar neck and the height of tarsal canal were measured.The models that screws passing into tarsal canals were built.The maximal length and angle that screws were inserted in the middle 1/3 and in the inferior 1/3 of medial wall of talus and run along to two directions were measured.At last,the data were analyzed with SPSS 13.0 software.Results The height of talar neck and tarsal canal had no significant difference between left side and right side in the same gender.The height of male talar neck was greater than the female' s.The models of screws passing into tarsal canals was gained.The length and angle in different insert points and different directions of screw fixation were also gained.When the screws were inserted from the middle 1/ 3 of the medial wall of talus,the angle of screw fixation was much wider than that when the screws were inserted from the inferior 1/ 3 of the medial wall of talus.At last,the safe range of the length and angel of screw fixation was calculated.Conclusion Damage of the blood of talus during internal fixation should be avoided.The middle 1/3 of the medial wall of talus is the most favourable choice to the fixation.Combining the digital technology with internal fixation for talar neck fracture could promote the operation' s security and feasibility.
3.Correlation between the stability of carotid artery plaque and cystatin C,homocysteine

Objective To investigate the relationship betweenthe stability of carotid artery plaque and cystatin C,homocysteine. Methods A total of 1 50 cerebral infarction patients with carotid artery plaque treated in Hospital of the First Electricity Authority in Guangdong were selected as the research subjects.According to Color Doppler ultrasound scanning,the patients were divided into the observation group (86 unstable plaque patients)and control group(64 stable plaque patients).Peripheral venous blood from the patients of both groups were collected for the detection of cystatin C,homocysteine,serum total cholesterol and fibrinogen levels and the results of detection were compared between the two groups.Results The levels of cystatin C,homocysteine,serum total choles-terol and fibrinogen in the observation group were significant higher than those of the control group(t =48.760,20.530,29.860, 33.720,P <0.05).The level of cystatin C was weak positive correlated with homocysteine(r =0.21,P <0.003).Multivariate lo-gistic regression analysis showed cystatin C,homocysteine,serum total cholesterol and fibrinogen were independent risk factors for carotid artery plaque instability in patients with cerebral infarction.Conclusion Cystatin C,homocysteine,serum total cholesterol and fibrinogen are important risk factors for carotid plaque instability,the cerebral infarction is closely related to the carotid artery plaque stability,so controlling the risk factors is the focus of prevention and treatment of cerebral infarction.
4.Association between left ventricular hypertrophy and peritoneal transport properties in peritoneal dialysis patients

Objective To investigate the association between left ventricular hypertrophy and peritoneal transport properties in Peritoneal Dialysis patients.Methods Sixty-nine Continuous Ambulatory Peritoneal Dialysis (CAPD)patients were enrolled in current study.All patients underwent echocardiography for left ventricular mass index (LVMI).Transport status was categorized as high transport,high average transport,low average transport and low transport based on modified peritoneal equilibration test (PET).The data collected included hemoglobin,albumin,blood urea nitrogen,creatinine,urea clearance (Kt/V) and creatinine clearance rate (Ccr),dialysis vintage,systolic blood pressure,diastolic blood pressure.Results Patients with high transport status were 41 cases,more than those with low transport status(28 cases).The dialysis age of high transport,high average transport,low average transport and low transport vintage were (39.2 ± 21.8),(26.6 ±15.6),(28.6 ± 14.4),(45.7 ± 35.0) years old respectively,and the difference was significant (F =4.128,P < 0.05).The dialysis age in the higher transport group was longer than that in high average transport group and low average transport group (P < 0.05).LVMI has significant positive correlations with D/Pcr at 4th hour,SBP and DBP (r =0.339,0.351,0.316,P < 0.01) and the negative correlation with albmin (r =-0.292,P <0.05).Left ventricular hypertrophy(LVH) in all patients was 63.8%,ant it was higher in high transport group than that low average transport and low transport group (x2 =5.455,5.091,P < 0.05) Conclusion High transport status is the most common in CAPD patients.There is high incident rate of LVH in this population.LVH has significant positive correlations with D/Pcr,higher SBP,DBP,and lower albumin.
5.Influence of platelet-rich plasma (PRP) on autologous fat grafting breast augmentation surgery

Objective To evaluate the influence of platelet-rich plasma (PRP) on autologous fat grafting breast augmentation surgery.Methods A lot of 40 patients were divided randomly into the treatment group and control group, each group of 20 cases.In the treatment group cases PRP was used in addition to autologous fat grafting.Chest circumference and thickness of subcutaneous fat were measured before and 6 month after operation and 12 months later.The results were compared and the influence of PRP was evaluated after operation.Results Both groups of patients were satisfied with the results.Compared with the control group, patients in the treatment group showed the chest circumference and thickness of subcutaneous fat were significantly increased 12 months after operation.Conclusions The efficacy of PRP is significant in enhancing fat graft, reducing fat absorption in 12 mouths after operation.The fat tissue mixed with PRP is a reliable reconstruction option for the breast augmentation.
6.Cardioprotective effect of losartan on subtotal nephrectomy renal failure rats

Objective To investigate the cardioprotective effect of losartan o n subtotal nephrectomy renal failure rats. Methods Renal failure rats were estab lished by subtotal nephrectomy, and then divided into RF group with subtotal nep hrectomy,RF+Los group with additional losartan(10 mg?kg-1?d-1) gavage. Rats underwent sham operation(sham group) with normal saline gavage as control. Hear t changes of experimental rats were observed 12 weeks after operation. Results T he RF group developed hypertension of arterial systolic blood pressure(SBP) and myocardiac myocytes hypertrophy. The levels of collagen type Ⅰ,type Ⅲ,and fi bronectin in endocardium and surrounding intracardial coronary arteries adventit ia of the RF group were significantly increased as compared to those of the sham group. The SBP and the levels of collagen type Ⅰ,Ⅲand fibronectin were marked ly decreased in the RF+Los group as compared to those of the RF group.Expressio n of angiotensin Ⅱsubtype 1 receptor (AT1R) mRNA in the myocardium of the RF+L os group was significantly down regulated as compared to that of RF group. Concl usions Losartan can prevent the cardiac ventricular remodeling with left ventric ular hypertrophy (LVH) and myocardial interstitial fibrosis by the depression of renin-angiotensin system through the down-regulation of myocardiac AT1R mRNA.
7.Influence of hemoglobin level(Hb) on survival and prognosis of elderly hip fracture at admission.

OBJECTIVE:
To analyze the relationship between hemoglobin(Hb) level on admission and survival prognosis of patients with hip fracture.
METHODS:
From February 2016 to October 2018, 249 elderly patients with hip fracture were surgically treated including 62 males and 187 females;the age ranged from 60 to 91(73.67±10.52) years;the time from injury to operation was (6.79±2.27) d. The clinical and laboratory examination results were collected. The Hb level at admission and the mortality at 30, 90, 180 and 360 days after operation were observed. According to the Hb level at admission, the patients were divided into Hb<120 g/L and Hb≥120 g/L groups. The survival conditions of the two groups at 30, 90, 180 and 360 days after operation were compared and analyzed. Logistic regression was used to analyze the effect of Hb level on death 30, 90, 180 and 360 days after operation.
RESULTS:
The mortality rates at 30, 90, 180 and 360 days after operation were 5.22%, 9.24%, 16.87% and 20.48% respectively. The level of Hb at admission was a risk factor for prognosis and death 30, 90, 180 and 360 days after operation(P<0.05). The OR(95% CI) were 2.431(1.475-4.006), 2.625(1.468-4.695), 2.276(1.320-3.925) and 2.082(1.221-3.551) respectively.
CONCLUSION
The level of Hb at admission can affect the survival and prognosis of elderly patients with hip fracture. We should further study how to manage the level of Hb before operation.

8.The effects of TCM YiShen decoction on the expression of PAI-1 in kidney tissue of mice with IgA nephropathy

Objective To explore mRNA and protein expression of PAl-1 in kidney tissue of mice with IgA nephropathy (IgAN) and the effects of YiShen Decoction. Methods The IgAN model was built by the method of oral intake of bovine serum albumin(BSA)together with the injection of staphylococcus enterotoxin B(SEB)through caudal vein. The mRNA expression of PAI-1 was measured by fluorescent quantitative polymerase chain reaction (FQ-PER)and the expression of PAI-1 protein was measured by immunohistochemistry. Results No significant difference of mRNA and protein expression of PAI-1 was found between the low concentration and high concentration Yishen Decoction group. But the secretion and mRNA expression of PAI-1 in the low and hiigh concentration Yishen Decoction group was decreased more than that in the IgAN model group. Conclusion The abnormal expression of PAI-1 mRNA and protein played an important role in the onset and development of IgAN. The TCM Yishen Decoction could reduce the abnormal expression of the mRNA and protein of PAI-1 in kidney tissue of mice with IgA nephropathy.
9.Effect of fleabane injection on serum D-dimer, fibrinogen and hypersensitive C-reactive protein in patients with IgA nephropathy

Objective To observe the effect of fleabane injection on serum level of D-dimer, fibrinogen and hypersensitive C-reactive protein in patients with IgA nephropathy and to explore the mechanism of fleabane injection for treating IgA nephropathy. Methods 29 patients with IgA nephropathy were given fleabane injection together with routine treatment. Another group of 28 patients with IgA nephropathy were only treated with routine treatment as con-trol. Determinations of 24 hours urine protein output(24HPQ), serum level of D-dimer(D-D), Fibrinogen(Fib) and hypersensitive C-reactive protein (hsCRP) were carried out pre- and post-study. Results Significant decrease in 24HPQ , serum level of D-D, Fib and hsCRP were observed in both treatment group and control group(P <0. 01 ~0. 05), but more significant in treatment group as compared with control group ( P < 0. 05). Conclusion Flea-bane Injection plus routine treatment could significantly decrease 24HPQ in patients with IgA nephropathy, and this maybe contribute to regulation of D-D, Fib and hsCRP by fleabane injection.

Purpose To Investigate the effect of diet-induced hyperchole sterolemia on the kidney ofWistar rats. Methods Male Wistar rats were fed with normal chow supplemented with 5 % cholesteroland observed biochemical changes in plasma lipid concentration, urinary microalbumin excretion, renalfunction, lipid component in renal cortices and morphological changes at 30,60 and 90 days. ResultsTotal plasma cholesterol (TCh) and low density lipoprotein (LDL) concentration were significantly elevatedin the group E (P<0.05)at 30 days, and progressively increased thereafter, but during the entire study,there ere no differences in plasma urea nitrogen(BUN),creatinine(Cr), and endogenous creatinine clearance(Ccr) between the two groups. Quantitative urinary microalbumin excration was markely elevated in group E( P < 0.05 ). Cholesterol (Ch), phosphatidylcholine(PC) and phosphatidylethanolamine(PE) levels of t he renalcortices were sigificantly increased in the group E at 12 weeks. Progressive development in mesangialhypercellulary, increased mesangial matrix, glomerular capillaries collapes were observed in the group E. Noelectron dense deposits were observed in any of the glomeruli examined. There was a siginificant positivecorrelation for the urinary microalbumin, Ch in the renal corticres, and glomerular size with plasma TCh andLDL concentration. Conclusions The diet-induced hypercholesterolemia may cause lipid nephrotoxicity inWistar rats.
