2.Study on the Quality Standard for Xiaozhong Tablets

Objective To establish the quality standard for Xiaozhong Tablets (Radix Angelicae Sinensis, Pheretima, etc.). Methods TLC was performed to Radix Angelicae Sinensis, Pheretima, Ursolic acid. The content of Aconitine was identified by TLC. HPLC was used to determine Strychnine content. Results The characteristic for identification by TLC was distinct and hightly specific. The quantification method of Strychnine had the linear range of 0.52~1.56 ?g (r =0.9998). The average recovery was 99.9% (RSD=1.77%, n =5). Conclusion The methods of identification and quantification were accurate, realizable and reproducible. It can be used effectively for the quality control of Xiaozhong Tablets.
3.A STUDY OF THE EFFECT OF BEE-POLLEN ON IMPROVING SPORT PERFORMANCES

This essay is to study on the effect of bee-pollen and its suitable dosage on improving sport performances. Fifty athletes were our experimental subjects. The athletes were divided into experiment group and control group. 14 objective indexes were measured before and after taking pollen at each of the 4 times within 3 months on both of experiment group and control group of athletes. The results showed that bee-pollen (15 g per day) improved the cardiac function, muscular strength and enduranco significantly.279 mice were experimental too, they were divided into experiment groups and control group. These animals were put in water for one hour per day in a period of 3 months. They were tested for pole climbing and exhausted swimming after taking pollen by the first, second and third months. Results showed that bee-pollen increased muscular strength and swimming time after taking the pollen at a large dosage.After taking the bee-pollen the body-weight remained somewhat unchanged.There is no observed side effect after taking bee-pollen by athletes.

Objective To investigate the effects of 15-deoxy-?12,14-prostaglandin J2(15d-PGJ2) and DDP on the growth of human pulmonary carcinoma PLA-801D cells and the mechanisms of apoptosis.Methods The human pulmonary carcinoma PLA-801D cells were selected and added to each well of 96-well place and cultivated for 24 h.Then the cells were treated with different concentrations of 15d-PGJ2(0,5,10,20,40 and 80 ?g?L-1) or 15d-PGJ2 combined with DDP(3 mg?L) for 24 h.0 ?g?L-1 15d-PGJ2 group was control group.The morphological changes of cells were observed under inverted microscope.Microculture tetrazolium(MTT)dye was applied to detect the proliferation of the human pulmonary carcinoma PLA-801D cells treated with 15d-PGJ2 and DDP.Diphenylamine assay(DPA) was used to evaluate the activation.Flow cytometry assay(FCM) was used to detect the apoptosis proportion and the changes of cell cycle.Results When the human pulmonary carcinoma PLA-801D cells were treated with low-concentration 15d-PGJ2 alone(5,10 and 20 ?g?L-1),no significant difference was observed in the inhibitory rate of cell growth and the apoptotic indexes such as the apoptosis proportion,the percent of DNA fragmentation and the activity of caspase-3 compared with control group(P

Objective To study the relationship between activity and virulence of secretory acid proteinase and extracellular phosphlipase of different Candida albicans isolated from various specimens.Methods 190 strains of Candida albicans were isolated from various specimens(sputum,blood,thrush,wound,secretion of vaginitis) of patients with Candida albicans infection in hospital.Milk-plate medium and egg-yolk medium were used respectively to test the activities of secretory acid proteinase and extracellular phosphlipase of Candida albicans.The suspension of two forms of Candida albicans(5?106CFU?mL-1) were injected to vein of tail in mice respectively in virulence test.The mortality and mean survival time of mice were observed in 1 month.The virulence was appreciated with the mean survival time of mice.Results All of 190 strains,the positive detectable rates of secretory acid proteinase and extracellular phosphlipase were 83.68% and 85.26%,respectively.Animal experimental results showed the activities of secretory acid proteinase and extracellular phospolipase in hyphal forms were significantly higher than that in spore form(P
7.Establishment of ELISA for detection of hepatoma specific γ-glutamyltransferase and its clinical application

Objective To prepare monoclonal antibody (McAb) against γ-glutamyhransferase(GGT) firmly bound to datura stramonim (DSA) leetin from primary hepatic carcinoma (PHC) tissue and establish an avidin-biotin enzyme-linked immunosorbent assay (ELISA) for evaluating the diagnostic value of serum DSA-GGT for PHC. Methods Anti-DSA-GGT monoclonal antibodies were obtained by McAb technology and purified by protein G-sepharose affinity chromatography. The McAb was labeled with biotin and avidin-biotin ELISA for measurement of serum DSA-GGT was established. Using the avidin-biotin ELISA, serum DSA-GGT levels was detected in 39 patients with PHC, and 122 patients with non-PHC diseases. The distribution of serum DSA-GGT values of 119 healthy subjects were determined by P-P plots. Optimal cut-off value for the diagnosis of PHC was determined by receiver operating characterstic (ROC) curve. Results The protein levels of McAb in the ascites derived from 5 McAb hybridoma cell strains ranged from 2. 12 to 6. 70 mg, The biotin-labeled rate varied from 48. 6% to 72. 2% respectively. The minimum detection limit of serum DSA-GGT in avidin-biotin ELISA was 2 μg/L. Intra-assay and inter-assay coefficients of variation were 8.9% and 11.5% respectively. The distribution of DSA-GGT values of 119 healthy subjects showed Gaussian distribution and its Mean ± SD was ( 1.50±0. 51 ) μg/L. Optimal cut-off value (3.25 μg/L) in the diagnosis of PHC was determined by ROC curve. DSA-GGT was positive in 26 out of 39 patients with PHC and 10 out of 122 patients with non-PHC diseases were positive. The sensitivity and specificity of this assay for the diagnosis of PHC were 66. 7% and 91.8% respectively. Conclusions The convenient avidin-biotin ELISA method was successfully established in our laboratory and it showed a good reproducibility and reliability. It may be a potential tool in the diagnosis of PHC to achieve higher sensitivity and specificity.
8.Clinical implication of microproteinuria and urine enzyme in the detection of early renal damage by antibiotics

Objeetive To analyze the clinical significance of serial microproteinuria and urease detection in early diagnosis of antibiotics damage to kidney by observing the changes of urine microalbumin/creatinine ratio (mAlb/Cr),transferrin (TRF),IgG,α1-microglobulin (α1-M G),β2-microglobulin (β2-M G),retinol-binding pmtein(RBP) and N-acetyl-β-D-glucosaminidase(NAG).Methods A total of 161 children with pneumonia whose test results were normal of urine protein,blood urea nitrogen (BUN) and serum creatinine (Scr),and had no related history of kidney diseases were selected.All the patients were divided into three groups according to antibiotics for the treatment,the penicillins (penicillin G,amoxicillin and potassium clavulanate,ticarcillin and potassium clavulanate) group,the cephalosporins (cefazolin,cefuroxime,ceftriaxone,cefoperazone,ceftazidime) group and the macrolides (erythromycin,azithromycin) group.Changes of mAlb/Cr,TRF,IgG,α1-MG,β2-MG,RBP,NAG,BUN,Scr levels of the patients one week before and after use the antibiotics were observed,and statistically analyzed.Results In the penicillins group and macrolides group,the results showed that none of the serial microproteinuria and urease changed(all P > 0.05).In the cephalosporins group,the urine mAlb/Cr,TRF,β2-MG and NAG were higher than before using the antibiotics [(15.56 ± 5.98) mg/g vs.(21.08 ± 10.88) mg/g,(1.61 ± 0,14)mg/L vs.(1.66 ±0.14) mg/L,(0.25 ±0.09)mg/L vs.(0.28 ±0.11)mg/L,(4.62 ±3.80) U/L vs.(4.98 ±3.97) U/L,t =-5.11,-3.24,-2.29,-2.04,P < 0.05 ~ 0.001].The levels of BUN and Scr revealed no change in all the patients(all P > 0.05).Conclusion Combined detection of serial microproteinuria and urease has great clinical significance in judgment and warning of early renal damage by antibiotics.
9.Association of polymorphism of codon 72 in p53 gene with susceptibility and radiosensitivity of non-small cell lung cancer in Chinese population.

BACKGROUNDLung cancer is one of the most common cancers worldwide and its mortality rate has increased year after year. Molecular biology has contributed to make people's understanding to the disease at the gene level. A new idea will be given to the early diagnosis and treatment for lung cancer with the method of molecular biology which can be used to find the association between some genes and lung cancer. The aim of this study is to evaluate the relationship between polymorphism of codon 72 (BstU I single nucleotide polymorphism, BstU I SNP) in p53 gene and susceptibility and radiosensitivity of non-small cell lung cancer (NSCLC) in Chinese population.
METHODSThe BstU I single nucleotide polymorphic sites at condon 72 in exon 4 of p53 gene in 50 patients with NSCLC as well as 50 healthy controls were inspected by polymerase chain reaction-restricted fragment length polymorphism assay and the relationship between BstU I SNP and susceptibility and radiosensitivity of NSCLC was analysed by case-control test.
RESULTSThe allelic distribution of the three genotypes (A1/A1, A1/A2, A2/A2) in healthy controls was 32.0%, 42.0% and 26.0% respectively, which differed slightly from that of lung cancer patients, which was 28.0%, 32.0% and 40.0%. These allelic and genotype differences between control and lung cancer groups (A1/A1 OR=0.83, 95% CI 0.36-1.27 and A2/A2 OR=1.90, 95% CI 0.82-4.42) were insignificant. The patients with A1/A1 or A2/A2 genotype were sensitive to radiotherapy and the patients with A1/A2 were not sensitive to radiother-apy (Chi-square=9.2, P < 0.05), the effective rate to radiotherapy were 71.4%, 70.0% and 25.0% respectively.
CONCLUSIONSThere is no significant relationship between the BstUI SNP in p53 and susceptibility in NSCLC .The p53 BstU I SNP is closely associated with radiosensitivity of NSCLC in the Northern Chinese population.
10. Statin use and low-density lipoprotein cholesterol levels in patients aged 75 years and older with acute coronary syndrome in China

Objective:
To assess the use of statins and low-density lipoprotein cholesterol (LDL-C) levels at admission in hospitalized patients aged 75 years and older with acute coronary syndrome (ACS) in China.
Methods:
Data used in this study derived from the Improving Care for Cardiovascular Disease in China (CCC)-ACS project, a nationwide registry with 150 tertiary hospitals reporting details of clinical information of ACS patients. This study enrolled patients 75 years and older with ACS in CCC-ACS project from November 2014 to June 2017. Patients were divided into two groups according to the history of atherosclerotic cardiovascular disease (ASCVD). Pre-hospital statin use, LDL-C levels at admission and prescription of statins at discharge were reported.
Results:
A total of 10 899 patients 75 years and older with ACS were enrolled. The median age was 79 years and 58.7% (6 397 cases) were male. Among patients with history of ASCVD, 33.9% (1 028 cases) of them received statins before hospitalization. Among patients without history of ASCVD, 12.7% (996/7 871) received statins before hospitalization. The mean level of LDL-C was (2.4±0.9) mmol/L and LDL-C was <1.8 mmol/L in 24.7% (747 cases) of patients with history of ASCVD. The mean level of LDL-C was (2.6±0.9) mmol/L and LDL-C was <2.6 mmol/L in 51.7% (4 072 cases) of patients without history of ASCVD. At discharge, 91.2% (9 524/10 488) of patients were prescribed with statins in patients without contraindications for statin.
Conclusion
In elderly patients with recurrent ASCVD, there was an inadequate statin use before hospitalization and most patients did not reach the LDL-C target level when they had the recurrent events. In the elderly ACS patients without history of ASCVD, more than half of the patients had an ideal LDL-C level. It seems that ideal LDL-C level for primary prevention of ACS in elderly people needs to be reevaluated with further studies.
