This study is designed to obtain recombinant human acetylcholinesterase (rhAChE) and apply it in screening acetylcholinesterase inhibitors. The rhAChE was overexpressed in HEK293 cells transfected by plasmid of pCMV-AChE with the cationic liposome and rhAChE was found to be secreted into cell culture medium. AChE activity was assayed according to modified Ellman method to obtain kinetic parameters. IC so50 values for donepezil compounds of rhAChE were calculated to determine their activities of inhibition. The results showed that Km value was 151.9 micromol.L-1 donepezil inhibited rhAChE in a mixed competitive-noncompetitive way (Ki= 16.03 nmol.L-1, Ki = 18.36 nmol.L-1) and that most new compounds tested exhibited high activities of inhibition on rhAChE. The study suggests that rhAChE is available to be applied in screening AChE inhibitors in vitro.

OBJECTIVE To study the best surgical technique for treatment of chronic hypertrophic rhinitis(HR).METHODS Eighty six cases with HR were divided randomly into 3 groups.Thirty cases were treated by transnasal endoscopic submucous inferior turbinate resection(group A),26 cases by partial inferior turbinectomy(group B)and 30 cases by bipolar radiofrequency ablation(group C).The epithelium of the inferior turbinate of the every case in 3 groups was examined with histomorphology and ultrastructure techniques at 6 months after operation. The efficacy and the features of in the 3 groups were compared. The MTR was determined in the patients of the 3 groups. The histomorphology and ultrastructure of the inferior turbinate were observed at 6 months after operation.RESULTS All the cases with HR in 3 groups were followed up for 3 to 6 months after operation.Effective rates of group A,group B and group C were 100 %,100 % and 96.3 % respectively, while there was no significant difference among the three groups.Compared to the group B,surgical techniques applied in Group A and group C have more advantages because ultrastructure of inferior turbinate mucosal cilia and nasal mucociliary transport function were nearly normal at 3 to 6 months after operation. The advantages include operating easily, wound healing up fast.MTR between pre-and post-operation was significant difference in group B.Epithelial tissue remained intact in group A and group C,while epithelial tissue in group B was instead of fiber tissue at 6 months after operation. The ultrastructure of inferior turbinate mucosal cilia in group A and group C remained intact at 6 months after operation.CONCLUSION The transnasal endoscopic submucous inferior turbinate resection and bipolar radiofrequency ablation for treatment of chronic hypertrophy rhinitis are effective. Submucous minimally invasive surgery of the inferior turbinate deserves to be recommended.

With the social development, the importance of medical records has become increasingly prominent. This paper analyzes the methods and shortcomings of the current medical record quality control and proposes a new mode under two-level randomized blind method combining with the experience of our own hospital. This new mode, through reconstructing the work team, adjusting work pattern and task allocation, supervising problems rectification, combining with experts′ supervising terminal record, inspecting link record randomly, handling dissent, evaluating connotation quality and assessing performance, effectively reduced the defect rate of medical records, improved the archiving rate and promoted the quality of medical records, which is worth promoting.

Objective To investigate the effects of the fibrin scaffold on the differentiation and the proliferation of neural stem cells and astrocytes. Methods Neural stem cells and the gliocytes derived from spinal cord were cultured in vitro respectively. The purified neural stem cells or gliocytes were seeded separately onto the fibrin scaffolds as experimental group and the glass slides modified with poly-L-lysine(PLL)as control group. At different time in culture the neural stem cells were immunofluorescence stained with antibodies against the marker of neurons I.e. Neurofilament(NF).The length of NF-positive neuritis was masured and the average value was calculated in the culture well (n=4). The gliocytes were immunofluorescence stained with antibodies against the marker of astrocytes I.e. Glial fibrillary acidic protein (GFAP ). The total number of the cells and the GFAP-positive cells were counted from 5 different fields of vision in the culture well (n=4), then the average ratio of GFAP-positive cells was calculated. The differentiation of neural stem cells, the extension of neurites and the proliferation of astrocytes on the fibrin scaffolds were compared with those on the slides. The protein of GFAP was detected by Western blotting to analyse the mature degree of astrocytes. All above experiments were repeated 3 times respectively. Results Immunofluorescence staining showed that the NF-positive neurites in the fibrin scaffold group were longer than those in the control group, whereas GFAP-positive cells were fewer than those in the control group. The expression of GFAP in the cells on the scaffold was lower than that in the control group. Conclusion The fibrin scaffold could promote differentiation of the neural stem cells to neurons and extension of the neurites. Meanwhile, the scaffold could inhibit proliferation and mature of the astrocytes.

Objective To investigate the serum levels of tumor markers CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF in gas-tric cancer. Methods The serum levels of CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF were detected in 46 healthy con-trols,45 atrophic gastritis patients and 39 gastric cancer patients.Serum levels of CEA,CA724 were measured by ELC meth-od,PGⅠ,PGⅡ by time resolved fluorescence immunoassay (TRFIA)and SF by immunoturbidimetry(ITM).Results ①Compared to the healthy controls, the statistical contrast of serum levels of CEA,CA724,PGⅠ,PGⅡ and PGI/PGII except SF was significant in gastric cancer patients(P<0.05),only PGⅠ,PGⅡ,SF had significant difference in atrophic gastritis patients (P<0.05).Compared to atrophic gastritis patients, the serum levels of CEA,CA724 were significantly higher, but PGⅠwas lower in gastric cancer patients(P<0.05).②When CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF were used to di-agnose gastric cancer individually, the sequence of the area under ROC curve was CEA,SF,CA724,PGI/PGII,PGⅡ and PGⅠ.Only the areas of PGⅠ,PGⅡunder ROC curve had significant statistical difference (P<0.05).③The sensitivity,speci-ficity,PPV and NPV were different when these indexes were used to diagnize gastric cancer individually or incorporatedly. Conclusion The serum levels of tumor markers CEA,CA724,PGⅠ,PGⅡ,PGI/PGII and SF had important reference value for the diagnosis of gastric cancer although the diagnostic value was different individually or incorporatedly;the content of serum PG and the ratio of PGI/PGII were closely related to the gastric mucosa.

Objective To explore the applicability and best cut-off value of the Montreal Cognitive Assessment Scale Beijing Version (MoCA-BJ) for mild cognitive impairment (MCI) screening in Xian City. Methods Community residents over 50 years old in Xiancheng District were enrolled as the research object,a random cluster sampling method was used to sample 1195 community elderly people in Xiancheng District. The subjects were tested for cognitive function with the Mini-mental State Examination (MMSE),and were divided into the normal group and the MCI group according to the diagnostic criteria, and the MoCA-BJ test was performed again.Groups were grouped according to education level, and the receiver operating curve (ROC) of each group was drawn to determine the MoCA-BJ cut-off value corresponding to the maximum Youden index. Results MMSE and MoCA-BJ scores were significantly correlated, the correlation coefficient was 0.791 P<0.001;the best cut-off values of MoCA-BJ in each group were 19 (illiterate),21 (elementary school),and 25 (junior high school and above). The consistency test Kappa value is 0.686 P<0.001,and the consistency is good. Conclusion MoCA-BJ can be effectively used in the screening of MCI population in this area.
