NIT-1 cells were exposed to various concentrations of glucose for 24,48,and 72 hours. The content of triglyceride in NIT-1 cells increased in dose-and time-dependent manners (P < 0.05).Long-term exposure of NIT-1 cells to high glucose concentrations caused an inverse "v"-like induction of HSL mRNA and protein expressions, which increased from beginning, and then decreased, along with similar changes of lipolysis. These results suggest that the adaptation of HSL may play an important role in regulating the intracellular triglyceride pool and the development of glucolipotoxicity in pancreatic β cells.

OBJECTIVE:To observe the efficacy and safety of Xingpi yang'er granule in the treatment of primary nocturnal en-uresis in children. METHODS:97 children with primary nocturnal enuresis were randomly divided into control group (46 cases) and observation group(51 cases). Control group received Racemic anisodamine tablet 0.5 mg/(kg.d),orally,once a day+Vitamin B1 tablet 10 mg,orally,3 times a day+Oryzanol tablet 10 mg,orally,3 times a day + Meclofenac hydrochloride capsule 0.1 g 30 min before going to bed, orally, once a day. Observation group was additionally received Xingpi yang'er granule 0.4 g for 5-6 years old and 0.6 g for 7-14 years old,3 times a day. They were treated for 4 weeks. Clinical efficacy, kidney Yang deficiency score,wake-up threshold, bladder capacity, bladder volume,BVWI,enuresis frequency,AVP,cAMP,cGMP,cAMP/cGMP be-fore and after treatment, recurrence rate and the incidence of adverse reactions in 2 groups were observed. RESULTS:The total ef-fective rate in observation group was significantly higher than control group(P<0.01). After treatment, the kidney Yang deficiency score,wake-up threshold,BVWI,enuresis frequency and cGMP in 2 groups were significantly lower than before, and observation group was significantly lower than control group;bladder capacity, serum AVP,cAMP and cAMP/cGMP in 2 groups were signifi-cantly higher than before, and observation group was significantly higher than control group(P<0.01). After 3 months, there was no significant difference in the recurrence rate in total effective children(P>0.05);after 6 months, the total effective rate recur-rence rate in observation group was significantly lower than control group(P<0.01). And there was no significant difference in the incidence of adverse reactions(P>0.05). CONCLUSIONS:Based on conventional treatment,Xingpi yang'er granule shows good efficacy and safety in the treatment of primary nocturnal enuresis in children, which can improve children's kidney Yang deficiency score,wake-up threshold and bladder capacity and serum AVP.

Objective To investigate the prevalence and risk factors of restless leg syndrome (RLS) in hemodialysis patients after kidney transplantation failure. Methods Patients of hemodialysis after kidney transplantation failure were investigated by face-to-face interviews, from March to July,2015,at four dialysis units in Beijing.RLS was diagnosed according to the International RLS Study Group (IRLSSG) criteria. The severity of RLS was assessed using International RLS Rating scale.Besides, three validated sleep disorder questionnaires (Hamilton anxiety and depression scale,Epworth sleepiness scale and Pittsburgh sleep quality index) were completed by the patients at the same time. Results Ninety-four hemodialysis patients after kidney transplantation failure were enrolled;46 patients (48.94%) met the diagnosis of RLS,the average age was 53.44±l 1.89 years, and the median time of RLS onset after kidney transplantation failure was 46 months. The International RLS Rating scale scores of the patients were 17.26±7.81;76.0% patients were above moderate. As compared with the non-RLS patients, patients with RLS used more erythropoietin (44/48 vs.46/46),less ferrila (30/48 vs.19/46),and few hypnotic medicine (10/48 vs.3/46),with significant differences (P<0.05).The serum ion, serum ferritin and serum Vitamin B12 of patients with RLS were significantly lower as compared with non-RLS patients (P<0.05);and poorer sleep quality and higher depression scale scores in the patients with RLS were noted as compared with those in the non-RLS patients (P<0.05).Conclusion The prevalence of RLS in hemodialysis patients after kidney transplantation failure is high, low iron protein content, low serum iron content and low vitamin B12 levels may be risk factors for RLS.

Objective:To explore the feasibility and safety of long-distance urological nephrotomy with the support of 5G communication technology by using the domestic robot. Methods:Clinical data of the patients with remote robot-assisted laparoscopic nephrectomy, which were completed from March to April 2021 by the Affiliated Hospital of Qingdao University (as the host hospital where the main operating system located) were retrospectively analyzed. There were 3 patients, including 2 males and 1 female. The average age was 61 (49-73) years, and the average body mass index was 23.73 (20.00-27.76) kg/m 2. One patient had a ASA classification of grade 2, and the other 2 patients had grade 3. All patients met the surgical criteria for non-functional nephrectomy. The chief surgeon who performing the telesurgery was located at the Affiliated Hospital of Qingdao University. The surgeon remotely controlled the bedside operating system (slave system) in 3 local hospitals located in other cities in Shandong Province (network communication distances of 82.5, 141 and 229 km, respectively) by manipulating the master system located in Qingdao. Images and operating instructions during surgery were transmitted using 5G wireless communication technology. Intraoperative network conditions, robot operation, and patient perioperative data were summarized. Results:All 3 tele-nephrectomies were successfully completed. The average network signal latency time was 27.3 (23-30) ms, with no packet loss, and the average total latency time was 177.3(173-180) ms. The mean resection time was 79.3 (52-111) min, and the average intraoperative blood loss was 31.1 (15.6-41.9) ml. There were no network related adverse events occurred during the operation, and the robot-related adverse events occured 3 times, all three of which were characterized by inconsistent master and slave movements of the manipulator arm and the bedside robotic arm. None of these adverse events affected the successful performance of the telesurgery. The mean postoperative exhaust time was 60.5 (38.5-78.0) h. The mean postoperative VAS score at 24 hours was 3.7 (3-4). The Clavien-Dindo classification were all grade I. No significant abnormality was found on the 30th day after surgery, and the patients recovered well at the follow-up until 6 months postoperatively. Conclusions:It is safe and feasible to perform remote robot-assisted laparoscopic nephrectomy based on 5G communication technology with no serious adverse events or surgical complications. However, the conclusion needs to be further verified by large sample and multi-center prospective study.
