1.Nitoprusside, dopamine and dobutamine for treatment of refractory heart failure

Objective To observe the clinical effect of nitroprusside,dopamine and dobutamine for treatment of refractory heart failure. Methods 60 patients with refractory heart failure were randomly divided into control group and treatment group. Clinical symptoms, signs, echocardiography and other changes were observed. Results The total effective rate was 86.67% ,control group was 66.67% ,the total effective between the two groups was statistically significant(P < 0.05). After treatment, left ventricular ejection fraction (LVEF) improved significantly compared with the control group, the difference was statistically significant(P < 0.05). Conclusion The Sodium nitroprusside, dopamine and dobutamine for treatment of refractory heart failure was safe and effective method.

Objective To find the effect of Rhodiola herb on the phenotypic transfer of epithelial cell in renal tubule of adriamycin-induced nephropathy rats.Methods The rat nephropathy model is established by adriamycin.For Rhodiola herb treated group,feed with Rhodiola herb 0.672 g/kg?d from the 4th week,for the control group and neohropathy group,feed with water [3 mL/(kg?d)].Kill the rats per batch at the week of 4th,8th,12th,16thand 20th week,observe the pathological change of nephridial tissue,test the expression of ?-SMA-protein of myofibroblast in the tuhulointerstitial.Results A semi-quantitative analysis of ?-SMA of rats at the 16th week in the nephropathy group is 6.24?0.57,which is obviously higher than that of Rhodiola herb treated group,3.84?0.28(P
3.Relationship between maternal serum lipid levels and newborn weight in pregnant women with single positive diabetic screening test

Objective To investigate the relationship between maternal serum lipid levels and newborn weight in pregnant women with single positive diabetic screening test. Methods On the basis of 75 g oral glucose tolerance test (OGTT), 406 pregnant women during 24-28 gestational week whose 50 glucose challenge test (GCT)≥7.8 mmol/L were separated to gestational diabetes mellitus (GDM) group with 115cases, gestational impaired glucose tolerant (GIGT) group with 60 cases,and GCT (+) group with 231 cases.The serum levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol(LDL-C), apolipoprotein A1 (ApoA1) and apolipoprotein B (ApoB) were measured at the same time. Results Serum TG concentration was significantly higher in GDM group than that in GIGT group and GCT(+) group(P<0.01).The level of HDL-C, ApoB in GDM group were lower than those in GIGT group and GCT(+) group (P<0.01). In the 231 women with positive diabetic screening but normal glucose tolerance who delivered at term, the incidence of macrosomia was significantly higher in those with TG levels higher than 3.5 mmol/L (25.0%, 11/44) than in mothers who TG levels were lower than 3.5mmol/L (11.2%, 21/187) (P=0.017). Multiple regression analysis (stepwise) showed that the independent risk factors of macrosomia was the level of TG and ApoB (F=7.677,P=0.006;F = 7.842,P = 0.001 ).Conclusion Among pregnant women with single positive diabetic screening test,maternal serum TG and ApoB are independently associated with neonatal birth weight.

Objective To examine the effects of acute hypobaric hypoxia on abilities of learning and memory,the water content in brain,the level of nitric oxide(NO) and endothelin-1 (ET-1) in hippocampus of mice.Methods The acute hypobaric hypoxia environment were made by putting the mice in a hypobaric chamber simulated at altitude of 6 000 meters for 8 hours.The capabilities of learning and memory of mice were detected by Morris water maze test.The content of water in hippocampus were examined by measuring the ratio of dry/wet weight,and the level of NO and ET-1 in brain was detected by colorimetric method.Results Morris water maze test showed that the mean escape latency of mice in the hypobaric hypoxia group were longer than that in the normobaric normoxia group((44.60±7.80) s vs (26.39±8.44)s,P<0.01),and the target quadrant residence time were decreased((19.78±2.74) s vs (22.98±6.14)s,P< 0.05).Compared with the normobaric normoxia group (NO:(2.37 ± 1.07) μmol/gProt,ET-1:(38.87 ± 6.17) ng/L),the water content in brain of mice in the hypobaric hypoxia group was increased (P< 0.05),meanwhile,both the level of NO ((4.48 ± 1.45) μmol/gProt) and ET-1 ((52.09±6.75)ng/L) in brain were increased too(P<0.01).Conclusion Hypobaric hypoxia can decrease the abilities of learning and memory of mice,and these changes might be related with the increased water content and the increased level of NO and ET-1 in hippocampus of mice.
6.Effects of cyclic tensile stress on metabolism and apoptosis of chondrocytes inrat

Objective To investigate the effect of cyclic tensile stress (CTS) on the metabolism inand apoptosis of rat chondrocytes.Methods Primary rat chondrocytes were cultured on a Bioflex plate for one day and then stretched cyclically for 24 hours at a frequency of 0.5 Hz using a Flexcell-5000T apparatus.The cells were divided into 5 groups according to their stretching ratio:0% (the control group),2%,6%,10% and 14%.After the stretching,Col Ⅱ,Aggrecan,MMP-13 and ADAMTS-5 mRNA were measured using qPCRs,and the NO and PGE2 levels were measured using ELISA kits.Moreover,TUNEL staining and Annexin V-FITC/PI were used to analyze the apoptosis of chondrocytes.Results Compared with the control group,the average levels of Col Ⅱ and Aggrecan mRNAdecreasedin 10% and 14% groups [(0.738±0.11) and (0.58±0.13),(0.75±0.11) and (0.55±0.09)].In those groups,the MMP-13 [(2.47±0.47) and (2.88±0.36)] and ADAMTS-5 mRNA level [(2.39±0.33) and (2.75±0.49)],the NO [(6.96±0.96) and (8.28±0.82)] and PGE2 level [(6.83±0.66) and (7.15±0.71)] had increased significantly.In the 6% group the average levels of Col Ⅱ(1.76±0.30) and Aggrecan mRNA (1.93±0.14)of 6% group were significantly higher than the control group,but the NO level of the former (3.07±0.20) was significantly lower than the control group (3.89 ± 0.33).The apoptosis rate of chondrocytesin 2% and 6% groups were (0.065±0.013) and (0.063 ± 0.147),without significant differences to that of the control group (0.045 ± 0.008).However,compared with the control group,apoptosis in the 10% and 14% groups [(0.135 ±0.026) and (0.184±0.020)] increased significantly.Conclusion The effect of cyclic tensile stress on chondrocyte metabolism and apoptosis was magnitude-dependent.Ten percent and 14% CTS can increase the catabolism and apoptosis of chondrocytes.Ten percent and 14% strain can increase the catabolism and apoptosis of chondrocytes.Cyclic 6% strain can increase the anabolism of chondrocytes,but 2% strain has no impact on metabolism or apoptosis.
7.Evaluation of the blood perfusion modes in retroperitoneal benign and malignant masses by real-time contrast-enhanced ultrasonograpby

Objective To analyze the blood perfusion modes of retroperitoneal benign and malignant masses by real-time contrast-enhanced ultrasonography (CEUS). Methods Forty-five patients with retroperitoneal masses, including 18 benign and 27 malignant masses, were performed with CEUS. The perfusion of contrast agent, entering style and the vascular patterns of contrast agent were described. And the diagnosis value of perfusion characteristics was evaluated in retroperitoneal benign and malignant masses. Results The perfusion of CEUS showed significant differences in the entering style of contrast agent and the vascular patterns in malignant masses in comparison to benign masses. In the entering style of contrast agent, center enhancement of malignant masses was 70. 37% (19/27), periphery enhancement of benign masses was 72.22% (13/18) ,there was statistical significane in the result( P = 0.0069). To the vascular patterns level,there were 2 with level 0 and 11 with level 1 in benign group, 10 with level 2 and 9 with level 3 in malignant group, there was statistical significane between malignant group and benign group too( P=0.0206). Conclusions CEUS is benefit to improve the accuracy of diagnosis of retroperitonea malignant and benign masses.
8.A comparison of clinical efficacy between covered stent-grafts and bare stents in transjuglar in-trahepatic portosystemic shunt

Objective To compare the clinical efficacy between covered stent and uncovered stent in transjuglar in-trahepatic portosystemic shunt (TIPS) .Methods Thirty patients with liver cirrhosis (portal hypertension), who received TIPS, were retrospectively studied.All patients were divided into two groups covered-stent group(n =20) and uncovered-stent group (n=10).For each patient, portal pressure was measured before and after operation, and the patency of shunt was evaluated by color Doppler ultrasound after operation.The mortality, recurrent bleeding rate and incidence of hepatic encephalopathy were analyzed by Fisher exact probability test.Results The TIPS treatment was successful in all patients, the portal pressure in the covered-stent group reduced from (3.78±0.50) kPa before operation to (2.21±0.44) kPa and that of the uncovered-stent group reduced from (3.67±0.48) kPa to (2.13±0.35) kPa.Twenty-six cases were postoperatively followed-up (17 cases in covered-stent group, 9 cases in uncovered-stent group).The follow-up period varied from 7 days to 62 months (median follow-up period was 23 months).Thirteen patients died of upper gastrointestinal bleeding and hepatic failure.The difference of mortality between covered-stent group (8/17) and uncovered-stent group (5/9) did not reach significant (P>0.05).The recurrent bleeding rate between the covered-stent group (5/17) and the uncovered-stent group (3/9) was not different too (P>0.05).The incidence of hepatic encephalopathy in the covered-stent group (4/17) was not different from that of the uncovered-stent group (2/9) (P> 0.05).The patency rates of 6 months and 12 months reached 100% in the covered-stent group, which were higher than those in the uncovered- stent group 77.8% (7/9) and 55.6% (5/9) (P<0.05) .Conclusions The patency rate of shunt at 12 months after TIPS was higher in the covered-stent group than the uncovered-stent group, while the mortality, recurrent bleeding rate and incidence of hepatic encephalopathy were not significantly different between the two groups.

Objective To evaluate the effect of silencing ACAT1 gene on colon cancer cells proliferation,migration,invasion and colon cancer development by using the small interference RNA (siRNA) in colon cancer cell line HT-29.Methods Acyl coenzyme A cholesterol acyltransferase 1 (ACAT1) gene was silenced in HT-29 cell lines using Hiperfect transfection reagent.The expression level of ACAT1 was detected by real time PCR.CFSE and transwell assays were used to evaluate the effect of ACAT1 gene interfering on cells proliferation,mi gration and invasion.Result ACAT1 mRNA expression decreased obviously after siRNA interference.Compared with pre-transfection,proliferation,migration and invasion of colon cancer cells have been significantly inhibited (P < 0.05).Conclusion ACAT1 gene interference reduced proliferation,migration and of invasion of HT29 cells,which provide a new potential target for colon cancer treatment.
10.Analysis of malaria situation and discussion of control strategy in Shandong Province,2013

Objective To understand the malaria situation of Shandong Province in 2013,so as to provide the evidence for formulating targeted prevention and control strategy and measures. Methods The data of malaria cases of Shandong Province in 2013 were collected from the Information Management System for Infectious Diseases Report and Information Management Sys-tem for Parasitic Diseases Control and Prevention. The data of epidemiological characteristics of malaria situation and the diagno-sis and treatment of malaria cases were analyzed by Microsoft Excell2007. Results There were 131 malaria cases reported in 2013,all of them were imported cases,and 127 cases(96.95%)were imported from African countries. A total of 116 cases (88.55%)were falciparum malaria cases. Totally 97.71%of the cases were male and the average age of malaria cases was 39 years. A total of 61.83%of the cases were peasants and 65.65%of the cases only received junior high school education. The dis-tribution of malaria cases was concentrated in Tai’an City(32 cases),Yantai City(19 cases)and Weihai City(17 cases),total-ly acounting for 53.13%. There was no significant seasonal variation in the reporting time of the cases. The median time from on-set to seeing doctor was four days and the median time from seeing doctor to being diagnosed was one day. Totally 35.88%of the cases were misdiagnosed when the first visit to a doctor. All of the cases were laboratory confirmed and 100%of them received the standard treatment after diagnosis. Conclusions All of the malaria cases were imported cases in Shandong Province in 2013. To control the imported malaria in Shandong Province,it is necessary to further strengthen the multi-sectoral cooperation, health education,malaria screening and professional training.
