Objective To clone and construct eukaryotic expressing vector of bcr-abl fusion gene and to express the gene in the mammal COS-7 cell lines. Methods bcr-abl fusion gene was amplified from human chronic myeloid leukemia (CML) K562 cell lines by RT-PCR and the fragment of cDNA was retrieved,purified and cloned into the pEGFP-N3 eukaryotic expressing vector. After the selection of the positive clone and by restriction enzyme analysis and DNA sequencing, the correct plasmid was transfected into COS-7 cell lines and observed the transient expression. Results A 874 bp DNA fragment was amplified by RT-PCR. The sequence analysis showed it was consistent with bcr-abl gene of GeneBank. RT-PCR, Western blotting analysis provided strong evidences that bcr-abl gene was expressed successfully in transfected COS-7 cells.Conclusion The eukaryotic expressing vector of bcr-abl fusion gene was constructed, it will lay the foundation for further study of bcr-abl gene in the diagnosis and treatment of CML.

Nerve agents (NAs) belong to the class of organic phosphorus compounds which are acetylcholinesterase ( AChE) inhibitors, including soman, sarin, tabun,VX, etc.NAs are extremely toxic and considered as the most danger-ous chemical warfare agents.The current standard treatment for poisoning by nerve agents consists of the combined adminis-tration of anticholinergic drugs such as atropine sulphate, AChE reactivators such as pralidoxime, obidoxime and HI-6 and diazepam for anticonvulsant effects, but oximes are therapeutic antidotes against nerve agent intoxications which exert the therapeutic purposes primarily by reactivating the NAs-inhibited AChE.In this paper, the mechanism of nerve agents, the main working procedure of anti-NAs drugs, the chemical structure of classic reactivator, the corresponding antitoxic action, in vivo and in vitro effects and metabolic kinetics are reviewed.

17.The abnormal results of METLP were found in 6(100%) patients with other myopathy.Most of them showed a high concentration of lactic acid post-aerobic exercise(L2),and sustainable to after rest,or even more higher(L3).3(21.4%)patients without myopathy showed abnormal results of METLP.Conclusions In all the indexes of METLP,the value of L2/P1 is the reliable index for diagnosis mitochondrial myopathy or no-myopathy;the value of L3/L2 is the most important index in differential diagnosis of mitochondrial myopathy and other myopathy.METLP plays a certain role in the diagnosis of the mitochondrial myopathy,but the clinical and pathological data ought to be combined.

Objective To analyses the CT characteristics of primary malignant ovarian tumors and metastatic ovarian carcinoma and its diagnosis and differential diagnosis.Methods CT findings in 31 patients with pathologically-proved malignant ovarian tumors,encountered during 1999-04~2003-11 were retrospectively analyzed.The observation of the lesion was focused on the following aspects:location,internal density,septa,wall,intra-cystic and extra-cystic vegetation,the composition of the soft tissue,ascites,peritoneum,mental implantation and metastatic lymphadenopathy.The characteristic of each in both primary malignant ovarian tumor and metastatic ovarian tumor were compared.Results Among the 24 primary malignant ovarian tumors,13 patients had lesions in both sides of ovary,type Ⅱa:n=3,Type Ⅱb:n=11,Type Ⅱc:n=7,Type Ⅲ:n=3.Among the metastatic ovarian carcinomas,Type Ⅱa:n=6,Type Ⅱc:n=1.Conclusion Type Ⅱb and Ⅱc are commonly seen in primary malignant ovarian tumors.Type Ⅱa is always seen in metastatic ovarian carcinoma.The metastatic tumors often happen in the right peritoneum,epiploon,and liver."arborization and silk screen likely"are the characteristic representation of the metastatic ovarian carcinoma.

Objective To enhance the cognition of the clinical and X-ray diagnosis of duodenum filled syndrome in children.Methods The clinical and X-ray features of duodenum filled syndrome cases in 78 cases were analysed.Results There were different degrees of obstruction in the horizontal segment of duodenum in all 78 cases.72 cases showed typical pressed strip tracks which were vertical and look like penhdders.The obstruction of duodenum was relaxed when the body position was changed in all cases but in 6 cases who with complications.Conclusion The symptoms of obstruction in duodenum filled syndrome can be relaxed when the patient’s position is in prone,lying on liftside and chest-knee situation.

Objective To establish a whole-body dynamic inhalation exposure system for toxicological studies on highly toxic chemicals, and to evaluate the safety and applicability of the system.Methods The safety and standardization of the laboratory were ensured after positive and negative pressure protection and airtight protection were finished.By modifying and optimizing the key technological units of the exposure chamber, the relationships between aerosol concentrations in the chamber and the push rate, exposure time and different monitoring points were investigated.Results and Conclusion Multi-protection was achieved, including the independent exposure chamber, negative pressure experiment and positive pressure protection under working conditions.The laboratory meets the demands of safety and specifications.The exposure aerosol concentrations in the chamber are uniform, stable and controllable while the air is dynamically flowing.The whole-body dynamic inhalation exposure system can meet the need for toxicological studies on highly toxic chemicals.

OBJECTIVE To establish a model for chlorpyrifos(CPF)whole-body dynamic inhalation exposure in SD rats and investigate the injury effects after acute exposure by CPF. METHODS By optimizing the aerosol parameters ,the animal acute dynamic inhalation exposure of CPF was established. Absorption sampling-gas phase detecting technology was used to monitor the concentration of CPF in the whole-body dynamic-inhalation exposure cabin by exploring the relationship between the concentration , particle size of CPF aerosol and the CPF inhalation time in the exposure cabin via a particle size detector. Using Bliss method,specific pathogen free SD male rats were allocated to the environment of CPF exposure at different lethal concentrations and time points. The symptoms and deaths of these SD male rats in different groups were recorded within the following 10 d. Based on the median lethal concentra?tion time(LCt50),the values of plasma cholinesterase(ChE)were checked at different time points after being exposed at different doses. RESULTS The mean concentrations of CPF aerosol at nine time points was 160.6 mg · m-3,the relative standard deviation value was 6.9%;the geometrical mean of aerosol particle size was 1.1 μm,and the geometric standard deviation was 1.8. The results met the technical requirements of Organization for Economic Cooperation and Development regarding acute inhalation exposure. Under these equipment conditions,the LCt50 of CPF acute inhalation of SD male rats was 1654.2 mg · m-3 · h,suggesting that plasma ChE inhibitory rate was higher with the increase in the exposing dose,and that there was a significant difference as compared with the controls(P<0.05).
