Objective To evaluate an ideal way to prepare the extracellular matrix of urethra. Methods An orthogonal design [L9(34)] was used in the experiment.Urethras were obtained from 37 rabbits, among which 27 segments were randomly selected and were decellularized following the orthogonal design in 9 groups. The whole experiments were repeated for 3 times. After the decellularization process, the acellularity of the ECM was examined by haematoxylin-eosin staining. The optimum way was found out through comparing the numbers of the remained cellular elements by computer image analysis. An ideal way was found by statistic analysis. Then the ECM was obtained from 10 pieces of urethras by the optimum methods. The scanning electronic microscopy was used to confirm the decellulary matrix.Subsequently, the ECM was used as a graft for replacement. In 10 rabbits, the urethral defect were replaced with the urethral ECMs. At sacrifice,10 days,3 weeks,6 weeks and 24 weeks, the grafts was taken out,and the regeneration was confirmed by the haematoxylin-eosin staining. Results ECM resulting from different dedellularization process in the urethras are different in the numbers of remaining cellular elements. There are no cellular elements in the 7th and the 9th group of the tissues. The cellular elements was not found by the scanning electronic microscopy in the ECM getting from the optimum methods. In the animals with replacement, histologic examination showed complete regeneration 24 weeks post operation. Conclusions The best way to prepare the ECM of urethra is A 3B 2C 3.

Objective This study was to investigate the diagnostic value of CT-guided percutaneous needle lung biopsy ( PN-LB) in different types of lung cancer and compare the pathological results of CT-guided PNLB and surgical specimens. Methods We performed a retrospective analysis of 324 cases of lung cancer, all confirmed by CT-guided PNLB after imaging manifestation of lung-occupying lesion and treated by surgical resection. We compared the pathological results of CT-guided PNLB and surgical speci-mens. Results The total coincidence rate of the pathological result from CT-guided PNLB with that from surgical specimens was 85.8% (278/324), with even a higher coincidence rate (96.4%) in adenocarcinoma, followed by squamous cell carcinoma (86.1%), and small cell lung cancer (45.5%).Large cell lung cancer and adenosquamous carcinoma were liable to be diagnosed as adenocarcinoma by CT-guided PNLB ( 7/10 and 6/10 ) .McNemar-Bowker test revealed no significant differences in the coincidence rate in adenocarcinoma, squamous cell carcinoma, and small cell lung cancer (κ=0.837, P=0.215). Conclusion CT-guided PNLB has a high coincidence rate with surgical specimens in the pathological diagnosis of lung cancer, but the coincidence rate varies with different pathological types of the tumor.

Objective To evaluate the effect of recominant human growth hormone(rhGH) in the prevention and treatment of endotoxemia(ETM) in murine experimental obstructive jaundice (OJ) Methods Twenty OJ rats received subcutaneously given rhGH of 0 75*!IU/kg daily for 2 weeks (rhGH group). Blood endotoxin(ET) was measured, and intestinal mucosa was observed under electron microscope at 5th week. Results In rhGH group, ET level 〔(0 40?0 02)*!EU/ml〕 was significantly lower than OJ rats〔(0 77?0 03)*!EU/ml, n =20〕 ( t = 6 237, P 0 05). Under electron microscope bowel villi in OJ group were distroyed, epithelial cells were degenerated and necrotic, whereas in rhGH group the ultrastructural pathology was much less evident and similar to that in sham operation rats. Conclusion rhGH has significant effect on protecting the injured mucosa barrier in OJ,and decreases ETM significantly.

Objective To investigate the effects of rutaecarpine on the cerebral injury and calcitonin gene-related peptide (CGRP) level during focal cerebral ischemia-reperfusion injury in rats. Methods Rats were intravenously given three doses of rutaecarpine (50, 100 and 300?g/kg) or vehicle 30 minutes before experiment. Then the rats subjected middle cerebral artery occlusion (MCAO) for 2 hours, and neurological deficits scores were performed at 6, 12 and 24 hours after reperfusion. After the last test the animals were sacrificed, the infarct volumes were determined by TTC staining, and CGRP levels were measured by radioimmuoassay. Results Rutaecarpine significantly reduced infarct volume, and improved cerebral function in dose-dependent manner compared with vehicle. Rutaecarpine significantly inecreased brain CGRP levels after reperfusion as well. Conclusion Rutaecarpine has marked protective effects on ischemic brain damage in rats possibly by increasing CGRP release in the brain.

Objective To study how and why the lipo-alkaloids changed in decocting the flowers of Aconitum kusnezoffii (FAK). [WT5HZ]Methods The alkaloids in ethanol extract of FAK, FAK decoction, and the residues of decocted FAK were analyzed and compared by electrospray ionization tandem mass spectrometry (ESI-MSn). Results No lipo-alkaloids were detected in FAK decoction, however, triester-lipo-alkaloids become the dominant components in the residues of FAK decoction. Conclusion Besides hydrolysis reactions, ester-exchange reactions happen for diester-aconitines and triester-aconitines in the decocting of FAK. It is the first time to report that C_8-acetyl is displaced by long chain fatty acyl for triester-aconitines in the ester-exchange reactions.
