Objective To evaluate the changes and clinic countermeasures of cough and expectoration in the patients with lung cancer after airway stent placements. Methods With the fibrobronchoscope, metal stents were placed in tracheas, main bronchia or right intermediary bronchia. It was compared that the degrees of cough and expectoration difficulties, the volumes and mucin content of sputa and lung functions before the placements, within 3 d and 1 to 2 months after operation. Results Within postoperative 3 days, the patients coughed more frequently, but VC, FEV_(1) and PEF were improved significantly. In postoperative 1 to 2 months, the degrees of cough were close to preoperative degrees, lung functions kept on improved conditions, but expectoration difficulties, volumes and mucin contents of sputa increased markedly, especially in the patients with tectorial membrane stents. Proper stents and reasonable treatment including use of antitussive, apophlegmatisant and antibiotics could decrease the degrees of cough within postoperative 3 days, and expectoration difficulties, volumes and mucin contents of sputa in postoperative 1 to 2 months. Conclusion To the patients with malignant central airway constriction, metal stent placement would improve ventilative functions but bring difficulties for cough and expectoration. The side complications would be relieved through corresponding treatments.

To provide the basis for preparation of diagnostic kits and vaccines in Toxoplasma gondii infection, the gene coding for the qualified recombinant p30 protein (SAG1) of this parasite was amplified by PCR, and the amplified gene was cloned into prokaryotic expression vector pET-30a(+) to construct the recombinant plasmid, and then transformed to E.coli DH5α. The positive recombinant plasmid was screened by PCR and double enzymes digestion, and the nucleotide sequence of p30 gene was determined by automated DNA sequencing. Meanwhile, the identified recombinant plasmid was transformed to E.coli BL21(DE3) with the expression of p30 on bacteria induced by IPTG and the expressed protein was identified by SDS-PAGE. The protein obtained was then further purified and refolded, and its biological activity was checked by Western blotting. It was shown that the size of the amplified gene was 750 bp with molecular weight of 30 ku, and this protein could specifically react with monoclonal antibody against p30 protein.

OBJECTIVE:
To investigate the correlation betwen varying degrees of non-acute tinnitus and anxiety and depression.
METHOD:
Seventy-seven outpatients with non-acute tinnitus as their in chief complaint were submitted to Tinnitus Handicap Inventory(THI), Self-Rating Anxiety Scale(SAS), and Self-Rating Depression Scale (SDS).
RESULT:
THI and its three subscales were found to have significant correlations with SAS and SDS. The group (THI ≥ 38) had more anxiety and depression than the mild (THI < 38). Significant correlations were also observed between THI, SAS and SDS in the group with THI ≥ 38, in contrast with the group of THI < 38.
CONCLUSION
Patients with THI ≥ 38 suffered from severe anxiety and depression than the mild. Doctors should pay more attention to these patients, especially their psychological disorders.

Objective To report a three-generation Chinese family with freckle and to make a genetic linkage analysis in this family.MethodsGenetic linkage analysis was carried out in this family using microsatellite markers distributed over chromosome 4q and 1.Two-point logarithm of odds(LOD)scores were calculated using the Linkage program package(version 5.1),and haplotype was analyzed with Cyrillic version 2.01 software. Results Freckle was inherited in an autosomal dominant pattern with a penetrance of99.9% in this family;linkage to chromosome 4q was ruled out however, supportive evidence was obtained for linkage to microsatellite markers D1S2635 and D1S2844 in chromosome 1q with a maximum LOD score of 1.50.Haplotype analysis in this family localized the locus of freckle to a 12 Mb region flanked by D1S2624 and D1S2799.Conclusions Freckle is a genetically heterogeneous disorder. The causative gene may be located in a 21.2 cM region on chromosome 1q22-24.

Objective: To report a case of azoospermia with a karyotype of 45,X,der(Y)t(Y;13)(q11.2;q12),-13,accompanied with slight bilateral gynecomastia and multiple nodules. Methods: The karyotype was identified by karyotyping and FISH,and the breakpoints of the Y chromosome and the copy number of the BRCA2 gene in 13q12 determined by PCR-STS and DNA polymorphic analysis. The testis and nodule tissues of the patient were obtained for biopsy. Results: FISH confirmed SRY and centromere of the Y chromosome on the questionable 13 chromosome and the karyotype to be 45,X,der(Y)t(Y;13)(q11.1;q12),-13.ish der(Y)(SRY+,DYZ3+,wcp13+).PCR-STS showed the deletion of regions AZFa,b and C,with a breakpoint located inYq11.1 below sY82.No deletion of the BRCA2 gene was observed. The patient was diagnosed with Sertoli cell-only syndrome by testicular biopsy and with angiolipomata by pathological examination of the nodule tissue. Conclusion: The patient's phenotype of complete masculinization could be attributed to presence of the SRY gene, and his azoospermia with small testis to the absence of a fragment from Yq11.1 to Yqter. However, the molecular mechanism of angiolipoma remains unknown.

Background Stromal cell-derived factor-1_?(SDF-1_?)has been demonstrated to be essential for stern cell mobilization/homing.Recent evidence indicates that SDF-1_? has been expressed in injured carotid arter- ies.Besides, high SDF-1_? plasma levels are clinically associated with stable coronary artery disease. Objective To investigate whether SDF 1 involves in mobilization of endothelial progenitor cells(EPC)and reendothelialization after vascular injury. Methods SDF-1_? was detected by RT-PCR and Western blot in carotid arteries of mice at different time points after wire-induced injury.SDF-1_? determination in peripheral blood samples and BM was per- formed by SDF-1_? enzyme-linked immunosorbent assay(ELISA)kit.EPC in peripheral blood collected at different time points after vascular injury were quantified by flow cytornetry. In subgroup, blocking SDF-1 rnonoclonal anti- body was injected, peripheral blood EPC were quantified after vascular injury and reendothelialization of injured ar- teries was determined 14 days later. Results Expression of SDF-1_? was evident at day 1,and peaked at day 3 after arterial injury.A rise in plasmatic concentration of SDF-1_? and a significant reduction of SDF-1_? in bone marrow concentration was noticed at all time points following injury. The amount of circulating EPC was increased shortly after induction of vascular injury and persisted up to 7 days(P
