Chinese Journal of Rehabilitation Theory and Practice 2013;19(10):970-971
Objective To explore the clinical effect of hydrochloride phenazopyridine on relieving the pain after cystoscopy. Methods 60 male patients undergoing cystoscopy were divided into observation group and control group according to the block randomization method with 30 patients in each group. 1 h after cystoscopy, the observation group was given orally hydrochloride phenazopyridine 0.2 g, 3 times a day for 0.6 g totally. The control group was not treated with oral medication for pain. The pain degree was evaluated with Numerical Rating Scale (NRS), and the side effects were recorded. Results There was no significant difference in NRS score immediately after cystoscopy between 2 groups (P=0.725). The NRS score was lower in the observation group than in the control group 24 h after cystoscopy (P=0.002).Conclusion Hydrochloride phenazopyridine can effectively relieve the postoperative pain of cystoscopy.

AIM:To select an optiomal formulation of liposome entrapped alkanin prepared with ethanol injection in the combination of orthogonal design and central composite design/Response surface method.METHODS:Five influencing factors, including ratio of alkanin to liposome and soyabean lecithin to cholesterol, volume ratio of ethanol phase to water phase, soybean concentration and mannitol concentration were chosen as indices by orthogonal design. Then the method utilized the central composite design, which fitted multivariate linear and second/third order nonlinear regression equation,to find out the optimal formulation.RESULTS:Third order regression equation gained made the good prediction about entrapment(85.4%)and alkanin load(5.16%),and exhibited no significant difference between theoretic value and measured value.CONCLUSION:The combination of orthogonal design with central composite design is a feasible and convenient method to optimize the prescription of alkanin-liposome.

Objective:To generate rabbit polyclonal antibody against human Argonaute2 (Ago2) protein and to identify its functional characterization for determination of differential expression and cellular localization of Ago2 protein in various cell lines. Methods:DNAstar software was applied for searching the high antigenicity region of Ago2 gene sequence termed k-Ago2.Prokaryotic expressing plasmid was constructed and transformed to E.coli BL21 (DE3) to induce expression by IPTG.The fusion protein was injected into rabbits subcutaneously to produce polyclonal antibodies after purification by gel regaining.ELISA was operated to detect antibody titer.Western blot was used to identify the specificity and sensitivity of the antibodies and detect the differential expression of Ago2 protein in various cell lines.Meanwhile, immunofluorescence experiments were arranged to show cellular localization of Ago2 protein. Results:The prokaryotic expressing plasmid was constructed correctly.K-Ago2 protein was expressed and purified, and then rabbit polyclonal antibodies against Ago2 were generated after immunization with k-Ago2 protein. The titer detected by ELISA was 1∶19 000.Western blot results demonstrated the high specificity of the antibodies. Finally,we successfully observed the differential expression and cellular localization of Ago2 protein in various cell lines. Conclusion:The polyclonal antibody against Ago2 protein has been achieved successfully. It will be propitious for the intensive study of the RNAi mechanism and even profound clinical application.

Journal of Shanghai Jiaotong University(Medical Science) 2001;21(2):170-172
Objective The purpose of this study is to assess the prognostic significance of over expression of P53, C-erbB-2 in breast cancer. Methods In 48 patients with breast cancer the expression of P53, C-erbB-2, PCNA and ER was determined by the S-P staining immunohistochemical technique. Results The positive rate of P53, C-erbB-2, PCNA and ER were 54%, 43.7% 52%, 41.6% seperateiy. Expression of P53 and C-erbB-2 increased with the class of the tissuess. There is significant difference between grade Ⅰ and Ⅱ , Ⅲ (P<0.05) as well as between lymph node metastasis (+) and lymph node metastasis (-) patients (P<0.05). Conclusion The over expression of P53, C-erbB-2 may be a useful prognostic indicator in breast cancer.

Objective To explore the related neurobiochemical mechanism by comparing the concentration change of dopamine (DA),dihydroxy-phenyl acetic acid (DOPAC),glutamate (Glu),and γ-aminobutyric acid (GABA) in the brain tissues in schizophrenia (SZ) developmental model rats and chronic medication model rats. Methods A total of 60 neonatal male Spragur-Dawley (SD) rats were randomly assigned to 3 groups at the postnatal day 6:an SZ developmental rat model group (subcutaneous injection with MK-801 at the postnatal day 7-10,0.1 mg/kg,Bid),a chronic medication model group (intraperitoneal injection at the postnatal day 47-60,0.2 mg/kg,Qd),and a normal control group (injection with O.9% normal saline during the corresponding periods).DA,DOPAC,Glu,and GABA of the tissue homogenate from the medial prefrontal cortex (mPFC) and hippocampus were examined with Coularray electrochemic detection by high performance liquid chromatogram technique. The utilization rate of DA and Glu was calculated. Results Compared with the normal control group, the concentration of DA and DOPAC in the mPFC and the hippocampus in the SZ developmental model group significantly decreased ( P < 0.05 ),and the GABA concentration and Glu utilization rate in the mPFC also decreased (P < 0.05 ).Compared with the chronic medication model group, the DA concentration of the mPFC in the SZ developmental group decreased ( P < 0.05 ),and the DOPAC concentration and the utility rate of DA in the hippocampus also decreased (P <0.01,P <0.05,respectively).Conclusion The activities of DA,Glu and GABA system decrease in the mPFC and the DA system function reduces in the hippocampus of SZ developmental rats.
