1.Expression of p53 and E-cadherin in gastric adenocarcinoma and their relationship with prognosis

Purpose To investigate the protein expression of p53 and E-cadherin in gastric adenocarcinoma and the effects of them and the clinicopathological parameters on prognosis of patients.Methods Expression of p53,E-cadherin in 150 cases of gastric adenocarcinoma was detected by tissue microarray and immunohistochemistry.The effects of them and the clinicopathological parameters on the prognosis of patients were studied by Kaplan-Meier method to calculate and compare the cumulative survival rate and postoperative mean survival time.Multivariate analysis was also performed by COX regression.Results The positive rate of p53 and E-cadherin expression in 150 cases of gastric adenocarcinoma was 31.3% and 91.3%,respectively.74 cases were followed up, and the 1-,3-,and 5-year survival rates were 83.8%,70.3% and 63.5%,respectively.The invasion, lymph node status and pTNM stages were prognostic factors;multivariate analysis revealed that p53,age and lymph node status were independent prognostic invasion, lymph node status and pTNM stages are important prognostic factors in gastric adenocarcinoma. Nevertheless, E-cadherin and histological classification are not prognostic factors.
3.A new selaginellin derivative from Selaginella pulvinata.

Abstract: A new selaginellin derivative named as selaginellin S (1) was isolated from the whole plants of Selaginella pulvinata (Hook. et Grev.) Maxim. (Selaginellaceae), together with a known one (selaginellin M, 2). Compounds 1 and 2 were separated and purified by silica gel and Sephadex LH-20 column chromatography. Their structures were determined on the basis of extensive spectroscopic analysis including IR, MS, 1D and 2D NMR experiments, as well as ECD calculations. Compound 1 is a key intermidiant in the biosynthesis pathway of selaginellins. Compound 2 is first reported in this plant.

4.Radiographic Study of the Modified Femoral Tunnels of Anterior Cruciate Ligament Reconstruction

Objective To modify anterior cruciate ligament (ACL)reconstruction to achieve better structure;to measure the tunnel's position and shape using three-dimensional computed tomography (CT)so as to show the change of the femoral bony tunnel.Methods Forty-five patients were diagnosed with an ACL tear and subsequently underwent the modified ACL reconstruction,using a minor-diameter drill to drill the femoral tunnel and then expand it to oval-shaped tunnel with the raspatory between July 2014 and 2015.All procedures were conducted by the same experienced surgeon (Ao Yf.).In all patients,CT was performed at one week after the ACL reconstruction to evaluate the femoral tunnel's positioning,then the CT-images were imported into the image processing software MIMICS to reconstruct the femoral bony tunnel to measure the tunnel's diameters of the section and the shape.The tunnel's position was evaluated using the quadrant method.Results In all the 45 patients,the graft could pass the tunnel.And no posterior tunnel wall blowout,neurological or vascular deficit was documented during the operation.According to the different methods of modification (6 to 7,6 to 8 and 7 to 8),the long diameters of the sections of the femoral bony tunnels were 8.34-± 0.52 mm,10.65 ± 0.72mm,9.27-± 0.11 mm,without significant difference from the theoretical value (8.16 mm,10.66 mm and 9.14 mm).However,the short diameters of the sections were 6.57--0.12 mm,6.74-± 0.13 mm,and 7.52 ± 0.05 mm,showing significant difference from the theoretical value (6mm,6mm and 7 mm).The average centre of the femoral tunnel was located at 26.4 ± 4.8% of the femoral length and at 30.1 ± 5.6% of the femoral height.Conclusion In this modified ACL reconstruction,a minor-diameter drill is used to drill the femoral tunnel and then expand it to oval-shaped tunnel with the raspatory,The graft was matched well with the bony tunnel intraoperatively.The three-dimensional CT shows that the femoral tunnels are well located within the anatomical ACL footprint,and its entrance is closer to the natural state.
5.MuItipIe xenobiotic metaboIism-associated cytochrome P450 isoforms might be invoIved in earIy activation of rat hepatic steIIate ceIIs in vitro

OBJECTIVE To investigate the role of several xenobiotic metabolism-associated CYP450 isoforms in the auto-activation of hepatic stellate cells(HSCs) invitro. METHODS HSCs were isolated from adult Wistar rats and cultured on plastic as an in vitroauto-activation model. Positive expression of α-smooth muscle actin(α-SMA)was used as an activated marker of HSCs and detected by immunocytochemical staining in HSCs cultured for 1,2,5 or 11 d. The expressions of CYP450 isoforms were analyzed by real-time RT-PCR in the HSCs. RESULTS The immunocytochemical stai-ning showed no α-SMA expression in HSCs cultured for 1 d,while its expression gradually increased during culture since day 2. ln terms of α-SMA expression,HSCs cultured for 1,2,5,and 11 d were classified as the quiescent,early,middle and later stages of activation,respectively. The RT-PCR results revealed that CYP1B1,CYP2B1/ 2,and CYP2E1 mRNA were expressed at high levels in the early stage of HSCs activation(at day 2),which were 2.1-,1.6-,and 23.9-fold those in the quiescent HSCs(day 1),respectively. Further study revealed that mRNA expressions of these up-regulated CYPs in the early stage of activation were diminished at the subsequent two stages. The levels of CYP1A1 and CYP1A2 mRNA decreased constantly throughtout the activation process. CONCLUSION Multiple xenobi-otic metabolism-associated CYP450 isoforms might be involved in the auto-activation of rat HSCs invitro.

7.Location and expression of apo(a) and apoB in peripheral blood of patients with cardiovascular disease

Objective:To explore the relationship between the severity of cardiovascular disease with the expression of apolipoprotein(a) [apo(a)] and apolipoprotein B(apoB) in peripheral blood and their location in peripheral blood cells.Methods: In this report,we selected 4 patients with angiography which indicated that three coronary arteries were narrowed and 5 control patients with normal angiography.Arterial blood was collected and analyzed for lipid parameters in plasma.The mRNA expression of apo(a) and apoB in peripheral white blood cells and platelets were determined by RT-PCR and their protein expression by western blot.Moreover,the expression and location of apo(a) and apoB in white blood cells were determined by confocal microscopy and computer 3D analysis.Results: In plasma,levels of high density lipo-protein-cholesterol(HDL-C) and apolipoprotein A-Ⅰ(apoA-Ⅰ) in cardiovascular disease(CVD) patients were significantly less than those in the control patients[(0.62?0.05) mmol/L,(0.78?0.08) mmol/L vs(0.81?0.15) mmol/L,(0.9?0.07) mmol/L,P0.05).Studies with confocal microscopy indicated that proteins of apo(a) and apoB were co-expressed by a few cells of leukocytes and the ratio of apoB/apo(a) in cardiovascular disease patients was significantly less than that in the control patients(optical density value 1.60?0.12 vs 4.40?0.35,P
8.Preparation and in Vitro/in Vivo Evaluation of Indomethacin Multi-dosage Mini Tablets for Colon Target Delivery

Objective Enzyme triggered multi unit colon targeting mini tablet of indomethacin were prepared,in order to improve the target treatment of colon disease.Methods Different proportion of enteric layer and chitosan layer were screened to optimize the prescription.The colon targeting mini tablets were prepared by direct compression method.The drug release properties were investigated in different release medium.Rats were used to investigate the distribution of tissue in vivo.The Beagle dogs were used to study the pharmacokinetics and bioavailability.Results The optimum chitosan layer prescription:coating liquid concentration was 2%,plasticizer three citric acid ethyl ester (TEC) was 15%,an anti sticking agent amount of talc was 30%,coating weight was 5%;Enteric layer prescription:coating liquid solid content was 20%,plasticizer content of TEC was 5%,anti sticking agent talc powder dosage was 40%,coating weight was 3%.The chitosan multi unit colon targeted preparation seldom released in rat stomach and small intestine,released slowly in colon.The pharmacokinetics parameters in Beagle dogs were:Cmax =(3.25 + 0.672) mg·L-1,tmax =(2.00 + 0.014) h,AUC(0.∞) =(10.2 +0.871) mg·L-1 ·h,MRT (0-∞) =(2.82 + 0.180) h,CL =(2.46 + 0.202) L·h-1 ·kg-1.The release time of mini tablets for colon targeted was significantly prolonged and preserved stable blood concentration.Conclusion The enzyme triggered multi unit colon targeting mini tablet of indomethacin showed good target to colon and sustained release effect,providing an important reference for the development of preparation of indomethacin for the treatment of colon disease.
9.Effect of Acupuncture on the Infracted Area and Apoptosis of Rats with Brain Contusion

Objective To observe the effect of acupuncture on the infracted area and apoptosis of rats with brain contusion.Methods Feeney's method was used to establish brain contusion model.The 40 rats were randomized into sham-operation,model,acupuncture and naloxone (NX) group with 10 in each.Acupuncture was performed for the rats of acupuncture group and NX was injected intraperitoneally for the rats of NX group.The treatment was done once everyday,7 days altogether.Then the brain histotomy was prepared after all the treatment finished.Computer image analysis system was used to detect the infracted area.TUNEL method was adopted to detect the apoptosis.Results The brain contusion in the model group was severe,there were lots of apoptotic cells and the infracted areas were bigger than the injured areas (P0.05).Conclusion Acupuncture can decrease the infracted areas and apoptosis of the rats with brain contusion,being effective in anti-secondary brain injuries.
10.The anatomical study of the pterygopalatine fossa under endoscopy via the nasal cavity approach.

OBJECTIVE:
To provide a basic anatomical study of clinical endoscopic surgical approach, we investigated the pterygopalatine fossa anatomy through lateral wall of the nasal cavity approach under endoscopy.
METHOD:
To observe important symbols and the neurovascular structure and its relations with the surrounding structure of pterygopalatine fossa, we dissect ten fresh cadaveric heads via lateral wall of the nasal cavity approach under endoscopy.
RESULT:
(1) The anatomical relations between pterygopalatine fossa and the surrounding structure were complicated, and internal maxillary artery and its branches were variate greatly. (2) Sphenopalatine foramen, infraorbital canal, foramen rotundum and pterygoid canal were the important symbol of bone of pterygopalatine fossa, and pterygopalatine fossa could be used as an access into the infratemporal fossa and sphenoid sinus.
CONCLUSION
(1) An intimate knowledge of the constant anatomical symbols of the pterygopalatine fossa and the surrounding structure would maintain a sense of direction and improve surgery safety. (2) The pterygopalatine fossa could be exposed fully under endoscopy, and the vision was clear. Besides, the important nerves and vessels would be well protected, and operative approach was flexible according to the scope of pathological changes. (3) The near anatomic region could be accessed and the pathological changes could be treated through the endoscopic surgical approach.
