The key constructional components for a bioartificial liver(BAL) include the source of hepatocyte cell line, the semi-permeable membrane or bioreactor and the delivery system. The most important part of a BAL is cell line. The properties of the hepatocyte cell line will directly affect the support efficacy of a BAL. This article reviewed the recent progress in researches on hepatocyte cell line used in BAL.
2.On Harmonious Physician-Patient Relationship and Hospital Culture Construction

3.Protective effect of cataractogenic lens injury on RGCs in optic nerve axotomy eye in vivo and its mechanism

Background It has been reported that murine Müller cells conditional medium can promote the survival of retinal ganglion cells (RGCs) and the regeneration of axons,and the survival rate of RGCs improve in the optic nerve axotomy eyes with cataractogenic lens injury in vitro.However,the interaction of Müller cells with pricking of lens in protecting RGCs is unclear.Objective The aim of this study was to investigate the role of Müller cells on survival of RGCs in the optic nerve axotomy with cataractogenic lens injury.Methods Forty-eight clean adult Wistar rats were randomized into sham operation group,optic nerve axotomy group and lens injury combined with optic nerve axotomy group.The optic nerve was exposed only in the rats of the sham operation group,optic nerve was completely transected at 3 mm behind the eyeball in the rats of the optic nerve axotomy group,and lens puncture and optic nerve axotomy were performed in the eyes of lens injury combined with optic nerve axotomy group.The rats were sacrificed at day 7 and day 14 after operation to prepare the retinal specimens.The RGCs were examined and counted by hematoxylin-eosin staining.Müller cells labeled by glial fibrillary acidic protein (GFAP) were counted using immunohistochemisty.Results The number of RGCs was (52.98 ± 1.90) /field and (51.81 ±3.09) /field on the 7th and 14th day in the sham operation group,without significant difference between them (t =0.910,P =0.378).The number of RGCs was significantly lower on the 14th day ([22.67±1.94] /field) than that of the 7th day ([36.61±1.69] /field) in the optic nerve axotomy group (t=15.312,P=0.000).Also,the number of RGCs was (50.76±2.77) /field and (35.69±1.80) /field on the 7th and 14th day in the lens injury combined with optic nerve axotomy group,showing a significant difference between the two timepoints (t =12.920,P =0.000).In addition,the RGCs number in the lens injury combined with optic nerve axotomy group was significantly higher than that in the optic nerve axotomy group both on 7 days and 14 days after operation (7 days:t =102.840,P =0.000; 14 days:t =164.020,P =0.000),and the number of RGCs was lower in the lens injury combined with optic nerve axotomy group than that of the sham operation group on day 14 (t =187.040,P =0.034).None of GFAP-labeled Müller cell was seen in sham operation group at both on 7 days and 14 days after operation,but a significant difference was found in the optic nerve axotomy group between the two timepoints ([29.38 ± 2.04]/field vs.[19.07 ± 2.14]/field ; t =-9.868,P=0.000).No significant difference in the number of the GFAP-labeled Müller cells was found in the lens injury combined with optic nerve axotomy group between 7 days and 14 days after operation([48.96±2.80] /field vs.[46.73±1.50]/field,t=1.987,P=0.067).In postoperative 7 days and 14 days after operation,the number of GFAP-labeled Müller cells increased in the lens injury combined with optic nerve axotomy group compared with the optic nerve axotomy group (7 days:t =-15.997,P=0.000; 14 days:t=-29.938,P=0.000).Conclusions In optic nerve axotomy with cataractogenic lens injury eye,the punctural injury of lens induce the activity of Müller cells and further promote the survival of RGCs in the cataratogenic lens injury combined with optic nerve axotomy rat eyes.
4.Influence of White Prepared Lateral Root of Aconite on Blood Biochemical Indicators in Normal and Kidney Yang Deficient Rats

Objective To explore the differences of blood biochemical indicators between normal and kidney Yang deficient rats administered orally different doses of white prepared lateral root of aconite.Method Eighty Sprague-Dawley(SD) male rats were divided randomly into normal group and kidney Yang deficient model group,forty rats in each group.After producing kidney Yang deficient model by injecting hydrocortisoni natrii succinas in intramuscular,normal and model rats were respectively and randomly divided into little,low,medium,great dose group and blank group,eight rats in each group.Those rats were administered orally with different concentrations of white prepared lateral root of aconite every day for two weeks.Twenty-four hours after the last oral administration,the blood were obtained and used for testing adrenocorticotrophic hormone(ACTH),cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),glucose(GLU),cholesterol(CHO),alanine aminotransferase(ALT),urea nitrogen(BUN).Results Before taking medicine,model rats' cAMP/cGMP and ACTH were lower than normal rats',and they showed a trend of rise with adding dose.Before taking medicine,model rats' GLU was lower than normal rats'.With adding dose,model rats' GLU increased,however,normal rats' GLU reduced.When taken medium dose white prepared lateral root of aconite,model rats' CHO was higher than normal rats'.Taking medicine made both groups ALT rise,but normal rats' went up more quickly than model rats'.Model rats' BUN was higher than normal rats',and taking low dose or little dose medicine could lead to incline of reduction.Conclusion White prepared lateral root of aconite caused different blood biochemical effects on normal and kidney Yang deficient rats,and more remarkable toxicity effect on normal rats.
5.Computer-aided design for occlusal rest of the removable partial denture

Objective:To explore the route of automatic computer-aided design for occlusal rest of the removable partial denture.Methods: A new module for the design of occlusal rest was developed.The three-dimensional data of a partially dentate dental cast was obtained and imported into the new module.The contour of the occlusal's tissue surface was identified.Then,the tissue surface and polished surface were constructed respectively and connected to each other to form an integrated entity.Results: The surface of the occlusal rest was smooth and the curvature matched that of the surrounding area of the abutment tooth.The accuracy was satisfying.Conclusion: Design of occlusal rest was much more convenient and time-saving using the newly developed module.The future development of special software package for the design of removable partial denture will benefit from it.
6.Effects of inhaled nitric oxide on erythrocyte deformability in septic acute lung injury

Objective To investigate the effects and related mechanism of iNO on erythrocyte deformability in septic acute lung injury(ALI).Methods Thirty anesthetized and mechanically ventilated young piglets(3-4weeks old)were allocated to 4 groups:(1)normal(group C,n=6):intraperitoneal injection of normal saline(NS).(2)normal+iNO(group CNO,n=4):NS followed by mechanical ventilation for 4 h,and then treated with iNO at 10 parts per million in volume(ppm).(3)model(group M,n=10):i.p.Escherichia coli at 5 × 109 cfu/ml,5 ml,and(4)NO(group MNO,n=10):i.p.Eschefichia coli at 5×109 cfu/ml lollowed by iNO-at 10 parts per million after ALI.Blood viscosity,malondialdehyde(MDA)in erythrocyte,Ca2+-Mg2+-ATPase and Na+-K+-ATPase of erythrocyte membrane were measured at basdine,establishment of ALI,12 and 24 h during the treatment.Results For normal age-matched piglets,there was little change of whole blood viscosity at high shear rate(110 s-1),which is a measure of erythrocyte deformability,so was the level of MDA in erythroeyte and the activity of erythroeyte membrane Ca2+-Mg2+-ATPase,Na+-K+-ATPase when iNO was provided for 12 or 24 h.At the time ALI was established,whole blood viscosity at high shear rate and MDA levels in erythroeyte increased significantly compared to the baseline level(P<0.05),while the activity of erythrocyte membrane Ca2+-Mg2+-ATPase and Na+-K+-ATPase decreased significantly(P<0.01,vsbaseline).Exposure of iNO for 12 h resulted in decreased whole blood viscosity at high shear rate and MDA levels in erythroeyte(P<0.05,vs group C),and higher activity of erythrocyte membrane Ca2+-Mg2+-AT-Pase and Na+-K+-ATPase.When iNO was administered for 24 h,there was little difference of whole blood viscosity,activity of erythrocyte membrane Ca2+-Mg2+-ATPase,Na+-K+-ATPase and MDA levels in erythroeyte between the C and NO groups.Conclusion In the piglets with septic ALI,there was deterioration of erythrocyte deformability.Inhaled NO for 12 h may alter the deterioration,however,this effect is transient.
7.Extrapericardial adipose-derived stem cells differentiate into myocardial cells

BACKGROUND:Adult stem cells are capable of proliferation and differentiation, which are an important part of tissue engineering. Although several studies have demonstrated that human adipose-derived stem cells obtained from elective liposuction procedures represent the characters of mesenchymal stem cells and can be induced to differentiate into myocardial cells, less is known about the characters of extrapericardial adipose-derived stem cells and their differentiation into myocardial cells.
OBJECTIVE:To explore the culture of extrapericardial adipose-derived stem cells in vitro and their ability to differentiate into myocardial cells.
METHODS:The extrapericardial adipose tissues were obtained and then isolated, cultured and passaged in vitro. The expressions of cellsurface specific antigens (CD44 and CD90) were detected by immunofluorescence assay at different time periods, and the fluorescence intensity was compared. After induced and cultured with myocardium induction medium for an optimal period,α-actin and cardiac troponin T expressions were detected by immunocytochemical detection and the phenotypic identifications were performed.
RESULTS AND CONCLUSION:During the in vitro culture, extrapericardial adipose-derived stem cells adhered to culture flask wal and proliferated strongly. The cells appeared to be shuttle-shaped and exhibit a vortex alignment. They were positive for CD44 and CD90, and the fluorescence intensity reached the peak at passages 3 and 4, but negative for CD34 and CD54. After induced and cultured with myocardium induction medium, the differentiated cells were muscle-like cells, the expression ofα-actin and cardiac troponin T were positive, and the number of cells reached the peak at passages 3 and 4. These findings suggested that the extrapericardial adipose-derived stem cells from human represent the morphological and cellular immunological characters of mesenchymal stem cells, and they can be induced to differentiate into myocardial cells. In addition, there is the best induction period.
8.Construction of inlay axial surface using B-Spline curve network

Objective: To realize the requirement of adjacent area accuracy,axial surface physical contour and smoothness in the process of inlay computer-aided design(CAD).Methods:A compromised method was brought forward that some assistant 3-D constraint lines from occlusion to gingiva were used to lead the inlay contour on axial corner and other characterized areas,and two perpendicular 3-D constructing lines at adjacent area were used to construct accurate 3-D configuration of this area.The axial surface was completely defined by the network of specified curves which were two constructing lines and four borders.Results:Inlay axial surface was smooth,the contour was consistent with the residual tooth body and the 3-D configuration in adjacent area was accurate.Conclusion:It is feasible to realize the inlay axial surface accuracy,smoothness,and consistency using the method of special distributed 3-D B-Spline curve network.
9.Posterior-anterior approach surgical treatment for severe cervical spondylotic myelopathy

[Objective]To explore the therapeutic effect of the posterior-anterior approach surgical treatment for severe cervical spondylotic myelopathy.[Method]Totally 32 patients with severe cervical spondylotic myelopathy operated by posterior-anterior approach surgical treatment from September in 1998 to January in 2005 were retrospectively analyzed.In the 32 patients,20 males and 12 females.Their age averaged 53.8 years(ranged,41~78 years),invasional segments:C3、42 cases,C4、5 9 cases,C5、611 cases,C6、7 10 cases.Four cases with lesions involving one intervenebral space,the other 12 and 16 cases involving two and three intervertebral spaces respectively.All cases had the posterior cervical open-door laminoplasty decompression in first,and had the anterior cervical decompression and bone grafting or fusion with titanium Cage internal fixation after a week.To follow up all the postoperative cases,neural function was recorded.According to JOA score,preoperative score and postoperative follow-up score were compared,improvement rates were caleulated respectively.[Result]All cases were regularly followed up.The duration of follow-up averaged 13.6 months(ranged,12~18 months).The JOA of before operation was(6.51?1.10),the JOA of first period of time after operation was(9.47?0.5),and follow-up JOA was(12.42?0.79).T-test was used to compare pre-operative and follow-up JOA scores.The statistical results were significantly different(P
10.Simultaneous determination of benzophenones and xanthone in leaves of Aquilaria sinensis by RP-HPLC-UV.

This study is to develop a sensitive method by using reversed-phase high performance liquid chromatography coupled with UV detector (HPLC-UV) to simultaneously determine four bioactive compounds, iriflophenone 3-C-beta-D-glucoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside in the leaves of Aquilaria sinensis. An Agilent Zorbax SB-C, column (4, 6 mm x 250 mm, 5 microm) was used, and the gradient elution was performed with mobile phase of 0.1% aqueous phosphoric acid and acetonitrile at a flow rate of 1 mL x min(-1). The detection wavelength was 280 nm, and the column temperature was 25 degrees C. The four marker compounds were well separated with good linearity (R2 > 0.9990), precision, stability and repeatabili y. The-recovery rates were in the range of 98.80%-101.39%. For 15 branch of the leaves, the contents of iriflophenone 3-C-beta-D-gluoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside were between 0.41-14.48, 0.72-3.85, 4.30-29.07, 0.24-5.06 mg, respectivley. This method is precise, accurate and reliable, which provides an efficient way for the quality control of the leaves of A. sinensis. This will promote the comprehensive usage of this plant.
