1.Effect of concentrations of serum on differentiation of adipose-derived adult stem cells into chondrocytes

[Objective]To study the differentiation of adipose-derived adult stem cells(ADASCs)into chondrocytes induced by TGF-?1 in the presence of different concentrations of serum in vitro.[Method]Adipose tissue-derived adult stem cells were isolated from rabbits subcutaneous adipose tissue using enzymatic digestion.The adipose tissue was minced and digested with collagenase type I,and the released cells were collected by density centrifugation and then placed in culture.After being passaged three times,ADASCs were induced to differentiate into chondrocytes by chondrogenic culture media containing 1% or 10% of FBS for 2 weeks.MTT,alkaline phosphatase toluidine blue staining and type Ⅱ collagen immunohistochemistry staining were then tested in both groups.Gray value was analyzed by Leica patho-image analysis system to observe if there was any difference in differentiative status between the two groups.[Result]MTT showed that the cells of 10% FBS group had higher proliferation activity than those of 1% FBS group(P
2.Osteogenic differentiation of adipose-derived stem cells from rabbits in vitro

[Objective]To observe the differentiation of adipose derived stem cells(ADSCs)into osteogenic cells in rabbit in vitro.[Method]ADSCs were prepared by collagenase Ⅰ digestion of subcutaneous fat from the nape of Japanese white rabbit after being excised and finely minced.Cells were identified by stro-1 immunocy to chemistry,and collagen Ⅰ immunocytochemistry,alkaline phosphatase assay and Von Kossa stain after osteogenic differentiation was performed.[Result](1)ADSCs were stro-1 positive.(2)The expression of collagen Ⅰ,alkaline phosphatase and calcium deposit of ADSCs were all positive under the influence of osteogenic differentiation medium.[Conclusion]ADSCs have similar characteristics to bone marrow stromal cells,but are much more easier to have high number upon harvest and bring less pain will occur during taking ADSCs than taking BMDCs.

[Objective]To investigate the effect of reconstituted bone xenografts(RBX) on tendon-to-bone healing by means of imaging,histological and biomechanical studies.[Method]Anterior cruciate ligament(ACL) reconstruction was performed bilaterally in 25 skeletally mature rabbits using long digital extensor tendon grafts.RBX were implanted into the treated knee of each rabbit,with the contralateral knees as controls.Every three rabbits were killed at 2,6 and 12 weeks postoperatively for routine histological studies.The samples were processed through Micro CT and subsequent HE and toludine blue staining.The remaining 16 rabbits were sacrificed at 6 and 12 weeks.Their femur-tendon graft-tibia complexes were harvested for subsequent mechanical testing.[Result]At 6 and 12 weeks after operation,the values of BMD in the RBX group were higher than those in the control group(P

Aim To study the effect of tetramethylpyrazine(TMP) on binding of 125I-VEGF to VEGF receptor. Methods The mice sera were collected after peritoneal injection with big-dose TMP,low-dose TMP,protamine and NS. A reversed-phase high performance liquid chromatography(RP-HPLC) method was used to determine the TMP in mice serum. The culture medium of ECV304 was treated with the mice sera in different groups. 7.Application of stroke standardized patient combined with the subjective-objective-assessment-plan assessment recording method in the teaching of Physiotherapy

Objective:To investigate the effectiveness of stroke standardized patient (SP) combined with the subjective-objective-assessment-plan (SOAP) problem-based medical recording method in the practice teaching of Physiotherapy. Methods:A randomized controlled study was conducted, and 116 students from the undergraduate class of rehabilitation therapy were selected as research subjects. The students in the control group received traditional teaching, and those in the experimental group received the teaching model of stroke SP combined with the SOAP assessment recording method. The effectiveness of teaching was compared between the two groups at the end of the course. SPSS 24.0 was used for statistical analysis; the chi-square test was used for categorical data, and the paired samples t-test was used for comparison between groups. Results:Compared with the control group, the experimental group had significantly higher scores of practical skills (85.97±3.19 vs. 78.14±3.86, P<0.05), basic theoretical knowledge (86.00±3.74 vs. 84.07±5.24, P<0.05), and writing of rehabilitation treatment records (85.74±4.76 vs. 66.86±4.16, P<0.05). Compared with the control group, the experimental group had significantly higher scores of the dimensions of knowledge confidence (12.55±1.30 vs. 8.79±1.59, P<0.01), skill confidence (10.86±1.51 vs. 7.45±1.27, P<0.01), and communication confidence (11.69±1.71 vs. 7.83±1.44, P<0.01). Conclusion:In the practice teaching of Physiotherapy, stroke SP combined with the SOAP assessment recording method can effectively improve the professional quality and clinical thinking ability of students, enhance their professional confidence, make up for the shortcomings of traditional teaching methods, and promote the teaching reform of rehabilitation medicine.
8.Relationship between cerebral hemorrhage and depression occurrence after craniocerebral injury surgery

Objective To explore the relationship between cerebral hemorrhage and the occurrence of depression after craniocerebral injury surgery.Methods From March 2015 to March 2017,70 patients with craniocerebral injury in our hospital were selected as the subjects of study.They were divided into the observation group (n =30) and the control group (n =40) according to whether there was cerebral hemorrhage after operation.The National Institutes of Health Stroke Scale score (NIHSS),Hamilton Depression Scale score (HAMD),Glasgow Coma Score (GCS) and the levels of interleukin (IL)-1,IL-6 and high sensitivity C-reactive protein (hs-CRP) in patients with different degrees of cerebral hemorrhage were compared.The correlation between HAMD score and other indexes was analyzed.The influencing factors and independent risk factors of depression were analyzed.Results The levels of NIHSS,HAMD score and serum IL-1,IL-6 and hs-CRP in the observation group were higher than those in the control group after operation (t =13.655,36.587,45.274,46.334,70.489,P < 0.05),while the GCS score was lower than that in the control group (t =15.517,P < 0.05).Compared with mild and moderate cerebral hemorrhage,the NIHSS,HAMD scores and serum IL-1,IL-6 and hs-CRP were higher in patients with severe cerebral hemorrhage (F =8.825,10.254,23.615,18.745,7.245,P < 0.05),while the GCS score was lower (F =7.459,P < 0.05).The correlation analysis showed that the postoperative HAMD score was positively correlated with NIHSS score and serum IL-1,IL-6 and hs-CRP (r =0.289,0.364,0.333,0.258,P < 0.05)and negatively correlated with the GCS score (r =-0.276,P < 0.05).Univariate analysis showed that the level of culture,personality,cerebral hemorrhage degree and serum IL-6 and hs-CRP were the influencing factors of postoperative depression in patients with craniocerebral injury (t orx2 =6.076,6.300,8.361,30.224,99.575,P < 0.05).Multivariate analysis showed that cerebral hemorrhage and hs-CRP were an independent risk factor for postoperative depression (P < 0.05).Conclusions Cerebral hemorrhage after craniocerebral injury is closely related to the occurrence of depression,which may be related to the increase of stress-induced inflammatory factors after cerebral hemorrhage.
