1.Determination of the Dissolution of Qingyangshen Dispersible Tablets

OBJECTIVE:To establish a method for the determination of the dissolution of Qingyangshen dispersible tablets so as to provide method and parameters for quality control of this kind of tablets.METHODS:With water as dissolution medium in a rotating speed of 100 r?min-1,the dissolution of Qingyangshen dispersible tablets was determined by HPLC at a wavelength of 259 nm.RESULTS:Similar dissolution rates were obtained for 3 batches of Qingyangshen dispersible tablets,with accumulative dissolution rates of all samples at 45 minutes reached above 90%,and a good reproducibility was noted.CONCLUSION:The quality of Qingyangshen dispersible tablets is up to the standard stated in Chinese Pharmacopoeia(2005 Edition).
3.Current status and analysis of intestinal nematode infection in Jiangsu Province

Objective To understand and analyse the epidemic status of human intestinal nematode infection in Jiangsu Province, so as to provide basis for making control measures. Methods The typical thirty-nine spots in thirteen counties among the sampling spots of National Investigation on Distribution of Human Parasites in 1990 were selected and investigated according to the Methods of National Investigation Scheme on Human Principal Parasites in 2004. Results The total prevalence was 3.88%, which decreased by 46.92% compared with the investigation in 2002 (7.31%). The infection rates of hookworm, Ascaris and Trichuris were 1.14%, 1.06% and 1.73%, respectively, and the proportion of light infection was 91.75%. The rate of multiple infections was 9.40%. The proportion of hookworm infection was 78.06% in Southern Jiangsu and the proportion of Ascaris or Trichuris was 89.03% in Northern Jiangsu. The infection rate of Enterobius was 3.72% among children aged less than 12 years. Conclusions The prevalence of intestinal nematodes has decreased to lower level in Jiangsu Province. The difference of prevalence in different regions has a relationship with the disease control and social-economic, culture and health levels. The principal control work should be still put in the northern part of Jiangsu Province. The preschool children and the middle and old age peasants are high-risk population. The different control measures should be taken in accordance with the situation of different regions in the future.
4.A survey on the quality of life and symptoms of community patients with advanced cancer

Objective To investigate the quality of life(QOL) and symptoms of community patients with advanced cancer,and analyze their correlation.Methods 94 advanced cancer patients in 3 communities of Yangpu district in Shanghai were surveyed by self-designed questionnaire,FACT-G and MDASI.The correlation between QOL and symptoms was analyzed.Results QOL of advanced cancer patients was poor,and the main influencing factors were gender,age,stage of cancer,KPS score,marital statusand economic burden.There was significant negative correlation between symptoms and QOL.Fatigue,amnesia,uneasy sleep,insensible feeling,distress and dry mouth were the symptoms patients suffered mostly.Conclusions For patients with advanced cancer,evaluation of the influencing factors of QOL and better control of symptoms could improve their QOL.
5.Analysis of family clustering on principal human parasitic infections in Jiangsu Province

Objective To understand the status and characteristics of principal human parasitic infections in rural areas of Jiangsu Province in order to provide basis for decision-making of practical control measures. Methods The survey data of principal human parasites in 58 sample sites of 19 counties (cities or areas) in Jiangsu Province were analysed on family clustering by G test, according to the methods of National Investigation Scheme on Principal Human Parasites. Results The infection of Ascaris lumbricoides, hookworm, Trichuris trichinra and Clonorchis sinensis in the province had obvious family clustering, while those in each county (city or district) appeared different states in some-points with the changing of parasitic infection rates and infection degrees. Conclusions The infection rates and infection degrees of principal human parasites decrease obviously in Jiangsu Province. The family clustering is disappearing with the dropping of them. Chemotherapy in family and health education should be strengthened to consolidate the control results in future.
6.Changes of Ascaris lumbricoides infection in rural residents of Jiangsu Province and its influencing factors

Objective To investigate and analyze the changes of Ascaris lumbricoides infection in rural residents of Jiangsu Province during 13 years and its influencing factors in order to provide the basis of making practical policies for ascariasis control in future. Methods The data were analyzed about the rates of Ascaris infection and mass chemotherapy, per capita income, and the rates of the use of running water and hygienic toilets in the southern part, the middle part and the northern part of Jiangsu Province from 1990 to 2002, and the main influencing factors for Ascaris infection were explored. Results The prevalence rate of Ascaris infection decreased continuously during 13 years, from 39.51% to 2.14%, and the decrease rate was 94.58%. In 2002, the infection rate was the lowest, only 0.41%, decreased by 98.92% in the middle part of Jiangsu Province, and the infection rate was the highest, 5.09%, decreased by 89.97% in the northern part of Jiangsu Province. The accumulated rates of mass chemotherapy were 159.00% in middle part of Jiangsu Province,103.00% in the southern part of Jiangsu Province and 105.00% in the northern part of Jiangsu Prvince.The per capita income, the use of running water and hygienic latrines increased gradually from the northern part to the southern part and there were significant differences in the 3 survey regions. Conclusion The Ascaris infection rate is influenced by mass chemotherapy, economics and health conditions in rural residents, especially the mass chemotherapy.
7.Protection against myocardial ischemia-reperfusion injury in rats by a novel selective lymphotoxin binding p55TNFR

Objective To investigate the effect of a novel lymphotoxin with selectively binding to p55 tumor necrosis factor receptor (p55TNFR) on myocardial ischemia-reperfusion injury in rats in order to explore the mechanism.Methods A total of 40 SD rats were randomly (random number) assigned into four groups (n =10 in each),namely sham operation group (group A),I/R group (group B),wild type rhLTα treatment group (group C),and p55TNFR selective rhLTα (rhLTα-Q107E) treatment group (group D).After I/R model rats were established,various therapeutic agents or saline were given by continuous intravenous infusion for 24 h via a micropump.After 24 hours of treatment,serum myocardial zymogram,such as aspartate aminotransferase (AST),lactate dehydrogenase (LDH) and creatine kinase (CK),as well as superoxide dismutase (SOD),malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) activities were determined.Myocardial infarction size (MIS) was measured by nitro blue tetrazolium chloride (NBT) staining.Results Compared to sham operation group,MIS,AST,LDH,CK,MDA were increased,while the activities of SOD and GSH-Px were decreased.However,all the effects were significantly reversed by treatment with rhLTα-Q107E (P < 0.05) but not rhLTα (P > 0.05).Conclusions The rhLTα-Q107E plays a role in the protection against myocardial ischemia-reperfusion injury in rats by the mechanism of scavenging oxygen free radicals and increasing the activity of endogenous antioxidant system.
8.Suppression of MDR1 gene expression and reversal of cisplatin resistance in renal carcinoma cells by RNA interference

Objective:To investigate the inhibitory effect of RNA interference (RNAi) on the expression of multidrug resistance (MDR1) gene and analyze the altered sensitivities of human renal carcinoma cell line to cisplatin.Methods:Three small interfering RNA (siRNA) sequences targeted MDR1 gene were synthesized and transfected into renal carcinoma A498 cells. The expression level of MDRl mRNA was measured by RT-PCR to identify the most effective siRNA sequence. The recombinant plasmid was packed by lentivirus and transfected into A498 cells. RT-PCR was used to screen the A498 cells with the optimal silencing efficacy. The MDR1 protein expression level in the cloned cells was verified by Western blotting. The inhibitory effect of cisplatin on the proliferation of A498 cells was assessed by MTT assay and the IC_(50) value was calculated. Results:The 3 siRNA sequences suppressed MDR1 gene expression at different degrees. The siRNA 1 sequence silenced MDR1 gene more effectively with a significant reduction of 67%. The MDR1 protein expression greatly decreased in screened A498 cells compared with non-transfected cells (P<0.01), and the IC_(50) value of cisplatin on screened A498 cells was significantly decreased by 83.37% (P<0.01). Conclusion: The RNAi could effectively inhibit the expression of MDR1 gene and increase the sensibility to cisplatin in human renal carcinoma A498 cell line, which make it possible to reverse the resistance of renal carcinoma to chemotherapy.
9.Triangular flaps and fascia-tissue flaps rotation for correction of severe inverted nipple in shallow and deep areola of breast

Objective To explore a new methodology for surgical treatment of severe retracted nipple in women. Methods 20 patients with inverted nipple and 4 recurred patients were involved in this study. Firstly, the site of neonipple tip was marked in the central part of the inverted nipple and its mean diameter usually was 1.2-1.5 cm. Then two shallow and deep triangular pedicled flaps were designed, respectively, in both superior and inferior areas near areola. With temporary traction of the nipple apex provided by a stay suture, the fibrotie bands underneath the nipple base might be cautious-ly released. Moreover, the shallow skin flaps should be about 0.5 cm in thickness and their blood sup-ply was from the subdermal arterial rete of the areola, which were used to cover and reconstruct the neck area of neonipple after a clockwise rotation and advancement simultaneously. While the deep fas-cia tissue flaps were revolved and advanced either horizontally to the opposite pedicle or upward to the inner tip through the tunnel underneath the nipple base in order to improve the height or width of the neonipple neck and prevent flattening as the supporting tissue and their blood supply was from some small perforating branch arteries in the deep part of mammary gland. Finally, purse-string suture was necessary in the base of neonipple which played a key role in avoiding recurrence of nipple inversion. Four vertical diamond-shaped excision-suturation treatment in neck area could make improvemts on the height of those stout and short nipples. Results In all 24 cases corrected by shallow and deep triangu-lar flaps rotation, after 3-6 months' follow-up, there were no complications related to surgery such as infection, hematoma, permanent sensory disturbance, or nipple necrosis, and postoperative recovery was rapid and uneventful. Especially, follow-up data revealed no evidence of recurrence of inversion and all patients were satisfied with their results. Conclusions Triangular flaps and fascia-tissue flaps in shallow and deep areola rotation is effective and easy to be popularized in correction of inverted nip-ple. This technique can improve both the diameter and height of the nipple, and certainly lower the re-currence rate of nipple inversion and achieve good aesthetic results.
10.Study on the action mechanism of basic fibroblast growth factor on vascular smooth muscle cells in spontaneously hypertensive rats

Objective To explore the action mechanism of basic fibroblast growth factor (bFGF) on vascular smooth muscle cells (VSMCs) in spontaneously hypertensive rats (SHR).Methods Ts were obtained from SHR and SD rats.The aortic VSMCs were cultured in vitro by tissue explant method.VSMCs were treated with different concentration of exogenous bFGF (0 ng/ml,20 ng/ml,40 ng/ml,60 ng/ml,80 ng/ml,100 ng/ml) for 48 h,then cell proliferation was detected by 3 (4,5 dimethylthiazol)2,5 diphenyltetrazolium bromide (MTT) colorimetric assay.VSMCs from SHR in control group were treated with bFGF (100 ng/ml) for 48h.VSMCs from SHR in treatment group were treated with bFGF (100 ng/ml) plus Proteinkinase C(PKC) inhibitor (staurosporine) for 48 h.Results After treatment with different concentration (0 ng/ml,20 ng/ml,40 ng/ml,60 ng/ml,80 ng/ml,100 ng/ml) of bFGF for 48 h,the values measured by MTT colorimetric method were 0.402 ± 0.103,0.605 ±0.090,0.696 ± 0.131,0.812 ± 0.080,0.901 ± 0.065,1.056±0.078 respectively in aortic VSMCs from SD rats,and 0.404±0.065,0.507±0.078,0.608±0.057,0.704 ± 0.107,0.812 ± 0.097,0.908 ± 0.032 respectively in aortic VSMCs from SHR.Compared with control group,the values measured by MTT colorimetric method were decreased in treatment group (P<0.05).The proliferative effect of bFGF in aortic VSMCs from SHR was attenuated after administration of PKC inhibitor staurosporine.Conclusions Exogenous bFGF administration promotes VSMCs proliferation in SHR and SD rats in a concentration-dependent manner.PKC plays an important role in the signal transduction mechanism in VSMCs proliferation by exogenous bFGF.
