Objective:cDNA microarrary technique was employed to detect the effects of evodiamine on functional regulation of gene expressing in murine marrow-derived dendritic cells. Methods:Immature dendritic cells (iDC) derived from bone marrow of BALB/c mice were induced by GM-CSF for ten days in vitro and received corresponding treatment for 24 h.Then the cell were randomized into the control groups (Ⅰ),evoamine group (Ⅱ),LPS group (Ⅲ) and evodiamine+LPS group (Ⅳ).After 24 h’s treatment, total RNA of the cells was collected, and cDNA microarrary (GEArray S Series Mouse Dendritic & Antigen Presenting Cell Gene Array:MM-604) was used to screen the functionalg correlation genes of dendritic cells. Results:There were seven genes up-regulated and two genes down-regulated two-folds above in Ⅱ/Ⅰ;thirty four genes up-regulated and twelve genes dwon-regulated two-folds above in Ⅲ/Ⅰ;forty six genes up-regulated and seven genes down-regulated two-folds above in Ⅳ/Ⅱ;twenty four genes up-regulated and two genes down-regulated two-folds above in Ⅳ/Ⅲ.The function of these genes involve the secret of the cytokines and the expression of their receptors, antigen uptake, antigen presentation, cell surface receptors and signal transduction. Conclusion:The effects of evodiamine on dendritic cell involve in multi-regulation and expression of genes, these genes contribute to the functions, differentiation and maturation of the dendritic cells. The work provides a potentiality to seek the target of medicine.

Objective To assess the diagnostic value of the latent membrane protein 1(LMP-1)gene detection for diagnosing na-sopharyngeal carcinoma(NPC).Methods The related researches on the detection of LMP-1 gene in the nasopharyngeal swab secre-tion for diagnosing NPC by PCR were entirely collected by using the computer retrieval or manual inquiring. Two estimators screened the literature according to the criteria of inclusion and exclusion. The quality evaluation was performed by adopting the QUADAS scale. The heterogeneity test was conducted by using Meta-Disc 1.4 software.According to the heterogeneous character-istics,the corresponding effect model was selected for calculating the pooled sensitivity and specificity and the summary receiver op-erating character(SROC)curve was drawn. Results 139 related articles were retrieved,in which 6 articles were finally included. 394 cases of NPC were definitely diagnosed by the pathological golden standard,802 cases were in the control group. The pooled sensitivity and specificity of the LMP-1 gene for diagnosing NPC were 0.90[95%CI (0.87,0.93)]and 0.98[95%CI (0.96,0.99)] respectively. The area under SROC(AUC)was 0.973 7.Conclusion LMP-1 gene has the higher diagnostic value for NPC and could be used for screening and auxiliary diagnosis of NPC.

Objective: To evaluate the quality of life, peripheral blood and immune function of patients with malignant tumors treated by PSP.Methods: We searched MEDLINE,Pubmed and other databases by computer. We assessed the identified studies in order to include high quality studies. Results:A total of 7 into the study,701 cases of patients.Meta analysis showed that polysacchariopeptide treatment for patients with certain malignant tumors after chemotherapy might improve the quality of life.Polysacchariopeptide group combined with chemotherapy drugs might improve the white blood cells, hemoglobin, platelet compared to chemotherapy drugs or batyl alcohol drugs combined with chemotherapy.Polysacchariopeptide drug combined with chemotherapy may increase the CD_3~+ ,CD_4~+ ,CD_8~+ and CD_4~+/CD_8~+ ratio, and the NK cells was no significant difference. Conclusion:PSP combined with chemotherapy to treat malignant tumors has a certain effect. However, most of trials included in the review may be of low quality, therefore,it may be neccessity to conduct multi-center randomized-controlled trials of high quality.

Objective:To detect the effects of polysaccharopeptide(PSP) on MyD88-dependent signaling pathway in EAC tumor-bearing mice, and explore the immunomodulatory mechanism of PSP.Methods: Ehrlich′s ascites carcinoma(EAC) C57BL/6 mice were used to establish the animal model for solid tumor.Mice were divided into two groups:WT group and MyD88-/-group.After 22 days of treatment, quantitative real-time PCR( Q-PCR) ,Western blot and were used to detect the related gene and protein expression of TLR4 pathway in spleens,ELISA were used to detect the terminal effect factors secretion eyeball blood from each group. Results:Related genes and proteins of TLR4 pathway in spleen were up-regulated significantly from two groups.Compared with WT group, related genes and proteins in MyD88-dependent pathway(MyD88,TRAF6,NF-κB,AP-1)was down-regulated in MyD88-/-group(P<0.05).Meanwhile,There was no significant change of the related genes and proteins of MyD88-independent pathway(TRAM,TRIF)in MyD88-/-group( P>0.05 ) .The terminal effect factors secretion of IL-2, IL-6, IL-10, TNF-αand IFN-γin MyD88-/-group were decreased significantly compared with WT group(P<0.05).Conclusion:The immunoregulatory effect of PSP on EAC tumor-bearing mice may be implement through the regulation of MyD88-dependent signaling pathway.

Objective:To investigate the effects of Acanthopanax Senticosus polysaccharides (ASPS) on TLR4 signaling pathway in Lewis tumor-bearing mice, and to explore the immunomodulatory mechanism of ASPS.Methods:Lewis lung carcinoma cells and C57BL/6 mice were used to establish the animal model for solid tumor. The tumor-bearing mice were divided into five groups:normal saline (NS) group,Adriamycin (ADM) group,ASPS low-dose group,ASPS middle-dose group and ASPS high-dose group.Mter 25 d treatment, the weight of tumor, tumor inhibition rate and immune organ index were measured.ELISA were applied to detected the cytokines in peripheral blood of tumor-bearing mice.Quantitative real-time PCR (Q-PCR) and Western blot were selectively used to detect the gene and protein expression of TLR4 signaling pathway in splenocytes. Results:The tumor inhibition rate, immune organ index and the secretion of TNF-α,IL-1β and IL-6 were increased by ASPS,compared with NS group (P<0.05).The gene and protein expression of TLR4,MyD88,TRAF6,NF-κB p65 and AP-1 were also induced by ASPS (P<0.05).Meanwhile,ASPS had no obvious effects on the secretion of IL-12p70 and the expression of TRAM (P>0.05).Conclusion:TLR4 signaling pathway may be involved in the immunomodulatory effects of ASPS on Lewis tumor-bearing mice.
