Aim To study the molecular basis of protection on myocardial ischemic injuries of total flavones of choerospondias axiaris fructus(TFC). Methods The microarray analysis for the protein expression in myocardium provide a strong tool to explore the key protein candidates involved in the pathogenesis of ischemic injury.Surface enhanced laser desorption/ionization(SELDI) mass spectrometry with protein chip IMAC3,SAX2 and NP20 was used to compare the differentially expressed protein in TFC-treated and untreated ischemic myocardium in rats and the results were analysized with Proteinchip Software 3.0.2.Results Seven differentially expressed proteins were indentified in TFC treated myocardium. These differential effects correlated with the expression of five downregulated proteins and two upregulated proteins, and four of them were discovered on the IMAC3 chip and one of them was discovered on the SAX2 chip. Conclusions The myocardial protection of TFC may be mediated by the differential expression of these proteins which could be the key protein candidates for further investigation.

Objective To investigate the distribution and clonality of TCR V? repertoire T cells in peripheral blood mononuclear cells with chronic myeloid leukemia(CML)at chronic phase(CP)and complete remission(CR).Methods The CDR3 of TCR V? 8 subfamily genes were amplified in mononuclear cells of peripheral blood from CML patients in CP(8 cases)and CR(8 cases)phases for observation of the usage of TCR V? repertoire by RT-PCR.The positive PCR products were further labeled with fluorescence and analyzed by genescan technique for the CDR3 size for evaluation of the clonality of the detectable TCR V? T cells.A total of 10 cases of healthy adults served as controls. Results The mean values of detectable TCR V? subfamilies were(3.25?0.89)in patients with CML-CP,predominantly in V?1,2 and V?3,whereas(3.5?0.76)of 8 V? subfamily T cells were expressed in patients with CML-CR,predominantly in V?1,2,3 and V?8.There was no significant difference between the above groups and the healthy adults(3.5?0.52).In addition,a higher frequency expression of V?8 could be found from peripheral blood in CR phase(5/8)rather than from that in CP phase(2/8)and from that of the healthy adults(3/10).Genescan analysis showed that the majority of PCR products from both CML and healthy adults displayed clonal expansion, predominantly in V?1,V?2 and V?3.Clonal expanded V?7 subfamily T cells, however, could be identified from two patients in CR phase but not in all of 10 healthy adults. Conclusion The similar distribution and clonal pattern of TCR V? repertoire T cells can be found in peripheral blood from the three groups, suggesting that the cellular immunosuppression of CML patients might not be mainly involved in the alteration of TCR V? repertoire. In CML-CR groups, the expression and clonal pattern of some V? repertoires are different from those in other groups, which needs further studies.

Objective To study the effects of fibroblast growth factor 2 (FGF2) on the proliferation and gene expression profiles of rabbit articular chondrocytes in vitro after different time periods of stimulation. Methods The chondrocytes were isolated and cultured in vitro, and the 3rd generation cells were harvested.Cells were divided into three groups. In the group 1 (FGF2 short-time action group),chondrocytes were cultured in medium with FGF2 for one day,and then transferred to fresh culture medium without FGF2 and cultured for another 6 days.In the group 2 (FGF2 long-time action group),chondrocytes were cultured in medium with FGF2 for 7 days.In the Group 3 (control group),chondrocytes were cultured in culture medium without FGF2 for 7 days. After culture for 1,3,and 7 days, the proliferation of chondrocytes in the all groups was detected respectively.Following extraction of mRNA,the gene expressions of BMP2,BMP4,SOX9 and COL2A1 of the chondrocytes in the all groups were determined by quantitative real-time polymerase chain reaction (qRT-PCR).The content of type Ⅱ collagen was measured via immunofluorescence staining. Results Compared with the control group,FGF2 promoted the proliferation of chondrocytes in the short-and long-time action groups and there was no significant difference between the two FGF2-treated groups. The results of qRT-PCR indicated that different treatment induced different gene expression profile.Particularly, compared with the control group and the FGF2 long-time action group, the expression of BMP2,BMP4,SOX9 and COL2A1 in the short-time action group were significantly upregulated at the 7th day.Immunofluorescence intensity of type Ⅱ collagen in the group 1 was stronger than that in the control group and group 2.Conclusions Different administration of FGF2 for different time periods induced different responses of chondrocytes.Short-term FGF2 stimulation was more beneficial to maintain the phenotype of chondrocytes and the synthesis of extracellular matrix.

Objective To observe the expression of PAX-2 and PTEN in different types of endometrial lesions, and to study their relationship with endometrial intraepithelial neoplasia (EIN). Methods 60 cases of endometrial hyperplasic lesions and 70 cases of endometrial carcinoma were enrolled. All cases were reclassified by using the diagnostic criteria of EIN, and PAX-2 and PTEN were stained to compare the difference among them. Results The deletion rates of PAX-2 in benign hyperplasia, EIN and endometrial carcinoma were 39.5 % (15/38), 72.7 % (16/22) and 78.6 % (55/70), respectively, and there was a statistical difference (χ2= 21.664, P= 0.000). The deletion rates of PTEN in benign hyperplasia, EIN and endometrial carcinoma were 47.4%(18/38), 54.5%(12/22) and 75.7%(53/70), respectively, and there was no statistical difference (χ2=2.878, P=0.411). Conclusion The staining of PAX-2 could be considered as a reliable adjuvant diagnostic method in the diagnostic criteria of EIN, however, the loss of PTEN just should be regarded as a suggestion of EIN, not a confirmed diagnostic basis.

Objective To investigate the relevance between expectations before treatment, new symptoms and satisfaction after treatment of the pelvic organ prolapse (POP) patients. Methods Made a collection of 75 cases of POP patients at Peking University Third Hospital, who were affected by the POP symptoms and came to our clinic for treatment from January to December in 2013. Prospectively investigate the patients′expectations before treatment, which were the most troubling symptoms to be solved. According to treatment we divided the patients into surgery and pessary groups. Two groups were followed up with the degree to achieve the desired goals using patient global impression of improvement (PGI-I), new symptoms and satisfaction after treatment, try to find the relevance between expectations before treatment, new symptoms and satisfaction after treatment. Results There were 47 (63%, 47/75) patients in the surgical group and 28 (37%, 28/75) patients in the pessary group. The top three problems for patients were friction when walking (25%, 19/75), dysuria (23%, 17/75) and the feeling of vaginal prolapse (19%, 14/75). The follow-up rate was of 93% (70/75), follow-up time was (5 ± 4) months. Satisfaction score after treatment of surgical group was higher than that of pessary group [(4.9±0.4) versus (4.0±1.3) scores, P<0.01]. There was no statistically significant difference between two groups of PGI-I score [(6.7±0.6) versus (6.6±0.9) scores, P=0.886]. The top three new symptoms after treatment were increased secretion, urinary incontinence and dysuria. PGI-I and satisfaction scores was relevant (P=0.021). The availability of new symptoms and satisfaction scores was relevant (P=0.001). Conclusion When achieving higher expectations to the treatment and no more new symptoms, the satisfaction score after treatment is higher.

Objective:To explore the effect of repetitive high-frequency magnetic stimulation (H-F rTMS) of the dorsolateral part of the prefrontal cortex (DLPFC) combined with smoking-related cues on nicotine addicts′ cigarette craving, the concentration of exhaled CO and sleep quality. Methods:Sixty nicotine addicts were randomly divided into groups A, B and C, each of 20. All were given H-F rTMS five times a week for two weeks, while those in groups A and B watched smoking and non-smoking pictures for ten minutes, respectively. Before and after the intervention, all of the subjects self-reported their cigarette cravings using a visual analogue scale. Exhaled CO (CO ppm) was measured and the Pittsburgh sleep quality index (PSQI) was evaluated. Results:After the intervention the average craving score, CO ppm and PSQI score had improved significantly in all three groups. The average craving score and CO ppm of group A were both significantly better than in the other two groups. Conclusions:rTMS can significantly improve cigarette craving, CO ppm and sleep quality of cigarette adicts. Viewing smoking-related pictures as an addition to rTMS can even better the effects of rTMS.
