ObjectiveExplore the clinical effect of U- 100 double- frequency laser lithotripsy in postoperative residual intrahepatic calculus under choledochoscopy.MethodsSeverty-four patients of postoperative intrahepatic residual stones with U-100 double-frequency laser lithotripsy under choledochoscopy were analyzed retrospectively.ResultsSixty-three (85.1％) of 74 cases were treated successfully in the first operation,8cases ( 10.8％ ) obtained satisfactory outcome in the second operation,and 3 cases (4.1％) in the third.All lithotripsies were performed successfully.ConclusionsU-100 double-frequency laser lithotripsy in postoperative residual intrahepatic calculus under choledochoscopy provides a safe and effective method.

To prepare HIV-1 Vif and hAPOBEC3G and to produce their antibodies, the full length gene fragment of HIV-1 Vif was amplified by PCR from a plasmid of HIV-1 NL4.3 cDNA, and the APOBEC3G gene was obtained by RT-PCR from the total RNA of H9 cells. The resulting DNA construct was cloned into a prokaryotic expression vector (pET-32a). Recombinant pET-vif and pET-APOBEC3G were expressed respectively in Eserichia coli BL21 (DE3) as an insoluble protein. The vector also contained a six-histidine tag at the C-terminus for convenient purification and detection. To express and purify the HIV-1 Vif and hAPOBEC3G in E. coli cells, the accuracy of inserted gene and specificity of proteins were detected by the two enzyme digestion method, SDS-PAGE, and Western blotting. Rabbits were then immunized by Vif or APOBEC3G protein and serum samples were tested by indirect ELISA to determine the level of antibodies. Immunoenzyme and immunofluorescence assays were performed to identify the specificity of polyclonal antibodies. The titer of the anti-Vif antibodies was 1:204800, and that of the anti-APOBEC3G antibodies was 1:102400. Thus the antibodies could detect the antigen expression in the cells, demonstrating that fusion proteins with high purity and their corresponding polyclonal antibodies with high titer and specificity were achieved.

Objective To explore the relationship between emotional intelligence and job performance by investigating the level of them in pediatric nurses,and to provide a basis for improving job performance and the implementation of“quality care.Methods 335 pediatric nurses were investigated by using emotional intelligence scale and job performance scale.And the result of the investigation was analyzed statistically.Results The average score of the emotional intelligence of the pediatric nurses were (3.63±0.52),the highest score was interpersonal latitude (3.87± 0.69),the lowest score was management of emotions latitude (3.25±0.90); titles,the form of post and degree of nurses had a certain influence on the emotional intelligence.The score of job performance was(3.65±0.75).By Pearson correlation analysis,the emotional intelligence score and job performance score were obtained a significant positive correlation,self-motivation latitude and job performance were the most related,the management of emotional latitude job performance was the lowest.Conclusions Nursing managers should take corresponding measures to improve nurses' emotional intelligence and job performance levels,and promote the implementation of quality care effectively.

Objective To observe the anti-influenza effect of a Chinese medicinal herb-Antiviral Agent No.1. Methods To study t he anti-viral effect of Antiviral Agent No.1 by means of the technique of cel l culture and using Ribavirin as a positive control. Results In MDC K cu lture, Antiviral Agent No.1 was found to be a potential inhibitor of influenza A 3 virus in a concentration-dependent manner, with a TC50 of 60.53 mg*m l-1. When drug was added 2 hours post virus infection, the EC50(TI) was 5.14 mg*ml-1(11.78); while drug was added 2 hours before infection, t he EC50(TI) was 5.20 mg*ml-1(11.65). Conclusions Antiv iral Agent No.1 had a significant anti-influenza effect on type A3 in MDCK.

Objective To retrospectively investigate the causes , location and the duration of hospital stays of the hospitalized patients with hard-to-heal wounds so as to provide data guidance for the prevention and research of these diseases. Methods Clinical data of hospitalized patients with hard-to-heal wounds in plastic surgery center of General Hospital of Guangzhou Military Command were collected from June , 2011 to December, 2013. Whether ulceration was recovered in the patients with skin tissue defect after 2 months treatment was regarded as the standard to screen the patients with hard-to-heal wounds. The causes , location, age and the duration of hospital stays of the hospitalized patients with hard-to-heal wounds were investigated by retrospective case-control method. Chi-square test and t-test were used in analyzing the investigation. Results 2 136 cases, aged from 20 to 86, were treated in plastic surgery center of the hospital. 120 cases have hard-to-heal wounds, which constituted 5.62% of all hospitalized patients. (1) Metabolic disease was the main causes of wound (43.3%), followed by wound infection and tumor (20.0% for each) (χ2 = 62.917, P < 0.01). ( 2 ) The peak age for patients with hard-to-heal wounds was 40 to 60 years old patients , followed by patients′age from 60 to 80 years. (3) The mostly like hard-to-heal wounds was on limbs (61.6%), especially on the foot (38.3%) (χ2 = 17.546, P = 0.002). (4) The average days for hospitalization of in the plastic surgery center were (7.41 ± 8.98), while the average days for hospitalization of the patients with years were (33.21 ± 28.27)(t = -9.968, P < 0.05). Conclusion The average diagnostic age of patients with hard-to-heal wounds is the middle and old aged patients. Chronic skin ulcers, which often occurs in a limb, seriously affects a person′s ability to move , which can prolong hospital stays , causing serious burden for the families of patients and the society.

Objective To summarize the clinical experiences of mini-incision urological surgery.Methods The clinical data of 43 patients who received mini-incision surgery were reviewed retrospectively.The subjects included 5 adrenal tumor excisions,9 nephrectomy,13 unroofing of solitary renal cyst,4 pyeloplasty.12 pyelolithotomy and ureterolithotomy.Results All surgical procedures were successful in the 43 cases.The length of the incision ranged from 3 to 8 cm.The average operation time was 80 minutes and average blood loss was 100 ml. No patients needed blood transfusion during the operation.No serious complications such as the surrounding organ damage happened.The postoperative hospitalization was 5-7 d. Conclusion Mini-incision approach for urological operation has the advantages of minimal invasion,safety,rapid recovery and no requirement for special equipments.Its easy to be popularized in the primary hospital.

Objective:To explore the role of long non-coding RNA rhabdomyosarcoma 2 associated transcript(lncRNA RMST)and microRNA-24-3p(miR-24-3p)in proliferation,migration and invasion of oral squamous cell carcinoma cells(OSCCs).Methods:The RMST expression in head and neck squamous cell carcinoma(HNSC)was analyzed by UALCAN data base.qRT-PCR was used to detect the expression of RMST and miR-24-3p in 5 human OSCC cell lines.CAL27 cells were in vitro cultured and allocated into con-trol group,pcDNA3.1 group(vector),pcDNA3.1-RMST+mimics-NC group(overexpression of RMST+negative control of miR-24-3p)and pcDNA3.1-RMST+miR-24-3p mimics group(overexpression of RMST+miR-24-3p).Dual luciferase reporter assay was per-formed to analyze the interaction between RMST and miR-24-3p.MTT assay,wound-healing and transwell assay were applied to de-tect the proliferation,migration and invasion of the cells.Western blot and qRT-PCR were conducted to measure the expression of E-cadherin,N-cadherin and c-myc in the cells.Results:RMST level were down-regulated in HNSC tissues and OSCC cell lines(P＜0.01).While miR-24-3p level was elevated in OSCC cell lines(P＜0.01).miR-24-3p was identified and confirmed as a potential binding partner to RMST.miR-24-3p level was lower in pcDNA3.1-RMST group than in pcDNA3.1 group(P＜0.01).The prolifera-tion,migration and invasion abilities of CAL27 cell in pcDNA3.1-RMST+mimics-NC group were lower than in pcDNA3.1 group(P＜0.01).While those of CAL27 cells in pcDNA3.1-RMST+miR-24-3p mimics group were higher than in pcDNA3.1-RMST+mimics-NC group(P＜0.01).Compared with pcDNA3.1 group,pcDNA3.1-RMST+miR-24-3p mimics showed higher expression of E-cad-herin,lower expression of N-cadherin and c-myc(P＜0.01).Compared with pcDNA3.1-RMST+miR-24-3p mimics group,pcD-NA3.1-RMST+miR-24-3p mimics group showed lower expression of E-cadherin,higher expression of N-cadherin and c-myc(P＜0.01).Conclusion:Down-expressed RMST may suppress the proliferation,migration and invasion progression of OSCC cells through targeting miR-24-3p.

Apolipoprotein B mRNA-editing enzyme catalytic polypeptide-like 3G (APOBEC3G) cDNA was amplified from total RNA prepared from nonpermissive H9 cells by RT-PCR. APOBEC3G cDNA is 1155nt long, encoding 384 amino acids. The APOBEC3G gene was then cloned into the eukaryotic expression vector pEGFP-C3. The generated pEGFP-3G construct was then transfected into CD4+ HeLa cell to determine the expression and the subcellular localization of GFP-APOBEC3G fusion protein. Under CLSM the localization of the expressed GFP-APOBEC3G in the cytoplasm of CD4+ HeLa cells was observed.
