1.Quality Standard for Ehong Capsules

Objective: To establish the quality standard for Ehong capsules.Methods: A TLC was adopted to identify safflower, pueraria and prepared Radix Polygoni Multiflori.The contents of hydroxyl-safflor yellow A and puerarin in Ehong capsules were determined by a dual wavelength HPLC.A Wondasil C 18-WR (250 mm× 4.6 mm ,5 μm) was used as the analytical column, the mobile phase was composed of methanol acetonitrile-0.7% phosphoric acid aqueous solution (16∶3∶81), the flow rate was 1.0 ml·min-1 ,the detection wavelength was 403 nm for hydroxy safflower yellow A and 250 nm for puerarin.The column temperature was 30℃.Results: The TLC spots were clear and well-separated without negative interference.The calibration curves of hydroxyl-safflor yellow A and puerarin were linear over the ranges, the average recovery was 101.02% and 100.03% , and the RSD was 1.43% and 2.40% (n =6), respectively.Conclusion: The method is fast and accurate with good specificity, which can be used for the quality control of Ehong capsules.
2.Research Progress in Quality Evaluation of Gypsum

Gypsum as a common mineral medicine is widely used in clinics, which is mainly used for the treatment of all kinds of fever. Through reviewing the historical document records of gypsum, the effects of the mineral resources and production process on the quality, and the quality studies were investigated in order to guide the production process,the preparation process and decoction pieces classification of gypsum, and further expand the effective application of gypsum in clinics.
3.Effects of the bone grafting pedicled with femoral quadratus for the osteonecrosis of femoral head

Objective To analyze the clinical effect of the bone grafting pedicled with femoral quadratus for the osteonecrosis of femoral head.Methods The clinical operation data were analyzed from 355 patients with osteonecrosis of femoral head underwent by the surgery of the bone grafting pedicled with femoral quadrates in the First Affiliated Hospital of Zhengzhou University from August,2006 to June,2013.According to association research circulation osseous (ARCO) classifying,41,126,115,62,6 and 5 patients were in stage Ⅰ-C,Ⅱ-A,Ⅱ-B,Ⅱ-C,Ⅲ-A,and Ⅲ-B.The results were evaluated by the hundred forked method.Results All the patients obtained the outpatient followed-up after operation,the follow-up time was 1-8 (4.3 ± 1.8) years.The pain of the hip,the function of the joint,the motion of hip joint,and the X ray evaluation became better than preoperations.There were statistically significant differences in the healing time among all the age patients (P < 0.05),along with the age growth,the healing time gradually extended.Compared with that before operation,the scores of all the stage patients after operation were higher (P < 0.05).The rate of excellent and good results were 98.5%,98.0%,92.3%,89.7%,83.5%,81.3% in stage Ⅰ-C,Ⅱ-A,Ⅱ-B,Ⅱ-C,Ⅲ-A,and Ⅲ-B,the excellent and good rate was 93.8% of all the 355 patients.Conclusion Bone grafting pedicled with femoral quadratus is effective surgical method for the osteonecrosis of femoral head.
4.Study on Grading of Gypsum Pieces

Objective:To comprehensively evaluate the intrinsic and external quality, and explore the grading of gypsum pieces. Methods:The characteristics, contents of main components and trace elements and X-diffraction exclusive characteristic peaks of gyp-sum pieces from different origins were detected. Combined with the market grading, the grading standard for gypsum pieces was further improved. Results:According to the tested indices, gypsum pieces were graded into two classes:excellent pieces and common decoc-tion pieces. Conclusion:The study shows reference value for guiding the market grading of gypsum pieces.
5.Study on the Quality Standard for Tiaojing Zhuyun Granules

Objective:To establish the quality standard for Tiaojing Zhuyun granules .Methods:A TLC method was used for the qualitative identification of main ingredients of Cornus officinalis and Angelica sinensis, and an HPLC method was used for the determi-nation of loganin in Tiaojing Zhuyun granule with SinoChrom ODS-BP (250 mm ×4.6 mm,5μm) as the analytical column .The mobile phase was composed of acetonitrile-0.1%phosphric acid solution (14∶86) at a flow rate of 1.0 ml· min-1 .The detection wavelength was set at 240 nm and the column temperature was 30℃.Results:The corresponding spots in the granule were clear without any inter -ference from the negative control .The calibration curve of loganin was in good linearity over the range of 50 .40-1008 .00 ng ( r =0.999 9).The average recovery was 98.02%(RSD=2.51%,n=6).Conclusion:The method is simple,feasible and reproducible, which can be used for the quality control of Tiaojing Zhuyun granules .
6.Quality Standard for Qingchang Mixture

Objective:To establish the quality standard for Qingchang mixture.Methods:TLC was adopted to identify angelica sinensis,paeoniae radix and phellodendri cortex.HPLC was used to determine the content of ferulic acid on a Wondasil C18 (250 mm×4.6 mm,5 μm) analytical column,the mobile phase was composed of acetonitrile-0.1% phosphoric acid solution(17∶83),the flow rate was 1.0 ml·min-1, and the detection wavelength was 316 nm and the column temperature was 30℃.Results:The TLC spots were clear and well-separated without any negative interference.The calibration curve of ferulic acid was in good linearity over the range of 39.6-316.8 μg·ml-1(r=0.999 9),the average recovery was 98.75% and RSD was 1.21%(n=9).Conclusion:The method is fast and accurate with good specificity,which can be used for the quality control of Qingchang mixture.
7.Effect of brain tissue extracts from traumatic brain injury rats treated with mild hypothermia on umbilical cord mesenchymal stem cells in vitro

Objective To simulate the chemical microenvironment of traumatic brain injury (TBI) under mild hypothermia, and investigate the effect of such microenvironment on umbilical cord mesenchymal stem cells (UCMSCs) in vitro.Methods Eighteen SD rats were allocated to shamoperated group, TBI group and mild hypothermia group according to the random number table, with 6 rats per group.Rat models of TBI were made by electric cortical contusion impactor.After systemic mild hypothermia (33℃) for 4 h, brain tissue homogenate extracts were harvested.Polyacrylamide gels mimicking the elastic modulus of brain were manufactured.Human UCMSCs were isolated and cultured on the gels, added with brain tissue extracts from each group.After 24 h, the apoptosis level of UCMSCs was checked, and the medium was changed with normal one.Cell growth and morphological changes in each group were given dynamic observation.Seven days later, cell immunofluorescence was implemented, with the differentiation level of each group estimated.Results Apoptotic rate in TBI group was 73.47%,significantly higher than 10.42% in sham-operated group (P <0.01).While the apoptotic rate was 28.57% in mild hypothermia group, indicating mild hypothermia significantly reversed the apoptosis of cells in TBI group (P < 0.01).Cell immunofluorescence demonstrated rate of neuronal differentiation of UCMSCs in sham-operated group, TBI group and mild hypothermia group was 16.48%, 2.59% and 11.83% respectively.Mild hypothermia resulted in significantly improved neuronal differentiation of UCMSCs after TBI (P < 0.05).Conclusions More apoptosis and lower neuronal differentiation ability are observed in UCMSCs in the chemical microenvironment after TBI.However, mild hypothermia significantly reverses the elevation of apoptosis and restores the neuronal differentiation capacity of UCMSCs after TBI.

Objective:To establish the quality standard for Huayu pills. Methods:TLC was adopted to identify salvia miltiorrhi-za, rhubarb charcoal and panax notoginseng; the contents of tanshinoneⅡA , chrysophanol and physcion in Huayu pills were deter-mined by RP-HPLC with Wondasil C18 (250 mm × 4. 6 mm,5μm) as the analytical column, the mobile phase was composed of metha-nol-0. 1% phosphoric acid solution (85 ∶15), the flow rate was 1. 0 ml·min-1,the detection wavelength was 254 nm, the column temperature was 35℃, and injection volume was 10 μl. Results:The TLC spots were clear and well-separated without any negative in-terference; the calibration curve of tanshinoneⅡA , chrysophanol and physcion was linear over the range of 0. 212-2. 12 μg ( r =0. 999 9),0. 159-1. 59 μg(r=0. 999 9) and 0. 072 6-0. 726 μg(r=0. 999 9), the average recovery was 99. 55 %, 99. 56% and 100. 60%, and RSD was 1. 57%,2. 26% and 2. 28%(n=6), respectively. Conclusion: The method is fast and accurate with good specificity, and can be used for the quality control of Huayu pills.
9.Rapid Determination of CaSO4 ·2H2 O in Gypsum by Raman Spectra

Objective:To develop a method for the rapid determination of CaSO4 ·2H2 O in gypsum by Raman spectra. Methods:Totally 40 batches of gypsum from different origins were used as the training set, compared with the results by ENTA titration, and OPUS software was used to establish a quantitative analysis model of CaSO4 ·2H2 O in gypsum by Raman spectra. Results:The estab-lished quantitative analysis model could provide a good prediction result rapidly when the content was between 97. 93% and 99. 81%. Conclusion:The method is accurate, fast and simple, which can be developed as an analysis method for CaSO4 ·2H2 O in gypsum.
10.Effect of Different Processing Methods on the Chemical Constituents in Wheat Bran

Objective: To explore the effect of different processing methods on the content changes of ferulic acid and total flavonoids in wheat bran and its processed products by HPLC and UV spectrophotometry.Methods: Ferulic acid was determined by HPLC with SHIMADZU C18 (250×4.6 mm,5 μm) as the analytical column, the mobile phase was composed of acetonitrile-0.1% phosphoric acid solution (16∶84), the flow rate was 1.0 ml·min-1, the detection wavelength was 320 nm and the column temperature was 35℃.Total flavonoids was determined by UV spectrophotometry at the wavelength of 258 nm(±2 nm).Results: The content of ferulic acid in the crude bran, the toasted to brown bran, the toasted to dark brown bran, the over toasted bran and the honey toasted bran was 2.07, 2.33, 1.68, 1.11 and 0.83 μg·g-1, and total flavonoids was 5.96, 7.15, 9.73, 13.61 and 9.42 mg·g-1, respectively.Conclusion: The higher the toasted processing degree is, the lower content of ferulic acid and the higher content of total flavonoids are in wheat bran.
