1.Clinical effect of different scheme of moxifloxacin and levofloxacin for elderly multi drug resistant pulmonary tuberculosis

Objective To explore the clinical effect of different scheme of moxifloxacin and levofloxacin for elderly multi drug resistant pulmonary tuberculosis.Methods 136 cases of elderly patients included in the study were randomly divided into the observation group 1 and observation group 2 with 68 cases in each group according to the sequence in group.The observation group 1 used moxifloxacin regimen for treatment of pulmonary tuberculosis while the observation group 2 used levofloxacin regimen.The two groups were treated for 18 months and observed the clinical curative effect,and the sputum negative,pulmonary lesions absorption,empty changes and adverse reactions.Results The total efficiency of the observation group 1 was 92.6%,which in the observation group 2 was 72.1%,the total efficiency of the two groups had a significant difference (x2 =9.917,P =0.002) ;the sputum negative conversion rate of the observation group 1 at the end of 3 months was significantly higher than that of the observation group 2 (x2 =4.115,P =0.043),no significant difference was found at the other time points (all P > 0.05) ; after treatment,the obvious absorption + absorption of lung lesion in the observation group 1 was 59 cases,accounting for 86.8%,that in observation group 2 was 48 cases,accounting for 70.6%,two groups of lung lesions absorption had significant difference (x2 =5.303,P =0.021) ; the pulmonary cavity closure + reduced in the observation group 1 was 52 cases,accounting for 76.4%,that in the observation group 2 was 41 cases,accounting for 60.3%,with significant difference between the two groups (x2 =4.115,P =0.043) ;there was no significant difference in adverse reaction of the two groups (P > 0.05).Conclusion Moxifloxacin anti tuberculosis regimen has a better curative effect in treatment of elderly patients with multi drug resistant pulmonary tuberculosis,which should be expanded the application.
2.Determination of Thiamazole Content in Yingqi Ling Tablets by HPLC

[Objective] To develop a method for the determination of thiamazole content in Yingqi Ling Tablets. [Methods] High performance liquid chromatography (HPLC) was used. The chromatographic conditions were: C18 Gravity Column (4.6mm ? 250mm), methanol - water (10:90 ) as mobile phase, flow rate being 1.0mL/min and the detection wavelength at 258nm. [Results] The calibration curve was linear in the range of 0.16 - 0.64?g. The average recovery was 101.01% (relative standard deviation sr = 2.21%). Relative standard deviation of precision test was 1.04% and the content of the sample was 0.4841 mg per tablet (sR = 0.78%). [ Conclusion] HPLC is effective for the determination of thiamazole content in Yingqi Ling Tablets.

4.Roles of TNF-?,Endotoxin in Upper Gastrointestinal Bleeding in Cirrhosis Patients and the Preventive and Treatment Mechanism of Rhubarb

Objective To explore the role of tumor necrosis factor (TNF-?) and endotoxin(LPS) in upper gastrointestinal bleeding in cirrhosis patients and the preventive and treatment mechanism of rhubarb. Methods 45 liver cirrhosis patients with upper gastrointestinal bleeding were treated with rhubarb, and its efficacy was compared with cemetidine's efficacy. Liver function, and serum levels of TNF-? and LPS were measured before and after the therapy. Results Patients’symptoms and liver function were improved, and serum levels of TNF-? and LPS significantly decreased after treated with rhubarb. Conclusions TNF-? and endotoxin might play an important role in upper gastrointestinal bleeding in cirrhosis patients. Rhubarb has protective and treatment effect on upper gastrointestinal bleeding in cirrhosis patients by reducing TNF-? and LPS release.
5.Diffuse hyperpigmentation with guttate depigmentation macules: clinical analysis of 10 cases

Objective To assess the clinical feature and diagnosis of diffuse hyperpigmentation with guttate depigmentation macules.Methods A retrospective study was carried out among 10 patients with diffuse hyperpigmentation with guttate depigmentation macules collected at the Institute of Dermatology,Chinese Academy of Medical Sciences and Peking Union Medical College from 2005 to 2012.The clinical manifestations,pathological findings and disease outcomes in these patients were analyzed.Results Of the 10 patients,6 were male,and 4 were female.The median age at onset was 7 years (range,4-25),and there were only 3 adult patients among these patients.None of the patients had a family history of pigmentary disturbance.Typical clinical manifestations included densely distributed,guttate hypopigmented macules arising on diffuse and uniform hyperpigmentation.Lesions could slowly spread over the body surface without the trend towards spontaneous regression.Pathologically,there was a slight increase in pigmentation of the epidermal basal layer,as well as melanins and melanophages scattered around blood vessels in the superficial dermis,with or without focal vacuolar degeneration of the basal cell layer.Conclusions Diffuse hyperpigmentation with guttate depigmentation macules,a rare pigmentary disturbance that clinically manifests as both hyperpigmentation and hypopigmentation and is pathologically characterized by postinflammatory hyperpigmentation,often affects children.Once the lesions occur,there is no trend towards regression.No effective treatment is available for this entity at present.
6.Effect of downregulation the expression of HDAC1 on cells differentiation of HL-60 cells.

This study is to investigate the effect of downregulation histone deacetylases 1 (HDAC1) gene by the technology of RNA interference on the differentiation of HL-60 cells line. The optimal segment targeting HDAC1 gene was designed and transfected into HL-60 cells by Lipofectamine 2000. The HDAC1 mRNA and protein level were detected by RT-PCR and Western blotting. The morphologic change of HL-60 cells was detected by an optical microscope with Wright-Giemsa. Cell differentiation was tested by NBT reduction assay. Expression of CD13, CD33 and CD14 was measured by flow cytometry. The results indicated that HDAC1 mRNA and protein were markedly suppressed by the siRNA targeting HDAC1 in a concentration-dependent manner. HDAC1 siRNA promoted cell differentiation. HL-60 cells became more mature in morphology after transfected to HDAC1 siRNA at a concentration of 30-60 nmol x L(-1) for 24 h. NBT reduction ability of HDAC1 siRNA with 30 nmol x L(-1) was 0.31 +/- 0.09, compared with negative control (0.20 +/- 0.02) (t = -3.1, P < 0.01), and with 60 nmol x L(-1) was 0.25 +/- 0.02 in comparison with negative control (0.21 +/- 0.04) (t = -2.12, P < 0.05). But it has no change in HDAC1 siRNA > or = 120 nmol x L(-1). After transfection with 60 nmol x L(-1) HDAC1 siRNA to HL-60 cells, the expression of CD13 was (96.50 +/- 0.70)% in compared to siRNA-NC (3.39 +/- 0.68) % (t = 164.9, P < 0.000 5), CD33 was (66.73 +/- 0.50) % in compared to siRNA-NC (96.80 +/- 1.70) % (t = 43.4, P < 0.000 5). CD14 was (0.53 +/- 0.00) % by comparison with siRNA-NC (0.49 +/- 0.02) % (t = -0.97, P > 0.1). HDAC1 siRNA promoted cell differentiation in indicated concentration. HDAC1 might be one of the targets of gene therapy for leukemia.

8.Protective effects of plant of rehmannia root extract on mitochondrial respiratory function of heart, brain and kidney in hypoxia rat

Object To observe the protective effects of rehmannia root extract (Reh) on mitochondrial respiratory function in hypoxia rat. Methods Rat was killed rapidly and the body was put at room temperatare for global ischemia of 15 min. Treatment of po Reh 2 and 4 g/kg was started 2 h before hypoxia. The mitochondrial respiratory parameter of myocardium, brain and kidney was measured by oxygen electrode method. The protective effect of Reh on mitochondrion was observed in 2h po. Bepridil (Bep), an Ca 2+ channel blocker, ip 5 mg/kg 30 min prior to hypoxia served as positive control. Results In myocardium and kidney, hypoxia injury was showed similar, ST 3 and RCI value was increased as compared with the control group in a dose dependent manner, showing a selective stronger protection on renal mitochondrial than that on heart and brain. Conclusion Reh is effective in protecting metochondrial respiratory function in heart, brain and kidney from hypoxia and with the most potent effect on kidney.

Objective To evaluate the effects of kurorinone combined with thymosin on the treatment of chronic hepatitis B.
