Objective To investigate the weight loss mechanism of sleeve gastrectomy.Methods SD rats were fed with high-fat diet to induce obesity,and then the obesity rats were randomly divided into experimental group(underwent SG) and control group(underwent sham surgery).Eight weeks after operation,the body weight,food intake and peripheral blood concentration of Ghrelin,GLP-1,PYY3-36 of rats were checked.Results Compared with controll group,in experimental SG model,food intake was decreased(P 0.05).Conclusions The weight loss of sleeve gastrectomy in DIO rats's decline is lasting and obvious.The postoperative effect of decreased Ghrelin and increased GLP-1,PYY3-36 is the main mechanism for weight loss.

Group-specific component(Gc), one of the polymorphic proteins of human plasma,has been isolated and purified from human plasma of Gc 2-2 phenotype by a procedure including DEAE-Sephadex-A50,Sephadex G100 and DEAE-Sephadex-A50 chromatography. The purified Gc identified by PAGE,SDS-PAGE and immunoelectrophoresis was homogeneous and reacted specifically with the commercial anti-Gc serum (DAKO) kit. The molecular weight of the purified Gc was about 58 kd.

Anti-GC serum was successfully prepared in two New Zealand rabbits immunized with GC protein which was isolated and purified from GC2-2 serum previously in our laboratory. The results of identification showed that the specificity of the home made anti- GC serum and the commercial anti- GC serum (DAKOPATTS) were identical. The titer of the home made anti-GC serum was 128. Three common phenotypes, GC1-1, GC2-1 and GC2-2could be identified by immunoelectrophoresis with the home made anti-GC serum. The concentration of GC protein as low as 3.1 ?g/ml could be detected by double immunodiffusion. In addition the anti-GC serum does not cross react with other human serum proteins.

Objective:To establish a mouse model with the main characteristics of asthma and to study the pathogenesis of its airway inflammation.Methods:Mice were given latex by the intraperitoneal sensitization/intranasal challenge.BALF histology were examined 24 hours following tracheal challenge;cytokines and serum total and specific IgE were examined in one month and the expression of IL-5?IFN-? were performed by ELISA ?RT-PCR.Results:After the latex challenge,in experimental group,the number of total BALF cells and the percentage of eosinophils elevated;the histological analysis showed the elevation of mucus production and goblet cell development within bronchial epithelium,spasm and constraction of bronchia ,marked peribronchial infiltration of eosinophils and lymphocytes; the elevation of total and specific IgE;reduction of IFN-? and increase of IL-5 in the BALF and lung.Conclusion:Latex sensitization and challenge can lead to airway eosinophilia,allergen-specific IgE induction and production of Th2 type cytokines, which were the main characteristics of asthma.

A PAGE IEF immunoassay was established to detect the ORM, Pi, AHSG and GC phenotypes slmultaneously in human bloodstains. The cumulative discriminating power and probability of paternity exclusion were 0. 9878 and 0. 6648 respectively. Human sera diluted 100 times and bloodstains kept at room temperature for 4 weeks could be typed for these four blood groups correctly. The phenotypes of ORM, AHSG and GC could be determined correctly in bloodstains kept at room temperature within 24 weeks. This provides a good approach for individual identification of human bloodstains.

Objective To investigate the association between the peroxisome proliferator-activated receptor-or (PPARα) polymorphism rs1800206 (c.484C ＞ G,L162V) and the risk of metabolic syndrome (MES)Methods There were 1184 subjects aged 40-60 years recruited from our hospital between March 2010 and March 2011.The PPARα polymorphism 484C ＞ G was genotyped using TAQMAN assay by real-time PCR. The relationship between PPARα polymorphism and MES risk were investigated.Results The genotype frequencies were 91.4％,8.4％ and 0.2％ for the PPARα CC,CG and GG,respectively.This SNP was in Hardy-Weinberg equilibrium(P =0.845).Compared with the most common CC genotype,the variant genotypes(CG + GG) had higher fasting glucose((5.82 ± 1.59) mmol/L vs (5.49 ± 1.17) mmol/L,t =2.630,P =0.009) and LDL-C levels((3.53 ± 1.03) mmol/L vs (3.36 ± 0.65) mmol/L,t =2.376,P =0.018) and lower HDL-C levels ((1.56 ± 0.35) mmol/L vs (1.65 ± 0.43) mmol/L,t =2.430,P =0.015).Furthermore,the 484G variant genotypes(CG + GG)was associated with the risk of MES after adjusting for age,gender,education,BMI and lifestyle (smoking and alcohol status) (adjusted OR =1.89 ;95％ CI =1.09-3.23,P =0.012).Condusion PPARα polymorphism rs1800206 is a significant independent predictor of the MES in Southern Chinese.

Objective To investigate the role of platelet rich plasma (PRP) on free fat graft's survival in a rat model.Methods Twenty-four rats were used in the experiment.For each rat,three groups of free fat grafts were injected into the subcutaneous layer of dorsum.Group A was the control group with 0.5 ml fat and 0.12 ml normal saline.Group B received 0.5 ml fat and 0.1 ml PRP with 0.02 ml normal saline.Group C was given combination of 0.5 ml fat and 0.1 ml PRP which was activated by 0.02 ml calcium chloride.Eight rats were euthanised using pentobarbital sodium each time,1,2 and 3 months after fat transplantation.Specimens were acquired and stained with haematoxylineeosin,and then the histological changes examined using light microscopy.All the data were statistically analysed using paired t test to determine if there were differences between the study groups.Results Compared the volume of the fat grafts at 1 month,there was no deference between the groups(P＞0.05).However,the diameter of group C was significantly higher than group A (P＜0.05).On histological examination,larger vascular density was observed in group C compared with group A at 1 month (P＜0.05).And reduced inflammatory reaction was also observed in group C at 3 months(P＜0.05).While other histological parameters were not statistically significant (P＞0.05).Conclusions PRP does not enhance the survival of fat graft in the Wistar rat model.

Objective To investigate the effect on NO、IL-10、CRP and bacterial translocation rate by glutamine enriched parenteral nutrition solution in purulent appendicitis in children.Methods72 cases of acute suppurative appendicitis complicated with peritonitis were selected and randomLy divided into control and treatment groups.Patients (36 cases) in the control group were given antibiotics and parenteral nutrition support for 7 days, 36 cases in the experiment group were given glutamine enriched parenteral nutrition solution for 7 days.NO、IL-10、 CRP and bacterial translocation rate were compared after the treatment.ResultsCompared with before treatment, two groups of patients with content of FT3, FT4 was higher(P<0.05), CRP, NO and IL-10、 content in serum was lower (P<0.05), 16Sr and BG positive rate was lower (P<0.05).Compared with the control group, the serum FT3, FT4 was higher(P<0.05), CRP, IL-8 and IL-6 content in serum was lower (P<0.05), 16Sr and BG positive rate was lower (P<0.05).ConclusionGlutamine enriched parenteral nutrition solution can significantly improve thyroid hormone levels,reduce serum CRP, bacterial translocation rate, and adverse reactions were mild.
