AIM To compare the inhibition of pravastatin on HMG CoA reductase with enalapril and verapamil in SDR and SHR liver. METHODS The liver microsomes were prepared by ultracentrifugation, the HMG CoA reductase activity was determined by radioactivity in vitro. RESULTS Pravastatin inhibited HMG CoA reductase, and its IC 50 value was 0 28 g?L -1 . 3.Epithelial-mesenchymal transition of renal tubular epithelial cells under anoxia

Objective To explore whether anoxia can induce expression changes in connective tissue growth factor(CTGF)in renal tubular epithelial cells(TECs)and epithelial-mesenchymal transition of TECs.Methods Rat renal TECs(NRK-52E)anoxia models were established.NRK-52E cells were exposed to anoxia for 4 h.The real-time RT-PCR,Western blotting,immunohistochemical staining were used to detect the expression of CTGF at 6,12,24,48,and 72 h in NRK-52E cells.Morphological changes and cytoskeleton remodeling in NRK-52E cells under anoxia were examined by a laser confocal microscope and BODIPYFL staining respectively.Results Under anoxia,NRK-52E cells became round,enlarged and cytoskeleton was remodeled.The expression levels of CTGF mRNA and protein were up-regulated at 6 h,reached their peak at 48 h:the expression of CTGF mRNA protein was 29.33±0.21 and 1.30±0.02 respectively.Under anoxia,NRK-52E cells underwent an epithelial-mesenchymal transition process,including cytoskeleton remodeling,and morphological changes.Conclusion Anoxia can change the expression of CTGF and other fibrosis-associated genes in NRK-52E cells,and CTGF played an important role in fibrosis process and epithelial-mesenchymal transition development in NRK-52E cells.
4.Endoscopic esophageal varices ligation under topical anesthesia,sedation with consciousness or intravenous anesthesia:a comparison between three anesthetic methods

Objective To compare the efficacy and safety of topical anesthesia,conscious sedation and intravenous anesthesia during endoscopic varices ligation(EVL).Methods Patients underwent EVL were divided into 3 groups to receive different anesthetic methods,namely topical anesthesia,conscious sedation and intravenous anesthesia,with 50 subjects in each group.The changes of vital signs,the tolerance to stimulation of the procedure,the time of operation,the rate of complication were recorded and compared between 3 groups.Results The procedure of EVL were completed in all patients.In topical anesthesia group,40(80%)patients had nausea and vomiting,9 cases(1 8%)tried to pull out the endoscopy.The mucosa at the entrance of the esophagus were injured in 10 cases(20%).Massive bleeding occurred in 4 patients(8%)during operation because of nausea and vomiting.In conscious sedation group,only 7 patients(14%)had mild nausea and vomiting,and no complication of variceal bleeding occurred.The mucosa at the entrance of the esophagus was injured in 5 cases(10%).In intravenous anesthesia group,no patient had nausea or vomiting.The respiratory rate,heart rate and mean artery pressure decreased during the procedure,but without significant difference(P>0.05).The operation time in intravenous anesthesia group WaS shorter than that in other two groups,but without significant difference(P>0.05).Conclusion EVL can be completed under 3 different anesthetic methods,while EVL under conscious sedation is more effective and safe.
5.Study on Three Principal Degraded Impurities of Mitiglinide Calcium

OBJECTIVE:To isolate and purify three principal degraded impurities of mitiglinide calcium (impurity A,B,C) and identify their structures,establish HPLC method for content determination of impurity A,B,C. METHODS:Mitiglinide calci-um was used as raw material and reacted with acid;3 impurities were then separated by HPLC and their structures were elucidated by IR,MS,1H NMR,13C NMR,LC-ESI-MS and ORD. 3 impurities of 3 batches of mitiglinide calcium were determined,and the determination was performed on Agilent Extend-C18 column with mobile phase consisted of 0.01 mol/L sodium acetate solution-ace-tonitrile-triethylamine(60:40:0.1,pH=3.0)at the flow rate of 1.0 ml/min. The detection wavelength was set at 210 nm and sam-ple size was 20 μl. The response tests of 3 impurities and mitiglinide calcium were conducted. RESULTS:After treated with acid, impurity A,B,C had been obtained,and their purity were 99.05%,98.87%,99.98%,respectively after isolation and purifica-tion;after identifying the structure, 3 impurities were S-2-bezylsuccinic acid, S-2-bezylsuccinic acid-4-methyl ester, methyl (2S)-2-benzyl-3-(cis-hexahydroisoindolin-2-ylcarbonyl) propionate;methodological study of content determination of impurities were all up to the requirement. The linear range of impurity A,B,C were 0.387 5-3.875,0.395-3.95 and 0.392 5-3.925 μg/ml(all r were 1.000 0). The response value of impurity A,B,C and mitiglinide calcium were 2.316 1,2.636 1,2.617 8 and 2.620 4,re-spectively. CONCLUSIONS:The structures of 3 principal degraded impurities of mitiglinide calcium have been identified and con-firmed;the content of them can be determined by HPLC main component self-comparison method.
6.Studies on Renal Histological Parameters in SD Rats and Beagles

[Objective] To investigate the renal histological parameters in SD rats and beagles and to supply evidence for renal toxicological and pathological diagnosis. [Methods] Forty SD adult rats and 20 adult beagles (half males and half females) , were respectively executed at 12 weeks and 12 months old. Then, kidney tissue was fixed in 10% neutral formalin, embedded in paraffin, made into slices at 5?m and stained by HE. The larger diameter and shorter diameter of glomerulus in the renal cortex and outer stripe of rats and beagles were determined by Spot Advanced image analysis software. The differences of the above parameters appearing in different species and genders were compared. [Results] The larger diameter was about 96 ?m, and the shorter diameter 76 ?m in the renal cortex of the beagles, and the larger was about 120?m and the shorter 94 ?m in the outer stripe of the beagles. The differences of the larger and shorter diameters were insignificant between the male and female beagles, but the larger and shorter diameters in the outer stripe were longer than those in the cortex. The larger diameter and shorter diameter were about 87 ?m and 64 ?m respectively in the renal cortex of SD rats, and the larger was about 88 ?m and the shorter 64 ?m in the outer stripe of SD rats. The differences of the larger and shorter diameters were insignificant between the male and female beagles, as well as in the outer stripe and the cortex. [Conclusion] The nephron in the outer stripe is larger than that in the cortex of beagles but no difference of the nephron exists in SD rats. The nephron in beagles is larger than that in SD rats, and there is no difference in the nephron between male and female beagles and SD rats.
7.Influence of collective motion excitation on COPD patients at stable stage

Objective To investigate the influence of collective motion excitation on the anxiety/depression and the quality of life of COPD patients at stable stage? Methods Ninety-six patients with chronic obstructive pulmonary disease(COPD)at stable stage were divided into the intervention group and the control group,both treated with regular medical treatment and general nursing?Collective motion excitation was implemented in the intervention group twice per week in 12 weeks? The Hamilton Anxiety Scale (HAMA),the Hamilton Depression Scale(HAMD)and the Short Form-36 Health Survey(SF-36)were used to assess all patients before and after the 12-week nursing? Result Compared to the control group,the intervention group demonstrated significant improvements in HAMA ,HAMD and SF-36 scores at the endpoint of 12 weeks(P < 0?05),and no difference before the nursing intervention(P > 0?05)? Conclusion The anxiety/depression and the quality of life can be improved with COPD patients at stable stage through collective motion excitation?
8.Morphometric Analysis of Liver Tissue in SD Rats and Beagle Dogs

Objective To investigate the parameters of liver tissue in SD rats and beagle dogs and to supply evidence for toxicologic pathological evaluation.Methods Forty SD rats and twenty beagle dogs,male in half,were used for the observation.SD rats were sacrificed at the age of 12 weeks and the beagles at the age of 12 months.After complete necropsy examination,all livers were fixed in 10 % neutral buffered formalin solution,embedded in paraffin,sliced at a thickness of 5 ? m and stained with hematoxylin and eosin(HE).The distance of central vein and portal area,and that of central veins in the neighbored lobules were measured by means of an image analyzer Spot Advanced.The difference of the above distance between beagles and rats,and that between male and female were compared.Results The distance of central veins in the neighbored lobules was about 883 ? m in the beagles and about 496 ? m in SD rats,and the distance between lobule center and portal area was about 511 ? m in beagle dogs and about 410 ? m in SD rats.There was no difference between the male and the female.Conclusion The distance of central veins in the neighbored lobules in beagle dogs is longer than that in SD rats,so does the distance between lobule center and portal area.The sex of the animal has no influnce on the size of the lobules.
9.Characteristics of human embryonic stem cells cultured on human- and mouse- origin feeder layers

BACKGROUND: Whether human embryonic stem cells (hESCs) cultured on different feeder layers can maintain identical or similar characteristics remains unclear.OBJECTIVE: To compare the characteristics of hESCs cultured on human- and mouse- origin feeder layers.DESIGN, TIME AND SETTING: The in vitro cytology observation was performed at the Kunming Institute of Zoology, Chinese Academy of Sciences between September 2007 and February 2009.MATERIALS: Two ICR pregnant mice with 12.5-13.5 embryonic days were provided by Animal Center of Kunming Medical College. Immortalized human adult fibroblast (HAFi) cell line was presented by School of Medicine, Johns Hopkins University (USA). hESCs line was provided by Kunming Institute of Zoology, Chinese Academy of Sciences.METHODS: Mouse embryonic fibroblasts (MEF) were harvested from ICR mice by trypsinization method. HAFi were conventionally cultured. After γ ray treatment, two kinds of cells were incubated on 6-well gelatin-coated plate with density of 2.5×10~4/cm~3. hESCs were cultured on HAFi or MEF feeder cells containing β-mercaptoethanol DMEM/F12 and basic fibroblast growth factor.MAIN OUTCOME MEASURES: Morphology, expressions of specific molecular markers, Oct-4 positive rate, as well as cell doubling time of hESCs cultured by two methods were compared.RESULTS: ①BG02 cells on MEF and HAFi shared the similar morphology and characteristic pluripotency markers, which expressed SSEA-3, SSEA-4, TRA-1-60, TRA-1-81 and Oct-4, but not SSEA-1. However, the proportion of positive Oct-4 cells in hESCs colonies maintained on MEF was lower than that of HAFi (P < 0.05) with shorter doubling time (P < 0.05).CONCLUSION: hESCs cultured on MEF and HAFi represent some differences in the growth and pluripotency characteristics.
10.Creation and analysis of the model of sleeve gastrectomy with modified jejunoileal bypass

Objective To construct and analyze the model of the sleeve gastrectomy with modified jejunoileal bypass (SG/MJIB) on non-obese Goto - kakizaki (GK) rats. Methods GK rats were randomly divided into SG/MJIB, sham-SG/MJIB, pair-fed (PF) and controls group. Before and after surgery, the changes of weight, food intake, fasting glucose, oral glucose tolerance test (OGTT), insulin tolerance test (ITT), fasting plasma Insulin and the histological changes in islet were dynamically observed for 16 weeks. Results From the 4th week postoperative, the weight changes of SG/MJIB significantly decreased compared with sham-SG/MJIB and PF group (P<0.01). Fasting glucose concentration of SG/MJIB animals was lower than sham-SG/MJIB, PF and controls group (P<0.05). The OGTT of SG/MJIB rats was obviously improved compared with the sham-SG/MJIB, PF and controls group (P<0.01). Two weeks after operation, glucose tolerance was better improved in SG/MJIB than preoperative, the area under the curve (AUC) of blood glucose concentration decreased by about 38.9% (P<0.01). Postoperative insulin levels in SG/MJIB group were obviously lower than sham-SG/MJIB group throughout the experiment(P<0.05). In the 16th week, the morph of pancreatic islet of SG/MJIB was obviously improved. In the SG/MJIB group, the number of positive β-cell and mature acinus was significantly increased, while sham surgery groups had no obvious changes as mentioned above. Conclusions SG/MJIB was directly linked to the reduction in glucose levels in GK rats, independently weight loss and caloric intake, and it can be served asa stable long-lasting hypoglycemic surgery model to research the mechanism for the treatment of type 2 diabetes.
