TGF ? plays a crucial role in the renal fibrosis and Smads mediate the intracellular signal transduction of TGF ?.Overexpression of Smad7,which can mediate TGF ? by the feedback action,is able to change the physiopathology equilibrium between the R Smads response and the I Smads inhibition to the fibrosis induced by TGF ?.It may help to understand the mechanism of renal fibrosis and Smads TGF ? signaling transduction, and it may also provide a new approach to the renal fibrosis.
2.Quantitative and qualitative research in cognition of clinical nurse specialist

Objective To study the clinical nurses and nursing students' cognition of clinical nurse specialist (CNS) and provide the basis for promoting the development of CNS.Methods Quantitative and qualitative research were adopted in this study.One hundred and ninety clinical nurses and nursing students were investigated with a questionnaire about the cognition of CNS,and the qualitative research was conducted on 9 clinical nurses selected by purposive sampling method.Results Most participants realized the importance of CNS and 78.4% of them wished to be an excellent CNS.The cognition was different between the two groups,especially in terms of ability,working performance and status.Conclusions We should improve the clinical nurses and nursing students' cognition of CNS,and cultivate them with the core competence of CNS.Meanwhile,improving the status and confirming the role must be done to promote the development of CNS in China.
3.Transcript assembly and quality assessment.

The transcript assembly is essential for transcriptome studies trom next-generation sequencing data. However, there are still many faults of algorithms in the present assemblers, which should be largely improved in the future. According to the requirement of reference genome or not, the transcript assembly could be classified into the genome-guided and de novo methods. The two methods have different algorithms and implementation processes. The quality of assembled transcripts depends on a large number of factors, such as the PCR amplification, sequencing techniques, assembly algorithm and genome character. Here, we reviewed the present tools of transcript assembly and various indexes for assessing the quality of assembled transcripts, which would help biologists to determine which assembler should be used in their studies.

4.Correlation between traditional Chinese medicine syndromes in primary immunoglobulin A nephropathy and A267G in 5'-untranslated region within exonal of megsin gene

OBJECTIVE: To observe the correlation between traditional Chinese medicine (TCM) syndromes ("deficiency of qi and yin" and "deficiency of liver yin and kidney yin") and A267G in 5'-untranslated region within exonal of megsin gene, and to search the substantial genetic basis for micro-differentiation of TCM syndromes in primary immunoglobulin A nephropathy (IgAN). METHODS: A total of 120 IgAN cases meeting the diagnostic criteria were enrolled. The sequence of single nucleotide polymorphism (SNP) of A267G in 5'-untranslated region within exonal of megsin gene was tested. The correlation between SNP and TCM syndromes was observed. RESULTS: There were 83 cases carrying GG genotype, 34 cases carrying GA genotype and 3 cases carrying AA genotype in 120 cases of primary IgAN. There was a high proportion of "deficiency of liver yin and kidney yin" in IgAN cases with AA and GA genotypes, and a high proportion of "deficiency of qi and yin" in IgAN cases with GG genotype (P<0.01). Odds ratio in TCM syndrome distribution between GG genotype and GA plus AA genotype was 9.800, and 95% confidence interval was 3.969-24.199. The discrepancy also resided in IgAN patients with different genders and ages. CONCLUSION: A267G in 5'-untranslated region within exonal of the megsin gene may be one of the substantial genetic basis for differentiating "deficiency of liver yin and kidney yin" syndrome and "deficiency of qi and yin" syndrome in primary IgAN.
5.Relationship between UGT1A1 * 28 gene polymorphism and delayed diarrhea caused by FOLFIRI treatment

Objective To analyze the relationship between UDP-glucuronosyltransferase 1A1 (UGT1A1)gene polymorphism and delayed diarrhea caused by FOLFIRI treatment.Methods Two hundred and one blood samples were taken from patients with metastatic digestive tract tumor before chemotherapy by FOLFIRI and then the UGT1A1 * 28 genetic polymorphism was performed.All the cases treated with FOLFIRI were chosen to be observed and recorded by situation of the delayed diarrhea during chemotherapy,and to analyze the relationship between UGT1A1 * 28 genetic polymorphism and grade 3 and 4 delayed diarrhea.Results The distributions of the genotypes in 201 metastatic digestive tract tumor patients were as follows:UGT1A1 * 28 wild-type genotype TA6/6 (155,77.11%),heterozygous genotype TA6/7 together with homozygous genotype TA7/7 (46,22.89%).In the 201 cases,the incidences of grade 1 and 2 delayed diarrhea in the patients carrying wild-type genotype and mutant type were respectively 45.16% (70/155),39.13% (18/46).The incidences of grade 3 and 4 delayed diarrhea were respectively 9.68% (15/155),19.57% (9/46),with no statistical difference (x2 =3.318,P =0.190).Conclusion The UGT1A1 * 28 polymorphism TA6/7 or TA7/7 can not increase the risk of grade 3 or more severe delayed diarrhea for the patients with metastatic digestive tract tumor after receiving FOLFIRI treatment.
6.Preparation,Characterization and Transdermal Ability Study in vitro of Ibuprofen Nano-powder

OBJECTIVE:To prepare and characterize Ibuprofen (IBU) nano-powder,and to investigate its transdermal ability in vitro. METHODS:Using chloroform-ethanol(7:3,V/V)as organic phase,deionized water as aqueous phase and polysorbate 80 as surfactant,the emulsification method was used to prepare IBU nano-powder. Laser granulometric analysis,Fourier transform in-frared spectroscopy(FT-IR),X-ray diffraction(XRD),differential scanning calorimetry(DSC)were used to characterize IBU na-no-powder. IBU nano-powder was compared with bulk drug in respects of saturation solubility,dissolution rate and transdermal rate in vitro. RESULTS:The optimum condition was as follows that the concentration of polysorbate 80 was 5 mg/mL;the volume ra-tio of water phase-organic phase was 40:1;the concentration of IBU was 250 mg/mL;homogenate speed was 5000 r/min;homog-enate time was 2 min. Prepared IBU nano-powder was polyporous crumbly coralliform,and its chemical structure kept stable;the nano-powder changed from crystal to amorphous state;the particle size was 179.6 nm,and drug-loading amount was 8.99%;satu-ration solubility,dissolution rate and transdermal rate of IBU nano-powder were 148,1.23 and 4.08 times of bulk drug. CONCLU-SIONS:The prepared IBU nano-powder shows good water-solubility and percutaneous permeability.
7.Effect of propolis on cariogenesis of main cariogenic streptococci and their fluoride-resistant strains

Objective To evaluate the effects of propolis on the growth of S.mutans ATCC 25175(S.m),S.sobrinus 6715(S.s) and their fluoride-resistant strains(S.m-FR,S.s-FR),and the change of their glucosyltransferase(GTF).Methods S.m and S.s were induced with sodium fluoride by minimum inhibitory concentration(MIC) to induce fluoride-resistant strains(S.m-FR and S.s-FR).Fluid dilution method was used to observe the effects of different levels of propolis on growth of S.m,S.m-FR,S.s and S.s-FR.Chemistry enzyme analysis was used to detect the influence of propolis on the enzyme activity of GTF.Results MIC of propolis for S.m,S.m-FR,S.s and S.s-FR were respectively 0.39,0.78,0.20 and 0.39 g/L.MBC of propolis for them were respectively 0.78,1.56,1.56,and 1.56 g/L.GTF of S.m,S.m-FR,S.s,and S.s-FR was decreased gradually with the increase of concentration of propolis.There were significant differences among total sample groups,and each experimental group and positive control group as well(P

9.LRIG1 expression and its possible suppressor role in oral verrucous carcinoma

Objective:To explore leuncine-rich repeats and immunoglobulin-like domains protein 1 (LRIG1) expression in oral verrucous carcinoma(OVC) and its possible mechanism of tumor suppression.Methods:Paraffin specimens of OVC (n =15) and oral squamous cell carcinoma(OSCC,n =30) and corresponding adjacent normal tissues,and 15 normal mucosa tissues(NM) as control from injured patients were collected.The expression levels of LRIG1 and Bcl-2 were examined using immunohistochemical SP method.The correlation between the expression levels of LRIG1 and Bcl-2 was determined using two tailed Pearson's correlation.Results:LRIG1 was significantly lower in OVC tissue than that in NM tissue,but higher than in OSCC tissue.The expression levels of Bcl-2 in OVC were significantly higher than that in NM,but lower than in OSCC.The expression level of LRIG1 was negatively correlated with expression level of Bcl-2 in NM,OVC and OSCC tissue.Conclusion:LRIG1 may inhibit the expression of Bcl-2 and the development of OVC.
10.Comparison of the efficacy between cyclophotocoagulation and trabeculectomy for primary acute angle-closure glaucoma with persistent ocular hypertension

Background Transscleral diode laser cyclophotocoagulation (TDCP) was used to later stage of glaucoma in the past.However,the indications of TDCP are expanding gradually over time and there are more and more reports about its application in different stages and different types of glaucoma.Objective This study was to compare the clinical efficacy of TDCP and trabeculectomy for primary acute angle-closure glaucoma (PAACG) with persistent ocular hypertension.Methods Ninety-five PAACG patients with persistent ocular hypertension were retrospectively analyzed.The patients were divided into TDGP group (62 eyes of 56 cases) and trabeculectomy group (44 eyes of 39 cases) based on the difference of treating fashions.The clinical outcomes were compared between the two groups with the evaluating indexes of visual acuity,intraocular pressure (IOP) before and 1 week,3 months and the end of following-up as well as the number of drug used and the re-treatment.Results The demography was matched between the two groups (all at P>0.05).The number of eyes with visual improvement was significantly different among the groups (x2 =30.927,P =0.002) in the TDCP group.Compared with before treatment,the number of visual improvement in 1 week after treatment was significantly raised (x2 =16.778,P =0.002).No significant differences were seen in the eye number of visual improvement in various timepoints after treatment (all at P>0.008).Also,the eye number of visual improvement was significantly different among the different timepoints (x2 =44.345,P<0.001),and statistically significant change was found between the preoperation and postoperative 1 week (x2 =23.725,P<0.001).However,there were no significant differences in the eye number of visual improvement in various timepoints after operation each other (all at P>0.008).The IOP was insignificantly altered among the groups at various timepoints (Fgroup =3.836,P =0.053),but the change was significantly different over time (Ftime =757.078,P<0.001).The postoperative IOP was significantly higher than that of preoperation in both groups (all at P<0.001).The proportion of eyes using lowering-IOP drugs was 48.4% in the TDCP group,and that in the trabeculectomy group was 27.3%,showing a significant difference between them (x2 =4.796,P =0.029).The retherapy was performed in 20 eyes (32.3%) and 2 eyes (4.5%) in the TDCP group and the trabeculectomy group,respectively.Conclusions The short-term lowing-IOP effecacy of TDCP is similar to trabeculectomy,but the longterm lowing-IOP efficacy of TDCP is not good.Compared with the trabeculectomy,TDCP shows more simple operation,less time-consuming during the surgery and milder tissue damage,but the eyes continuing to use anti-glaucomatous drugs or re-therapy are increased.
