1.Effects of Enteral Nutrition on Clinical Curative Effect in Elderly Patients with Severe Craniocerebral Diseases

Objective To investigate the best time to establish the enteral nutrition (EN) in elderly patients with se?vere craniocerebral diseases. Methods Eighty-one elderly patients were divided into groups A, B and C. The EN support were given 24 h, 25-48 h, and 49-72 h after surgery. Changes of serum biochemical indexes, complications and GCS score were detected and statistical analysed. Results The serum levels of albumin and hemoglobin were significantly increased in group B than those of group A and group C. The total number of lymphocytes was significantly higher in group A and group B than that of group C (P<0.05). The amount of diarrhea was significantly fewer 7-day after operation in group B than that of group A (3.6%vs 26.9%). The amount of stress ulcer was significantly fewer in group B than that of group C (0 vs 22.2%). The amounts of lung infections (10.7%vs 38.5%) and stress ulcer (3.6%vs 25.9%) were significantly fewer 14-day after oper?ation in group B than those of group A. The scores of GCS were significantly higher 14-day after operation in group B than those of group A and group C (10.2±1.3 vs 9.5±1.2 vs 9.4±1.2, P<0.05). Conclusion The establishment of EN support 25 to 48 hours after surgery may meet the need of nutritional and metabolic support, reduce complications, be conducive to the consciousness recovery in elderly patients with severe craniocerebral diseases.
2.Effect of bFGF on expression of endothelial cell integrin ?_1 subunits

AIM: To study the effect of basic fibroblast growth factor(bFGF) to the expression of integrin ? 1 subunit of endothelial cells. METHOD: The expression of integrin ? 1 subunit of endothelial cells was determined before and after bFGF treatment with Western Blot and immage analysis method.RESULTS: The mean gray value of immunostain by immage analysis method is 166.11?9.86 in the experimental group and 175.32?5.12 in the control group, suggested that bFGF may upregulate expression of integrin ? 1 subunits of endothelial cell. CONCLUSION: bFGF may play an important role in angiogenesis by way of inducing the overexpression of endothelial cell integrin ? 1 subunits.
3.Bacteriological investigation of sea waters around an island of Xi-Sha archipelago

Objective To explore the bacterial species and total number of bacteria in seawater around an island in Xi-Sha archipelago and evaluate the drug sensitivity of the bacteria. Methods We studied the bacteria in various sea districts of the island. We collected 80 samples of seawater and cultured to identify bacterial species and their drug sensitivity. Results Vibrio ranked first on the isolation rate (45.98%), and from the near to distant sea districts, its isolation rate presented a typical stepwise upward tendency. The isolation rates of Sphingomonas paucimobilis and Moraxella species presented a typical stepwise downward tendency in contrast to Vibrio alginolyticus. CDC group EO-2 was mainly found in near sea districts but not along the seashore or in distant sea districts. The average bacteria number was 337.44?174.88 cfu/ml. The quantities of bacteria presented an obvious gradient downward trend from the near to distant sea districts. Vibrio cholerae, Samenella. dysenteridae, E.coli and Staphylococcus aureus were not detected. The four main kinds of bacteria found by us were all highly susceptible to 16 kinds of antibiotics, and we found no resistant strains to Tetracycline, Gentanycin. Chloromycin, and Amikacin. Conclusions The quality of seawater in Xi-Sha was fine in so far as bacterial contamination was concerned. Main bacterial species found included Vibrio alginolyticus, Sphingomonas paucimobilis, Moraxella species and Comamonas terrigena etc. The isolation rate of the different bacterial species presented obvious gradient variations. In sea districts far off the seashore, Vibrio alginolyticus accounted for 77.27% of the all bacteria. All the main bacterial species were highly sensitive to antibiotics.
4.CT evaluation of lumbar drainage improving subarachnoid hemorrhage

Objective To analyze the safety and effectiveness of using lumbar drainage (LD) in patients with subarachnoid hemorrhage (SAH).Methods Data from 44 patients with SAH were collected.Of these patients,the LDs group consisting 19 patients in whom a lumbar drain had been placed for cerebro-spinal fluid (CSF) drainage,whereas the control group was composed of 25 patients who received no form of CSF drainage or treated solely with an external ventricular drainage (EVD).Incidence of new cerebral infarction of the two groups was measured on sequential CT and complications of LDs group.Results This retrospective review demonstrated a statistically significant beneficial effect in LDs group on CT,reducing the incidence from 32% to 5%.A similar degree of benefit was observed in two group patients with hydrocephalus.Complications with lumbar drains were rare and yielded no permanent sequelae.Conclusion With rare complications and definite safety,LDs are effective to reduce vasospastic cerebral infarction of SAH patients when appropriate patient selection and good technique are employed.
5.Putative Phosphorylation Sites On WCA Domain of HA2 Is Essential For Helicoverpa armigera Single Nucleopolyhedrovirus Replication

Protein phosphorylation is one of the most common post-translational modification processes that play an essential role in regulating protein functionality.The Helicoverpa armigera single nucleopolyhedrovirus (HearNPv) orf2-encoded nucleocapsid protein HA2 participates in orchestration of virus-induced actin polymerization through its WCA domain,in which phosphorylation status are supposed to be critical in respect to actin polymerization.In the present study,two putative phosphorylation sites (232Thr and 250Ser) and a highly conserved Serine (245Ser) on the WCA domain of HA2 were mutated,and their phenotypes were characterized by reintroducing the mutated HA2 into the HearNPV genome.Viral infectivity assays demonstrated that only the recombinant HearNPV bearing HA2 mutation at 245Ser can produce infectious virions,both 232Tbr and 250Ser mutations were lethal to the virus.However,actin polymerization assay demonstrated that all the three viruses bearing HA2 mutations were still capable of initiating actin polymerization in the host nucleus,which indicated the putative phosphorylation sites on HA2 may contribute to HearNPV replication through another unidentified pathway.
6.Dexmedetomidine intervention effects on aquaporin -1 and inflammatory cytokines in myocardial tissue of septic rat

Objective To study the regulation of aquaporin-1(AQP-1)changes in the heart of septic rats, compare the correlations of the AQP-1 with myocardial cytokines tumor necrosis factor-α(TNF-α),interleukin-6 (IL-6),and myocardial tissue water content,and to investigate the dexmedetomidine protective effect on myocardia in septic rats and its possible mechanism. Methods According to the random number table methods,90 male Sprague-Dawley(SD)rats were divided into sham operation group,sepsis model group and dexmedetomidine group, 30 rats in each group. The rat sepsis model was established by cecal ligation and puncture(CLP). In the sham operation group,the animal abdomen was only opened and closed without CLP. Half hour before operation in dexmedetomidine group,dexmedetomidine 1μg/kg(2μg/mL)was injected into the vein,while in the model and sham groups,saline 5 mL/kg was subcutaneously injected into the rat after the operation. At 2,12,24,48,72 hours after operation,6 rats were sacrificed and their hearts removed at one time point in a group. Enzyme linked immunosorbent assay(ELISA)was used to detect the content of AQP-1 and the levels of the TNF-α,IL-6 in the myocardial tissue homogenate at all time points,the myocardial tissue water content was detected by dry wet weight,and the correlations between AQP-1 and TNF-α,IL-6 and between AQP-1 and myocardial tissue water content were compared. Results From 2 hours after operation,the levels of the AQP-1,TNF-αand IL-6 in model group were significantly higher than those in the sham operation group;with prolongation of time,the level of AQP-1 and myocardial tissue water content were decreased, but the levels of TNF-α and IL-6 were persistently increased. From 2 hours after operation in dexmedetomidine group,all the above indexes except myocardial tissue water content at 72 hours after operation were significantly lower than those in the model group〔AQP-1(ng/g):9.29±0.15 vs. 9.73±0.26,TNF-α(pg/g):109.47±8.41 vs. 128.13±7.36,IL-6(pg/g):232.95±20.56 vs. 279.71±22.24,myocardial tissue water content:(74.82±6.37)%vs.(75.62±6.39)%,all P<0.05〕,but still higher than those of the sham operation group. The correlation analyses for the septic group showed that the change of AQP-1 was positively correlated to the myocardial water content in early stage(r=0.418,P=0.001)and later stage(r=0.235,P=0.022),and the changes of the AQP-1 in early stage (at post-operative 2 hours)were positively correlated to the concentration changes of the cytokines TNF-α(r=0.235,P=0.021)and IL-6(r=0.345,P=0.003),but in the later stage(at post-operative 72 hours)were negatively correlated with the changes of TNF-α(r=-0.408,P=0.037)and IL-6(r=-0.276,P=0.002). Conclusions In the early stage of septic rats,there is obvious myocardial injury,resulting in the over expression of AQP-1 and the occurrence of myocardial edema,dexmedetomidine can play a role in myocardial protection in such rats and its mechanism is possibly related to the reduction of the expression of AQP-1 and the levels of inflammatory cytokines, and in turn the alleviation of myocardial cell edema.
7.Clinical study of Beling Weitong granule combined with proton pump inhibitors in senile gastroesophageal reflux disease

Objective:To explore the clinical effect of Beling Weitong granule combined with proton pump inhibitors in senile gastroesophageal reflux disease(GERD).Methods:A total of 186 patients with GERD treated in the Second People′s Hospital of Dujiangyan from January 2020 to December 2021 were retrospectively selected as the study subjects, of which 93 patients received proton pump inhibitor (prilosec) orally (control group) and 93 patients received Beling Weitong granule combined with prilosec orally (combination group). Both groups were treated for 8 weeks. Symptom score, reflux symptom-based questionnaire (GERDQ) score, clinical efficacy, serum levels of gastrin-17 (G-17), motilin (MTL), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) before and after treatment and safety were compared between the two groups.Results:After treatment, the symptom score and GERDQ score in the combination group were lower than those in the control group: (8.28 ± 2.57) scores vs. (15.68 ± 3.69) scores, (8.15 ± 2.23) scores vs. (10.50 ± 2.45) scores, there were statistical differences ( P<0.05). The total effective rate in the combination group was higher than that in the control group: 96.77% (90/93) vs. 87.10% (81/93), there was statistical difference ( χ2 = 5.87, P<0.05). After treatment, the levels of serum G-17 and MTL in the combination group were higher than those in the control group, the levels of IL-6 and TNF-α were lower than those in the control group: (7.48 ± 0.96) pmol/L vs. (6.55 ± 0.83) pmol/L, (206.96 ± 37.83) pg/L vs. (184.75 ± 35.69) pg/L, (9.38 ± 1.92) ng/L vs. (14.13 ± 2.04) ng/L, (9.41 ± 1.85) ng/L vs. (12.01 ± 2.11) ng/L, there were statistical differences ( P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups ( P>0.05). Conclusions:Beling Weitong granule combined with proton pump inhibitor in the treatment of senile gastroesophageal reflux disease is effective, and it is safe.
8.Effect of miR-203a-3p overexpression on pulmonary fibrosis after LPS-induced acute lung injury in rats and its mechanism

Objective:
To investigate the effects of miR-203 a-3 p overexpression on pulmonary fibrosis after acute lung injury induced by lipopolysaccharide(LPS) in rats and its possible mechanism.
Methods:
The rat model of pulmonary fibrosis after acute lung injury was established by multiple low-dose injections of LPS. The successfully modeled rats were randomly divided into model group, agomir-negative control group(agomir-NC group) and miR-203 a-3 p agomir group(agomir group), and another control group was set, with 15 rats in each group. One day before modeling and one week after modeling, miR-203 a-3 p agomir was injected by caudal vein for intervention. Two weeks after modeling, the wet/dry specific gravity(W/D) and hydroxyprolinic acid(Hyp) content of lung tissue were determined. The pathological changes and the degree of pulmonary fibrosis of lung tissue were observed by HE staining and Masson staining. The levels of interleukin-1β(IL-1β), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in broncholveolr lavage fluid(BALF) were determined by ELISA. The expression levels of miR-203 a-3 p and follicle-statin like 1(Fstl1) mRNA in the lung tissues of rats were detected by qRT-PCR. The protein expression level of Fstl1 in lung tissue of rats was detected by Western blot. The targeting relationship between miR-203 a-3 p and Fstl1 was detected by dual luciferase reporting system.
Results:
Compared with control group, the degree of lung injury and pulmonary fibrosis in model group were relatively serious, the W/D value and Hyp content of lung tissue increased(P<0.05), the levels of IL-1β, IL-6 and TNF-α in BALF increased(P<0.05), the expression of miR-203 a-3 p in lung tissue decreased(P<0.05), while the mRNA and protein expression levels of Fstl1 increased(P<0.05). Compared with model group, the degree of lung injury and pulmonary fibrosis were improved in agomir group, the W/D value and Hyp content of lung tissue decreased(P<0.05), the levels of IL-1β, IL-6 and TNF-α in BALF decreased(P<0.05), the expression of miR-203 a-3 p in lung tissue increased(P<0.05), while the mRNA and protein expression levels of Fstl1 decreased(P<0.05). The luciferase reporter gene assay confirmed that Fstl1 was the target gene of miR-203 A-3 p.
Conclusion
Overexpression of miR-203 a-3 p improves pulmonary fibrosis after LPS-induced acute lung injury, and the mechanism may be related to the targeted down-regulation of Fstl1 expression.
