1.Comparison of the Efficacy and Safety of Levonorgestrel Intrauterine System and Mifepristone in the Treatment of Perimenopausal Dysfunctional Uterine Bleeding

OBJECTIVE:To compare the efficacy and safety of levonorgestrel intrauterine system and mifepristone in the treat-ment of perimenopausal dysfunctional uterine bleeding. METHODS:200 patients with perimenopausal dysfunctional uterine bleed-ing were randomly divided into observation group (100 cases) and control group (100 cases). Patients in the observation group placed levonorgestrel intrauterine system (containing levonorgestrel 52 mg),patients in control group received Mifepristone cap-sule 10 mg/d,oral,the treatment course for 2 groups were 3 months. Clinical efficacy,endometrial thickness,menstruation(PBAC score),hemoglobin level before and after treatment and incidence of adverse reaction in 2 groups were observed. RESULTS:After treatment,the total effective rate in observation group was significantly higher than control group,incidence of adverse reactions was significantly lower than control group,the differences were statistically significant(P<0.01). Before treatment,there were no significant differences in the endometrial thickness,PBAC score and hemoglobin level between 2 groups;after treatment,endome-trial thickness and PBAC score were significantly lower than before,and observation group was lower than control group;hemoglo-bin level was significantly higher than before,and observation group was higher than control group,the differences were statistical-ly significant(P<0.01 or P<0.05). CONCLUSIONS:The efficacy and safety of levonorgestrel intrauterine system are significant-ly superior to mifepristone in the treatment of perimenopausal dysfunctional uterine bleeding.
2.Perspective on the novel methods for DNA assembly.

In 2010, the artificial synthesis of Mycoplasma mycoides triggers the new era of synthetic biology. This great breakthrough is achieved mainly thanks to the powerful DNA recombinant ability of yeast. In recent years, except for the methods used for large DNA assembly on the basis of in vivo homologous recombination, various different DNA assembly methods in vitro, based on the concept of DNA ligation or polymerization, have also been developed, such as Biobrick\BglBrick, SLIC and Gibson one-step assembly. Application of these new technologies has greatly accelerated the construction of synthetic part libraries, biosynthetic pathway and even microbial chromosomes. In fact, all DNA assembly methods are derived from the combinations of DNA joining and organizational schemes. This review describes the brief introduction of the main in vivo and in vitro DNA assembly protocols developed so for, which will benefit the construction of different types of synthetic functional devices and also biosynthetic pathways in the research of synthetic biology in China.

3.Voxel-based comparison of whole brain gray matter of patients with mild Alzheimer's disease with normal aging volunteers

Objective To detect gray matter abnormalities of whole brain in patients with mild Alzheimer's disease(AD)by voxel-based morphometry(VBM).Methods Thirteen patients with mild Alzheimer's disease and sixteen normal aging volunteers underwent 3D SPGR scanning.For every subject,data was transferred to PC to be normalized,segmented and smoothed using SPM99.Non-dependent samples T-tests were conducted to compare gray matter density voxel to voxel between the two groups.Results Significant reductions in gray matter density were found in the bilateral hippocampi and nucleus amygdalae,bilateral insulae,bilateral medial thalami,bilateral rectus gyri,right superior temporal gyrus,right caudate nucleus,right prefrontal lobe,right basal forebrain and portions of right occipital lobe.Conclusion VBM reveals significant gray matter reductions of numeral cortices in mild Alzheimer's disease.It can be a useful method to evaluate the anatomical changes in the progress of the disease.
4.The Value of Measurement of Fractional Anisotropy on the Splenium of the Corpus Callosum in Diagnosis of Patients with Mild Alzheimer’s Disease

Objective To detect abnormal anisotropy on the splenium of the corpus callosum in patients with mild Alzheimer’s disease (AD).Methods Normalized hippocampal volume and fractional anisotropy on the splenium of the corpus callosum were measured in 13 patients with mild Alzheimer’s disease and 19 normally aging volunteers. Non-dependent samples t-tests were conducted to compare them between the two groups. In addition, the sensitivity in the diagnosis of mild Alzheimer’s disease was calculated.Results The splenium of the corpus callosum showed reduced anisotropy in mild Alzheimer’s disease. The normalized hippocampal volume also decreased in patients. The FA of the splenium of the corpus callosum was more sensitive in the diagnosis of mild Alzheimer’s disease. Conclusion The decreased anisotropy on the splenium of the corpus callosum reflects the degeneration of the fibers, and it is promising in the early diagnosis of Alzheimer’s disease.
5.Biocompatibility of Fe-Cr-Mo soft magnetic alloy with chromium plating

BACKGROUND:Studies have demonstrated that Fe-Cr-Mo soft magnetic aloy has desired magnetic properties and machining properties. Surface Cr6+ plating treatment can significantly enhance the corrosion resistance in oral environment, but its biosecurity needs further testing. OBJECTIVE: To evaluate the biocompatibility of Fe-Cr-Mo soft magnetic aloy with chromium plating. METHODS:Logarithmic growth phase L929 cel suspension was obtained and seeded in 96-wel plates at a cel concentration of 6×107/L. Pure titanium extract, Fe-Cr-Mo soft magnetic aloy original extract, Cr6+-plated Fe-Cr-Mo soft magnetic aloy and PVC extract were added respectively. After 5 days of culture, cel morphology and adherent circumstances were observed. The absorbance value was detected using cel counting kit-8 assay. The relative growth rate of cels in each group was calculated. The cytotoxicity grades of materials were evaluated. RESULTS AND CONCLUSION:In the pure titanium extracts group, the cels with normal morphology showed good adherent growth and no cytotoxicity. In the Fe-Cr-Mo soft magnetic aloy original extract group, the cel morphology and growth status were both good, with occasionaly individual cel lysis. Scattered reddish brown particles were visible in the culture solution, showing no or very mild cytotoxicity. In Cr6+-plated Fe-Cr-Mo soft magnetic aloy extract group, the cels grew wel and showed no or very mild cytotoxicity. In PVC extract group, more than 70% of cels were vacuole-shaped and presented with pyknosis or dissolution, there was a large number of cel debris, and cel growth was inhibited in over 50% cels, showing moderate cytotoxicity or above. The cytotoxicity of Cr6+-plated Fe-Cr-Mo soft magnetic aloy extract was grade 0-1. These results demonstrate that Fe-Cr-Mo soft magnetic aloywith chromium plating has good biocompatibility.
6.Study on the effects of different pacing modes of cardiac resynchronization therapy on dogs with heart failure

Objective To evaluate advantages and disadvantages of different pacing modes of cardiac resynchronization therapy (CRT).Methods Twelve dogs with heart failure were performed in every dog at random,and the pacing modes employed in the test included right atrium-different sites of ventricle,and ventricular sites included right ventricular bifocal (RV-Bi),biventricular (Bi-V),left ventricular (LV).The pacing frequency was 180 times per minute,and the results were measured before pacing and after 15 minutes when the pacing became stable in Color Doppler echocardiography,including left ventricular enddiastolic diameter (LVEDd),left ventricular ejection fraction (LVEF),interventricular mechanical delay (IVMD),interventricular septum and left ventricular posterior wall motion delay (SPWMD),left ventricular 12-segment peak time standard deviation (Ts-SD).Results (1)Compared with before pacing,at the RV-Bi,Bi-V,and LV pacing modes,LVEDd,IVMD,SPWMD,and Ts-SD decreased,LVEF increased,the difference was statistically significant [(42.42 ± 3.94) mm vs (34.00 ± 4.07) mm,(34.17 ± 3.95)mm,(33.75 ±4.18)mm; (28.08 ±4.01)mm vs (13.00 ±3.64) mm,(11.95 ±2.54)mm,(12.08 ±3.51) mm; (75.00 ± 10.22)mm vs (51.75 ±9.84) mm,(20.66 ±7.41) mm,(20.75 ±7.56) mm; (25.08±4.16)mm vs (14.91 ± 3.31)mm,(7.50 ±4.24) mm,(7.41 ±3.39)mm;(32.91 ±4.46)mm vs (41.50 ±4.16)mm,(42.00 ±4.63) mm,(42.41 ±4.99)mm,P <0.05].(2)Compared with RV-Bi pacing mode,at the Bi-V,LV pacing modes,SPWMD and Ts-SD decreased,the difference was statistically significant(P < 0.05); there was no significant difference among LVEDd,IVMD,and LVEF (P >0.05).(3)There was no significant difference in LVEDd,IVMD,SPWMD,Ts-SD and LVEF between LV and Bi-V pacing (P > 0.05).Conclusions The hemodynamic effects of RV-Bi and LV pacing modes were similar to that of Bi-V pacing,and they can be used as CRT biventricular pacing alternative modes; however,the mechanisms of improving ventricular synchronization are not identical in above pacing modes.
7.Effects of right ventricular septal pacing modes on acute cardiac function and ventricular synchronization in dogs with heart failure

Objective To evaluate effects of right ventricular septal pacing modes on treatment of heart failure.Methods Twelve dogs with heart failure were performed in every dog at random,and the pacing modes employed in the test included right atrium-right ventricular apex (RVA),right atrium-right ventricular septal(RVS),and right atrium-biventricular (Bi-V) ; The pacing frequency was 180 times per minute.The results were measured before pacing and after 15 minutes when the pacing became stable in Color Doppler echocardiography,including left ventricular end-diastolic diameter (LVEDd),left ventricular ejection fraction (LVEF),interventricular mechanical delay (IVMD),interventricular septum and left ventricular posterior wall motion delay (SPWMD),and left ventricular 12-segment peak time standard deviation (Ts-SD).Results Right ventricular septal pacing mode:(1)Compared with parameter before pacing and RVA pacing,LVEDd,IVMD,SPWMD,and Ts-SD decreased and LVEF increased,and the difference was significant(P <0.05).(2)Compared with Bi-V pacing,LVEDd,IVMD,and SPWMDandTs-SD increased and LVEF decreased,and the difference was significant (P < 0.05).Conclusions Right ventricular septal pacing could improve ventricular synchrony and cardiac function partly,and the effect was better than right ventricular apical pacing but less than biventricular pacing,also could not be a alternative models of cardiac resynchronization therapy for heart failure.
8.Clinical research on the treatment of moderate bronchial asthma with inhaled inactivated-mycobacterium phlei

Objective To introduce inhaled inactivated-mycobacterium phlei on prevention and treatment of moderate bronchial asthma to observe the clinical effect. Method This study was a prospective and controlled study. The patients diagnosed with asthma in our out-patient from March 2009 to December 2010 were collected, who met the following conditions were included in the study: age≥ 14 years; met the criteria of moderate chronic persistent bronchial asthma in Global Initiative for Asthma (GINA) in 2008; suspended receiving systemic corticosteroids, Montelukast, ketotifen and other anti-inflammatory and anti-allergic drugs in one month; no significant respiratory tract infections; and other serious illnesses or abnormalities known.A total of 100 patients with asthma were selected, including 37 males and 63 females, age (32.11 ± 12.95 )years. The patients were randomly(random number) divided into two groups: A group(treatment group; 16males and 34 females, age 33.56 ± 14.23 years) and B group (control group; 21 males and 29 females,age 30.66 ± 11.50 years); 50 in each group. No significant difference was noted between the two groups on age and gender composition. The patients in A group were treated with inhaled inactivated-mycobacterium phlei F. U. 36 Injection 1.72 μg/mL × 2 that adding 3 mL normal saline, once a day for 5 days. The patients in B group were treated with salmeterol xinafoate and fluticasone propionate powder for inhalation (50/100 μg), twice daily for sustainable use. The patients in the two groups were observed for one month. During this course, the patients in the two groups could inhale the salbutamol sulphate aerosol as need to relieve symptoms. And the number of using was recorded. Pulmonary function test and asthma provocative test were carried out on the Day O, 6 and 31. ACT scores were measured before and after the treatment. Results On Day 6 and 31 after treatment, the negative conversion rates of asthma provocative test of the patients in A group were 82% and 78% respectively, B group were 84% and 90% respectively. Provocative test of the patients in the two groups were negative conversion significantly before and after treatment. There was no significant difference between the two groups by chi-square test (P > 0. 05 ). Completely random designed data was analyzed by analysis of variance. The analysis showed that the accumulated doses of methacholine of the patients in the two group increased significantly ( P < 0. 05 ), but no difference between the two groups.There was a improvement trend on forced expiratory volume in one second( FEV1 )of the patients in A group after treatment, but no difference. FEV1 of the patients in B group increased significantly higher ( P <0.05), which was significantly higher than A group on the 31th day (P <0. 05); Peak expiratory flow (PEF) of the patients in the two group increased significantly on Day 6 and 31 after treatment (P <0.05 ).On Day 31, B group was significantly higher than A group ( P < 0. 05 ); Scores of asthma control test (ACT)of the patients in the two group were significantly increased, and the number of using of salbutamol sulfate aerosol was significantly reduced (P <0.01 ). B group was obvious than group A (P <0.05 ). During treatment, there were only two adverse reaction cases of transient low fever; most obvious was on the third day.Conclusions Inhaled inactivated-mycobacterium phlei would inhibit the airway hyperresponsiveness of the patients with moderate bronchial asthma in short time, improve the symptoms, reduce the acute exacerbation, and reduce the use of rescue medication, which has the roles of prevention and treatment of moderate asthma in a certain period of time.
9.Verification of a molecular pathology detection method of sentinel lymph node in breast cancer patients:a comparison between RT-LAMP and GeneSearch

Objective To verify the reliability of our previously established reverse-transcription loop-mediated isother-mal amplification ( RT-LAMP) method for the detection of sentinel lymph nodes metastasis in breast cancer patients .Meth-ods Sentinel lymph nodes of breast cancer patients were analyzed by RT-LAMP and FDA-approved GeneSearch methods respectively, and the consistency of the two methods was assessed with a kappa concordance test.Results One hundred and thirty-four cases of sentinel lymph node samples were collected from seven hospitals in Beijing .Using the GeneSearch assay as the gold standard, the sensitivity, specificity and consistentcy of RT-LAMP were 96.2%(25/26),96.3%(104/108) and 96.3%(129/134), respectively.Statistical analysis showed that the two methods were consistent (Kappa=0.8857, P<0.001).Conclusion RT-LAMP is highly consistent with GeneSearch ,efficient,simple and inexpensive, promising a good prospect of application to intra-operative detection of sentinel lymph nodes metastasis for breast cancer patients.
10.Effects of a loading dose of clopidogrel hydrogen sulfate on hemorheology and neurological function in patients with acute progressive cerebral infarction

Objective:To investigate the effects of a loading dose of clopidogrel hydrogen sulfate on hemorheology and neurological function in patients with acute progressive cerebral infarction.Methods:A total of 110 patients with acute progressive cerebral infarction who received treatment between January 2018 and January 2019 in the Affiliated Hospital of Shaoxing University, China were included in this study. They were randomly assigned to receive either conventional treatment (control group, n = 55) or treatment with a loading dose of clopidogrel hydrogen sulfate based on conventional treatment (study group, n = 55) for 2 weeks. Hemorheological parameters, National Institute Health of Stroke Scale (NIHSS) scores and adverse reactions were compared between the study and control groups. Results:After treatment, the levels of hemorheological parameters in both groups were obviously decreased. Plasma viscosity, high-shear whole blood viscosity, low-shear whole blood viscosity, fibrinogen level and hematocrit in the study group were (1.01± 0.37) mPa/s, (4.23 ± 0.35) mPa/s, (8.36 ± 1.64) mPa/s, (1.23 ± 0.28) g/L, (40.08 ± 5.04) %, respectively, which were significantly lower than those in the control group [(1.84 ± 0.52) mPa/s, (5.44 ± 0.39) mPa/s, (10.54 ± 1.79) mPa/s, (2.30 ± 0.37) g/L, (44.36 ± 5.12) %, t = 5.485, 8.594, 9.523, 7.789, 11.236, P = 0.019, 0.006, 0.004, 0.007, 0.001]. After treatment, NIHSS scores in the study group were significantly lower than those in the control group [(3.11 ± 1.17) points vs. (6.43 ± 2.25) points, t = 10.416, P = 0.003). There was no significant difference in the incidence of adverse reactions between study and control groups [5.45% (3/55) vs. 9.09% (5/55), χ2 = 0.539, P = 0.463). Conclusion:The loading dose of clopidogrel hydrogen sulfate is highly effective in the treatment of acute progressive cerebral infarction and it can greatly improve hemorheological parameters and neurological function.
