1.Observation of the curative effects of Edaravone for treatment on Parkinson disease

Objective To explore the curative effects of Edaravone for treatment on Parkinson disease(PD).Methods 30 patients with PD were classified into two groups:18 patients with early PD(courses of disease≤3.5 years) and 12 patients with late PD(courses of disease≥4 years).Edaravone was administered intravenous,30 mg twice daily for 14 d.Then,the drug was discontinued and a follow-up examination carried-out for 3 months.The clinical disability was assessed by using unified PD rating scale(UPDRS) at baseline and 14 d,1 month and 3 months after therapy.The adverse reaction was also observed during the course of treatment.Results In the early PD group,compared to before therapy,the scores of Mentation Behavior and Mood had significantly decreased for 14 d,1 month and 3 months after Edaravone therapy,the score of activities of Daily Living and Motor examination had significantly decreased(P

Objective To investigate the effect of aprotinin on nuclear factor-?B p65 expression in brain regions around the hematoma of intracerebral hamorrage (ICH)in rats. Methods ICH models were induced utilizing the local injection of collagenase into the basal ganglia with sterile technique and divided into ICH group, aprotinin group(ATG) and Pyrrolidine dithiocarbamate(PDTC) group. The expression levels of NF-?B p65 protein and mRNA were detected by immunohisochemistry or RT-PCR at 6 h,24 h,72 h,7 d after injection. Results The amount of NF-?B positive cells increased greatly at 24 h following ICH, peak at 72 h, then decreased in each group. There was no markedly different from ICH group to ATG group, but PDTC group were singnificantly decreased after ICH 24 h~7 d than groups ICH and ATG(P
3.Arthroscopic Reconstruction of Anterior Cruciate Ligament with Hamstring Tendons and Button-Suture Plate

Objective To study the efficacy of arthroscopic reconstruction of anterior cruciate ligament(ACL)with hamstring tendons and button-suture plate.Methods Thirty-two cases of ACL rupture received arthroscopic reconstruction with hamstring tendons and button-suture plate.The Lachman test and Pivot shift test were performed before and after operation to assess the stability of the knee joint.The knee function was evaluated according to Lysholm rating scale.Regular MRI,anterioposterior and lateral films of knee joint,patella axial radiography were conducted before the operation,to exclude the possibility of osteal avulsion at both ends of ACL.Results The 32 cases were followed for 3.5-29 months,in which 25 cases were more than 12 months.The knee stabilities of all cases recovered.The preoperative Lachman test results were positive in 32 cases,and the Pivot shift test results were positive in 28 cases.Lysholm rating score was 51.8?5.6.The postoperative Lachman tests result were negative in 30 cases,and weakly positive in 2 cases.The Pivot test showed negative results in all the cases.The Lysholm rating score was increased to 90.7?2.5 after the operation.Three cases experienced joint effusion and were treated with puncture aspiration.Conclusions The short-term clinical effect of arthroscopic reconstruction of ACL with hamstring tendons and suture plate is favorable.
4.THE RELATIONSHIP BETWEEN THE RATIO OF ZINC TO COPPER IN THE HAIR AND ARTERIOSCLEROSIS

The content of zinc and copper in the hair of 321 persons was detected by an atomic absorption spectropolarimeter. The subjects included young healthy group (35 men, 30 women), middle aged healthy group (72 men, 18 women), arteriosclerosis group (81 men, 20 women), cerebral embolism group (23 men, 8 women) and its control group (23 men, 8 women). The ratio of zinc to copper in the hair of the persons suffered from cerebral embolism was obviously higher than that of healthy controls. By comparing the function of the susceptible factors in cerebral embolism with Feisher's determination analysis method, diastolic pressure was found to be 75.02 percent, the ratio of zinc to copper 16.32 percent. The diagnostic coincidence was found to be above 82 percent.
5.Study of growth, invasion and metastasis on ovarian epithelial cancer cell line with CCL18 over-expression by mediated in vitro

Objective To investigate the effect of growth,invasion and metastasis on ovarian cancer cell line SKOV3 with chemokine (C-C motif) ligand 18 (CCL18) over-expression by mediated in vitro.Methods The restructuring plasmid of CCL18 expression was constructed and SKOV3 cells was transfected with plasmid DNA in vitro.The growth curve,cell cycle,cell migration,invasion and adhesion capacity in SKOV3-CCL18 cells and control cells were detected by methyl thiazolyl tetrazolium (MTT) assay,flow cytometry,transwell chamber,migration invasion and fibronectin adhesion method,respectively.Results (1) CCL18-pEGFP-N1 plasmid was successfully constructed and transmitted to the SKOV3 cells,the stable growth of the subculture SKOV3-CCL18 cells was screening and completed.(2) Compared with SKOV3-CCL18 cells and SKOV3 or SKOV3-pEGFP-N1 cells,there was not differences statistically significant in growth curve (all P > 0.05) ; but the rate of the SKOV3-CCL18 cells in a proliferative field (S + G2 + M) was significantly higher than that in SKOV3 or SKOV3-pEGFP-N1 cells (32.80% versus 27.06%,32.80% versus 26.98%,respectively; all P < 0.05).(3) The invasion,migration and adhesion capacity of SKOV3-CCL18 cells (being 0.49 ± 0.18,1.16 ± 0.25 and 0.39 ± 0.10,respectively) in vitro were significantly higher than those in SKOV3 (being 0.23 ± 0.13,0.36 ± 0.10 and 0.16 ± 0.03,respectively) or SKOV3-pEGFP-N 1 cells (being 0.19 ± 0.05,0.38 ± 0.23 and 0.13 ± 0.11,respectively; all P < 0.05).Conclusion The CCL18 over-expression in epithelial ovarian cancer SKOV3 cells could lead to strengthen ability of invasion,migration and adhesion in vitro.
6.Analysis on clinical and CT imageological features in 276 cases of intracranial tuberculosis

Objective To investigate the clinical and head CT imageological features features in 276 cases of intracranial tubercu‐losis .Methods The clinical and CT features in 276 inpatients with confirmed intracranial tuberculosis were analyzed .Results These 276 cases of intracranial tuberculosis all were complicated by pulmonary tuberculosis ,among them blood‐borne type accoun‐ted for 49 .6% ; young and middle‐aged cases were predominant (72 .5% ) ; the majority manifested by the symptoms of fever (70 .0% ) and headache(66 .2% ) ;the summer and autumn had the higher incidence rate ,which were 167 cases(80 .7% ) .The posi‐tive detection rate of head CT plain scanning was 63 .4% ,which of enhanced scanning was 98 .6% .Among them ,236(85 .5% ) cases were tuberculous meningitis lesions ,158 cases(57 .2% ) were tuberculous encephalitis ,144 cases (52 .2% ) were tuberculoma and 133 cases (48 .2% ) were tuberculous vasculitis .Conclusion The clinical menifestations of intracranial tuberculosis are atypical ,but the head CT plain scannig displays multiple abnormalities .Moreover ,the enhanced scanning increases the positive detection rate . The lesions of meninges ,brain vessels and brain parenchyma often occur together .Therefore ,the diagnosis basis should be combined with clinic ,laboratory tests and head CT ,especially the clinical application of head CT enhanced scanning should be paid attention to .
7.Effects of 1-3-n-Butylphthalide on the blood-brain barrier following whole brain irradiation in rats

ObjectiveTo investigate the effects of 1-3-n-Butylphthalide on the blood-brain barrier (BBB) following whole brain irradiation in rats.Methods144 male Sprague Dawley rats were randomly divided into sham-irradiation group,irradiation group,1-3-n-Butylphthalide group,and irradiation plus 1-3-n-Butylphthalide group.Whole-brain irradiation was given as a single-dose of 10 Gy using 4 MV X-ray.The rats were injected intraperitoneally with 1-3-n-Butylphthalide at 0.3 mg/kg,1.0 mg/kg,3.0 mg/kg once per day.The changes of the BBB were assessed by Evans blue (EB) assay.The expression of vascular endothelial growth factor (VEGF) in the brain tissue was determined by immunohistochemistry. The circulating endothelial cells (CECs) isolated from right ventricular blood were counted.MRI was evaluated with the T1-weighted images,T2-weighted images and MRI enhancement images induced by Gd-DTPA.The data were compared among the groups through Student-Newman-Keuls test.ResultsCompared with the sham-irradiation group,the EB content,the expression of VEGF in the brain tissue and the CECs were significantly increased in the irradiation group (2.81∶ 7.82,P =0.002;5.83∶ 10.26,P=0.003;3.16∶6.14,P =0.002).The signal intensity of T1-weighted images was significantly decreased while T2-weighted images and the enhancement rate significantly increased in the irradiation group (P =0.004 -0.018 ).Compared with irradiation group,the EB content,the expression of VEGF and the CECs were decreased significantly in the irradiation plus 1-3-n-Butylphthalide group ( 7.80∶ 3.86,P =0.007 ; 10.83 ∶ 5.26,P =0.008 ;6.36∶ 3.64,P =0.009 ).However,the changes in the MRI were significantly attenuated ( P =0.008-0.026,and 0.006 -0.038,respectively).Conclusions Following whole brain irradiation,1-3-n-Butylphthalide can decrease the permeability of the BBB in rats via decreasing VEGF expression and decreasing the CECs.
8.Protective effect of DL-3-n-Butylphthalide on radiation injury of rat brain tissue

Objective To investigate the protective effect and its mechanism of DL-3-n-Butylphthalide on the brain damage in rats following whole brain irradiation.Methods A total of 120 male Sprague Dawley rats were randomly divided into sham-irradiation group,irradiatien group and DL-3-n-Butylphthalide group.The model of whole-brain irradiatien was established by exposuring rat brain to 4 MeV X-rays with a single-dose of 10 Gy.The rats were intraperitoneally injected with DL-3-n-Butylphthalide at the dosages of 0.3,1.0,and 3.0 mg/kg once a day.The contents of malondialdchyde and super oxide dismutase activity were measured,while the expressions of apoptosis-associated genes and the ultrastructural changes in hippocampus were examined by immunohistnchemisty staining and electron microscope,respectively.Results After irradiation,the content of malondialdehyde and the expression of apoptosis gene bax in rat brain tissue increased while the activity of super oxide dismutase(SOD) and the expression of anti-apoptosis gene bcl-2 decreased.Apoptosis was also observed in the neurons of hippocampus CA1.Compared with irradiation group,the content of malondialdehyde and the expression of bax gene in the DL-3-n-Butylphthalide group wen significantly reduced ( t =-3.89--1.96,2.72-3.48,P < 0.05 ),while the activity of SOD and bcl-2 gene were significantly elevated ( t =2.94-3.76,-3.18--2.08,P < 0.05),and the injury degree of neuron structure in the DL-3-n-Butylphthalide group was slighter than that in the irradiation group.Conclusions DL-3-n-Butylphthalide executes protective effects in a dose-dependent manner againest the radiation injury in rats brain by reducing the induction of malondialdehyde,raising the activity of SOD and inhibiting the generation of apoptosis.
9.Roles of Toll-like receptor 4 in cerebral ischemia

In recent years,studies have shown that inflammation is an important factor in secondaryischemic injury.The exploration of the related mechanisms of the occurrence of inflammation has become ahot spot in the field of cerebral ischemia.Toll-like receptor 4 (TLR4) signaling pathway is one of theclassical inflammatory pathways.Studies have shown that TLR4 is involved in the early inflammatoryresponse after cerebral ischemic injury and the late nerve repair.This article reviews the research progress ofthis signaling pathway in cerebral ischemia injury,so as to seek a therapeutic target against inflammatoryinjury caused by cerebral ischemia through the analysis of its potential research direction.
10.Changes of plasma fibrinogen level among acute ischemic stroke subtypes according to TOAST criteria and effects of Songling Xuemaikang.

OBJECTIVETo investigate the changes of plasma fibrinogen level among acute ischemic stroke (ACI) subtypes according to Trial of Org10172 in Acute Stroke Treatment (TOAST) criteria and effects of Songling Xuemaikang.
METHODThe 160 patients with acute ischemic stroke were divided into two groups randomly: treatment group 85 cases (Songling Xuemaikang + Shuxuetong + Aspirin enterie coated tablets), control group 75 cases (Shuxuetong + Aspirin enterie ccoated tablets). The plasma fibrinogen was detected before and after treatment.
RESULTCompared with OC subtype, Fbg was higher in LAA, CE and SAO subtypes (P < 0.05). Compared with UE subtype, Fbg was higher in LAA, CE and SAO subtypes (P < 0.05). There was a significantly difference between LAA and SAO (P < 0.05). In LAA, SAO, CE of treatment group,the Fbg level were lowered significantly at the 15th day compared with pretherapy (P < 0.05). There was a significantly difference of Fbg between treatment group and control group In LAA, SAO and CE subtypes at the 15th day (P < 0.05).
CONCLUSIONFbg produces a marked effect at the pathomechanism of LAA, SAO and CE subtypes. Songling Xuemaikang can depress the plasma fibrinogen level of ACI, and be an effective adjunctive therapy on ACI.
