2.The protective effects of melatonin on acute ischemia/reperfusion induced myocardium damage in rats in vivo

Aim To investigate the protective effects of MLT(melatonin) on acute ischemia-reperfusion induced myocardium damage in rats in vivo. Methods 36 anesthetized rats were separated randomly into 3 groups: ① Control group (n=12), without LAD (left anterior descending coronary artery) ligation and MLT; ② I/R(ischemia reperfusion) group(n=12), LAD was ligated for 10 min and reperfused for 15 min without MLT; ③ I/R+MLT group(n=12): MLT(10 mg?kg~(-1)) was in jected via peritonium 10 minutes before LAD ligation and reperfusion. ECG and haemodynamics were continuously monitored and recorded throughout the whole process, MDA(malondialdehyde, lipid peroxidation product, an index of myocardium damage) and SOD( superoxide dismutase ) activity were tested in the injuried myocardium. The hearts tissue of each group was also examinated by electron microscopy.Result The levels of MDA were significantly higher while the SOD activities were lower in I/R group compared with I/R+MLT and control group(P0.05). 3.Effect of Combined Hypoxia and High Fat Diet on eNOS/NO of Myocardium in Experimental Rats

Objective: To investigate the effect of combined hypoxia and high fat diet (HFD) on endothelial nitric oxide synthase (eNOS)/nitric oxide (NO) of myocardium in experimental rats with its possible mechanisms.
Methods: A total of 60 male SD rats were randomly divided into 3 groups, n=20 in each group. Control group, the rats were fed by normal diet with normal oxygen condition. Hypoxia group, the rats were fed by normal diet with simulated 5000m altitude oxygen condition. Combined hypoxia and HFD (H+HFD) group, the rats were fed by HFD and simulated 5000m altitude oxygen condition. All animals were treated for 4 weeks and peripheral blood and myocardium specimen were collected. Hemoglobin was examined by automatic blood cell analyzer, plasma malondialdehyde (MDA) was measured by TBA method, superoxide dismutase (SOD) activity was detected by WST-1 method, mRNA and protein expressions of eNOS were examined by real time PCR and Western blot analysis respectively, the myocardium nitrates and nitrites (NOx) was measured by nitrate reductase method.
Results: Compared with Control group, Hypoxia group and H+HFD group had increased mRNA and protein expressions of eNOS, H+HFD group had lower NOx levels than the other 2 groups P<0.05. Compared with Hypoxia group, H+HFD group showed obviously increased total cholesterol, LDL-cholesterol and decreased SOD activity, diseased MDA level P<0.05.
Conclusion: Upon hypoxia alone, H+HFD may further reduce NOx level of myocardium, it implies aggravated chronic hypoxia impairment, which might be related to dyslipidemia and lack of anti-oxidative ability in experimental rats.
4.Effects of non-wounded ischemic preconditioning on ischemia-reperfusion injury in isolated rat hearts

AIM: To observe the protective effect of non-wounded ischemic preconditioning on ischemic/reperfusion injury in isolated rat hearts. METHODS: 25 male SD rats, weighting (250?30) g, were randomly divided into three groups: control group (C, n= 8), anoxia/reoxygenation group (A, n= 8) and non-wounded legs ischemic preconditioning group (N-WIP, n= 9).Hearts were isolated from rats and perfused on a Langendorff apparatus with a normal Krebs-Henseleit buffer (saturation 95% O 2+5% CO 2) at a constant pressure (8.33 kPa) and temperature (37 ℃) in C group ; Following 15 min equilibration, hearts were subjected to 15 min of global ischemia and 15 min reperfusion (37℃) in A group ; Rats were subjected to non-wounded leg repeated-brief ischemic preconditioning, and then treated in procedure similar to A group in N-WIP group.The activities of superoxide dismutase (SOD) and Ca 2+ -Mg 2+ -ATPase, malondialdehyde (MDA) content of efflux from coronary vessel and myocardium, myocardium monophasic action potential and contractile force were measured before ischemia, 15 minutes after ischemia and 5, 15 minutes after reperfusion. RESULTS: Compared with A group, non-wounded legs ischemic preconditioning reduced the incidence of reperfusion arrhythmias ( P
