1.The effect of batroxobin and Buyanghuanwu Tang on the serum levels of IL-8 and leptin in patients with acute cerebral infarction

Objective To explore the effect of hatroxobin and Buyanghuanwu Tang on the serum]evels of in-terleukin-8 (IL-8) and leptin in patients with acute cerebral infarction (ACI). Methods 85 cases with cerebral in-farction were randomly.divided into control group of 43 cases and treatment group of 42 cases. 85 cases were given routine treatment, the treatment group were given hatroxobin ,20BU and Buyanghuanwu Tang, 14d. The serum levels of IL-8 and leptin were investigated before treatment and 14d after treatment. Results After treatment,the serum levels of IL-8 and leptin in the treatment group were improved than that of control group (P < 0.05). Conclusion The treatment of batroxobin and Buyanghuanwu Tang could improve the production of IL-8 and leptin and interrupt the Ieu-kecyte infiltrating ischemic region.
2.Effect of Chaihu Jiebiao Chongji in treatment of acute upper respiratory tract infection of 169 patients

Objective To explore the clinical effect of Chaihu Jiebiao Chongji in treatment of acute upper respiratory tract infection.Methods To analyse curative effect of Chaihu Jiebiao Chongji(ten grams of chaihu,guizhi,lianqiao,baishao and huangqin and thirty grams of yinhua) in treatment of acute upper respiratory tract infection and take empirical study of relieving fever and anti-inflammatory action in 169 cases.Results Cured of treatment group 44 cases , obvious effective of treatment group 62 cases, effective of treatment group 48 cases, ineffective of treatment group 15 cases,total effective rate 91.12%.Conclusion The effect of Chaihu Jiebiac Chongji in treatment of upper respiratory tract infection is satisfactory.
3.Open reduction fixation for treating ankle joint fractures:a wire anchor in the repair of triangular ligament injury

BACKGROUND:Ankle joint fracture combined with triangular ligament injury is a common type of injury, and is easy to have an impact on the anatomy of the ankle joint, leading to the occurrence of joint instability; the treatment is extremely easy to lead to a variety of complications. For injured triangular ligament, it is controversial whether it is necessary to repair in the clinic. OBJECTIVE:To investigate the effect of a wire anchor for repairing triangular ligament in ankle joint fracture fixation. METHODS:The clinical data of 36 cases of ankle fractures combined with triangular ligament injury, who were treated in the Department of Orthopedics of the Seventh People’s Hospital of Zibo City from May 2013 to May 2014, were retrospectively analyzed. The ankle fractures combined with triangular ligament injury were treated with open reduction plate fixation. The patients were folowed up for 12 months. The recovery of ankle joint function was evaluated with the American Orthopaedic Foot & Ankle Society (AOFAS) ankle and foot function score. Gravity stress X-ray examination was performed before and after treatment. Space of medial maleolus was measured. Adverse events were observed. RESULTS AND CONCLUSION:The patients were folowed up 3, 6 and 12 months after treatment. AOFAS score of the ankle joint was increased and significantly higher at 12 months than that before surgery (P< 0.05). Gravity stress X-ray examination revealed that the space of medial maleolus of the affected side was smaler at 12 months than that before treatment (P < 0.05). No injuries to nerves and blood vessels were found during surgery, and the incision was healed in the first stage, and no infection was observed. Adverse events such as fixator breakage and loosening were not visible. However, partial function of the ankle joint was limited after repair in triangular ligament injury. Results confirmed that open reduction and internal fixation combined with a wire anchor obtained good effects in the repair of ankle joint fracture with triangular ligament injury.
4.Total knee replacement for osteoarthritis of knee joint

[Objective]To estimate the results of the total knee replacement for osteoarthritis knee joint.[Method]Eighty-one patients(95 knees)with osteoarthritis of knee joint were selected for total knee replacement.The average age of the patients was 65.5 years with a range of 50~82 years,which involved 10 males and 71 females.Left knee was in 35 cases and right knee was in 26 cases.Bilateral knee replacement one time was 17 cases.The posterior crnciate ligament retaining prosthesis was 18 knees,posterior stabilizing prosthesis was 62 knees and mobile bearing prosthesis was 15 knees.[Result]All the patients were available at the follow-up,with a mean of 42 months(14~108 months).According to HSS system,80 knees(84.21%)had an excellent results;10 knees(10.53%),a good result;3 knees(2.86%),a fair result;and 2 knees(1.9%),poor result.The satisfactory rate was 95.23%.[Conclusion]Total knee replacement can decrease knee pain and improve knee function.The prosthesis choice,surgical technique and postoperative rehabilitation are key factors to the operation results.
5.Effect of patella resurfacing versus nonresurfacing in total knee arthroplasty on anterior knee pain

[Objective]To compare retrospectively the clinical outcome in total knee arthroplasty with and without patellar resurfacing.[Method]From October 1994 to October 2004,125 patients(148 knees)with osteoarthritis were enrolled in the clinical trial of total knee arthroplasty,patients received resurfacing(84 knees)or retention patellar(64 knees).Evaluation was done from preoperatively to final followed-up postoperatively.The patients were scored using American HSS Score for knee and the Feller Score for patalla.The data were analyzed using SPSS software.[Result]In resurfacing patella group,the preoperative and final follow-up HSS scores of patients were(39.6?9.8)and(90.9?8.2)points respectively,scores of patailar were(14.4?6.4)and(25.2?4.8),scores of anterior knee pain were(4.6?3.9)and(10.6?4.1).In nonresurfacing patella group,the preoperative and final follow-up HSS scores of patients were(38.8?9.8)and(90.2?8.9)points respectively,scores of patallar were(14.2?6.2)and(25.1?4.8),scores of anterior knee pain were(4.8?3.8)and(10.3?4.1).There were significant differences in both groups preoperatively and postoperatively in anterior knee pain score,but no significant difference between the two groups in anterior knee pain score.[Conclusion]Whether the patellar replaced or not,the most importance for avoiding anterior knee pain postoperatively is the reasonable selection of patient and correct operation.
6.Identification of Fengliaoxing Fengshi Dieda Medicinal Wines in Fake by GC and TLC

Objective To establish a method for the fake indentification of Fengliaoxing Fengshi Dieda medicinal wines by TLC and GC. Methods The column was Supelco SE-30 (30 m?0.25 mm?0.25 ?m). The flow rate was 1.4 mL/min, and gas was N2. The inlets temperature was 210 ℃;the detectors temperature was 300 ℃;the column temperature use program rising temperature. Results TLC can identify whether ephedra and erycibe were contained in the medicinal wines. GC can identify whether diclofenac sodium was contained in the medicinal wines. Conclusion The method is accurate and reproducible, and can be used for the identification of Fengliaoxing Fengshi Dieda medicinal wines.
7.Local drug injections for analgesia after total knee arthroplasty

0.05).[Conclusion]Periarticular drug injection during TKA and intra-articular drug injection after operation can relieve pain early after TKA,improve the range of motion of the knee and rehabilitation,no further side effect has been noted.
8.Clinical application of the GIEBEL system in high tibial osteotomy

[Objective]To discuss the results of the GIEBEL system in high tibial osteotomy to treat varus knee.[Method]Nineteen patients with medium varus knee were selected to be treated with osteotomy.Preoperatively,the mean varus angle was 12.29?,without deformity in other department of the knee joint.The main symptom was the pain in medial department.Accurate high tibial osteotomy with GIEBEL system fixation was performed in 31 knees of these patients.Weight-bearing extremity X-ray testing,varus angle,mechanical axle and anatomical axle were marked preoperatively and postoperatively.Joint space discrepancy between lateral and medial joint,the range of motion,Lysholm assessment and objective satisfactory survey were also performed to determine the outcome of the treatment.[Result]The mean follow-up of nineteen patients was 22 months.During this period,one case of temporary common fibular nerve paralysis occurred and healed at several days.All of the osteomy interface achieved bony healing.The corrected abnormality was 12.32?.The mechanism axle of the extremity maintained as the level just after operation during the follow-up and no degeneration of the joint space aggravated.The excellent and good rate of the treatment was 89.5%,and 84.2% of the patients expressed satisfactory for the operation.No difference of the range of motion was found before and after operation,but the Lysholm score,joint space discrepancy between lateral and medial joint and varus angle were improved after the osteotomy.[Conclusion]The GIEBEL system can fixate the bony interface of the high tibial osteotomy effectively,and possess the advantages of minimal invasion,stronger intensity and higher healing rate.
9.Bone marrow stem cells co-transfected with transforming growth factor beta 3 and bone morphogenetic protein 2

BACKGROUND: Bone morphogenetic protein 2 and transforming growth factor β are important factors in bone regeneration, increasing the expressions of bone morphogenetic protein 2 and transforming growth factor β can promote the osteogenic differentiation of bone marrow mesenchymal stem cells. OBJECTIVE: To construct the lentivirus vector carrying bone morphogenetic protein 2 and transforming growth factor β3, and to observe the expression of lentivirus vector in bone marrow mesenchymal stem cells. METHODS: The recombinant lentiviral vectors carrying transforming growth factor β3, bone morphogenetic protein 2 and green fluorescent protein were constructed with recombinant lentiviral technology, and then the recombinant lentiviral vectors were used to transfect the passage 3 rabbit bone marrow mesenchymal stem cells in vitro cultured (transfection group). The bone marrow mesenchymal stem cells transfected with single gene lentivirals (single gene transfection group) carrying transforming growth factor β3 and bone morphogenetic protein 2 or single lentivirals were as control (control group). At 1 week after trasfection, the total RNA and protein were extracted from each group for detection. RESULTS AND CONCLUSION: The green fluorescence bone marrow mesenchymal stem cells transfected with transforming growth factor β3 and bone morphogenetic protein 2 gene for 3 days could be observed under fluorescence microscope, and the transfection efficiency was over 90%. Reverse transcription-PCR and Western blot results showed the mRNA and protein expressions of transforming growth factor β3 and bone morphogenetic protein 2 in the transfection group were higher than those in the single gene transfection group and the control group. The results indicate that lentivirus can successful y transfect transforming growth factor β3 and bone morphogenetic protein 2 into the bone marrow msenchymal stem cells and achieve its high expression, and these two genes have the synergistic effect of promoting expression.
10.Construction and identification of pIRES2-EGFP-hBMP-2 bicistronic eukaryotic expression vector

BACKGROUND: Bone morphogenetic protein (BMP) and its derived scaffold or vector here been widely used in experiment and gradually applied in the clinic. However, the tracing method challenges the therapeutic effect, The discovery and application of enhanced green fluorescent protein (EGFP) can solve this problem.OBJECTIVE: To construct the bicistronic eukaryotic vector pIRES2-EGFP-hBMP-2 and to observe its expression in human embryo kidney (HEK) 293 cells.METHODS: hMBP-2 gene was extracted from human osteosarcoma by RT-PCR and inserted into PMD18-T vector. Following the DNA sequence verification, it was then sub-cloned into the eukaryotic vector pIRES2-EGFP. After restriction enzyme analysis the pIRES2-EGFP-BMP-2 was transfected into HEK 293 cells. Then its expression was observed using fluorescence microscopy and Western blot.RESULTS AND CONCLUSION: hBMP-2 gene was amplified and the eukaryotic vector plRES2-EGFP-hBMP-2 was constructed successfully. At 48 hours after tranfection of pIRES2-EGFP-hBMP-2 into HEK 239 cells, approximately 30% of cells emitted green fluorescence under a fluorescence microscope. Western blot results demonstrated that there was a specific BMP strap at Mr46×10s side, which indicated that the target gene could be expressed in constructed pIRES2-EGFP-hBMP-2 vector.
