1.The Clinical Epidemiological Analysis of 1433 Cases of Type 2 Diabetes and Their Diabetic Complications

Objective To investigate the frequency of type 2 diabetes and diabetic complications. Methods The clinical and laboratory data of 1433 cases of patients with type 2 diabetes mellitus were retrospectively analyzed. Results The frequency of diabetic complications of patients with type 2 diabetes mellitus were 88.28%(1626/1433). The rates of complicated hypertension, coronary heart disease, peripheral neuritis, diabetic renopathy, diabetic retinopathy, hyperlipoidemia, urethritis, diabetic acidosis, pneumonia, fatty liver, diabetic foot gangrene, cholecystitis, skin infection, brain vessel disease and vegetative nerve disease were 46.9%, 33.2%, 29.7%, 25.4%,22.0%,13.0%,7.5%,4.2%,4.0%,4.0%,3.5%,3.5%,3.3%,2.9%,and 2.0%, respectively. Conclusion The frequency of diabetic complications of patients with type 2 diabetes mellitus were much high, and the distrubution of diabetic complications was different in different ages, course of disease and syndromes.
2.Observation of clinical efficacy on prescriptions of traditional Chinese medicine in 60 cases of type 2 diabetes

Objective To explore the treatment perscription with the method of integrated traditional and western medicine in type 2 diabetes.Methods With the methods of differential diagnosis of diseases in western medicine and differential symptoms and signs in traditional Chinese medicine(TCM),the diseases were separated into different patterns of syndrome and treated with propotional prescription of TCM.The curative effect was evaluated with the scores in the integration of symptoms and the changes on the levels of blood glucose and glycated-hemoglobin.Results Clinical curative effects were achieved using the prescription of TCM and the simple western medicines.The curative effects were much better on lowering integration of symptoms and the level of pre-meal blood glucose with the prescription of TCM than those of the simple western medicine.Conclusion The clinical curative effects are significant on relieving the clinical symptoms of patients with the prescription of TCM than those of the simple western medicine.
3.The clinical analysis of primary central nervous system lymphoma in 23 patients

ObjectiveTo explore the clinical characteristics and the relationship between rational therapy and prognosis in primary central nervous system lymphoma (PCNSL).MethodsClinical data of 23 patients pathologically confirmed as PCNSL and treated in our hospital from January 2005 to December 2007 were collected and analyzed retrospectively.SPSS 13.0 statistical analysis software was used to analyze the correlation between therapy,clinical characteristics and prognosis.Among the 23 patient,10 were male and 13 were female,with a median age of 50 (2-75) years old.Eighteen patients were undertaken lumbar puncture,and tumor cells in cerebral spinal fluid (CSF) was found in 4 patients.Tumorectomy was performed in 8 patient,while 15 patients biopsy.Among them,4 accepted whole brain irradiation,6 accepted associated chemotherapy based on a high-dose of methotrexate (MTX)and 13 accepted radiotherapy combined with chemotherapy.Results Kaplan-Meier analysis showed that in this serial patients,the median survival time was 45.0 months and the 3-year survival rate was 56.5%.Log-Rank test revealed that the whole survival time and the non-progression survival time of radiotherapy combined with chemotherapy (43.8 and 30.0 months,respectively) and high-dose MTX chemotherapy ( 39.7 and 29.7 months,respectively )were much longer than those of radiotherapy alone (25.7 and 19.8 months,respectively,all P < 0.05 ).Log-Rank test showed no significant difference between high-dose MTX chemotherapy alone and radiotherapy combined with chemotherapy (P >0.05 ),but the whole survival time was much shorter in high-dose MTX chemotherapy ( P < 0.05 ).ConclusionsThe detection of tumor cells in CSF can confirm the diagnosis,but pathological biopsy is the gold standard.The efficacy of associated chemotherapy based on a high-dose of MTX combined with radiotherapy is much better than radiotherapy or chemotherapy alone.
4.Effect of psoralen on TopoIIα expression in breast cancer stem cells

Objective To explore the effect of psoralen on topoisomerase IIα(TopoIIα) expression of breast cancer stem cells.Methods CD44+CD24-/low breast cancer stem cells were sorted from MCF-7/ADR by magnetic-activated cell sorting(MACS).We observed the growth characteristics of these stem cells through optical microscope and detected the growth-inhibitory effects of psoralen on breast cancer stem cells by CCK-8 assay and IC50 of adriamycin and adriamycin combined with psoralen to calculate the reversal index.The mRNA and protein expressions of Topo IIα were detected using RT-PCR and Western blot, respectively.Results Under the optical microscope, breast cancer stem cells presented spheres.IC10 and IC20 of psoralen on breast cancer stem cells were (6.77±0.23)μg/mL and (10.36±0.21)μg/mL.IC50 of adriamycin and adriamycin combined with psoralen on breast cancer stem cells was (90.03±3.56)μg/mL and (21.47±0.82)μg/mL, the reversal index was 4.19.Psoralen significantly raised the expressions of Topo Ⅱα at mRNA and protein levels.Conclusion Psoralen reversed the resistance of adriamycin by increasing the gene and protein expressions of breast cancer stem cells Topo Ⅱα and the drug targets.
5.Psoralen reverses glutathione-S-transferaseπ-mediated multidrug resistance in breast cancer stem cells

BACKGROUND:Breast cancer stem cells not only lead to theoccurrence of breast cancer, but also may cause breast cancer metastasis and recurrence. The relationship between stem cells and cell resistance is also gaining increasing attentions, and the focus on the stem cell treatment may result in unexpected results.OBJECTIVE:To explore the reversal effect of psoralen on glutathione-S-transferase π (GST-π) in human breast cancer MCF-7/ADR cells and its mechanism.METHODS:MCF-7/ADR cells were cultured and enriched in serum-free medium to obtain breast cancer stem cells.RT-PCR and western blot were used to detect the expression of GST-π at the levels of gene and protein in the MCF-7/ADR cells after treatment with 0, 4, 8, 12, 16 mg/L psoralen. To observe the activation of nuclear factor-κB,western blot was used. The expression of GST-π was detected by RT-PCR in 18 μmol/L SN50 group and 8 mg/L psoralen group. Cell counting kit-8 assay was used to detect the effect of doxorubicin on cell proliferation.RESULTS AND CONCLUSION:Compared with the control group, psoralen reduced the expression of GST-π at the mRNA and protein levels, and significantly inhibited the activation of nuclear factor-κB. It was suggested that psoralen could reverse the multidrug resistance of human breast cancer MCF-7/ADR stem cells by decreasing the expression level of GST-π. The mechanism may be achieved by inhibiting the nuclear factor-κB signal pathway.
6.Abdominal electroacupuncture for post-stroke constipation:a randomized controlled trial

Objective:To observe the effect of abdominal electroacupuncture(EA)on intestinal function in patients with constipation after stroke. Methods:A total of 100 patients with post-stroke constipation were divided into an observation group and a control group by the random number table method,with 50 cases in each group.The control group was treated with oral lactulose,10 mL each time,3 times a day,for 2 consecutive weeks.The observation group was treated with additional abdominal EA at Tianshu(ST25),Fushe(SP13),and Daheng(SP15)on the basis of oral lactulose,30 min for needle retention each time,once a day,for 2 consecutive weeks.The total effective rate after treatment,constipation symptom score,the score of patient assessment of constipation quality of life scale(PAC-QOL),serum motilin(MTL)level,and the score of mini-mental state examination(MMSE)were compared between the two groups. Results:After 2 weeks of treatment,the total effective rate was 93.5%in the observation group and 76.1%in the control group,and the difference between the two groups was statistically significant(P<0.05).The constipation symptom score,PAC-QOL score,MMSE score,and MTL level changed significantly after treatment in both groups(P<0.05).After treatment,the constipation symptom score and PAC-QOL score in the observation group were lower than those in the control group,while the MMSE score and MLT level were higher than those in the control group,and the differences between the two groups were statistically significant(P<0.05). Conclusion:On the basis of oral lactulose,EA at abdominal points can significantly improve the intestinal and cognitive function of stroke patients simultaneously.
7.Research progress on physical pain and social pain in non-suicidal self-injury and their influencing factors

Non-suicidal self-injury is a prevalent pathological behavior in clinical settings,significantly impacting both physical and mental health.Non-suicidal self-injury is closely associated with the abnormal perception of pain.Pain can be classified as physiological pain and social pain.In contrast to healthy people,individuals engaging in non-suicidal self-harm exhibit lower sensitivity to physiological pain but higher sensitivity to social pain.Their physiological pain thresholds are elevated,while their social pain thresholds are reduced.The alterations in pain sensitivity led them to alleviate emotions through a"pain to counter pain"approach.The abnormalities in pain sensitivity among individuals who engage in non-suicidal self-injury are closely linked to their social and psychological factors.Self-criticism,depressive symptoms,and adverse childhood experiences are all associated with the pain sensitivity of non-suicidal self-harm patients.
8.Development and challenges of gene editing technology.

Gene editing is a technique for modifying gene fragments. The novel gene editing technology focuses on the field of artificial nuclease cleavage technology, mainly ZFN technology, TALEN technology, CRISPR technology and base editing technology. The continuous improvement of gene editing technology has promoted the rapid development of agriculture, animal husbandry and biomedicine, but at the same time, technical defects and ethical controversy have brought enormous challenges to its own development. This article will briefly discuss the development and challenges of gene editing technology, as well as the views at home and abroad, and hope to inspire readers to recognize gene editing technology.

9.Association between gene polymorphism of calcium/calmodulin-dependent kinase 4 and efficacy of amlodipine in the treatment of hypertension in Chinese Han nationality.

OBJECTIVE:
To evaluate the association between single nucleotide polymorphisms of calcium/calmodulin-dependent kinase 4 (CAMK4) and the therapeutic effect of amlodipine in essential hypertensive patients in Chinese Han nationality.
METHODS:
A total of 108 mild-to-moderate essential hypertension patients in Chinese Han nationality were treated with amlodipine for 8 weeks at a dosage of 5 mg/d. Polymerase chain reaction-restriction fragment length polymorphism was performed to detect the genotypes (rs10491334). Blood pressure was measured and analyzed.
RESULTS:
The result of rs10491334 polymorphism of CAMK4 was consistent with Hardy-Weinberg equilibrium distribution and the frequencies of C allele and T allele were 88.89% and 11.11%, respectively. The systolic blood pressure and diastolic blood pressure before amlodipine treatment were not statistically different among different genotype carriers (P>0.05). The blood pressure was significantly reduced in all patients after amlodipine treatment (P<0.05). Systolic blood pressure was significantly decreased in patients with rs10491334 CC genotype and CT genotype compared with those patients with rs10491334 TT genotype. Total effective rates of CT and TT carriers were higher than those of the CC genotype carriers (P<0.01).
CONCLUSION
The CAMK4 gene polymorphism might be associated with the efficacy of calcium channel blocker in treating mild-to-moderate essential hypertension patients.

10.Comparative study of physical therapist allocation, education and training, and professional accreditation system in East Asia

ObjectiveTo compare the current development of physical therapists in China, Japan and South Korea. MethodsThe condition of configuration, education and practice of physical therapists in China, Japan and South Korea were browsed from the official websites and existing data. ResultsThe allocation, education and training mechanism, and the professional accreditation system needs to be optimized in China. ConclusionThe physical therapists should be allocated based on rehabilitation service settings and needing, trained based on the World Health Organization's Rehabilitation Competency Framework and the International Classification of Functioning, Disability and Health, and the professional system should be constructed based on the World Health Organization's Rehabilitation Competency Framework and national professional admission systems.
