1.The changes of specific binding sites of glucocorticoid after scalding in rats

The binding capacity (R_o) and apparant dissociation constant (K_d) of glucocrticoid binding sites in hepatic cytosol of normal and scalded rats were measured by pseudoscatchard analysis. Compared with control group, the levels of Ro of glucocorticoid receptor was significantly decreased 1 h after scalding, partialy recovered at 12h after scalding and fully recovered at 24 h after scalding. The levels of low-affinity glucocorticoid binding sites were also decreased 12 h after scalding, but had no statistical significance. The possible significance of these results has been discussed.
2.Rare Pancreatic Tumor of MSCT Diagnosis and Differential Diagnosis

Purpose To investigate the MSCT diagnosis and differential diagnosis of rare pancreatic tumors.Materials and Methods The MSCT features were analysied respectively of 31 cases with pancreatic tumors confirmed by surgery and pathology.Results There were serous cystadenoma in 7 cases,mucinous cystadenoma in 9 cases,mucinous cystadenocarcinoma in 1 case,solid-pseudopapillary tumor in 8 cases,islet cell tumor in 4 cases,carcinoid tumor in 1 case and intraductal papillary adenocarcinoma 1 case.The MSCT appearances were different each other.Conclusion MSCT is very helpful in diagnosis and differential diagnosis for the rare pancreatic tumors.
3.The Value of Multi-slice Spiral CT in Diagnosis of Extragastrointestinal Stromal Tumor

Objective To investigate multi-slice spiral CT manifestations and its diagnostic value of extragastrointestinal stromal tumor(EGIST).Methods CT data in 19 patients with EGIST proved pathologically were retrospectively reviewed.Results Of the 19 cases,the lesions located in reteoperitoneum in 8 cases,adrenal,greater omentum,omentum and mesentery in one case respectively,mesentery in 4 cases,and abdominal cavity in 4 cases.2 cases were benign,4 cases were potential malignant,and 13 cases were malignant.The size of tumors ranged 4.1~35 cm.The tumors were round or oval in 5 cases,lobulated in 5 cases and irregular in 9 cases.The tumors in 4 cases were uniform density on plain CT scans,1 case of uniform and moderate enhancement,1 case of non-uniform enhancement,2 cases of apparent enhancement as ring on contrast-enhanced CT scans;15 cases showed unhomogeneous density,on contrast-enhanced scans,moderate or remarkable enhancement was seen in the parenchyma of the tumor,while no enhancement was revealed in the necrosis and cystic areas of the tumors.Conclusion EGIST have certain imaging characteristics and MSCT was valuable in diagnosing and differential diagnosing EGIST.

6.Change of GFAP and NSE protein expression in brain of Alzheimer's disease model rats

Objective To find out the difference in expression of glial fibrillary acidic protein and neuron-specific enolase in hippocampi and olfactory bulb of Alzheimer's disease rats,and the relationship between olfactory and Alzheimer's disease.Methods Adult rats,the expression of GFAP and NSE in hippocampi and olfactory bulb was measured by immuhistochemical after a single intracerebroventricular injection of ?-amyloid peptide,and change of these protein in olfactory.Results Comparing the hippocampi and olfactory of model and control,the expression of GFAP in model group increased evidently,the expression of NSE descended animals.Conclusion There is significant difference in olfactory system between Alzheimer's disease model and control group rats.
7.Relationship between blocking of N-acetylglucosaminyltransferase V in hepatocarcinoma cells and cellular endoplasmic reticulum stress

Purpose:To study the connection between the blocking of N-acetylglucosaminyltransferase V(GnT-V) and endoplasmic reticulum stress(ER stress). Methods:The recombinant plasmid expressing siRNA of GnT-V was constructed and transfected into SMMC-7721. The key molecules such as GRP78,XBP1 and PERK during ER stress was analyzed in mRNA and protein levels. Results:GRP78 mRNA and protein levels were upregulated, the spliced forms of XBP1 mRNA and protein appeared and ER eIF2? kinase PERK was activated. Conclusions:These results suggest the blocking of GnT-V in SMMC-7721 induces cellular ER stress.
8.The Basic Characteristics of Low-affinity Glucocorticoid Binding Sites

Scatchard, pseudoscatchard and competitive analyses showed that there were two glucocorticoid (GC) binding sites in rat hepatic cytosol: high-affinity glucocorticoid binding sites (HAGS) and low-affinity glucocorticoid binding sites (LAGS). Pseudoscatchard analysis showed the dissociation constant (Kd) of LAGS was 4.1+1.4umol/L (n=4). The inactivation of LAGS in vitro was slower" than that of HAGS. LAGS and HAGS could not be separated by DEAE-cellulose chfomatography. HAGS were saturable and had steroid specificity for glucocorticoid just as the classic glucocorticoid receptor. Triamcinolone acetonide, RU486 and RU26988 competed equally well to LAGS while aldosterone, progesterone, estradiol and testosterone did not compete with LAGS. These results suggest that LAGS have steroid specificity for glucocorticoids. The biological actions of LAGS await fur ther study.
9.Dysregulation of glucose metabolism in tumorigenesis and tumor progression

Dysregulation of cell metabolism, especially glucose metabolism, is implicated in tumorigenesis and tumor progression.Cancer cells uptake a large amount of glucose and prefer to perform glycolysis in the cytosol even under normal oxygen condition, which fuels fast cell growth and proliferation.Dysregulation of glucose metabolism leads to tumorigenesis and promotes cancer development.Conversely, the initiation and development of cancer reprograms glucose metabolism to confer cancer cells the ability to survive and proliferate.Oncogenes, tumor suppressors or non-coding RNAs could regulate glucose metabolism.Meanwhile, the enzymes and metabolites involved in glucose metabolism could regulate the expression of factors related to cancer.Enzymes, the direct executor of cell metabolism, play a key role in dysregulation of glucose metabolism, tumorigenesis and tumor development.
10.Clinical Observation of Warm Acupuncture in Treating Diabetic Peripheral Neuropathy

Objective:To observe the clinical efficacy of warm acupuncture in the treatment of diabetic peripheral neuropathy and its effects on the conduction velocity of the tibial nerve and common fibular nerve.Methods:Fifty.two subjects were evenly randomized into two groups and treated by conventional therapy;in addition,the control group was given mecobalamin injection at 500 μg,once a day for four weeks,and treatment group was given warm acupuncture,6 times a week for 4 weeks.The clinical efficacy and the conduction velocity of the tibial nerve and common fibular nerve were observed and compared between the two groups.Results:After four-week treatments.the total effective rate Was 88.5% in the treatment group and 61.5% in the control group,with a higher rate in the treatment group than in the control group (P<0.05):the conduction velocity of tibial nerve and common fibular nerve Was higher in both groups(P<0.05),with higher conduction velocity in the treatment group than in the control group (P<0.05).Conclusion:Warm acupuncture is quite effective to treat diabetic peripheral neuropathy and to improve nerve velocity.
