1.Effect of elemene on radiosensitivity of A549 cells and its possible molecular mechanism

Objective To investigate the effect of elemene on the radiosensitivity of A549 cells and its possible molecular mechanism.Methods The effect of radiosensitivity was detected by colony forming assay.The protein expressions of DNA-PKcs,Bcl-2 and P53 were detected with Western blot.The correlation between the protein expression of DNA-PKcs and Bcl-2,DNA-PKcs and P53 was analyzed.Results Elemene had radiosensitizing effect on A549 cells,with the SERDo and SERDq 1.54 ± 0.20 and 1.43±0.15,respectively for 10 μg/ml elemene,and 1.63 ±0.32 and 1.75 ±0.19,respectively for 20 μg/ml elemene.Compared with irradiation group,the expression of DNA-PKcs was reduced significantly in 10,20 μg/ml elemene combined with radiation group ( t =7.52,8.33,P < 0.05 ),so was for Bcl-2(t =10.74,11.33,P <0.05).The expression of P53 protein increased significantly (t =-9.25,7.66,P <0.05).There was a remarkable negative correlation between the expression of DNA-PKcs and P53(r =-0.569,P <0.05),and a remarkable positive correlation between DNA-PKcs and Bcl-2 (r =0.755,P <0.05 ).Conclusions Elemene has radiosensitizing effect on A549 cells,which might be related to down-regulation of DNA-PKcs gene expression,up-regulation of P53 and down-regulation of Bcl-2.
2.Effect of β-elemene Combined with Radiotherapy on the Expression of KU70 mRNA and Apoptosis of Lung Adenocarcinoma Cell Line A549

Objective: To study the effect of β-elemene combined with radiotherapy on the expression of KU70 mRNA and apoptosis of lung adenocarcinoma cell line A549. Methods: A549 cells were divided into the control group (c), irridation group (IR), β-elemene group (0.1×IC50 and 0.2×IC50 I.e. 10 μg/mL and 20pg/mL β-elemene) and β-elemene combined with irridation group (0.1×IC50 + IR and 0.2×IC50 + IR). IC50 was obtained through MTT method and cell survival rate was analyzed by colony formation test. Cell cycle and apoptosis were detected with flow cytometry. The expression level of KU70 mRNA was detected by RT-PCR. Results: MTT assay showed that IC50 value of A549 cells was 120 μg/mL. The number of cell clones and survival rate of β-elemene groups were declined significantly. The radiosensitivity of A549 cells can be enhanced by β-elemene. The flow cytometry confirmed that the ratio of G_2/M and apoptosis were significantly in-creased under the effect of β-elemene, statistically different from those in the control group (P <0.05). The expression level of KU70 mRNA in β-elemene with radiotherapy group was declined significantly, statistically different from those in the con-trol group (P<0.05). Cendusion: The rediosensitization effect of β-elemene on A549 cells is associated with induction of cell apoptosis and down-regulation of KU70 mRNA expression.
3.Screening radiosensitizing-related genes mediated by element in lung adenocarcinoma A549 cells by using gene chip

Objective To screen radiosensitizing-related genes mediated by elemene in lung adenocarcinoma A549 cells by using gene chip. Methods MTT test was used to calculate the IC50 of elemene. ①The effect of radiosensitivity was detected by colony forming assay. A549 cells were divided into 2 groups: radiation group and radiation + elemene group. Oligonucleotide chip was used to screen the gene expression changes of A549 cells from these 2 groups. The up-regulated gene Egr-1 and the down-regulated gene CyclinDl were selected to undergo RT-PCR so as to confirm the reliability of the result. Results MTT test showed the elemene inhibited the proliferation of the A549 cells dose-dependently. The IC50 value of elemene on the A549 cells was 120 mg/L. ②10 mg/L elemene had radiosensiting effect on A549 cells.The values of SERDO and SERDq obtained from the survival curve were (1.54±0. 20) and (1.43±0. 15 )respectively. Gene chip screened 122 differentially-expressed genes, including 89 up-regulated genes and33 down-regulated genes. ③These altered genes could be related to cell structure, substance metabolism,cell proliferation, cell differentiation, signal transduction, material transport, DNA repair, apoptosis,immune response and so forth. The RT-PCR results of Egr-1 and Cyclin D1 were consistent with the genenchip analysis.Conclusions The mechanism of elemene enhancing the radiosensitivity of lung adenoearcinoma A549 cells is the result of participation and collaboration of multiple genes. Further study of the newly-discovered differentially-expressed gene helps find out new radiosensitizational targets of elemene.
4.Assessment of consistency of the whole tumor and single section perfusion imaging with 256-slice spiral CT: a preliminary study

Objective To determine the consistency between quantitative CT perfusion measurements of colorectal cancer obtained from single section with maximal tumor dimension and from average of whole tumor,and compare intra-and inter-observer consistency of the two analysis methods.Methods Twenty-two patients with histologically proven colorectal cancer were examined prospectively with 256-slice CT and the whole tumor perfusion images were obtained.Perfusion parameters were obtained from region of interest (ROI) inserted in single section showing maximal tumor dimension,then from ROI inserted in all tumor-containing sections by two radiologists.Consistency between values of blood flow (BF),blood volume (BV) and time to peak (TTP) calculated by two methods was assessed.Intra-observer consistency was evaluated by comparing repeated measurements done by the same radiologist using both methods after 3 months.Perfusion measurements were done by another radiologist independently to assess inter-observer consistency of both methods.The results from different methods were compared using paired t test and Bland-Altmnan plot.Results Twenty-two patients were examined successfully.The perfusion parameters BF,BV and TTP obtained by whole tumor perfusion and single-section analysis were (35.59 ± 14.59) ml · min-1 · 100 g-1,(17.55 ±4.21) ml · 100 g-1,(21.30 ±7.57) s and (34.64 ± 13.29)ml· min-1 · 100 g-1,(17.61 ±6.39)ml± · 100 g-1,(19.82 ±9.01)s,respectively.No significant differences were observed between the means of the perfusion parameters (BF,BV,TTP)calculated by the two methods (t =0.218,-0.033,-0.668,P > 0.05,respectively).The intra-observer 95% limits of consistency of perfusion parameters were BF-5.3% to 10.0%,BV-13.8% to 10.8%,TTP-15.0% to 12.6% with whole tumor analysis,respectively; BF-14.3% to 16.5%,BV-24.2% to 22.2%,TTP-19.0% to 16.1% with single section analysis,respectively.The inter-observer 95% limits of consistency of perfusion parameters were BF-8.0% to 8.3%,BV-10.9% to 11.5%,TTP -14.5% to 11.1% with whole volume analysis,respectively; BF-10.2% to 14.1%,BV-19.0% to 17.6%,TTP-22.0% to 24.0% with single section analysis,respectively.Conclusion There was no statistically different between the single section and whole volume analysis of tumor perfusion CT.The whole volume perfusion analysis apparently improves intra-and inter-observer consistency and can reflect the whole tumor angiogenesis more accurately and repeatedly.
5.Information needs of users in biomedical industry clusters

A large number of starring biomedical industry clusters are emerged in recent years with the develop-ment of biomedical industry. The information access approaches of users in biomedical industry clusters consisting of higher education institutions, scientific research institutions and enterprises, the types of biomedical information they need, the transformation ways of their achievements, and the major barriers of information access were thus in-vestigated in this paper with questionnaires. The different information behaviors were compared between the users of scientific research group and enterprise group. The characteristics of information need of users in biomedical indus-try clusters were summarized, and following suggestions were put forward, namely establishing information service platform, perfecting institution-oriented information service model, and improving information service system for bi-omedical industry clusters.
6.Analysis of clinical etiology in 270 children patients with severe pneumonia complicated with congenital heart disease

Objective To analyze the clinical features of severe pneumonia complicated with congenital heart disease (CHD) . Methods The clinical data of 270 children patients with severe pneumonia complicated with CHD (CHD group) were collected to analyze the relative medical history ,etiology ,bacterial drug resistance and clinical outcomes .Moreover ,636 age-matched children pa-tients with pure severe pneumonia were selected as the control group .Results In the CHD group ,250 cases were infants and 35 ca-ses suffered from repeated pneumonia .Compared with the control group ,the CHD group was earlier in onset and more prone to re-peated pneumonia .Among 270 cases ,totally 220 cases were checked out the pathogens ,but no statistically significant difference in the detection rates of pathogens between the CHD group and the control group was found (P>0 .05);179 cases were infected by bacteria ,in which 126 cases were Gram-negative bacteria ,the positive rate of bacterial infection and the detection rate of Gram-nega-tive bacterial were relatively higher than those in the control group ,the difference was statistically significant (P<0 .05);95 cases were infected by virus ,with respiratory syncytial virus being the most common pathogen ,30 cases were infected by fungi ,with Can-dida albicans being predominant ,the infection rates of virus and fungus had no statistical difference between two groups (P>0 .05);in the drug sensitive test ,the detection rate of extended spectrum beta lactamase (ESBLs)-producing positive bacteria in the CHD group was higher than that in the control group ,the difference was statistically significant (P<0 .05) .Main Gram-negative bacteria Pneumonia klebsiella pneumonia subspecies ,Escherichia coli and haemophilus inf luenzae were highly sensitive to imipenem ,mero-penem ,levofloxacin and ciprofloxacin ;main Gram-positive bacteria Staphylococcus aureus and streptococcus pneumoniae were highly sensitive to vancomycin and gentamicin .Especially ,pneumonia klebsiella pneumonia subspecies ,Escherichia coli and haemophilus influenzaewere100% resistanttopenicillin.TheaveragelengthofhospitalstayintheCHDgroupwas (20.9±12.5)d,207cases developed the respiratory failure ,88 cases appeared the heart failure and 205 cases were effective in treatment ;compared with the control group ,the CHD group had a longer length of hospital stay ,higher occurrence rate of heart and respiratory failure and lower treatment effective rate (P<0 .05) .Conclusion Because of the earlier onset ,more prone to repeated pneumonia ,more sensitive to bacterial infection ,esp .Gram-negative bacteria ,more ESBLs production ,longer hospital stay and more prone to complications ,com-pared with the control group ,children with severe pneumonia complicated with CHD require active treatment .
7.Value of high-sensitive troponin I detection in the diagnosis of early myocardial injury

Objective To explore the clinical value of high‐sensitive troponin I detection in the diagnosis of early myocardial inju‐ry .Methods Totally 240 type 2 diabetes mellitus (T2DM ) patients (study group) and 40 healthy people(healthy control group) were chosen as subjects .The serum level of HbA1c ,AST ,CK ,CK‐MB and hs‐cTnI were measured among the two groups ,respec‐tively .Results Hs‐cTnI level in T2DM group was significantly higher than in healthy control group(P< 0 .05) ,and AST ,CK and CK‐MB levels in two groups had no significant difference(P> 0 .05) .Hs‐cTnI level and positive rate in HbA1c ≥ 6 .5% group was significantly higher than in HbA1c ≤ 6 .4% group(P< 0 .05) ,and no significant difference of AST ,CK and CK‐MB levels and posi‐tive rate were observed in two groups(P> 0 .05) .There was a positive correlation between the level of HbA1c and that of serum hs‐cTnI in T2DM group(r= 0 .471 ,P< 0 .05) .Conclusion The hs‐cTnI level detection has important clinical value in the diagnosis of early myocardial injury in T2DM patients ,and HbA1c is positively correlated with hs‐cTnI .
8.The relationship between the cystic fluid characteristics of symptomatic sacral canal cyst and the interventional therapeutic prognosis

Objective To investigate the relationship between the cystic fluid characteristics of symptomatic sacral canal cyst and the interventional therapeutic prognosis.Methods A total of 114 patients with symptomatic sacral canal cyst were enrolled in this study.Clinically,all patients complained of discomfort at lumbosacral area.Among the 114 patients,86 were primary sacral canal cyst and 28 were recurrent sacral canal cyst.Under DSA guidance,percutaneous puncturing of the cyst was performed,2-5 ml cerebrospinal fluid (CSF) was aspirated and sent for laboratory tests.Then a small amount of nonionic contrast agent was injected into the cyst to determine whether the cyst was communicated with the subarachnoid space or not.Finally,double-needle method was used to aspirate the cyst fluid.Results Radiography showed that communication between the cyst and subarachnoid space was detected in 66 patients (group A),while no communication between the cyst and subarachnoid space was observed in 48 patients (group B).In patients with primary symptomatic sacral canal cyst,the differences in the sugar and chloride levels of CSF between group A and group B were statistically significant.In patients with recurrent symptomatic sacral canal cyst,the differences in the sugar,protein and chloride levels of CSF between group A and group B were also statistically significant.Statistically significant correlation existed between the single or multiple CSF changes and the interventional therapeutic prognosis.Conclusion Sacral canal cysts can be classified into two types:cyst-subarachnoid space communicating type and cyst-subarachnoid space non-communicating type.The characteristics of CSF in patients with primary symptomatic sacral canal cyst are different from those in patients with recurrent symptomatic sacral canal cyst.Multiple CSF changes,the increased sugar level and decreased chloride level in CSF are well correlated with the interventional therapeutic prognosis.
9.Comparative study of neurological soft signs in remittent schizophrenia and non-psychotic first-degree relatives

Objective To examine the difference of neurological soft signs (NSS) among the remittent schizophrenia patients,first-degree non-psychotic relatives of schizophrenia patients and healthy controls.Methods The Cambridge Neurological Inventory (CNI) (the Chinese version) was administered to 86 remittent schizophrenia patients (patient group),86 first-degree non-psychotic relatives of schizophrenia patients (relative group) and 86 healthy controls (control group).Results Compared with relative group,NSS total-score,the subscore of motor coordination and sensory integration were significantly higher in patient group (P<0.01).Compared with control group,NSS total-score,the subscore of motor coordination and sensory integration were significantly higher in patient group (P<0.01).Compared with control group,NSS total-score and the subscore of motor coordination were significantly higher in relative group (P<0.01).Conclusions The levels of NSS in remittent schizophrenia patients and their first-degree non-psychotic relatives are higher than normal control,and the patients have more NSS than their relatives.The motor coordination nay be a potential endophenotype for schizophrenia.
10.Safety of cleaning and disinfection of duodenoscope for endoscopic retrograde cholangiopancreatography

Objective To investigate the safety of cleaning and disinfection of duodenoscope used in endoscopic retrograde cholangiopancreatography (ERCP).Methods The duodenoscopes,which were used in Endoscopy Center of Digestive Disease Hospital,Heilongjiang Provincial Hospital from May 4 to May 31 2016,were cleaned manually and disinfected with sterilization powder.The samples from elevator and elevator channel were collected for bacterial culture to evaluate the safety of clinical application of duodenoscope.Results Among 60 high level disinfections,one was excluded because disinfectant concentration wasn't effective.Among 59 elevator samples no positive cultures were detected,while among 59 channel samples,4(6.8%) cultures were positive and all from the same duodenoscope,including 3 cases of Klebsiella pneumonia and 1 case of Pseudomonas aeruginosa.Conclusion Sterilization powder is effective for disinfection of duodenoscope used for ERCP.Residual organic material can't be excluded from elevator sample even if it is negative for culture.Duodenoscope with high level disinfection still has the risk of transmitted infection,and should be monitored regularly by culture and reprocessed in time.
