BACKGROUND:With the extensive application of cone-beam CT in oral and craniofacial surgery, a clear hyoid bone position indicator for normal population has important implications for the change of hyoid bone position before and after orthodontics. OBJECTIVE:To determine the cone-beam CT measurement range of hyoid bone position in populations aged 6-19 years from Shandong, so as to provide a reference marker for the change of hyoid bone position before and after orthodontics in local children and adolescents. METHODS:Totaly 254 healthy children (120 males and 134 females) aged 6-19 years from Shandong Province were subjected to cone-beam CT scan of the hyoid bone. Mimics10.01 was used to evaluate the linear and angular measurements of hyoid bone position, and then the normal value range was confirmed. Independent-samplet-test was used for analysis of gender difference and 95% confidence interval was calculated. RESULTS AND CONCLUSION: There were gender differences in a part of indicators of hyoid bone position among 14-15 years, 16-17 years, 18-19 years groups (P < 0.05). For children aged 12-13 years, the hyoid bone position vertical to the base of skul and upper jaw bone is lower in males than in females. For children aged 14-15 years and 18-19 years, the hyoid bone horizontal to the cervical spine is more forward in males than in females.

Purpose To investigate the effect of subcellular location of tumor BRCA1 on the sensitivity to ionizing radiation (IR) and PARP inhibitor. Methods siRNA of BRCA1 were first used to inhibit endougenous BRCA1 expression in MCF7 cells. Then, plasmids of pCMV-3xFlag-WT-BRCA1,pCMV-3xFlag-NES-BRCA1 and pCMV-3xFlag-NLS-BRCA1 were transfected in MCF7 cells.Immunofluorescence staining was used to detect BRCA1 subcellular location as well as the formation of Rad51 and γ-H2AX foci.Apoptotic cells were analyzed by flow cytometry, and colony formation assay was performed to evaluate the survival of cells. Results There were 47% cells with nuclear BRCA1,23% cells with cytoplasmic BRCA1 and 30% cell with mixed nuclear and cytoplasmic BRCA1 expression in WT-BRCA1 transfected cell. There were 87% cells with nuclear BRCA1 in NES-BRCA1 transfected cell, and 82% cells with cytoplasmic BRCA1 in NLS-BRCA1 transfected cell. There were 87%,84% and 13% Rad51 foci positive cells at 2 hours after 4 Gy radiation treatment and 22%,25% and 59% γ-H2AX foci positive cells at 24 hours after 4Gy radiation treatment in WT-BRCA1,NES-BRCA1 mutant and NLS-BRCA1 mutant transfected cell respectively.ABT-888 and radiation treatment induced more apoptosis and fewer colonies in NLS-BRCA1 transfected cell than WT-BRCA1,NES-BRCA1 mutant transfected cell. Conclusion Subcellular location of BRCA1 might affect homologous recombination repair of DNA double strand breaks and can be used to predict sensitivity to IR and PARP inhibitor.
6.Research advances in predictive markers for HBeAg seroconversion in chronic hepatitis B patients treated with nucleos(t)ide analogues
Journal of Clinical Hepatology 2020;36(6):1358-1361
Nucleos(t)ide analogues (NAs) are effective inhibitors for HBV replication and have become the preferred antiviral regimen for most patients with chronic hepatitis B (CHB). HBeAg seroconversion is an important index used to evaluate the durability and efficacy of antiviral therapy in HBeAg-positive CHB patients. The search for biomarkers that can predict HBeAg clearance or seroconversion after NAs treatment plays an important role in the selection of antiviral drugs, the adjustment of treatment regimens, and the achievement of individualized treatment. This article reviews the value of related markers, including HBV DNA, HBV RNA, anti-HBc, and HBcrAg, in predicting HBeAg clearance or seroconversion in CHB patients treated with NAs.
