Chinese Journal of Tissue Engineering Research 2015;(41):6654-6658
BACKGROUND:CT perfusion technology is a common non-invasive detection method, which can be used to quantitatively determine the ischemia severity and range at early stage of cerebral infarction and then judge whether ischemic brain tissues can survive or recover. OBJECTIVE:To assess the neurological function recovery of cerebral infarction rats undergoing neural stem cel transplantation using CT perfusion imaging. METHODS:A total of 60 Sprague-Dawley rats were randomly divided into control group, cerebral infarction group, transplantation group, with 20 rats in each group. Rat model of middle cerebral artery occlusion was made in the latter two groups. After 24 hours of modeling, PBS and 8×105 neural stem cels were administratedvia the tail vein into the rats in the cerebral infarction and transplantation groups, respectively. CT perfusion-weighted imaging was performed at 1, 3, 7, 14, 28 days after transplantation. Modified neurological severity scores were recorded at 1, 2, 3, 4 weeks after transplantation. Triphenyltetrazolium chloride staining was used to calculate infarct volume at 4 weeks after transplantation. Hematoxylin- eosin staining was adopted to observe pathological changes of brain tissues at 2 weeks after transplantation. RESULTS AND CONCLUSION: There were no abnormal hemodynamic changes in the control group at different time points. The transplantation group exhibited an increasing CT value with time, and the increased cerebral blood flow could improve the survival rate of neurons in the ischemic penumbra. The modified neurological severity score and infract volume in the transplantation group were both significantly lower than those in the cerebral infarction group (P < 0.05). Cel necrosis was improved obviously in the transplantation group. These results show that CT perfusion imaging can be used to observe the neurologic function recovery of cerebral infarction rats in aspects of morphology and hemodynamics.

Objective Based on the theory of the Balanced Score Card framework and using Delphi method and analytic hierarchy process to construct the clinical nursing staff performance appraisal index evaluation system and provide reference for clinical nursing staff post management. Methods According to the theory of Balanced Score Card, constructed the preliminary establishment of index system for performance evaluation of clinical nursing. Used Delphi method to interview 26 nursing experts to select clinical nursing performance evaluation indicator. In the end, applied hierarchical analysis to determine index weight at all levels. Results Conducted two rounds of expert consultation;distributed 2 rounds of questionnaires by 27 and 26;recycled by 26 and 25;the recycling rate was 96.3%and 99.1%. It showed that the active coefficient of experts was higher. The degree of experts′authority was 0.89, which meaned the higher authority. Through two rounds of expert consultation, the Kendall coordination coefficient was 0.199-0.388. The significant test indicated that it was statistically significant (P<0.05), which meaned the experts tend to agree. After two rounds of expert consultation, the final clinical nursing staff performance appraisal indicators included 4 first-class indexes, 10 secondary indexes, 24 third-class indexes. Each index weight was determined by the analytic hierarchy process. In primary index, the internal process, learning and growth, performance indicators and the weight of customer satisfaction were 0.3, 0.3, 0.3 and 0.1 respectively. In secondary index weight, the top three were the work′s quality, post, and quantity. In the third class index weight, the top three were undertaking clinical work, annual nurse competence grading and the person-time of critical patients′management or surgical patient management. Conclusions Based on Delphi method and analytic hierarchy process to determine the clinical nursing staff performance appraisal evaluation index, whic can provide theoretical basis for clinical nursing staff post management.

Due to the influence of potential psychological trauma, athletes in the competition may repetitively suffer poor performances, such as losing his usual form, slump, choking, and so on. These under-performing problems actually are symptoms of sports post-traumatic stress disorder (SPTSD). SPTSD refers to the pathological physical and mental after-effects of a trauma previously suffered by the athlete on that athlete's subsequent performances. This is a very common physical and emotional problem in athletics that impairs athletes' performance, affecting their competition results. By peak performance eye movement desensitization and reprocessing therapy, therapists can deal with this problem effectively and help athletes eliminate symptoms of sports post-traumatic stress disorder, improving the competition performance.

Objective To investigate the effect of umbilical cord blood dendritic cells(DCs)induced by gastric cancer antigen combined with cytokine induced killer(CIK)cells in gastric cancer cell lines SGC-7901 in vitro. Methods Mononuclear cells from umbilical cord blood were used to create DCs and CIKs. The cell surface antigen expression of the mature DCs such as CD83,CD86,CD11c and the cell surface antigen of CIKs such as CD3,CD56,CD4,CD8,CD16 were detected using flow cytometry. Sensitized DCs-CIKs,DCs-CIKs,CIKs as effective cells,and SGC-7901 as target cells,the killing activities of these effective cells were tested with LDH release,which the number ratio of cells between effective cells and SGC-7901 cells were 10 :1,20 : 1,40 : 1,respectively. Results The cell surface antigen expressions of the mature DCs,such as CD83 + CD86 + ,CD11c + CD83 + ,CD86 + CD11c + were(75. 4 ± 2. 1)% ,(79. 3 ± 1. 4)% ,(80. 2 ± 2. 6)% , respectively. The mature sensitive-DCs surface antigen expressions,such as CD83 + CD86 + ,CD11c + CD83 + , CD86 + CD11c + ,were(77. 7 ± 1. 5)% ,(82. 6 ± 1. 9)% ,(76. 9 ± 2. 6)% ,respectively. There was no sta-tistical significance about the surface antigen expression between DCs and sensitive-DCs(t = 1. 526,P ﹥ 0. 05;t = 0. 958,P ﹥ 0. 05;t = 1. 049,P ﹥ 0. 05). The CIKs surface antigen expressions,such as CD4 + ,CD8 + , CD3 + CD56 + CD16 + ,were(22. 8 ± 1. 3)% ,(77. 3 ± 1. 8)% ,(24. 5 ± 2. 1)% ,respectively. The results suggested that the killing effect of the three kinds of combination cells on gastric cancer cells was different. The number ratio of cells between sensitive-DCs and SGC-7901 cells were 10 : 1,20 : 1,40 : 1,which the killing activities of sensitive-DCs-CIKs against SGC-7901 were(37. 68 ± 1. 49)% ,(41. 67 ± 0. 90)% ,(42. 71 ± 0. 98)% ,respectively. The killing activity of sensitive-DCs-CIKs was the highest when the ratio of cells between sensitive-DCs and SGC-7901 cells were 40 : 1. The killing activities of DC-CIKs were(36. 77 ± 0. 46)% ,(38. 94 ± 0. 95)% ,(41. 15 ± 0. 89)% ,respectively. The killing activities of CIKs were(34. 74 ± 1. 01)% ,(37. 76 ± 0. 43)% ,(39. 65 ± 0. 79)% ,respectively. There were statistically significant differences among the three groups(F = 5. 92,P ﹤ 0. 05;F = 19. 13,P ﹤ 0. 05;F = 8. 88,P ﹤ 0. 05). Conclusion The tumor killing activity of CIK is enhanced obviously by umbilical cord blood DCs which is sensitized by gastric cancer tumor antigen. There is the highest killing activity when the number ratio of cells between sensitive-DC-CIK and SGC-7901 cells is 40 : 1.
