1.Effects of chemotherapeutic drugs on proliferation of tongue carcinoma cells

Objective To observe the effects of paclitaxel,mitomycin,5-FU and cisplatin on the proliferation of tongue carcinoma cells and to guide the reasonable using of chemotherapeutic drugs and avoids the side-effects.Methods The fresh tongue carcinoma tissues from surgery were collected and unicellular suspension were prepared.Then paclitaxel,mitomycin,5-FU and cisplatin were used to investigate the drug sensitivity on primary tongue carcinoma cells and a blank control group was set up.The inhibitory effects of chemotherapeutic drugs on the growth of tongue carcinoma cells and the cell cycle phases were observed by MTT colorimetry and flow cytometry.Results The inhibitory rates of paclitaxel,mitomycin and 5-FU on tongue carcinoma cell proliferation were 45.3%,37.3%,and 36.2%,respectively,and in control group it was 2.1%,the difference was significant(P

This article is to review the basic principles of renal perfusion in CT and MR,imaging method and perfusion characteristics in kidney diseases.It shows that in addition to the morphologic information,perfusion imaging can also provide functional and metabolic information about the renal neoplasms,which is very helpful for not only the differential diagnosis,but also the evaluation of tumor biological behaviors,it will have a bright future in clinical application.
3.Multi-glycosidorum triptery suppresses production of interleukin-4 by splenocytes in oxazolone-induced murine colitis

Objective To investigate the in vitro effect of multi glycosidorum triptery (MGT) on cytokine production by splenocytes of oxazolone (OXZ) induced colitis in murine model. Methods Six mg of OXZ (in 50% of ethanol) was administered in male SJL/J mice intrarectally to induce colitis and mice were sacrificed 3 days later. Isolated splenocytes were cultured for 24 hours in the presence of PMA and ionomycin, MGT of 0.1 mg/ml or 0.01 mg/ml was added to the culture medium of splenocytes. Production of IFN ? and interleukin 4 (IL 4) in the supernatant was measured by ELISA. 4.Electron beam CT three-dimensional reconstruction in the diagnosis of bowel obstruction

Objective: To evaluate the electron beam computed tomography(EBCT) three-dimensional reconstruction in the diagnosis of bowel obstruction.Methods: Seventy-eight patients suspected of bowel obstruction underwent EBCT with two-phase enhancement scanning and 32 of them were re-established by corona-MPR and sagittal-MPR.All the EBCT results were analyzed and compared with the clinical findings.Results: Seventy-three of the cases were diagnosed as bowel obstruction by EBCT,which were all confirmed by surgery or follow-up,with an accuracy rate of 100% for location diagnosis and 86.3% for etiology.Conclusion: EBCT 3D-reconstruction has great advantages in determining the location,etiology and blood-supply of bowel obstruction.
5.Preparation of bifunctional anti-human lingual cancer/anti-VEGF antibody

Objective:To develop bifunctional anti-human lingual cancer/anti-VEGF antibody.Methods:Antigen of lingual cancer was prepared from Tca 8113 cells and that of VEGF from endothelial cells of umbilical cord vein.The hybridoma cells of anti-human ligual cancer/anti-VEGF were prepared by the technique of secondary hybridoma.The antibody was identified by ELISA. Results:The specific conjugation rate between bifunctional antibody and lingual cancer tissue was 85.29%, and that between the antibody and vascular endothelial cells was 82.35%.Conclusion:The bifunctional antibody of anti-human lingual cancer/anti-VEGF has good targeting potential.

BACKGROUND: Generally speaking, neuroendocrine cells have been not observed in normal breast tissue but found in breast carcinoma tissue which was affected by local microenvironment and hormone level during differentiation of breast epithelial stem cells.OBJECTIVE: By detecting expressions of breast cancer resistance protein (BCRP), cytokeratin-8 (CK8), and chromogranin-A (CgA) in breast carcinoma tissue, to explore the possible mechanism of neuroendocrine cells observed in breast carcinoma tissue during differentiation of multi-lineage potential of bone marrow mesenchymal stem cells.METHODS: BCRP, CK8, and CgA were used as markers for SP stem cells, glandular epithelium differentiation, and neuroendocrine differentiation, respectively. Immunohistochemistry was used to detect the expressions of BCRP, CK8, and CgA in breast tissues of 89 subjects and analyze their correlation. RESULTS AND CONCLUSION: Both BCRP and CK8 expressions were observed in normal breast tissues, hyperplastic tissues, and breast carcinoma tissue. BCRP expression was increased in the breast carcinoma tissue; CK8 expression was decreased with the abnormal differentiation of breast tissue; CgA expression was only detected in breast carcinoma tissue. BCRP expression was significantly correlated with positive CgA expression (P < 0.01), but it was no correlation with positive CK8 expression in normal breast tissues, hyperplastic tissues, and breast carcinoma tissue (P=0.069). The results suggested that neuroendocrine cells were not observed in both normal breast tissues and hyperplastic tissues but in breast carcinoma tissue, which possibly correlated to differentiation of multi-lineage potential of bone marrow mesenchymal stem cells.
8.Dose effect of laser irradiation in photodynamic therapy for laryngocarcinoma in BALB/c mouse

Objective The dose and type of light and photosensitizer could seriously affect the curative effect of photodynamic therapy (PDT).The purpose of this study was to observe whether or not PDT with hematoporphyrin monomethyl ether (HMME) can cure laryngocarcinoma in the solid tumor model,and to define the proper laser amount for killing the cancer cells.Methods Forty eight BALB/c mouse models with subcutaneous Hep-2 laryngeal carcinomas were prepared.Mice were divided into six groups depending on the amount of laser received from 30 J/cm2 to 480 J/cm2 including a control group,tumor size in each group was between 8 mm and 10 mm.Tail vein injection were given with HMME prior to applying the laser light,and then illumination was carried out on the tumor at 3 h after HMME administration.Tumor volume,animal weight and histopathologic changes were observed after PDT.Results All mice apparently showed positive results via PDT,and the cancer had been cured in 120 J/cm2 and 480 J/cm2 groups.The laryngeal cancer lesions began to form scab 1 d after PDT and the scab became hard and black after 5 d.The tumor regression began simultaneously and completed around 30 d after PDT.Conclusions PDT may treat laryngeal cancers which sized less than 10 mm in mouse models.The optimum energy to destruct the laryngeal cancer cells may be 120 J/cm2.

Objective To study the chemical constituents from the root of Rumex gmelini.MethodsThe compounds were isolated and purified by chromatography,polyamide column chromatography,and preparation HPLC etc.Their structures were elucidated by physicochemical and spectroscopic evidences.Results Ten compounds were identified as:nepodin (Ⅰ),emodin (Ⅱ),citreorosein (Ⅲ),chrysophanol 8-O-?-(6'-acetyl) glucopyranoside (Ⅳ),chrysophanol 8-O-?-D-glucopyranoside (Ⅴ),resveratrol (Ⅵ),9,9'-dianthranone-2,2'-dimethyl-5,5'-bis (?-D-glucopyranose)-9,9',10,10'-tetrahydro-4,4'-dihydroxy-10,10'-dioxo (trivial name:rumoside A) (Ⅶ),emodin-8-O-?-D-glucopyranoside (Ⅷ),resveratrol-3-O-?-D-glucoside (Ⅸ),and rutin (Ⅹ).Conclusion Compound Ⅶ is a new compound,named rumoside A.Compounds Ⅴ,Ⅷ,and Ⅹ are separated from R.gmelini for the first time.Compounds Ⅲ and Ⅳ are the compounds which have been found in the plants of Rumex L.for the first time.
10.Pathological observation of adriamycin nephropathy in rats

AimTo establish the model of adriamycin- induced nephritic syndrome in rats and its pathological change wasobserved. MethodsThe model of nephritic syndrome in rats was established by intravenous injection of adriamycin in the dose of 5mg · kg-1 and 3 mg · kg-1 respectively at the first day and 15 th day. The amount of urine protein was measured at the 1、2、3、4、6、8 and 12 th week and the serum biochemical pa rameters of TG, TCH, BUN, Cr, TP and ALB were detected at the 6 th and 12 th week. Results The urine protein and the serum biochemic parameters were increased significantly, except the TP and ALB, which were decreased. Pathological changes at the sixth week was similar to minimal changes in glomerulonephritis, and at the 12 th week, sclerosis of glomeruli of kidney could be found. ConclusionAfter first injection,the changes of kidney were like those inminimal change glomerulonephritis of human being. But after the second injection. The kidney had the changes of focal segmental glomeralosclerosis.
