Objective To assess the potential immunological activity of anthocyanins from cherries on adjuvant arthritis (AA) rats and observe the effect on the ratio of T lymphocyte subpopulation and splenic lymphocyte and the levels of IL-6 in serum and PGE_2 in paws. Methods Sixty male Spraque Dauley rats were randomly allocated to six groups, they were normal group, model group, anthocyanins (300, 150, 75 mg/kg) -treated groups, and polyglycosides of Tripterygium wilfordii (TW)-treated group. Arthritis was induced by the complete Freunds adjuvant. The T lymphocyte subpopulation of CD3, CD4, and CD8 and CD4/CD8 value in peripheral blood were assayed by FACA Calibur flow cytometer. The splenocyte proliferation was assessed by means of MTT Assays. IL-6 levels in serum and PGE_2 in paws were determined by radioimmunoassay. 3.Differential Analysis of Two-dimensional Gel Electrophoresis Profiles of Gastric Carcinoma and Paired Normal Gastric Mucosa

Objective To establish two-dimensional electrophoresis profiles with high resolution and reproducibility from human gastric carcinoma and paired normal gastric mucosa tissues and to analyse the differential expression proteins between the two types of the tissues. Methods The total proteins of gastric carcinoma and paired normal gastric mucosa tissues were seperated by immobilized pH gradient-based two-dimensional gel electrophoresis (2-DE).After silver staining, the differential expression proteins were analyzed by using Imaging Master 2D analysis software. Results ⑴Average protein spots were 1049?67, 1097?73 in gastric carcinoma and paired normal gastric mucosa respectively,the matched spots were 835?48, 953?56 in both the two types of tissues respectively, and the average matching rate was 79.6%, 86.7% respectively. ⑵A total of 769?45 protein spots were matched between the electrophoretic maps of 5 gastric carcinoma and paired normal gastric mucosa tissues. The 81 differential protein spots were identified between the two types of the tissues. 17 protein spots were expressed only in the gastric carcinoma and 24 protein spots were expressed only in the normal gastric mucosa. The expression levels of 26 protein spots in the gastric carcinoma were higher than those in the normal gastric mucosa, and the expression levels of 14 protein spots in the gastric carcinoma were lower than those in the normal gastric mucosa. Conclusion In this study, the well-resolved,reproducible 2-DE patterns of gastric carcinoma and paired normal gastric mucosa tissues were established and some differential proteins between the two types of tissues were found. These data will be helpful to further screen specific biomarkers of gastric carcinoma.
4.Effect of gradient pressure socks combined with intermittent pneumatic pressure pump on lower extremity deep venous thrombosis after gynecological laparoscopy

Objective To study the effect of gradient pressure socks combined with intermittent pneumatic pressure pump on lower extremity deep venous thrombosis after gynecological laparoscopy.Methods Toally 45 patients to underwent gynecological laparoscopy during April 2013 to February 2015 were assigned as observation group and from April 2012 to March 2013 another 45 ones to undergo the same therapy were as control group.The latter was treated with routine nursing and the former with gradient pressure socks combined with intermittent pneumatic pressure pump apart from the routine nursing.The effective rate was conpared between two groups.Result The observation group was significantly more effective in the prevention of lower extremity deep venous thrombosis than that of the control group (P<0.05).Conclusion The gradient pressure socks combined with intermittent pneumatic pressure pump can be effective in the prevention of lower extremity deep venous thrombosis after gynecological laparoscopy and thus deserves clinical popularization.

Objective Study the special value of the brainstem auditory evoked potential(BAEP)and Brain Geography Diagram(BEAM/EEG)for diagnosis of Newborn HypoxicIschemic Encephalopathy(HIE),the correlation between BAEP,BEAM/EEG and HIG,and the relation with HIE linger effect,so as to evaluate clinical curative effect,find out the best treatment time,lower the occurrence of linger effect.Methods the BEAM/EEG and BAEP were detected synehronously by adopting 16 traots of BEAM/EEG and BAEP instruments for HIE before treatment,at thirtieth day,the third Month,the sixth Month,the twelfth Month and the twenty-fourth Month respectively.Results The BAEP,BEAM/EEG have special diagnostic value on nerve cell function recovery,examination of recovery period in particalar.The research discovers the both are not synehronous in recovery period,the BAEP recovers is later than BEAM/EEG;for heavy degree HIE,BAEP recovering is slow with bad therapy effect,BAEP index sign is more sensitive to BEAM/EEG to assess linger effect.Conclusions Simple BEAM/EEG damaye indicates the brain cortex iniury only,so the prognosis is good;while BAEP damage suggestes the patiemts have brain stem nerve cell injury simultaneously,with bad prognosis for the heavy degree abnormality.in combination with BAEP and BEAM/EEG in clinical practiae,investigation of recovery degree of HIE herve cells by kinds of electric physiology angles it would have important value to perfect the treatment of HIE.

Objective:To elucidate the effects of calcitonin gene-re la ted peptide(CGRP) on the gene expression of osteoprotegerin(OPG) and receptor ac tivator of nuclear factor-?B ligand(RANKL) in rabbit osteoblasts. Meth ods:Osteoblasts were cultured in media containing 10 -10~10 -7 mol/L of CGRP. After 24-hour incubation,semi-quanitative RT-PCR was perfor med to detect the expression of OPG and RANKL mRNA,and with ?-actin mRNA as th e internal control. Results:CGRP increased the mRNA expressio n of OPG with the maximal effect at the concentration of 10 -8~10 -7 mol/L. CGPR downgulated the mRNA expression of RANKL dose-dependantly.C onclusion:CGRP may regulate the activities of osteoclasts by regulating gene expression of OPG/RANKL.

Objective:To study the effect of sensory nerve on fracture of mandible and try to find the possible mechanism.Methods:Thirty-two China rabbits were used and divided into two groups for study.They all suffered from a standardized fracture in the left mandible,and half of them with inferior alveolar nerve section(IANS)or without.The callus were stained to observe the expression of substance P(SP)by immunochemistry and analysed by a computerized-graph analysing system.Results:Immunoreactivites of substance P occurred weakly on Day 7 and 14 after operation,and became stronger at late stage in IANS group.On the contrary,stronger immunoreactivites of substance P occurred at the early stage,less on day 21 posttrauma and stronger again on day 28 in control group.The strongest immunoreactivites at each stage occurred mainly in bone marrow on day 28 posttrauma in both groups.There was significant difference in the expressions of substance P in the callus between the two groups(P

9.Construction and identification of bicistronic retrovirus expression vector for Delta1 and EGFP gene coexpression

Objective To construct a bicistronic retrovirus expression vector to coexpress Delta1 and EGFP genes. Methods Delta1-IRES-EGFP bicistronic retrovirus vector was constructed by linking full-length Delta1 cDNA with IRES-EGFP retrovirus vector. Transfected with the retrovirus vector by electroporation, the ecotropic- and amphotropic-packaging cell lines GP+E86 and PA317 were selected by addition of puromycin. Positive cell clones were obtained and tested for the efficiency of the retroviral supernatants. Two positive cell clones with relatively highest titers were picked up and used for ping-pong amplification assay to improve the retroviral supernatants titer. At day 2 after transfection, GFP expression in NIH3T3 was detected by FACS, and Delta1 expression was determined by RT-PCR assay. Transfected or wild type NIH3T3 were co-cultured with myoblasts cell line C2C12 to test whether the exogenous Delta1 protein was functional or not. Results The bicistronic retrovirus expression vector coexpressing Delta1 and EGFP genes was successfully constructed and verified by PCR and DNA sequencing. A high-level expression of functional Delta protein was detected in transfected NIH3T3 cells. Conclusion Retrovirus is a safe, highly efficient gene transfer system.
10.Experimental study on protective mechanism of the combined treatment of pyrrolidine dithiocarbamate and mycophenolale mofetil on kidneys of diabetic rats

Objective To investigate protective effects of pyrrolidine dithiocarbamate (PDTC) and mycophenolate mofetil (MMF) on kidneys of diabetic rats and the possible mechanism. Methods Thirty-four streptozotocin-induced diabetic rats were randomly divided into 4 groups: diabetic aminals without treatment (D group), diabetic rats treated with PDTC (P group), diabetic rats treated with MMF (M group), diabetic rats treated with combined PDTC and MMF (P+M group), and normal rats were considered as control group (C group). Blood glucose, blood urea nitrogen (BUN), serum creatinine (Scr), and 24 h urinary protein (24 h UP) were detected at the end of treatment lasting for 8 weeks. Kidneys were weighed in order to measure kidney weight/body weight ratio (KI) after rats were killed. Pathological changes in the kidneys were observed by light microscope and electron microscope. Expressions of interleukin-18 (IL-18), intercellular adhesion molecule-1 (ICAM-1) and tumor necrosis factor-α (TNF-α) in renal glomerulus were detected by immunohistochemical staining.Results Blood glucose, BUN, Scr, 24 h UP, KI were significantly higher in D, P, M and P+M groups than those in C group (all P<0.05). BUN, Scr, 24 h UP, KI were significantly lower in P, M and P+M groups than those in D group (all P<0.05). 24 h UP was reduced in P+M groups compared with P,M groups (P<0.05). Pathological changes were attenuated in P,M and P+M groups compared with D group. Expressions of IL-18, ICAM-1, TNF-α in renal glomerulus were much higher in D group than those in C group (all P<0.05) and were reduced in P, M and P+M groups compared with D group (all P<0.05). Expressions of IL-18, ICAM-1, TNF-α in P+M group were lower than those in P group (all P<0.05), while expressions of IL-18, TNF-α were lower than those in M group (both P<0.05). Conclusion The protective effects on kidneys of diabetic rats produced by combined use of PDTC with MMF were much better than by the application of either PDTC or MMF alone. The mechanism appears to be related with suppressing expressions of inflammatory cytokines.
