Objective To evaluate the clinical significancy of laparoscopic intrafascial hysterectomy. Methods We reviewed 37 cases of laparoscopic intrafascial hysterectomy and 45 cases of open intrafascial hysterectomy. A controlled study on operative time, blood loss, time to first flatus, postoperative morbidity and complications was made. Results All operations in two groups were successfully completed, and no severe complications were seen. OBJECTIVE: Inter-oocyte cytoplasmic transfer has been performed recently as a means to improve the outcome of assisted reproductive methods and becomes a hotspot. To overview the recent advances and problems in this area in this article, and put forth a new concept of reverse cloning technique based on ooplasmic transfer. DATA SOURCES: An online search of PUBMED was undertaken to identify correlative English articles published from January 1998 to December 2006 by using the keywords of "ooplasmic transfer, mitochondria heteroplasmy,animal cloning". Meanwhile, relevant Chinese articles published between January 1998 and December 2005 were searched in Wanfang database with the same keywords in Chinese. STUDY SELECTION:The articles were primarily screened, and the citations at the end of each reference were reviewed. Inclusion criteria: The articles related to the research of ooplasmic transfer, cloning or reverse cloning. Exclusion criteria: Repetitive studies or Meta analytical articles. DATA EXTRACTION:Totally 126 relevant articles were collected, and 31 of them were in accordance with the inclusion criteria, while 95 of them were excluded because of repetition or obsolescence. In 31 articles met the criteria, 17 studies were relevant to the ooplasmic transfer, 13 studies related to animal cloning and one study related to reverse cloning. DATA SYNTHESIS: Exciting advances have been achieved in human gamete biology, and ooplasmic transfer have been widely applied. Many studies indicated that mitochondria in oocytoplasm affected obviously the fertilization of the oocytes and early embryo development. Lots of experiments have been done on the interaction of cytoplasm and nucleus as well as the mechanism of genomic reprogramming to further the investigation. Meanwhile, the techniques are progressed. Cloning techniques is excellent in application prospect, while there are some problems at the same time. Therefore the concept of reverse cloning technique was proposed based on ooplasmic transfer. CONCLUSION: The mitochondria in oocytoplasm has remarkable effect on the fertilization of the oocytes and early embryonic development. However, ooplasm transfer can lead to mitochondria DNA heteroplasmy, and more work must be done on potential problems. The reverse cloning technique based on the ooplasmic transfer has a good prospect.
4.Comparative study on the efficacy and safety of zoledronic acid and sodium ibandronate in treating bone metastasis in breast cancer patients

ObjectiveTo retrospectively evaluate the efficacy and safety of zoledronic acid and sodium ibandronate in treating bone metastasis in breast cancer patients.MethodsThe study included 47 patients who were treated with zoledronic acid and sodium ibandronate respectively from Aug.2006 to Mar.2011. KaplanMeier curve and Log rank test were adopted to detect the difference in survival time of skeletal related event (SRE) and survival rate between patients treated with different medicine,and x2 test was uscd to rcveal thc rate difference of pain killing effects and adverse effects.Results 1.The total effective rate of pain killing was 88.9% and 85% respectively.The difference had no statistical significance(P =0.467).2.The 1,2 and 3-year-survival rate in zoledronic acid group and sodium ibandronate group was 88.7%,44.4%,24.2%vs 94.7%,40.5%,5.8%.The difference had no statistical significance(P =0.744).3.The 2,3,and 4-year-survival rate in zoledronic acid group and sodium ibandronate group was 70.4%,40.7%,23.1% vs 85%,46.7%,17.5%.The difference had no statistical significance( P =0.994).4.The 1,2 and 3-year SRE-free overall survival rate of the group with metastasis first to bone only was 92%,50.8%,and 23.8%,while the rate of the group with first metastasis to visceral organs was 85.4%,21.4%,and 5.3% ( P =0.012).5.The 2 and 3-year overall survival rate of the group with metastasis first to bone only was 95.8% and 74.2% respectively,while the rate of the group with first metastasis to visceral organs was 56.5% and 10.1% ( P <0.001 ).The difference of 4 and 5 had statistical significance.The difference of zoledronic acid group and sodium ibandronate group in rate of adverse effects had no statistical significance.Conclusions Compared to sodium ibandronate,zoledronic acid has no superiority in either delaying the occurrence of SRE or improving overall survival rate.The -2are similar in incidence of adverse effects and pain control.The prognosis of patients whose first presentation is complicated by metastasis to other organs is poor.
5.Study on Formation Technics of Xuesaitong Drop Pills

OBJECTIVE:To study the optimum formation technics of Xuesaitong drop pill.METHODS:Parallel tests were conducted on the dosage of different base materials and the main drug with the forming percentage and the rate of qualified weight as the index of evaluation,the orthogonal test was conducted on the4factors,including the temperature of drops and the liquor condensate,the drug height in the drug storage tank and the dropping distance.RESULTS:The ratio of base materials and the main drug was2.5∶1.The optimum forming technics could be seen as follows,the height of the drug storage tank was3cm,the temperature of drops was90℃,the dropping distance was5cm and the temperature of the liquor condensate was12.5℃.CONCLUSION:There was a high rate of end product of dropping pill prepared with this optimum process,which was in conformity with the standard stated in the Chinese Pharmacopoeia.
6.Preparation Methods of Dripping Core Pills

OBJECTIVE: To develop a new form named dropping core pills which contain both water soluble and liposoluble constituents of the traditional Chinese medicine compounds. METHODS: Volatile, liposoluble and water-soluble constituents were extracted with supercritical fluid extraction-CO2,ethanol extraction, water decoction and membrane separation and other methods respectively. The dropping core pills were prepared with liposoluble and volatile constituents before being coated with water-soluble constituent. RESULTS: As compared with watered pills and dropping pills, dropping core pills had the advantages of both but overcome their shortages in that the dosage form was optimized, the amount of the reagent less and the resolving time was shorter. CONCLUSION: Dropping core pill was a new dosage form suitable for traditional Chinese Medicine compounds in which liposoluble and water-soluble components were both active compounds.
7.Expression of CXC chemokine receptor 5 in skin lesions of bullous pemphigoid and its significance

Objective To study the role of CXC chemokine receptor 5 (CXCR5) in the pathogenesis of bullous pemphigoid (BP) by detecting the expression of CXCR5 on follicular helper T (Tfh) cells in skin lesions of BP.Methods An immunohistochemical procedure was used to detect CXCR5 expression in biopsy specimens from skin lesions of 16 patients with BP and normal skin of 10 healthy individuals.Results CXCR5 was mainly expressed in the cytoplasm and membrane of Tfh cells in the basal cell layer of normal skin as well as in both basal cell layer and prickle cell layer of BP lesions.The number of CXCR5-positive ceils per high power field (× 400) was 11.16 ± 4.47 in normal skin specimens,significantly lower than that in lesional skin specimens (35.70 ± 12.20,t =6.07,P < 0.01).Conclusion CXCR5 may be involved in the pathogenesis of BP.

9.Human nerve growth factor beta-modified bone marrow mesenchymal stem cells from the rabbit mandible by transfection of lentiviral vectors

BACKGROUND:Central nerve damage and peripheral nerve injury are common clinical problems that have no ideal treatment. Nerve growth factor has an important role in neuronal repairing and growth. But its local injections may have shorts of inactivation and loss. OBJECTIVE:To construct human nerve growth factor beta recombinant plasmids, which are transfected into bone marrow mesenchymal stem cells from the rabbit mandible by lentiviral vectors, and to investigate the bioactivity of human nerve growth factor beta. METHODS:pDC316-hNGFβ-mCMV-EGFP plasmids were constructed via lentiviral vectors using Hind III+Not I digestion. Bone marrow mesenchymal stem cells from the rabbit mandible were isolated and cultured, and then transfected by recombinant plasmids. The expression of human nerve growth factor beta in transfected cells was detected by ELISA method. RESULTS AND CONCLUSION:pDC316-hNGFβ-mCMV-EGFP plasmids were proved to be constructed successful y by gene sequencing and enzyme identification. The transfected cells under a fluorescence microscope emitted green fluorescence, and the fluorescence intensity had no change with incubation time. The expression of human nerve growth factor beta was maintained at a level of 25μg/L at 7 days after celltransfection, and the bioacitivty was increased significnalty.
10.Clinical study on bacterial condition in amniotic cavity in the third trimester of pregnancy

Objective To study bacterial conditions in amniotic cavity of the third trimester and the delivery. Methods Patients underwent cesarean section due to various reasons were randomly enrolled in the study. Ten ml aqua amnii taken from using aspesis were cultured and the bacteria were identified and tested for drug sensitivity. All patients were not treated with antibiotics before and during operation,conventional antibiotics treatnent was used after operation. The patients were divided into 3 groups based on their status: 34 cases of premature rupture of membrane ( PROM ), thirty-four cases of labor without PROM, and 27 cases of pregnancy without PROM. The positive rate, species , distribution and drug sensitivity of bacteria in the 3 groups were analyzed. Results The positive rate was 61.8% (21/34) in PROM ,52. 9% ( 18/34 ) in labor without PROM,11.1% (3/27)in pregnancy without PROM. The positive rates were significantly different among the 3 groups (x2 = 17.29 ,P =0.000). However, there was no significant difference between the labor without PROM group and the pregnancy without PROM group ( x2 = 0.541 , P = 0.462 ). Staphylococcus epidermidis was the most common isolated bacteria, following by staphylococcus aureas, streptococcus, colon bacillus, and pseudomonas. There were no significant differences of bacterial species between labor without PROM and pregnancy without PROM( x2 = 11.9,P =0.535 ). The relative higher positive rate in the labor without PROM and PROM indicated that they were important inducement of bacteria infection. There were no significant difference on positive rate and bacteria species distribution between the PROM and labor without PROM group,which suggested that up-bound along vagina to amniotic cavity was the most common route of infection. Sensitive antibiotics should be given to the patients of PROM and labor without PROM targeted at staphylococcus,streptococcus, colon bacillus and monad. Antibiotics such as Cephems, Penicillins and Clindamycin were sensitive to various bacteria and safe to both mother and baby, thus were recommended. Quinupristin, Vancomycin,Furantoin, Fusidic ,Teicoplanin, Amikacin and Meropenem were also sensitive to various bacteria, but due to their side effects and the principle of ladder antibiotics using, they were recommended as second line antibiotics. Conclutions Up-bound along vagina to amniotic cavity is the most common route of infection. For rupture of membrane and labor without PROM patients, antibiotics should be given, and strict aseptic technique,washing cavity and incision, using antibiotics after operation are necessary to prevent infection.
