2.Optimization of Extraction Process of Effective Constituents from Rosae Laevigatae Frustus by Box-Behnken Response Surface Method

Objective To screen extraction process for Rosae Laevigatae Fructus.Methods With the content of total saponins and total phenolic acids from Rosae Laevigatae Fructus as the evaluation indexes, ethanol concentration, extraction time, solid-liquid ratio and extraction times as investigation factors, the test data were analyzed by Design-Expert8.0.5 software, and a multiple quadratic regression equation was set up to screen extraction process parameters of Rosae Laevigatae Fructus.Results The best extraction process parameters of Rosae Laevigatae Fructus were as follows:ethanol concentration was 62.79%;solid-liquid ratio was 13.12:1;the extraction time was 137.93 min;extraction times were 1.85 times. Considering the convenience for actual operation and the practicability of industrial production, the extraction process parameters should be ethanol concentration of 60%, revised solid-liquid ratio of 13:1, 2 hours' extracting time, and 2 extraction times.Conclusion Final selected parameters provide the basis for the extraction process of Rosae Laevigatae Fructus.
3.Effect of low power ultrasound on ovarian angiogenesis of rabbit

4.Relationship between dyslipidemia and blood pressure variability in patients with essential hyperten-sion

Objective:To explore the relationship between dyslipidemia and blood pressure variability (BPV)in pa-tients with essential hypertension.Methods:A total of 251 patients with essential hypertension (EH),who were treated in our department of outpatient from Jan 2011 to Dec 2012,were selected.There were 114 patients with EH complicated dyslipidemia were regarded as EH+dyslipidemia group and 137 pure EH patients reganed as pure EH group.All patients received ambulatory blood pressure monitoring (ABPM);BPV indexes were compared between two groups.Results:Compared with pure EH group,there were significant rise in standard variations (SD)of 24h systolic blood pressure (SBP)and diastolic blood pressure (DBP)[(10.59±2.04)vs.(14.96±4.12),(10.16± 2.11)vs.(12.34±3.19)],24h SBP coefficient of variation (SBPCV)[(0.13±3.07)vs.(0.91±2.21)];daytime (d)SBPSD and DBPSD [(11.27±1.97)vs.(13.98±4.03),(8.94±2.01)vs.(11.34±3.12)],dSBPCV [(0.79±0.21)比 (1.01±0.31)];nighttime (n)SBPSD and DBPSD [(7.96±3.14)vs.(13.22±5.37),(8.13±1.97) vs.(9.95±3.47)],and nSBPCV and nDBPCV [(0.78±0.18)vs.(0.09 ±0.43),(1.07±0.34)vs.(0.13 ± 0.37)]in EH+dyslipidemia group,P <0.01 all.Conclusion:There are relationship between dyslipidemia and blood pressure variability in patients with essential hypertension.
5.Preparation of Captopril Hydrophilic Gel Sustained-release Tablet and Influencing Factors on Its Release in Vitro

OBJECTIVE:To prepare captopril hydrophilic gel sustained-release tablet and study the influencing factors on its release METHODS:With HPMC as the matrix,the tablets were prepared by direct compression method and influencing factors on release were studied RESULTS & CONCLUSION:The in vitro release of prepared tablets conformed to Higuchi equation The HPMC matrix tablet could release in a sustained way when the proportion of HPMC was at least 15% in weight and the best proportion was 60%;The dissolution of Methocel K was slower than 60RT or 75RT;Taking lactose as the filler was better than starch or CaSO4;When the proportion of lactose increased,the dissolution sharply decreased;The tablet dissolved more rapidly with paddle method than with rotating basket methos,compression force and pH of dissolution medium affected the release very little
6.Expression of aquaporin 3 and aquaporin 9 in placenta and fetal membrane with idiopathic polyhydramnios

Objective To investigate the pathogenesis role of aquaporin 3 and aquaporin 9 in idiopathic polyhydramnios by detecting their expression and distribution in fetal membranes and placenta.Methods Twenty-one of term pregnancy women with idiopathic polyhydramnios were enrolled as patient group matched with 30 women with normal term pregnancy as control group.The expression and localization of aquaporin 3 and aquaporin 9 in fetal membranes and placenta were detected by real-time polymerase chain reaction and streptavidin peroxidase immunohistochemiscal staining.Results (1)The mRNA expressions of aquaporin 3 and aquaporin 9 were detected in amnion,chorion and placental tissue in both patient group and control group.Both aquaporin 3 and aquaporin 9 were demonstrated positive staining in the amnion epithelia,chorion cytotrophoblasts and placental trophoblast.(2)The ratio of aquaporin 3 and aquaporin 9 mRNA expressions in amnion in patient group comparing to those in control group were 5.00 and 3.25,while in chorion they were 2.03 and 2.08.When compared with those in amnion and chorion of control group,there was a significant difference(P<0.01).However,the relative change fold of aquaporin 3 and aquaporin 9 in placental trophoblast in patient group were decreased in comparison of those in control group,which also showed statistical difference(P<0.01).(3)The expression of aquaporin 3 and aquaporin 9 protein in anmion were 7.5 ±2. 0 and 11.1 ± 1.8 in patient group, while they were 5.3 ± 1. 6 and 5.6 ± 2. 3 in control group. In chorion, the expression of aquaporin 3 and aquaporin 9 protein was 7.5±2. 0 and 10. 0 ±1.6 in patient group, respectively, while in control group, they were 5.4 ±2.2 and 5.6±2. 1. When compared with those proteins in control group, it exhibited statistical difference (P<0.05). However, in placental trophoblast of patient greup,the expression of aquaporin 3 and aquaporin 9 protein were 3.5±1.4and 4. 0±2. 5, respectively, which were significantly decreased than 5.6±1. 3 and 7. 1±2. 9 in control group(P< 0. 05). Conclusions The alterations of aquaporin 3 and aquaporin 9 expressions in fetal membrane and placenta might be an adaptive response to idiopathic polyhydramnios. Further investigation should be needed to clarify the regulatory mechanism of aquaporin 3 and aquaporin 9 expressions.
7.Infertility in polycystic ovary syndrome treated with acupuncture and clomiphene: a randomized controlled trial.

OBJECTIVETo explore the best therapy for infertility caused by polycystic ovary syndrome (PCOS).
METHODSOne hundred and twenty patients were randomized into three groups, a clomi-phene group (group A), an acupuncture-moxibustion + Chinese medicine group (group B) and a clomiphene + acupuncture-moxibustion+ Chinese medicine group (group C), 40 cases in each one. In the group A, since the 5th day of menstruation, clomiphene was prescribed for oral administration. In the group B, on the 5th day of menstruation, warm needling therapy was applied at Zhongji (CV 3), Guanyuan (CV 4), Guilai (ST 29), etc. Additionally, the Chinese herbal medication for tonifying the kidney and activating blood circulation was provided. In the group C, the therapy as the group B was combined on the basis of the treatment as the group A. The treatment lasted continuously for 3 menstrual cycles. The endometrial thickness, endometrial type and cervical mucus score on human chorionic gon adotropin (HCG) day, and ovulatory cycle rate, clinical pregnancy rate and abortion rate after treatment were observed in the patients of the three groups.
RESULTS1) HCG day cervical mucus score, endometrial thickness and endometrial morphology (A type rate): the results in the group C were better than those in the group A (all P<0.01); the results in the group B were better than those in the group A (all P<0.05). The difference in the endometrial thickness was not significant in comparison between the group C and the group B (P>0.05). The cervical mucus score and endometrial morphology (A type rate) in the group C were better than those in the group B (both P<0.05). 2) The ovulatory cycle rates in the group A and group (C were higher than that in the group B (both P<0.05), the pregnancy rate in the group C was higher than that in the other groups (both P<0.05), and the early abortion rate in the group C was lower than that in the group A and group B (both P<0.01). 3) Follicle diameter from 18 mm to 20 mm and endometrial thickness: the differences were not significant between the normal pregnancy patients and the early abortion patients (both P>0.05). The endometrial morphology A type rate in the normal pregnancy patients was higher than that in the early abortion patients (P<0.05).
CONCLUSIONThe combined therapy of acupuncture, herbal medicine and clomiphene improves the pregnancy rate and reduces early abortion rate by effectively improving HCG day cervical mucus, endometrial thickness and morphology. The efficacy is apparently superior to the simple medication with clomiphene and the combined application of acupuncture and herbal medicine.

8.Bacteriostatic effect of extract of Ginkgo Ailoba leaves on Porphyromonas gingivalisinvitro

Objective To explore the antibacterial activity of extract of Ginkgo biloba leaves on Porphyromonas gingivalis in vitro ,and to provid pharmacological reference for developing a new type of antibacterial drugs in the treatment of periodontal disease.Methods This experiment was divided into negative control group,imipenem control group and different concentrations and forms of extract of Ginkgo biloba leaves groups.Solvent extraction method was used to extract the extract of Ginkgo biloba leaves, punching method and test tube method were performed to detect the antibacterial activity of extract of Ginkgo biloba leaves in anaerobic environment invitro and compared with Staphylococcusaureus and E.coli.By observing the antibacterial ring diameter and determination of the minimum bacteriostasis concentration (MIC),the antibacterial activities of extract of Ginkgo biloba leaves in vitro were measured.Results In the experiment of bacteriostatic ring,Porphyromonas gingivalis was treated with extract of Ginkgo biloba leaves,Ginkgo biloba leaf tablet and Ginkgo biloba soft capsule concentrate and 1∶4 diluent,the bacteriostatic ring diameters were decreased with the decreasing of the concentration.The maximum bacteriostatic diameter of Ginkgo biloba extract was 1 6.5 mm,and the maximum bacteriostasis diameters of Ginkgo biloba leaf tablet and soft capsule were 15.3 and 14.5 mm,respectively;the bacteriostatic diameter of the exact of Ginkgo biloba leaves was bigger than those of Ginkgo biloba leaf tablet and Ginkgo biloba soft capsule (P < 0.05), but there was no statistical difference of bacteriostatic diameter between Ginkgo biloba leaf tablet and Ginkgo biloba soft capsule (P> 0.05);E.coli and Staphylococcusaureus groups get the same results.When the concentration of extract of Ginkgo biloba leaves was more than 1.95 mg·L-1 ,there was no growth of Porphyromonas gingivalis but E. coli and Staphylococcus aureusa still grew;only the concentrations of exact of Girkgo biloba leaves were more than 6.25 and 12.5 mg· L-1 ,E. coli and Staphylococcus aureus didn’t grow;the bacteriostatic effect of extract of Ginkgo biloba on Porphromonas gingivalis was better than E.coli and Staphylococcus aureus . Conclusion Extract of Ginkgo biloba leaves has antibacterial effect on Porphyromonas gingivalis.
9.Analysis on Chemical Compositions and Antioxidant Activity of Different Processing Decoction Pieces of Rosae Laevigatae Fructus

Objective To investigate the honey and bran of processing for chemical compositions of Rosae Laevigatae Fructus.Methods With total polyphenols, total saponins, total polysaccharides and amino acids as the evaluation indexes, changes of the chemical compositions of these four chemical components in Raw product, honey products, bran products of Rosae Laevigatae Fructus were studied; in vitro activity of DPPH radical clearance for evaluation index, antioxidant activity of raw products, honey products, bran products of Rosae Laevigatae Fructus were compared.Results Raw products, honey products, and bran products of total saponin content were 3.332%, 4.880%, and 4.572%; total phenolic contents were 1.92%, 6.38%, and 7.30%; total polysaccharide contents were 35.99%, 40.38%, and 36.86%; the total amino acid contents were 0.67%, 0.76%, and 0.96%. Total polyphenol, total saponins, total polysaccharides, and total amino acids of Rosae Laevigatae Fructus after processing increased, in which the total polyphenol was most obvious; DPPH radical scavenging IC50 of raw products, honey products, and bran products were 0.47, 0.51, and 0.22 mg, respectively.Conclusion Compared with raw product, the content of chemical fractions in honey products and bran products of Rosae Laevigatae Fructus increase, and oxidation resistance is enhanced.
10.Building and use of the appraisal indicators system of hospitals brand effect

Literature review of the world on hospital brands, data collection in summary and comparison, and specialist consultancy were used to set the appraisal indicators, while the stratum analysis was used to calculate the weight coefficients of individual indicators, for building the appraisal indicators system for hospital brand effect. In addition, the indicators system and appraisal model were called into play to explore the use of hospital brands, offering scientific references for hospital management.
