Objective To construct a recombinant plasmid pLEGFP-mIFN? for developing the genetical therapy of tumor by stem cells. Methods PCR was used to amplify the mIFN? gene from plasmid pcDNA3.1-mIFN? in which the mIFN? was cloned.A new plasmid,pLEGFP-mIFN?,was then constructed by inserting the amplified mIFN? gene into pLEGFP-C1.Restriction analysis and sequencing were used to confirm the structure of pLEGFP-mIFN?.Neural stem cells were transfected by supernatant of PA317.Results A DNA fragment in size of 500 bp was amplified.Restriction analysis showed that the amplified gene was inserted into pLEGFP-C1 correctly.(Conclusion The) plasmid pLEGFP-mIFN? is constructed successfully, and it can transfect neural stem cells.
2.Factors influencing hemophilic children's ability in the activities of daily living
Chinese Journal of Physical Medicine and Rehabilitation 2013;(3):205-208
Objective To investigate the ability in the activities of daily living (ADL) of children with hemophilia and the factors which influence it.Methods Sixty boys aged 4-12 years with hemophilia were recruited.Their ADL ability was assessed using the functional independence score in hemophilia (FISH).Results The average FISH score of the sixty patients was 25.75 ± 5.71.The percentage of the patients who had experienced functional declines in self-care,transfer and locomotion abilities were 46.7%,55% and 75%,respectively.There was negative correlation between the patients' ages, the replacement therapy they had received and their FISH scores. The Spearman correlation coefficients were-0.371 and-0.388 respectively. The duration and frequency of the preventive therapy and appropriate exercise they had experienced were positively correlated with their FISH scores. The Spearman coefficients were 0.339.0.317 and 0.436 respectively. But there was no correlation between FISH scores and the level of coagulating factor in the subjects' blood, their frequency of bleeding in the most recent three months, muscle bleeding, the dosage of replacement therapy or the patient's place of residence. Conclusion Seventy-five percent of young, male hemophilia patients manifest a decline in ADL ability.Declines in locomotion occurred the most frequently. The ADL ability of young hemophilia patients probably declines with age.Long term preventive therapy and appropriate exercise would be helpful to maintain ADL ability.

ve To study the growth curve of Alexandrium tamarense and the toxicity of paralytic shelfish poisoning(PSP) selected in different growth phases, as well as to quantitatively analyze the toxins and compare the different efficiency of breaking cell wall and their virtues and defects between freezing-thawing method and supersonic method, and to provide theoretical basis for the selection of the methods for extraction of PSP. Methods Cells were collected by suction filter, cell wall was broken by freezing-thawing method and supersonic method, the toxicities of Alexandrium tamarense among different growth phases were detected and compared. Results The growth of Alexandrium tamarense typically showed three phases which included a lag phase(0~9 days), an exponential growth phase(10~18 days), and a stationary phase(19~22 days) . The maximum toxicity of cells in the stationary phase appeared, especially in day 21 when the population of cells reached to 9.43?10-6 MU/cell, although the cells in the exponential phase grew fastest. Toxicity of the cells in the lag phase broken by freezing-thawing method was more stronger than that by supersonic method, that meant the toxicity of cells was lowered after the supersonic treatment. In additional, the filtering method for collecting cells presented a lower recovery rate of average 69.0% . Conclusion As for the comparing of freezing-thawing method and supersonic method, it was found that, when cells were in the same phase, the supersonic method had remarkable advantages, but it also had the disadvantage of lowering the tox-icity of cells. When in the same condition of growth and method of fragmentation, cells in the lag phase were broken most easily, and cells in the stationary phase were more difficult to be broken than those in the other two phases. Toxicity of cells was larger and larger with the time of incubation. The toxicity of cells in the lag phase had reached a higher intensity, although toxicities of cells in the next two phases would increase a little.

Medical Journal of Chinese People's Liberation Army 1982;0(03):-
To study the repairing effect of NADH on PC12 cells damaged by chemical drugs, we determined the cytotoxicity of chemical drugs (cisplatin, doxorubicin and rotenone) on PC12 cells with cell proliferation test and MTT assay. At the same time, we analyzed the cyclin protein, GTPase binding protein and transglutaminase Ⅱ content with flow cytometry(FCM). NADH significantly inhibited the cytotoxicity of chemical drugs on PC12 cells. Cisplatin, doxorubicin and rotenone upregulated the expression of cyclin D1 and down regulated the expression of cyclin A and B1 in PC12 cells. On the contrary, NADH increased the expression of cyclin A and cyclin B1 and decreased the expression of cyclin D1. NADH could upregulate the expression of GTPase binding protein and transglutaminase Ⅱ, which were decreased by chemical drugs in PC12 cells. It is suggested that NADH could inhibit the cytotoxicity of chemical drugs to PC12 cells, which are associated with the regulation of cyclin proteins, GTPase binding proteins and transglutaminase Ⅱ.

Medical Journal of Chinese People's Liberation Army 1982;0(03):-
To investigate the melatonin receptor(MR) gene and protein expression in human HUT78 cell line, total RNA of HUT78 cells was isolated by single-step method of acid guanidinium-thiocyanate-phenol-chloform, and mt 1 and MT 2 mRNA were determined by reverse transcription-polymerase chain reaction(RT-PCR). The Envision method was applied to immunohistochemistry to identify and localize mt 1 and MT 2 protein. The mt 1 cDNA fragment of the expected size of 370bp was determined, but the MT 2 cDNA fragment of the expected size of 320bp was not determinable by RT-PCR. Sequencing result indicated that the positive product coincided with the cDNA of human mt 1. The mt 1 protein was observed by immunohistochemistry. These buffy positive granules were scattered with some areas stronger than the others, and were primarily located in cytoplasm and membrane, with rare location in nucleus; meanwhile the MT 2 protein was not observed. These results demonstrated the mt 1 mRNA and protein expression in HUT78 cell line. It is indicated that melatonin has directly immune-regulative effects on T lymphocyte, The changes of MR in physiological and pathological stages need to be investigated.
