1.Factors influencing hemophilic children's ability in the activities of daily living

Objective To investigate the ability in the activities of daily living (ADL) of children with hemophilia and the factors which influence it.Methods Sixty boys aged 4-12 years with hemophilia were recruited.Their ADL ability was assessed using the functional independence score in hemophilia (FISH).Results The average FISH score of the sixty patients was 25.75 ± 5.71.The percentage of the patients who had experienced functional declines in self-care,transfer and locomotion abilities were 46.7%,55% and 75%,respectively.There was negative correlation between the patients' ages,the replacement therapy they had received and their FISH scores.The Spearman correlation coefficients were-0.371 and-0.388 respectively.The duration and frequency of the preventive therapy and appropriate exercise they had experienced were positively correlated with their FISH scores.The Spearman coefficients were 0.339.0.317 and 0.436 respectively.But there was no correlation between FISH scores and the level of coagulating factor in the subjects' blood,their frequency of bleeding in the most recent three months,muscle bleeding,the dosage of replacement therapy or the patient's place of residence.Conclusion Seventy-five percent of young,male hemophilia patients manifest a decline in ADL ability.Declines in locomotion occurred the most frequently.The ADL ability of young hemophilia patients probably declines with age.Long term preventive therapy and appropriate exercise would be helpful to maintain ADL ability.
2.Analysis of transvaginal color Doppler ultrasound in diagnosis for 69 patients with endometrival lesions

Objective To analyze the sonographic characteristics of endometrial lesions to increase the diagnostic accuracy of the sonography. Methods 69 patients diagnosed as endometrial lesions by transvaginal color Doppler sonography(TVCDS) were retrospectively analyzed. The findings of TVGDS were compared with pathological diagnosis. Results The total diagnostic accordance rate of TVCDS was 91. 3% for endometrial lesions, including 93. 0% for endometrial polyps,91. 3% for endometrial hyperplasia,84. 2% for endometrial cancer, and 95. 0% for submucosal uterine fibroids respectively. In addition,the blood flow image rate and arterial resistance index(RI) had significant difference (P<0.05) between endometrial cancer and other benign lesions of endometrial, and all of these have obvious correlation with pathological findings. Conclusion TVCDS was an effective screening method for the diagnosis and differential diagnosis of endometrial lesions.
3.Construction and transfection of neural stem cells of recombinant plasmid pLEGFP-mIFN?

Objective To construct a recombinant plasmid pLEGFP-mIFN? for developing the genetical therapy of tumor by stem cells.Methods PCR was used to amplify the mIFN? gene from plasmid pcDNA3.1-mIFN? in which the mIFN? was cloned.A new plasmid,pLEGFP-mIFN?,was then constructed by inserting the amplified mIFN? gene into pLEGFP-C1.Restriction analysis and sequencing were used to confirm the structure of pLEGFP-mIFN?.Neural stem cells were transfected by supernatant of PA317.Results A DNA fragment in size of 500 bp was amplified.Restriction analysis showed that the amplified gene was inserted into pLEGFP-C1 correctly.(Conclusion The) plasmid pLEGFP-mIFN? is constructed successfully,and it can transfect neural stem cells.
4.The clinical value of intrahminal ultrasound in diagnosis of cesarean scar pregnancy

ObjectiveTo investigate the diagnosis value of intracardiac echocardiography in cesarean section scar pregnancy. MethodsCavity ultrasound was applied in patients of cesarean scar pregnancy with sonographic features and treatment. ResultsIn 52 patients, ultrasound diagnosed accurately in 45 cases, diagnosis accurate rate was 86.5% (45/52). Sonogram showed gestational sac simply type in 16 cases, mixed echo-type( or mass-based) of 36cases. In 52 cases of CSP, according to their clinical and sonographic features ,2 cases of uterine scar lined laparotomy and the uterus excision repair;three cases of laparoscopic removal of uterine scar pregnancy lesion repair. 6 patients to methotrexate;five cases of abdominal surgery;36 routine uterine arterial embolization in the ultrasound monitor after curettage. ConclusionUltrasound could apply in cesarean scar pregnancy timely,and accurate diagnosis was made;sonographic features according to the patients,help choose the appropriate clinical treatment,and assess efficacy.
5.A systematic review of the treatment of gastroesophageai reflux disease with mosapride in China

Objective To evaluate the efficacy and safety of mosapride in the treatment of gastroesophageal reflux disease (GERD). Methods Papers of PubMed,EMBASE,OVID,VIP Database and WANGFANG Data were searched and reviewed according to international Cochrane Collaboration.Results A total of 21 trials (1666 patients) were included.The treatment group was with mosapride and PPI,the control group was with single PPI or PPI and placebo.The Results of Meta analysis showed:for non-erosive negative reflux disease,there was no significant difference in efficacy between two groups.As for erosive esophagitis treated with mosapride (5mg):There was no significant difference in the 4-week healing rates between treatment group and control group (P =0.06).The 4-week symptom relief rate of treatment group was better than that of control group (P<0.01).The 6-week healing rate of treatment group was better than that of control group (P<0.01).The 6-week symptom relief rate of treatment group was better than that of control group (P<0.01).There was no significant difference in the 8-week healing rate of esophagitis between treatment group and control group (P =0.05).The 8-week symptom relief rate of treatment group was better than that of control group (P<0.01).For erosive esophagitis treated with mosapride 10mg,.The 8-week symptom relief rate of treatment group was better than that of control group (P=0.02).Conclusion According to the meta-analysis of these 21 trails,mosapride was safe and effective in the treatment of GERD.However due to the quality of the literatures,the above findings should be treated with caution.
6.Variation of serum ghrelin and leptin in patients with functional dyspepsia

Objective To investigate the alteration of serum ghrelin and leptin in patients with functional dyspepsia (FD) and their clinical significance. Methods Sixty patients with FD [-30 patients with postprandial distress syndrome(PDS) and 30 patinets with epigastric pain syndrome(EPS)] were enrolled. Thirty healthy subjects were severed as controls. The serum ghrelin and leptin were measured by enzyme linked immunosorbent assay and radioimmunosorbent assay, respectively. Results The concentrations of serum ghrelin and leptin were both significantly lower in patients with FD than in controls (P<0.01). Meanwhile, the concentrations of serum ghrelin and leptin in PDS patients were significantly lower compared with controls (P<0.01) and EPS patients (P<0.01). There was no significant difference in levels of serum ghrelin and leptin between controls and EPS patients(P>0.05). Conclusions The alterations of serum ghrelin and leptin in PDS patients are responsible for their reduction in FD patients. The PDS may he associated with abnormalities in gastrointestinal motility. There is an interaction between serum ghrelin and leptin in development of FD. The detection of serum ghrelin or leptin is helpful in classification and treatment of FD.
7.A study on function of knee and quality of life in patients with knee osteoarthritis

Objective: To investigate the effect of function of knee on quality of life (QOL) in patients with knee osteoarthritis (OA). Method: Forty patients with knee OA were recruited. Their function, pain and range of motion (ROM) of knee were assessed with Lequesne index, Lysholm knee score (LKSS), visual analogue scale (VAS) and protractor measure respectively. QOL was evaluated with arthritis impact measurement scale (AIMS). Result: The scores of AIMS, Lequesne and VAS in female were significantly higher than that in male. And the LKSS in female was significantly lower than that in male (P<0.05). There was a positive correlation between AIMS score and Lequesne index, VAS score and limit of ROM, but a negative correlation between AIMS and LKSS (P<0.05). Conclusion: The function of knee and QOL in female patients with OA were worse than that in male patients. Pain, limited ROM and weaken function in knee could effect QOL in OA patients to some extent. Health education and clinical treatment should be provided to improve the function of knee and QOL in OA patients.
8.Construction of CDK7 siRNA expression vector and its effect on cisplatin sensitivity of endometrial carcinoma cells

Objective:This study was aimed to investigate the influence of CDK7 siRNA on the sensitivity of endometrial carci-noma cell line HEC-1-A to cisplatin (DDP)-based chemotherapy. Methods:Different CDK7 siRNA fragments were synthesized based on the designs of the CDK gene sequence and were transfected into HEC-1-A. Real time reverse transcription polymerase chain reac-tion (RT-PCR) and Western blot analysis were employed to demonstrate the effects of transfection. The best CDK7 siRNA was chosen to specifically silence CDK7 expression in HEC-1-A.The sensitivity of the cells to DDP therapy before and after transfection was deter-mined by methyl thiazol tetrazolium (MTT) cytotoxicity assay, flow cytometry, and Hoechst/PI double-staining fluorescence microsco-py. Results:A total of four different CDK7 siRNA segments were designed and successfully transfected into HEC-1-A cells. The inter-ference effect in each group was confirmed by real time RT-PCR and Western blot assays. CDK7-423 was determined as the best per-forming CDK7 siRNA (over 70%) to transfect into HEC-1-A cells. MTT cytotoxicity test showed that IC50 of DDP decreased to a range from 45.122 μg/mL and 3.200 μg/mL after inhibition of CDK7 expression. DDP toxicity to the endometrial carcinoma cells sig-nificantly increased (P<0.05). Flow cytometry revealed that the average cell apoptosis rate significantly increased after the inhibition of CDK7 expression (11.66%to 37.57%, P<0.05). Similar results were observed using Hoechst/PI double-staining fluorescence microsco-py, and the number of apoptotic corpuscle demonstrated an apparent increase in the low CDK7-expressing group compared with the pa-rental cells. Conclusion:After the downregulation of CDK7 expression by CDK7 siRNA transfection, DDP chemotherapy sensitivity and apoptosis of endometrial carcinoma cells significantly increased. Further research is anticipated on the use of CDK7 as a new treat-ment target for endometrial carcinoma.
9.To Study the Clinical Manifestation and Endocrine Disorder of the Bulimia Nervosa

Objective Our aim is to study the clinical manifestation and endocrine disorder of the bulimia nervosa (BN). Methods 2 cases of BN were clinically analysed. Results Most of the patients were anorexia before the disease happened, with many aspects of endocrine disorders. Conclusions BN and amenorrea could result from hypothalamus function disorders. However, the alteration of corticoid,thyroid hormones may be results of emotion stress and body weight loss.
10.NADH REGULATES THE EXPRESSION OF TRANSGLUTAMINASE Ⅱ, CYCLIN PROTEINS AND GTPase BINDING PROTEIN IN PC12 CELLS DAMAGED BY CHEMICAL DRUGS

To study the repairing effect of NADH on PC12 cells damaged by chemical drugs, we determined the cytotoxicity of chemical drugs (cisplatin, doxorubicin and rotenone) on PC12 cells with cell proliferation test and MTT assay. At the same time, we analyzed the cyclin protein, GTPase binding protein and transglutaminase Ⅱ content with flow cytometry(FCM). NADH significantly inhibited the cytotoxicity of chemical drugs on PC12 cells. Cisplatin,doxorubicin and rotenone upregulated the expression of cyclin D1 and down regulated the expression of cyclin A and B1 in PC12 cells. On the contrary, NADH increased the expression of cyclin A and cyclin B1 and decreased the expression of cyclin D1. NADH could upregulate the expression of GTPase binding protein and transglutaminase Ⅱ, which were decreased by chemical drugs in PC12 cells. It is suggested that NADH could inhibit the cytotoxicity of chemical drugs to PC12 cells, which are associated with the regulation of cyclin proteins, GTPase binding proteins and transglutaminase Ⅱ.
