Objectives To evaluate the diagnostic value of real-time quantitative fluorescence polymerase chain reaction( Q-RT-PCR ) assay and immunofluorescence assay for diagnosis of hMPV. Methods Totally 1 283 children with acute respiratory infection admitted in Jiaxing Maternity and Child Health Care Hospital for treatment from November 2008 to May 2009 were recruited in this study. The hMPV positive stains were separated and sequenced in this area. The sequences between the local hMPV stains and Holland stains NLD00-1 were compared. The specific primers and fluorescent probe were designed according to the sequence of epidemic hMPV strain. The Taqman methodology was applied in Q-RT-PCR. Negative pressure suction was used to acquire nasopharyngeal secretions specimens. Both Q-RT-PCR and immunofluorescence with FITC labeled monoclonal antibody were used to analyze them, respectively. The McNemar, test was applied to analyze the correlation between the two methods. Results Totally 1 283 specimens were analyzed with Q-RT-PCR and immunofiuorescence simultaneously. Q-RT-PCR analysis showed there were 59 cases positive. Immunofluorescence analysis showed there were 55 cases positive. Fifty-two cases were positive in both assays. There were 7 cases positive in Q-RT-PCR assay but negative in immunofluorescence assay and 3 cases negative in Q-RT-PCR assay but positive in immunofluorescence assay. If Q-RT-PCR method was set as the golden standard, the sensitivity and specificity for immunofluorescence detection method were 88. 1%and 99. 8%, respectively. Positive predictive value and negative predictive value were 94. 5% and 99. 4%,respectively. There was no significant difference ( χ2= 0. 9, P > 0. 05 ) by McNemar' test between the two methods. Conclusion The diagnostic value of immunofluorescence assay is close to Q-RT-PCR assay.
4.An experimental study on the central effects of procaine.
Chinese Journal of Anesthesiology 1994;0(04):-
The analgesic, sedative and convulsive effects of procaine were determined by animal experiments. The analgesic ED50 of procain were 21.7mg/kg or 52.8ug/ each (iv or icv, hot plate) and 29.2mg/kg or 52.2ug/ each (iv or icv, electral stimulation) in mice.Procaine In subthreshold dose had additive hypnotic effect of phenobarbital in mice and rabbits, but could not de crease spontaneous activity in mice. The convulsive ED50 of procaine were 13.5mg/kg (iv) or 2.4mg/each (icv) in rabbits. There was no influence on the righting reflex in all the experiment animals when iv or icv procaine was given alone. These results suggest that the analgesic and sedative effects of procaine are weak, but may be potentiated when administered concomitantly With other potent drugs.
5.Effects of sodium oxybate on pharmacodynamics of ketamine in animals
Chinese Journal of Anesthesiology 1996;0(08):-
The interaction between sodium oxybate and ketamine were studied in conscious animals. Sodium oxybate increased the LD_(50) of ketamine, increased the incidence of sleep caused by ke tamine and prolonged the sleep duration and potentiated analgesic action of ketamine. Sodium Oxybate didn't effect the respiratory and circulatory function in rabbits. The results showed sodium oxybate po tentiated the anesthetic action of ketamine and reduced the side effect of ketamine. So It is suggested that sodium oxybate has the anesthetic synergism with ketamine in animals.

Objective To investigate the clinicopathological features of thymic neuroendocrine carcinoma and its differential diagnosis. Methods Clinicopathologic analysis and immunohistochemical staining were performed in 5 cases of TNC. Results For 5 cases TNC, 3 cases occurred in men, 2 cases occurred in women. The average age was 46.8 years. They were with chest symptoms before operation. 3 cases were Ⅱ and 2 cases were Ⅲ by clinic stages. Grossly , the tumors were gray in color with areas of hemorrhage and necrosis. Histopathology of well differentiated 2 cases and poorly differentiated 3 cases. Immunohistochemically, the tumor cells were positive for CK, NSE, SyN and CgA. 3 cases were positive for PCNA, Ki67, p53. Conclusions TNC has a pleomorphic appearance of histological features with well, moderate and poorly-differentiated areas. Immunohistochemical staining is helpful in the differential diagnosis of TNC from other tumors derived from the thymus. Neither grading as neuroendocrine carcinomas nor any individual histologic parameter showed a significant association with prognosis. Initial aggressive treatment, including complete surgical excision and adjuvant radiotherapy, appears to offer the best hope for prolonged survival. Adjuvant chemotherapy also should be considered, because the incidence of distant relapse is high.

Abstract Objective:To explore homology in mage-A family and find some common epitopes recognized by cytotoxic T lymphocyte(CTL) to oppose tumor escape. Methods:The amino acid sequence, differents open reading frame(ORF) and the known CTL epitope of themember of MAGE-A family, were analyzed using DNAstar software, and phylogenetic tree is also constructed. Results: MAGE-A family shareidentity nucleotide 57.6% - 98.0%,the phylogenetic tree showed that they are derived from a common ancestor at different time, as well asfound some CTL epitopes high similarity. Conclusion: MAGE-A family come from common an ancestor, but they bave many differences in thepattern of expression in tumor. This study can help to find the best CTL epitope vaccine to cure tumor.

Journal of Third Military Medical University 2003;0(07):-
Objective To explore the effect of prenatal inflammation on vascular remolding and blood pressure in mid-aged rats. Methods Time-mated pregnant Sprague-Dawley(SD)rats were randomly divided into 2 groups, received peritoneal injection of 0.79 mg/kg LPS on the gestation days 8,10,and 12,or same volume of the sterile saline at the same time points.Nine pups were randomly selected from each group for the later experiments, and the offspring were named as LPS groups for those having prenatal LPS exposure and control group for those having not.Their blood pressure was determined with a rat tail non-invasive instrument by tail-cuff method from 6 weeks old to 35 weeks old,and then their aortas were taken out for media thickness(MT),diameter of lumen(LD),and the ratios of MT/LD.The expression of proliferating cell nuclear antigen(PCNA)on the vessel was detected by ELISA.The serum level of NO and plasma endothelin-1(ET-1)were measured by nitrate reductase and radioimmunoassay respectively. Results Compared with control group,LPS groups had significantly raised blood pressure,a significantly higher ratio of MT/LD,obviously increased expression of PCNA,and markedly elevated serum ET-1.Conclusion The offspring whose prenatal rats were exposured to LPS result in vascular remolding, vasofunctional disturbances and hypertension.

Chinese Journal of General Practitioners 2002;0(01):-
Objective To investigate correlation between serum level of uric acid (SUA) and carotid atherosclerosis in the elderly. Methods A group of 113 elder subjects were examined by high-frequency Doppler ultrasound to measure their intima-media thickness (IMT) of carotid artery and severity of atherosclerotic plaque formation. Blood levels of biochemical factors such as SUA, total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), fasting blood sugar (FBS), creatinine (Cr) were determined, as well as 24h urine level of albumin (UALB). Correlation between SUA, IMT and the plaque was analyzed. Results SUA in quartile positively correlated with increasing IMT and severity of plaque in the elderly. SUA correlated with IMT of carotid artery, independent from age, gender, TC, TG, HDL-C, LDL-C and Cr, and correlated with severity of plaque adjusted for age, gender, history of hypertension, TC, TG, HDL-C, LDL-C, Cr, FBS and logarithm of UALB.Conclusions Level of SUA positively correlated with IMT, and increased level of SUA was a risk factor for carotid plaque, independent from other cardiovascular risk factors and renal impairment indicators, such as age, gender, history of hypertension, blood lipid level, Cr, etc.

Chinese Journal of Tissue Engineering Research 2007;0(25):-
Injectable bone substitutes can repair bone defect via percutaneous injection, including organic bone graft materials and inorganic bone graft materials. Furthermore, some materials considered as vector can composite with cells to generate tissue engineering materials, which are injected into bone graft to improve fusion rate. The development of injectable bone substitutes provides a new way to treat some relative diseases. Here, the article was divided three parts to introduce biological characteristics and clinical application of organic bone substitute materials, inorganic bone substitute materials, and bone tissue engineering vector materials. Although bone tissue engineering research achieves some effects initially, there are still a lot of problems, such as collection and preparation of extracellular matrix materials, composite and transplantation of cell and vector; all of them need to be further studied.
