6.Construction and expression of recombinant adenovirus carrying human endostatin gene
Chinese Journal of Tissue Engineering Research 2007;0(07):-
AIM:So far, endostatin is found playing the strongest effect to inhibit endothelial cells.Because endostatin protein is instable,it is difficult to prepare and apply.Gene therapy is used to assist endostatin to inhibit angiogenesis. This study constructed recombinant adenovirus of human endostatin gene using AdEasy-1 system to investigate the feasibility of expressing endostatin protein in ECV304 cells(human umbilical vein endothelial cells).METHODS:The experiment was performed at College of Life Science,Sun Yat-set University from March to October 2006.Endostatin was amplified from Pshuttle-Endostatin plasmid with PCR and subcloned into the pAdTrack-CMV shuttle vector after sequencing and identification. The resultant plasmid was co-transduced into E.coli BJ 5183 cells with pAdEasy-1 plasmid for homologous recombination. The recombinant plasmid was detected by restriction analysis and DNA sequence analysis. The linearized recombinant plasmid was transfected into AAV 293 cells with LipofectAMINE2000.The recombinant adenovirus was detected by GFP,PCR and restriction analysis. The virus was used to infect ECV304,and the expression of endostatin in vitro was detected by the techniques of RT-PCR and Western blot.RESULTS:The positive clones of the recombinants were constructed and confirmed by restriction endonuclease analysis and DNA sequence analysis.AAV293 cells were transfected. The titer of viral liquid was 2.06?1010 pfu/mL.The transcription of endostatin mRNA in ECV304 cells was checked by RT-PCR and endostatin protein could be detected in the culture by Western blot analysis.CONCLUSION:The recombinant adenoviral vector pAd-Endo carrying human endostatin is successfully constructed, and effective expression is found in ECV304 cells in vitro.
7.Role of PI3K/Akt in endothelial progenitor cells differentiation in vitro
Chinese Journal of Tissue Engineering Research 2007;0(24):-
BACKGROUND:The molecule mechanism underlying endothelial progenitor cells(EPCs) differentiation into mature endothelial cells remains poorly understood. It requires for the regulation of the expression of various genes inside cells by various signal pathways,PI3K/Akt maybe included.OBJECTIVE:To investigate the role of PI3K/Akt in EPCs differentiation.DESIGN,TIME AND SETTING:A group control in vitro experiment was performed at the Pathology Laboratory in Shengjing Hospital of China Medical University from September 2007 to March 2008.MATERIALS:Wistar rats aged 4-6 weeks,of either gender.METHODS:EPCs were isolated from rat bone marrow with density gradient centrifugation. Then difference-speed adherence screening method was used to obtain the second time adherent cells, which then were inoculated in culture flask. After 5 days of culture, cells were collected to be indentified with laser confocal microscopy. The differentiating EPCs were those that have positive results of both AC133 and vWF stainings.MAIN OUTCOME MEASURES:The mRNA and the protein expression of AC133,vWF,PI3K and Akt in EPCs were detected with Western blot and reverse transcription-polymerase chain reaction(RT-PCR) after 0,3,7,10 and 14 days of inoculation respectively.RESULTS:According to the RT-PCR and Western blot detection,AC133 showed the strongest expression at day 0,the weaker at day 3 and none at day 7,10 and 14(P
8.An experimental research on the analgesic effect and underlying mechanism of BTX-A for rat knee osteoarthritis
Chinese Journal of Rehabilitation Medicine 2017;32(6):649-653
Objective:To evaluate the analgesic effect of botulinum toxin type A (BTX-A) in osteoarthritis model and the changes of Nav1.8 protein expression in spinal ganglia changes.Method:Animal model of knee osteoarthritis (KOA) was established by intra-articular injection of 4% papain solution 0.3ml into SD rat right knee. After the formation of arthritis, they were randomly divided into two groups at the 2nd day:BTX-A group (n=10):intra-articular injection of 5μl BTX-A 0.1IU;WFI group (n=10):intra-articular injection of 5μl water. No papain or BXT-A was given to the sham group (n=10).At the 1st,3rd,5th day after injection,we tested the pain behavior, thermal pain threshold, and sodium channel 1.8 (Nav1.8) protein expression in spinal ganglion by using immunohistochemistry.Result:Analysis of spontaneous pain behavior showed abnormal gait caused by rat osteoarthritis.Comparing with the WFI group, abnormal gait caused by osteoarthritis improved significantly in BTX-A group. At day 5 it improved more significantly than day 1 and day 3.Thermal pain threshold of BTX-A group increased more than that of WFI group (P<0.05) at any time point.Abnormal high Nav1.8 protein in model rats decreased in spinal ganglia for BTX-A group. Conclusion:The intra-articular injection of BTX-A may play the analgesic effect in the model of KOA by down-regulating of the expression of Nav1.8 protein in spinal ganglia and reduction in the central sensitization to pain stimulation.

OBJECTIVE:To establish the method for simultaneous determination of 5 heavy metals in Vespae Nidus, and to dis-cuss the effects of difterent producing area and part on the contents of heavy metals in Vespae Nidus. METHODS: The contents of arsenic(As),mercury(Hg),lead(Pb),cadmium(Cd)and copper(Cu)were determined by atomic spectrophotometry(hydride generation method, cold vapor method, graphite furnace method and flame method). The difference in the contents of heavy metals in samples were compared among different producing areas by analyzing data. RESULTS:The linear range of As,Hg,Pb,Cu, Cd were 0-40 ng/mL(r=0.9930),0-0.9 ng/mL(r=0.9974),0-80 ng/mL(r=0.9955),0-8 ng/mL(r=0.9927),0-800 ng/mL(r=0.9966),respectively. Limit of quantitation were 4.473,2.35×10 - 3 ,8.380,2.128,54 ng/mL,limit of detection were 1.342,0.701× 10 - 3,2.514,0.632,16 ng/mL. RSDs of precision, stability and reproducibility tests were all lower than 5.0%. The recoveries were 84.26% -102.25%(RSD=7.71% ,n=6),90.90% -115.31%(RSD=9.38% ,n=6),92.16% -105.70%(RSD=6.19% ,n=6), 90.81%-113.99%(RSD=8.86%,n=6),91.63%-102.79%(RSD=5.09%,n=6). The phenomenon that heavy metals of sample from Shandong and Hebei exceed the standard was the most serious. Main heavy metal which exceed the standard was Pb. The con-tent of Pb in Chamber was higher than other parts. CONCLUSIONS: The method is simple, precise, stable and reproducible, and can be used for simultaneous determination of 5 heavy metals in Vespae Nidus. There are different contents of heavy metal in Ves-pae Nidus of different producing areas and parts.
