1.Effects of Glycosaminoglycan from Scallop Skirt on the Releasing of Vasoactive Substance from Endothelial Cells Injured by Hydrogen Peroxide

OBJECTIVE: To observation the effects of Glycosaminoglycan from Scallop Skirt (SS-GAG) on the production of vasoactive substance in the endothelial cells injured by hydrogen peroxide(H2O2). METHODS: In H2O2-induced en-dothelial cell oxidative damage model group which was established with human umbilical vein endothelial cell (HUVEC, ECV304) cultured in vitro, SS-GAG groups (at different concentrations) and the normal group, levels of 6-keto-PGF1a, Thromboxane-B2 and Endothelin (ET) in the culture fluid were detected by radioimmunity assay and the content of nitric oxide was determined by nitrate reductase method. 2.HPLC fingerprint of Xuefu Zhuyu Capsule

AIM:To establish a fingerprint method of analyzing Xuefu Zhuyu Capsule(Semen Persicae,Radix Angelicae sinensis,Fruclus Aurantii,Rhizoma Chuanxiong,etc.) by HPLC. METHODS: A HPLC in combination with PDA detector was applied with a HiQ C_(18)(4.6 mm?250 mm,5 ?m) column by gradient elution using methanol-1.0% acetic acid solution as the mobile phase.The flow rate was 0.5 mL/min and the detection wavelength was at 260 nm. RESULTS: Thitrty-six common peaks were obtained in the fingerprint of Xuefu Zhuyu Capsule with good separation,six of them were identified as adenosine,paeoniflorin,naringin,hesperidin,neohesperidin and ligustilide by comparing chromatogram with control compounds. CONCLUSION: The HPLC method with good precision and reproducibility can be used for the quality control of Xuefu Zhuyu Capsule.

4.Frequent epigenetic silencing of proapoptotic gene MAPK10 by methylation in B cell lymphoma

Objective To investigate epigenetic silencing of the proapoptotic gene MAPK10 by methylation in B-cell lymphoma. Methods We examined MAPK10 expression and methylation in seven cell lines derived from B-cell lymphoma by semi-quantitative RT-PCR and methylation-specific PCR (MSP), respectively. Methylation status was further examined in 24 diffuse large B-cell lymphoma (DLBCL), 15 follicular lymphoma (FL) and 12 reactive hyperplasia lymph nodes (LRH) both by MSP and bisulfite genomic sequencing (BGS). Results MAPK10 is silenced or downregulated in all seven B-cell lymphoma cell lines mostly due to promoter methylation. MAPK10 methylation was frequently detected in DLBCL (17 of 24, 71%) and FL (15 of 15, 100 %), but not in 12 LRH tissues. Conclusion MAPK10 is frequently inactivated by tumor-specific methylation, and thus, is a potential biomarker.

MMP-9 is one of the family of matrix metalloproteinases, which degrades extracellular matrix. MMP-9 is induced by many inflammatory factors, including IL-1?,IL-8 and TNF-?. Meanwhile, MMP-9 potentiates inflammatory factors by aminoterminal processing. The signal transduction by which inflammatory factors induce MMP-9 is also an important part in recent research. This review will give a summary in all these fields.

9.Structural equation model of relationship of coping style and social support with hope in maintenance hemodialysis patients

Objective To explore the relationship of coping style and social support with hope in maintenance hemodialysis patients.Methods A total of 177 maintenance hemodialysis patients were recruited from 3 dialysis centers in Guangzhou and were investigated with the self-designed questionnaire,the Herth Hope Scale,Simplified Coping Scale and Social Support Scale.Structural equation model was used to establish the model of relationship of coping style and social support with hope level.Results The average score of hope in maintenance hemodialysis patients was (31.83±3.69) points.The total score and all the sub-scores of social support were (42.24±4.09) points,(23.72±2.36) points,(10.82±2.26) points,(7.70±1.36) points respectively.And the average scores of positive coping style and the negative coping style were (2.15±0.28) points,(1.56±0.33) points,respectively.Social support had direct positive effect on hope level,and the positive and neg-ative coping style had indirect positive and negative effect on hope level respectively.Conclusions Social support has a direct effects on hope level in maintenance hemodialysis patients.Positive and negative cop-ing styles have indirect effects on hope level through social support.
10. Preparation and physicochemical evaluation of indomethacin sustained-release microspheres for ophthalmic delivery

OBJECTIVE: To obtain sustained-release indomethacin PLGA/Eudragit RS 100 microspheres for ophthalmic application and to evaluate its quality. METHODS: The microspheres were prepared by O/W solvent evaporation using PLGA and Eudragit RS 100. The morphological, drug content, entrapment efficiency, release behavior, particle size, residual organic solvent, Zeta potential of indomethacin PLGA/Eudragit RS 100 microspheres were determined; meanwhile, the cytotoxicity of blank microspheres was evaluated using PRE cells. RESULTS: Indomethacin microspheres containing PLGA and Eudragit RS 100 at the weight ratio of 1: 3 had even particle size distribution, and their surfaces were smooth under scan electronic microscope; and the mean diameters were (1676.4 ± 739.5) nm. The drug content and entrapment efficiency of the microspheres were (18.79 ± 0.19)% and (98.25 ± 2.11)%, respectively. Indomethacin was released slowly for a month from the microspheres, though burst release was observed within 24 h. The mean residual organic solvent was (267.33 ± 13.57) ppm. The zeta potential of all blank PLGA / Eudragit RS 100 microspheres was positive. The blank microspheres showed no sign of toxicity. CONCLUSION: Indomethacin PLGA / Eudragit RS 100 microspheres have a better clinical application prospect due to its high entrapment efficiency, narrow distribution, slow release and ocular safety.
