BACKGROUND:Exogenous neurotrophic factors or chemical induction can induce rat bone marrow mesenchymal stem cells to differentiate into neuron-like cells. However, exogenous inductors exert a short inducible action, and their chemical substances inevitably have a negative impact on cellviability to limit the application prospects of bone marrow mesenchymal stem cells to a certain extent.
OBJECTIVE:To investigate the effect of glial cellline-derived neurotrophic factor, green fluorescent protein gene transfection by adenovirus vector on biological characteristics of rat bone marrow mesenchymal stem cells, to observe the expression of glial cellline-derived neurotrophic factor and green fluorescent protein and the role of nutrition on bone marrow mesenchymal stem cells, and to explore the ability to differentiate into neuron-like cells induced by glial cellline-derived neurotrophic factor.
METHODS:The bone marrow mesenchymal stem cells at passage 3 were transfected by recombinant adenovirus (Multiplicity of infection=10, 50, 80, 100, 150, 200). The experiment had two groups according to target genes:bone marrow mesenchymal stem cells were transfected by Ad-GDNF-GFP in transfection group, and bone marrow mesenchymal stem cells were not transfected in control group. The expression of green fluorescent protein was detected by inverted fluorescence microscope. Transfection efficiency was calculated by flow cytometry. cells viability and the morphological changes of cells were compared respectively by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and inverted fluorescence microscope between the two groups. On days 5 and 10 after transfection, the expression of glial cel-derived neurotrophic factor mRNA was detected by PCR. On day 5, the expression of neuron-specific enolase was determined by immunofluorescence examination. On day 10, the expression of microtubule-associated protein 2 was identified.
RESULTS AND CONCLUSION:By the end of 12 hours after transfection, the green fluorescent protein expressed in cells, and the fluorescence intensity gradual y increased with time. When the multiplicity of infection was 100, the fluorescence intensity was strong and stable, and the transfection rate was nealy 90%on day 3 after transfection. cellviability in the transfection group was strengthened after transfection. On day 5 after transfection, bone marrow mesenchymal stem cells expressed neuron-specific enolase, and neuron-like protrusions gradual y extended. On day 10 after transfection, bone marrow mesenchymal stem cells expressed microtubule-associated protein 2 and glial cellline-derived neurotrophic factor mRNA, and exhibited neuron-like morphology and interconnected synpases. The recombinant adenovirus, Ad-GDNF-GFP, can highly transfect bone marrow mesenchymal stem cells when the multiplicity of infection is 100, and glial cellline-derived neurotrophic factor can promote the proliferation of bone marrow mesenchymal stem cells and induce bone marrow mesenchymal stem cells to differentiate into neuron-like cells.

Objective To investigate the effect of comprehensive nursing intervention on the quality of life of patients suffering from hypertension. Methods A total of 120 cases of essential hypertension were evenly randomized into observation group and control group. Both groups received conventional antihypertensive drug and nursing, and additionally, the observation group was given intervention of comprehensive nursing including dietary regulation, exercise training, emotional care and medication instruction. The intervention lasted 3 months. Before and after the intervention, traditional Chinese medicine ( TCM) symptom classifying list and World Health Organization ( WHO) QOL-BRIEF were applied to evaluate the therapeutic effect of the two groups. The changes of blood pressure, clinical manifestation and the quality of life were also compared between the two groups. Results (1) After intervention, the systolic pressure and diastolic pressure were both improved in the two groups ( P<0.05 or P<0.01 compared with those before treatment) , and the improvement in the observation group was superior to that in the control group (P<0.01). (2) After intervention, the scores of TCM symptom scores and QOL-BRIEF total scores were improved in the two groups ( P<0.05 or P<0.01 compared with those before treatment) , and the improvement in the observation group was superior to that in the control group ( P<0.01). Conclusion TCM comprehensive nursing intervention can effectively relieve hypertension and improve the quality of life of hypertension patients.
6. Regulation of dentin remineralization progress by non-collagenous protein analogues
Chinese Journal of Stomatology 2018;53(11):789-792
The use of various biomimetic methods to achieve remineralization of demineralized dentin and the formation of an organic matrix-inorganic mineral complex with a certain mechanical strength has been a research hotspot in recent years in the field of stomatology, and it also provides a new idea for the restoration of dentin defect. Dentin biomineralization is a process that simulates the mineralization of biological tissue in nature in which the remineralization of dentin collagen is induced and regulated by organic macromolecules. This review summarizes the process of remineralization of decalcified dentin regulated by non-collagenous protein analogues

ObjectiveTo observe the effect of Sanhuang Tangshenkang on the Wnt/β-catenin signaling pathway in the bone tissue of diabetic rats. MethodA high-sugar and high-fat diet was administered for 4 weeks, along with intraperitoneal injection of freshly prepared 2% streptozotocin (pH 4.5) at 30 mg·kg-1 body weight to induce a diabetes model in rats. The rats with diabetes were randomly divided into model group, low- and high-dose Sanhuang Tangshenkang groups (12.8, 38.4 g·kg-1), and Gushukang group (1.8 g·kg-1) according to the blood glucose level. Rats of the same age were fed on a regular diet and assigned to the control group. After 12 weeks of respective treatments with drugs or physiological saline, the fasting blood glucose (FBG) levels of the rats were measured using an automated biochemical analyzer. Enzyme-linked immunosorbent assay (ELISA) was used to detect fasting serum insulin (FINS), bone-specific alkaline phosphatase (BALP), and tartrate-resistant acid phosphatase (TRAP) levels. Micro-computed tomography (Micro-CT) was used to scan the femurs of rats to observe bone tissue microstructure and measure bone mineral density (BMD). Hematoxylin-eosin (HE) staining and safranin O/fast green staining were performed to observe pathological changes in the femoral bone tissue. Immunohistochemistry (IHC) and Western blot were used to detect the expression of Wnt3a, low-density lipoprotein receptor-related protein 5 (LRP-5), and β-catenin proteins. ResultCompared with the control group, the model group showed a significant increase in FBG, FINS, and TRAP levels (P<0.01), a significant decrease in BALP level (P<0.01), a significant decrease in BMD (P<0.01), and disorganized, elongated, and sparse bone trabecular structures with fractures and increased lipid droplets. Additionally, the expression of Wnt3a, LRP-5, and β-catenin proteins decreased (P<0.05, P<0.01). Compared with the model group, the low- and high-dose Sanhuang Tangshenkang groups showed a reduction in FBG and an increase in BALP (P<0.05). The low-dose Sanhuang Tangshenkang group also exhibited a decrease in FINS (P<0.05). All treatment groups showed a significant decrease in TRAP (P<0.01), varying degrees of improvement in BMD (P<0.05, P<0.01)), increased and denser bone trabeculae with more regular arrangements and reduced lipid droplets, and improved bone microstructure morphology. The average optical density values of Wnt3a, LRP-5, and β-catenin proteins were significantly increased in all drug-treated groups (P<0.05, P<0.01), and the expression of Wnt3a, LRP-5, and β-catenin proteins was elevated (P<0.05, P<0.01). ConclusionSanhuang Tangshenkang may regulate the imbalance of the Wnt/β-catenin signaling pathway by increasing the expression of Wnt3a, LRP-5, and β-catenin proteins in bone tissue, which may promote bone formation, reduce bone resorption, and lower blood glucose levels, thereby achieving the effect of preventing and treating diabetic osteoporosis.

Objective To compare the clinical outcomes of transcatheter aortic valve implantation (TAVI) in oncology and non-oncology patients with severe aortic stenosis (AS). Methods A computer-based search in PubMed, The Cochrane Library, EMbase, CBM, CNKI and Wanfang databases from their date of inception to December 2021 was performed, together with reference screening, to identify eligible clinical trials. Two investigators screened the articles, extracted data, and evaluated quality independently. RevMan 5.3 and Stata 12.0 softwares were used for meta-analysis. Results The selected 8 cohort studies contained 57 988 patients, including 12 335 cancer patients and 45 653 non-cancer patients. The results of meta-analysis showed that in patients with cancer, the 30-day mortality [OR=0.74, 95%CI (0.65, 0.84), I2=0%, P<0.000 01], stroke [OR=0.87, 95%CI (0.76, 0.99), I2=0%, P=0.04] and acute kidney injury [OR=0.81, 95%CI (0.76, 0.85), I2=49%, P<0.000 01] were lower than those in patients without cancer. The 1-year mortality [OR=1.46, 95%CI (1.15, 1.86), I2=62%, P=0.002] and late mortality [OR=1.51, 95%CI (1.24, 1.85), I2=61%, P<0.000 1] were higher in patients with cancer. Conclusion It is effective and safe in cancer patients with severe AS undergoing TAVI. However, compared with patients without cancer, it is still high in long-term mortality, and further study of the role of TAVI in cancer patients with AS is necessary.
