1.The relationship between electrophysiological and morphological properties of neurons in visual cortex of developing rat-intracellular staining study

Objective To investigate the relationship between electrophysiological and morphological properties of neurons in visual cortex of developing rat, speculate the coincided degree between electrophysiological and morphological change and realize the mechanism of normal visual development. Methods Whole cell patch-clamp recording and intracellular staining were used to acquire cellular microelectrode recording in visual cortex from Sprague-Dawley rats (4~28 days old). The histological process was made. Results The differences of electrical feature between pyramidal cells and non-pyramidal cells were significant. The morphological maturity degree is different in developing visual cortex. Conclusion The different function of pyramidal and non- pyramidal cells in local integrition is reflected by their electrical feature in the process of visual development. In critical period of visual development, the coincision degree of the electrophysiological and morphological change in visual cortex is larger than that in the subcortex constructure.
2.Effect of simvastatin on IL-6 and adiponectin secretion and mRNA expression in 3T3-L1 adipocytes.

In order to investigate the effects of simvastatin on secretion and mRNA expression of interleukin-6 (IL-6) and adiponectin in 3T3-L1 adipocytes, mouse 3T3-L1 adipocytes were stimulated with lipopolysaccharide (LPS). Production and mRNA expression of IL-6 and adiponectin in 3T3-L1 adipocytes were measured using enzyme-linked immunosorbent assay (ELISA) and reverse transcriptase polymerase chain reaction (RT-PCR), respectively. The results showed that simvastatin could significantly suppress LPS-induced IL-6 production and mRNA expression in adipocytes (P<0.05), but increase the LPS-induced adiponectin secretion and mRNA expression in a dose-dependent manner (P<0.05). It was suggested that simvastatin could exert beneficial effects on prevention of obesity-induced metabolic changes in adipocytes.
3.Clinicopathological study of the alveolar soft part sarcoma with 16 cases reports

Purpose To investigate the clinicopathological changes, immuniohistochemistry and molecular genetics phenotypic charac-teristics of the alveolar soft part sarcoma ( ASPS) . Methods 16 cases of ASPS were studied with clinicopathological, cytochemistry technique and immunohistochemical staining, two cases of ASPS were studied by FISH. Results There were 6 males and 10 females with the age 8~58 years (median age 31. 7 years). The tumors were located at limbs, shoulder and back, tongue, vocal cords, lung, cervix, and ureter. The clinical manifestations of the patients was a slowly growing mass. Histopathologically the tumor showed typical organ-like or acinar-like structure with sinus-like blood vessels and the fibrous septa formation. Sometime the clear or abundant eosino-philic granular cytoplasm of the tumors were obvious. The tumor cells had a crystalline substance formation by PAS staining. The tumor cells were positive for TFE3 and Cathepsin K by immunohistochemical staining. The ASPL-TFE3 gene fusion detection of tumor cells were present. Conclusion ASPS often located on the limbs of young patients. It may misdiagnosed as malignant epithelial tumors, primary or metastatic adenocarcinoma and paraganglioma when the tumor locate on a rare anatomical parts or an organs, such as tongue, vocal cords, cervix, ureter, etc. It is valueable that the typical alveolar-like structure of the tumor and the expression for TFE3 and Cathepsin K for the pathological diagnosis of ASPS. It is an important indicator that the ASPL-TFE3 gene fusion detection by FISH for the tumor.

The leading teachers' responsibility system in laboratory medicine course should be promoted.
6.Pathogenic Bacteria in Respiratory Infection in Newborns:Their Distribution and Drug Resistance

OBJECTIVE To investigate the distribution and drug resistance of pathogenic bacteria which caused respiratory infection among suscepted patients and offer scientific basis for reasonable usage of antibiotics.METHODS Oropharyngeal swabs among 709 cases of respiratory infection neonates were investigated by the routine methods and drug resistance was analyzed by K-B method.RESULTS Totally 438 bacterial strains were isolated from 709 neonates.most of these bacteria were Gram-negative bacilli(70.3%),among which Haemophilus influenzae and Klebsiella pneumoniae were accounted for 39.8% and 10.3%,respectively;fungi and Gram-positive cocci were accounted for 23.5% and 6.2%.CONCLUSIONS Most strains present higher resistance rates to penicillin and ampicillin;but cefoxitin,amikacin,vancomycin,imipenem and the third generation cephalosporins are revealed with higher sensitivity rates for pathogenic bacteria in newborns.
7.Male breast cancer:a report of 38 cases

Objective To explore the clinical characteristics,diagnosis,treatment,and prognosis of male breast cancer.Methods The clinical data of 38 cases of male breast cancer,who were treated in the two hospital during the past 10 years,were analyzed retrospectively.The diagnosis was mainly based on preoperative B-ultrasound,radiography with molybdenum target tube and fine needle aspiration.Among the 38 cases,radical mastectomy was conducted in 3 cases,modified radical mastectomy in 27 cases,simple mastectomy in 5 cases,and operation was refused in 3 cases.Postoperatively,radiotherapy was adopted in 4 cases,simple endocrinotherapy in 7 cases,radiotherapy plus chemotherapy in 9 cases,and chemotherapy plus endocrinotherapy in 8 cases.Results Median age at treatment was 58.5(53-82)years.TNM staging included stageⅠin 13 cases,stageⅡ in 19 cases,stageⅢ in 4 cases,and stageⅣ in 2 cases.Median follow-up was 73.8 months(2 months-10 years),and follow-up rate was 73.6%(28/38).During the follow-up,13 cases died including 8 cases died of local recurrence and metastasis,and 5 cases died of heart and brain disease.The 5-and 10-year overall survival rate was 71.6% and 55.3% respectively;5-and 10-year disease-specific survival of stageⅠ,Ⅱ was 92.6% and 78.9% respeoively;among 6 cases of stageⅢ,Ⅳ 4 were followed,and all 4 cases died with in 3 years.Conclusions Male breast cancer has a long course,poor prognosis and low survival rate.Combined therapy with modified radical mastectomy,as its basis,is the treatment of choice for male breast cancer.There is a certain correlation between the prognosis of male breast cancer and clinical stage of the disease.

10.E-cadherin-transfected neural stem cells transplantation for spinal cord injury in rats.

The effects of E-cadherin-transfected neural stem cells (NSCs) transplantation for spinal cord injury (SCI) in rats were investigated. Sixty SD rats were randomly divided into model control group, NSCs group, empty plasmid group and E-cadherin overexpression group (n=15 each). The animal SCI model was established by using the modified Allen's method. NSCs were cultured. Rats in NSCs group were subjected to NSCs transplantation. E-cadherin gene eucaryotic expression vector and pcDNA3.1-E-cadherin were respectively transfected into cultured NSCs, serving as empty plasmid group and E-cadherin overexpression group respectively. At 7th day after transplantation, neurological function of all rats was assessed by Tarlov score. After rats were sacrificed in each group, the number of BrdU and Nestin positive cells was counted by immunohistochemistry. Immumofluorescence method was used to detect the expression of neurofilament protein (NF) and glial fibrillary acidic protein (GFAP). As compared with model control group, the Tarlov score and the number of of BrdU and Nestin positive cells, and the expression of NF and GFAP in NSCs group, empty plasmid group, and E-cadherin overexpression group were increased significantly (P<0.05), and those in the E-cadherin overexpression group were increased more significantly than the other transplantation groups (P<0.05). It was suggested that E-cadherin could be conductive to nerve regeneration and repair probably by promoting the proliferation and differentiation of NSCs.
