Objective To explore the characteristics of DNA antigen deposited in the skin lesions of patients with lupus erythematosus (LE). Methods Thirty-two LE skin specimens were collected. The deposited immune complexes were obtained by cryoprecipitation methods. Then the samples were digested by protease. Finally DNA was extracted with phenol and chloroform. The size of DNA fragments was detected on agarose gel electrophoresis. Ten kinds of probes were used to analyze the origin of these DNA molecules by dot hybridization. Results DNA fragments were successfully isolated from 27 skin specimens with four kinds of different sizes including band-I (20 000 bp), band-Ⅱ (1 300 bp), band-Ⅲ (800-900 bp) and band-Ⅳ (100-120 bp). In 15 specimens most of DNA was identified as band-I. In 2 specimens band-Ⅰ, -Ⅱ and -Ⅲ were all noticed, while all four bands were detected in 10 specimens. There was a positive correlation between small-sized fragments (100-200 bp) and disease activity (P

Objective To evaluate the effects of different dialysis treatment on femoral artery elasticity in uremia patients by Echo-tracking technique. Methods 81 cases of uremia patients (42 peritoneal dialysis patients and 39 hemodialysis patients) and 31 cases of normal adults as control were recruited in this study. The Intima-media thickness (IMT), Internal systolic diameter (Ds), Internal diastolic diameter (Dd), Value difference between Dd and Ds (△D), Arterial stiffness (β), Pressure strain elastic modulus (Ep), Arterial compliance (AC), Pulse wave conducting velocity (PWVβ) and Augmentation index(AI) parameters were analyzed for evaluation the right femoral arterial elasticity. Results The parameters ofβ, Ep, PWVβin dialysis patients significantly increased (P<0.05) and their AC significantly decreased (P<0.05) compared with normal group. Theβand Ep in hemodialysis patients is higher that those patients not receiving dialysis (P<0.05). Theβand Ep in peritoneal dialysis patients is lower that those patients not receiving dialysis (P<0.05). Theβ, Ep and PWVβin hemodialysis patients are higher than those in peritoneal patients (P<0.05). And AC is lower than control group (P<0.05). In uremia patients, Vascular stiffness was positively correlated with age, duration of dialysis, IMT, pulse pressure, Dd, and negatively correlated with△D(P<0.05). Conclusions Femoral artery endothelial function in uremia patients was damaged earlier than morphological changes. Vascular intimal injury resulting from hemodialysis is more serious, and vascular elasticity decrease is correlated with age, dialysis duration and pulse pressure. Echo-tracking technique could provide quantification evidences for early diagnosis of the femoral artery endothelial dysfunction in uremia patients. The echo-tracking technique has certain significance in early intervention and improvement the prognosis, however, the influence factor of the accuracy of measurement should be consider during the procedure.

Objective:To discuss the relationship between blood-lipid change and acute cerebrovascular disease (ACVD) and tocompare the blood-lipid levels in the patients with cerebral hemorrhage (CH group) with those in the patients with cerebralinfarction (CI group). Methods:The blood-lipid levels in 300 patients with ACVD (ACVD group) and 100 patients withother system disease (control group) were tested with automatic biochemistry analysor and compared between the twogroups. Results:There were significant differences between ACVD group and control group as well as between CH group andCI group. Conclusion:Although there were blood-lipid metabolic disturbances in both CH and CI patients, their degrees weresignificantly different, which should be closely observed and controlled in clinic.

Journal of Medical Postgraduates 2001;14(3):207-209,212
Objectives:To explore the antiepileptic cellular mechanism of chronic vagus nerve stimulation (VNS). Methods: The rats were kindled by intraperitoneal injection of pentylenetetrazol (PTZ) (45mg/kg weight), and chronic VNS was applied to the kindled-rats. The effects of glutamate (Glu) on the intracellular free calcium ([Ca2+]i) of cortical and hippocampal neurons of normal, PTZ-kindled and VNS-treated rats were examined with fluorescence spectrophotometry respectively. The behavioral changes of all rats were observed. Results: The neuronal [Ca2+]i of the VNS-treated rats were significantly lower than that of the PTZ-kindled rats. VNS could significantly inhibit the augmentation of neuronal [Ca2+]i induced by Glu. Behavioral changes demonstrated that VNS could significantly reduce the severity and prolong the latency of seizures of PTZ-kindled rats. Conclusions: VNS could significantly inhibit the kindling effect of PTZ and reduce the excitability of neurons induced by Glu. VNS might exerted the antiepileptic effects by influencing the activation of neural receptors.

BACKGROUND:Astragaloside Ⅳ is a major component of Huangqi, promoting proliferation and differentiation of bone marrow mesenchymal stem cells;however, the mechanism has been less reported yet.OBJECTIVE:To explore the effect of Astragaloside Ⅳ on expression of multiple hematopoietic growth factors in bone marrow mesenchymal stem cells.METHODS:Bone marrow mesenchymal stem cells were isolated from adult Wistar rats by using the method of adhesive culture and clone, and they were then plated on 96-well plate and separately incubated with 100 uL Astragaloside Ⅳ(25,50,100,200 g/L)for 72 hours. The cells in the control group were cultured with an equal volume of DMEM-LG culture liuquid.Indirect immunofluorescence was used to detect the biological activity,MTT method was used to evaluate the effect of Astragaloside Ⅳ on proliferation and differentiation of bone marrow mesenchymal stem cells, and RT-PCR method was used to measure the expression of hematopoietic growth factors in bone marrow mesenchymal stem cells.RESULTS AND CONCLUSION:The 3~(rd)-passage bone marrow mesenchymal stem cells highly expressed CD44 but lowly expressed CD45.As compared with control group,Astragaloside Ⅳ promoted proliferation of bone marrow mesenchymal stem cells in a time/dosage-dependent manner,in particular, the 200 g/L Astragaloside IV and 72-hour intervention(P< 0.05).SCF expression was significantly increased in the drug group compared with control group(P < 0.01);however,TPO,GM-CSF,and TGF-β1 expressions were not changed significantly(P > 0.05).Moreover,interleukin-3 expression was not found in the bone marrow mesenchymal stem cells.Astragaloside Ⅳ promoted in vitro proliferation of bone marrow mesenchymal stem cells, possibly involving in SCF secretion.
