5.Comparison of Dynamics in Viremia Levels in Chickens Inoculated with Marek's Disease Virus Strains of Different Pathotypes

One-day-old unvaccinated chicks were inoculated with serotype I virulent strain GA and vaccine strain CVI988, serotype III vaccine strain HVT o f Marek's disease viruses(MDV). By using an indirect immunofluorescence assay of suspensions of peripheral blood mononuclear cells(PBMC) with monoclonal antibo dies(MAb) BA4 and BD8 specific to glycoprotein B of MDV, The dynamic course s of vir emia levels for each of 3 strains from 4 days to 45 days postinfection (PI) were determind. Viremia for virulent strain GA-infected chickens were detected from 4 days PI until days before death of infected chickens, it got its peak at abou t 14 days PI. Viremia in serotype 1 vaccine CVI988-inoculated chickens were mea sured between 4 to 22 days PI, it could be detected from 4 days PI and ended at 18 days PI, it got its peak at 8 days PI. For serotype 3 vaccine HVT-inoculated chickens, viremia could be detected beween 4 to 14 days PI, its peak happened a t about 6 days PI. Such IFA with MAbs BA4 and BD8 could be used to evaluate qu al ity of vaccination process in chicken flocks. The differential dynamic courses o f viremia for different strains may be used to diagnose virulent MDV infection i n vaccinated flocks. The viremia levels measured by IFA with the MAbs and co-cu ltivation assay for plague forming unit(PFU) were compared. The results indicate d that IFA assay of PBMC suspension was more sensitive and specific than co-cu ltivation assay, i.e. viremia levels measured by IFA were 30 to 100-fold higher than that by plague-forming assay in CEF for the same sample.
6.Acute action of glucocorticoid on catecholamine secretion of adrenal chromaffin cells in rats

By using the whole-cell clamp-patch technique, the effects of dexamethasone on calcium channel current (Ica) and nicotine receptor channel current (I_(NIC)) were examined. The acute action of glucocorticoid on adrenal medullary chromaffin cell (AMCC) of rats was a significant inhibition of I_(NIC), while no apparent effect was observed on Ica induced by electricity. The results suggest that the acute action of glucocorticoid on catecholamine release in rat AMCC may be directly related to nicotine receptor.
7.Effects of calcium channel blockers on nicotinic acetylcholine receptor current in rat chromaffin cells

AIM: To study the effects of calcium channel blockers (CCB) on nicotinic acetylcholine receptor current (I_(NIC)) in rat adrenal medullar chromaffin cells (RAMCs). METHODS: By using the whole-cell clamp-patch technique, we have investigated the effects of nifedipine(NIF)、?-conotoxin GVIA and ?-agatoxin IVA on I_(NIC) induced by nicotine(NIC) before and after RAMCs perfusion. RESULTS: After perfusing RAMCs for 5 min, different kinds of calcium channel blockers at different concentration showed significant inhibitory effects on I_(NIC) induced by 50 ?mol/L NIC. The peak inhibition rates of 10 ?mol/L NIF、400 nmol/L ?-conotoxin GVIA and 100 nmol/L ?-agatoxin IVA were (61.7?5.1)%,(29.3?7.4)% and (17.6?7.5)%, respectively. CONCLUSION: The acute effects of different kinds of CCBs on RAMC were that they obviously inhibited I_(NIC) induced by NIC. These results suggest that CCBs may inhibit catecholamine secretion by directly blocking nicotinic acetylcholine receptor channel. [
8.A novel chromene with anti-tumor activities from fungus Phomopsis sp.

A new chromene (1) and six known compounds identified as 6-hydroxymellein (2), 6-hydroxy-5-methylmellein (3) nectriapyrone (4), chermesinone A(5), chermesinone B(6), and pomopxanthone A(7), were isolated in our investigation of the cytotoxic constituents from the fermented rice substrate of endophytic fungus Phomopsis sp. HCCB03519. The structures of these com pounds were elucidated through spectroscopic data analysis. All compounds exhibited inhibitory activities against cancer cell lines. Compound 7 showed stronger inhibition against cancer cells than the positive control 5-Fu.

9.Effect of Local Delivery of Paclitaxel on Anastomotic Wound Healing of the Common Bile Duct in Guinea Pig

Objective To investigate the effect of local delivery of paclitaxel on bile duct scar formation and biliary stricture in guinea pig.Methods Models of end-to-end anastomosis of the common bile duct were established in 42 guinea pigs.The animals were randomly divided into paclitaxel and control groups.In the paclitaxel group,the drug solution(1000 ?mol/L,0.05 ml) was smeared onto the anastomotic wound before closing the abdomen.The animals were killed in 3 days,2 weeks,and 1 month after the operation(7 in each group at each time point).The sepcimens of the common bile duct were obtained for histological examination.Transmission electron microscopy was employed to observe the ultrastructure of paclitaxel-induced apoptotic cells.Results Histological examination showed inflammatory reaction and hyperplasia of the bile duct wall.The bile duct wall of the paclitaxel group was significantly thiner than that of the control group in 3 days and 1 month after the operation [3 days:(574.41?24.53)?m vs(802.98?24.42) ?m,t=-6.604,P=0.000;1 month:(1383.36?36.64) ?m vs(1518.56?34.89)?m,t=-2.672,P=0.020].Paclitaxel-induced cell apoptosis and injured cell organs were found by transmission electron microscopy.Conclusion Single local delivery of paclitaxel can inhibit biliary stricture in one month.
10.Changes of Neural Stem Cells in Subventricular in Developing Human Fetal Brain at Different Ages

Objective To observe the changes of neural stem cells(NSCs) in subventricular in developing human fetal brain at(diffe)-rent ages.Methods Thirty cases of embryoes at gestational age 24-28 weeks induced by labor with water bag were collected to determin distribution,shapes and growth modes in subventricular with hybridization in situ under light microscope.Results NSCs expressing Nestin mRNA existed in subventricular from human fetal brain at different ages,NSCs existed in subventricular of different fetal age included astro-NSCs,each had enations from 3 to 6,and all projections crowded together into a reticular plexiform,in which NSCs districuted.Nucli were round in shape,each had nucleoli from 1 to 4.NSCs had rarefaction chromatin,most NSCs existed in a single growth mode,symmetral cleavage growth mode and clony with 3 NSCs would be seen.There were no differences between positive Nestin mRNA NSCs in distribution and cell shapes,but had differences in growth mode.Conclusion NSCs exist in subventricular from different gestational ages and their growth mode are changing with difference of gestational age.
