1.Establishment of diabetic cystopathy guinea pig model and its urodynamic evaluation

BACKGROUND:Diabetic cystopathy is one of the most common chronic diabetic complications. The establishment of animal models of diabetic cystopathy wil provide experimental animal platform for relevant research.
OBJECTIVE:To establish a guinea pig model of diabetic cystopathy and to evaluate its urodynamic characteristics.
METHODS:Fifty short-hair Britain female guinea pigs were randomly divided into two groups, 42 as the experiment group and the other 8 as the control group. The experiment group was intraperitoneal y injected with streptozotocin to induce diabetes. The control group received injection of blank citric acid buffered solution. Diabetic guinea pigs were detected by urinary dynamics test at 9 and 12 weeks. Diabetic guinea pigs were further assigned into diabetic cystopathy subgroup and compensated subgroup. The urodynamic parameters of three groups were compared.
RESULTS AND CONCLUSION:Twenty of 42 guinea pigs were successful y induced diabetes by the injection of streptozotocin. At 9 weeks after the injection, bladder function compensation was present in six diabetic guinea pigs while bladder function was decompensated in another three diabetic guinea pigs. At 12 weeks, bladder function compensation was present in one diabetic guinea pig, while another eight guinea pigs were confirmed with diabetic cystopathy (88.89%). In the diabetic cystopathy subgroup, the residual urine volume was increased (0.72±0.08) mL, maximal detrusor pressure was decreased (0.63±0.05) kPa, maximum bladder capacity was increased (2.01±0.05) mL, and bladder compliance was increased (0.34±0.04) mL/kPa. There were significant differences compared with the compensated subgroup and the control group (P<0.001). Diabetic cystopathy occurs at 12 weeks after diabetic models are successful y established in guinea pigs, and urodynamic changes are mainly the increase of residual urine volume.
2.Clinical factors for operative effects in acetabular fracture: A Logistic regression analysis

Objective The aim of this study was to evaluate the clinical factors contributing to the operative effects in patients with acetabular fracture.Methods A total of 129 cases of patients with acetabular fracture were retrospectively analyzed from Jun 2006 to Jan 2011 in The Second Hospital of Shaoxing.Of the 129 acetabular fracture patients,96 cases were in the superior group and 33 patients in the inferior group according to Merled'Aubigne-Postel methods.The potential clinical factors related to the operative effects in acetabular fracture patients were evaluated by the method of logistic regression analysis.Results The superior rate was 74% in this study.Single factor analysis demonstrated that the diaplastic quality(x2=21.53,P <0.01),the femoral cartilage defect(x2 =17.50,P <0.01),the inra-aticular free bone(x2=12.18,P =0.0005),the type of fracture (x2 =7.24,P =0.0071) and the timing of operation (x2 =4.36,P =0.037) were correlated with operative effects in acetabular fracture patients.The binary logistic regression analysis indicated that non-anatomic diaplastic (OR =4.17,P =0.01)and the femoral cartilage defect(OR =3.11,P =0.03) were independent risk factors for operative effects in patients with acetabular fracture.Conclusions The prognosis of patients with acetabular fracture,which was complex,was affected by multiple factors.And introperatively,the anatomic reestablishment should be performed in order not to affect the post operative hip function.
3.Expression of miR-181b in Colitis-associated Colon Carcinogenesis

Background:miR-181b is related to the progression of many tumors,however,its expression in colitis-associated colon cancer is not clear. Aims:To investigate miR-181b expression and its potential target genes in the process of colitis-associated colon carcinogenesis. Methods: Colitis-associated colon cancer model was established by combined administration of azoxymethane(AOM)and dextran sulfate sodium(DSS)in mice. The expression of miR-181b in colon tissue at different time points was measured by real time fluorescent quantitative PCR(qPCR). Target genes of miR-181b were screened preliminarily by genome microarray combined with miRNA target gene prediction softwares TargetScan, PicTar and miRDB. Expressions of target genes in colitis-associated colon cancer mice were detected by qPCR. Results:The expression of miR-181b was gradually elevated during the development of colitis-associated colon cancer. However, AOM or DSS alone did not change expression of miR-181b. Genome microarray combined with miRNA target gene prediction softwares showed that Ipmk,E2f5,Klf6,Prkcd,Bai3,Hic2 might be the target genes of miR-181b. qPCR showed that expressions of Ipmk,Prkcd,Bai3,Hic2 were significantly decreased in colitis-associated colon cancer than in control group. Conclusions:Expression of miR-181b in colon is upregulated with the development of colitis-associated colon cancer,but is irrelevant with simple chronic inflammation.
4.Prevalence and associated risk factors of pre-hypertension and hypertension in young and middle-aged population in Nanjing

Objective:To investigate the prevalence and associated risk factors of pre-hypertension and hypertension in young and middle-aged population in Nanjing.Methods:Subjects of the study were those who underwent physical examination in the physical examination center of Nanjing Drum Tower Hospital from 2009 to 2016. The prevalence and risk factors of pre-hypertension and hypertension in young (aged 18-44 years old) and middle-aged people (aged 45-59 years old) were analyzed.Results:A total of 142 857 participants aged 18-59 years old were analyzed. Among them, 64 220 cases in the pre-hypertension group and 13 912 cases in the hypertension group. The prevalence of hypertension was 9.74% (12.51% in males and 5.82% in females). The prevalence of pre-hypertension was 44.95% (53.31% in males and 33.15% in females). In the middle-aged group, the prevalence of pre-hypertension and hypertension were 51.68% and 15.13%, respectively, which was higher than that in the young group (37.95% and 4.13%, respectively). The prevalence of pre-hypertension and hypertension in 2013-2016 was 45.37% and 10.65%, respectively, which was higher than that in 2009-2012(44.52% and 8.78%). In addition, the prevalence of abnormal blood glucose metabolism, abnormal blood lipid metabolism and abnormal glucose and lipid metabolism in the pre-hypertension group was higher than that in the normal blood pressure group, but lower than that in the hypertension group ( P<0.001). A logistic regression analysis indicated that age, overweight or obesity, hyperglycemia, hypertriglyceridemia and hypercholesterolemia were risk factors of pre-hypertension in male. Age, overweight or obesity, hyperglycemia, hypertriglyceridemia, hypercholesterolemia and hyper-low density cholesterolemia were associated with hypertension in male and with pre-hypertension and hypertension in female. Conclusions:Middle age, overweight/obesity, elevated fasting plasma glucose, elevated triglyceride and elevated total cholesterol were risk factors of pre-hypertension and hypertension in both men and women. Intervention on the related risk factors should be conducted as early as possible.
5.Application of quality control circle in reducing the incidence of complications in patients with peritoneal dialysis

Objective To explore the effect of quality control circle (QCC) on reducing the incidence of complications in the patients with peritoneal dialysis (PD). Methods Eighty-nine patients who underwent peritoneal dialysis after discharge were divided into QCC group (45 cases) and control group (44 cases) according to random sampling method. Both groups were followed up regularly, QCC activity was applied in patients of the QCC group on the basis of the QCC activity procedure. The intervention time was 1 year. The blood pressure, standard rate of urea clearance index, peritonitis incidence, catheter drift rate, infection incidences of external orifice of catheter, admission rate of complications and peritoneal dialysis withdrawal incidence between 2 groups were compared. Results After interference, the systolic and diastolic blood pressure in QCC group were significantly lower than those in control group:(124.11 ± 13.07) mmHg (1 mmHg=0.133 kPa) vs. (132.68 ± 12.09) mmHg and (72.13 ± 10.51) mmHg vs. (78.91 ± 11.79) mmHg, and there were statistical differences (P<0.05). The standard rate of urea clearance index in QCC group was significantly higher than that in control group:95.56%(43/45) vs. 81.82%(36/44), and there was statistical difference (P<0.05). After interference, the peritonitis incidence and admission rate of complications in QCC group was significantly lower than those in control group: 2.22% (1/45) vs. 13.64% (6/44) and 0 vs. 9.09% (4/44), and there were statistical differences (P<0.05). There were no statistical differences in catheter drift rate, infection incidences of external orifice of catheter and peritoneal dialysis withdrawal incidence between 2 group (P>0.05). The Spearman correlation analysis result showed that peritonitis was negatively correlated with education level (r = -0.28, P < 0.05), and standard rate of urea clearance index was positively correlated with education level (r = 0.21, P < 0.05). Conclusions The QCC management model can reduce the incidence of complications in patients with peritoneal dialysis.

Objective To observe expression of membrane-type 1 matrix metalloproteinase-1 (MT1-MMP) and matrix metalloproteinase-2 (MMP-2) and its relation to clinicopathological characteristics in cutaneous squamous cell carcinoma (SCC) and basal cell epithelioma (BCE). Methods The expression of MT1-MMP and MMP-2 was determined in situ hybridization and ABC immunohistochemical technique respectively in 48 cutaneous SCCs and 41 BCEs. Results The expression of MT1-MMP and MMP-2 in SCC was significantly higher than that in BCE (P 0.05). Conclusions Overexpression of MT1-MMP and MMP-2 might be related to lymphatic metastasis in SCC. Expression levels of MT1-MMP and MMP-2 may not be responsible for the differentiation in aggressiveness of BCE.

Objective: To observe the effect of extracorporeal shock wave therapy (ESWT) on proliferation, cell cycle and intercellular adhesion molecule-1 (ICAM-1) expression in human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were culturedin vitro at the concentration of (1×105/ml) and the cells were divided into 2 sets of groups:CSWT group, the cells were treated by different energy of (0.03, 0.09, 0.18, 0.24) mJ/mm2 respectively and corresponding Control group, in which the cells had no CSWT. HUVEC proliferation was detected by CCK colorimetric method, cell cycle was measured by lfow cytometry, mRNA and protein expressions of ICAM-1 were examined by RT-PCR and Western blot analysis respectively. Results: Compared with Control group, (0.09 mJ/mm2) CSWT group had promoted HUVECs proliferation,P<0.05 and the other CSWT groups were similar to corresponding Control groups,P>0.05; (0.09 mJ/mm2) CSWT group showed decreased proportion of G0/G1 stage and increased S and G2/M stages, allP<0.05; while (0.03 mJ/mm2) CSWT group only increased the proportion of G2/M stage,P<0.05 and the other CSWT groups were similar to corresponding Control group,P>0.05. Compared with Control group, (0.09 mJ/mm2 ) and (0.03mJ/mm2) CSWT groups showed increased mRNA expression of ICAM-1 (9.27±0.95) vs (1.02±0.27),P<0.001 and (7.08±0.60) vs (1.02±0.27),P<0.01; (0.09 mJ /mm2) CSWT group had elevated protein expression of ICAM-1,P<0.05. Conclusion: ESWT especially at (0.09 mJ/mm2) may accelerate cell cycle transition from G0/G1 stage to S and G2/M stages, promote HUVECs proliferation and increase ICAM-1 expression which may play important roles in ESWT facilitated angiogenesis in vitro.
9.Effects ofTianqi-Yizhi granules on oxidative stress in the brain tissue, and learning and memory in Alzheimer's disease model rats

Objective To investigate the effects ofTianqi-Yizhi granules on oxidative stress in the brain tissue, and learning and memory in Alzheimer's disease model rats.Methods A total of 90 male SD rats were randomly divided into 6 groups by random number table method: sham operation group, model group, huperzine A group and groups of low-, medium- and high-doseTianqi-Yizhi granules, with 15 rats in each group. The AD rat model was prepared by the left lateral ventricle injection of amyloid-β1-42. One week after modeling, the rats in the groups of low-, medium- and high-doseTianqi-Yizhi granules received intragastric administration of 0.8, 1.6 and 3.2 g/kgTianqi-Yizhi granules, respectively; the rats in the huperzine A group received intragastric administration of 0.02 mg/kg huperzine A solution; and the rats in the sham operation and model groups received intragastric administration of equivalent volume of normal saline for 30 days. Learning and memory were evaluated using the dark avoidance test. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and malonaldehyde (MDA) level in the brain tissue were determined. ResultsIn comparison with the model group, the latencies to step into the dark chamber in the high- and medium-dose Tianqi-Yizhi granules groups were significantly longer (239.05 ± 48.42 s, 214.35 ± 74.52 svs. 97.39 ± 30.69 s; allP<0.01), the numbers of errors significantly decreased (1.93 ± 3.25, 2.27 ± 3.09vs. 6.62 ± 3.45; allP<0.05), the activities of SOD (177.27 ± 63.10 U/mg, 164.53 ± 72.58 U/mgvs.72.56±21.04 U/mg; all P<0.01) and GSH-Px (2 899.36 ± 362.27 U/g, 2 407.68 ± 472.14 U/gvs. 1 397.64 ± 442.17 U/g; allP<0.01), and MDA level (24.75 ± 9.94 nmol/mg, 27.74 ± 5.82 nmol/mgvs. 37.56 ± 17.23 nmol/mg; allP<0.01) in the brain tissue significantly increased. ConclusionTianqi-Yizhi granules could attenuate oxidative stress in the brain tissue, and improve learning and memory in AD rats.
10.Effects of Tianqi Yizhi Granules on Mitochondria of Brain Tissues and Energy Metabolism ;in Alzheimer Rats

Objective To investigate the action mechanism of Tianqi Yizhi Granules for Alzheimer disease. Methods Ninety SD rats were divided into sham-operation group, model group, positive medicine group, high, medium and low dosage of Tianqi Yizhi Granules groups. SD rats were injected with aggregated amyloid β1-42 protein (Aβ1-42) into their left ventricle to establish AD models. Treatment groups received gavage with Huperzine A or Tianqi Yizhi Granules. The sham-operation group and model group received gavage with the same volume of normal saline. The mitochondrial membrane potential, activity of complex Ⅰ, Ⅱ, Ⅲ and Ⅳ of mitochondrial respiratory chain, enzymatic activity of Na+-K+ATP, energy charge, and space leaning ability were detected. Results Compared with model group, Tianqi Yizhi Granules could significantly improve mitochondrial membrane potential, activity of complex Ⅰ, Ⅲ and Ⅳ of mitochondrial respiratory chain, enzymatic activity of Na+-K+ATP, space leaning ability, and energy metabolism of brain in AD rats. Conclusion Tianqi Yizhi Granules showed mitochondrial protective capacity, and could improve energy metabolism of brain in AD rats, and then improve AD rats’ space learning ability.
