1.Experimental study of the differentiation of HPV16 subgenes-immortalized human endocervical cells induced by ATRA
Journal of Chongqing Medical University 1987;0(01):-
Objective:To study the differentiation-inducing effects of all-trans-retinoic acid (ATRA) on HPV16 subgenes-immortalized human endocervical cells (H8 cell) and the mechanism. Methods:After H8 cells were treated with ATRA,the aiti-proliferation of ATRA on them was tested by MTT assay, morphological changes observed by LM and TEM,cell cycle analyzed by FCM,expression of Ki67 tested by immunocytochemistry, the activity of telomerase tested by PCR-ELISA,and expressions of HPV and E6 tested by fluorescence immunocytochemistry. Results:ATRA could inhibit proliferation of H8 cells and induce their benign morphodifferentiation. According to FCM,H8 cells accumulated in G1 phase after being treated with ATRA,and expression of Ki67 and activity of telomerase reduced. Conclusion:ATRA could induce the differentiation of H8 cell line, which might be achieved by inhibiting proliferation, blocking cell cycle, and decreasing activity of telomerase.

6.Effects of incretin class of antidiabetic drugs on lipid metabolism in patients with type 2 diabetes and its mechanism
Chinese Journal of Endocrinology and Metabolism 2017;33(5):435-437
Chronic hyperglycemia is often accompanied by abnormal lipid metabolism, hypertension, low-grade systemic inflammation and oxidative stress. These factors increase the risk of cardiovascular disease(CVD)in type 2 diabetes mellitus(T2DM)patients.Comprehensive treatment of T2DM should emphasize the improvement of abnormal lipid metabolism, prevention of weight gain and reduction of the CVD risk in addition to proper glycemic control.Incretin, as a new hypoglycemic drug, has more advantages in improving glucose and lipid metabolism, finally to reduce cardiovascular risk.Through possible mechanisms including direct influence on liver lipid metabolism, change of fat mobilization and delay of gastric emptying, incretin shows positive influence on the lipid metabolic markers such as low-density lipoprotein-cholesterol, high-density lipoprotein-cholesterol, triglycerides, and total cholesterol, while it improves glucose control. Thus, incretin plays an important role in the comprehensive management of type 2 diabetes mellitus.

Chinese Journal of Dermatology 2011;44(5):350-351
Objective To investigate the role of vascular endothelial growth factor (VEGF) and transforming growth factor-α (TGF-α) in the angiogenesis of condyloma acuminatum (CA). Methods Tissue specimens were obtained from the lesions of 30 patients with CA and foreskin of 15 normal human controls. Reverse transcription (RT)-PCR and immunohistochemical method were utilized to measure the protein and mRNA expressions of VEGF and TGF-α in the specimens. Microvessel density (MVD) was determined by staining with anti-CD34 polyclonal antibodies. Results A statistical elevation was observed in the expression of VEGF and TGF-α mRNA as well as MVD in CA specimens compared with the normal control specimens (P < 0.001, 0.001, 0.01). The expressions of VEGF and TGF-α protein were observed in 90% and 86.7% of the CA specimens, and in 40% and 26.7% of the control specimens (both P < 0.01). MVD was positively correlated with the expression of VEGF and TGF-α protein in CA tissues (both P < 0.01). Conclusion The overexpression of VEGF and TGF-α in CA tissue may accelerate angiogenesis in, and participate in the development and progression of, CA.
