1.Evaluation on the effects of different dialysis treatment on femoral artery elasticity in uremia patients by Echo-tracking technique

Objective To evaluate the effects of different dialysis treatment on femoral artery elasticity in uremia patients by Echo-tracking technique. Methods 81 cases of uremia patients (42 peritoneal dialysis patients and 39 hemodialysis patients) and 31 cases of normal adults as control were recruited in this study. The Intima-media thickness (IMT), Internal systolic diameter (Ds), Internal diastolic diameter (Dd), Value difference between Dd and Ds (△D), Arterial stiffness (β), Pressure strain elastic modulus (Ep), Arterial compliance (AC), Pulse wave conducting velocity (PWVβ) and Augmentation index(AI) parameters were analyzed for evaluation the right femoral arterial elasticity. Results The parameters ofβ, Ep, PWVβin dialysis patients significantly increased (P<0.05) and their AC significantly decreased (P<0.05) compared with normal group. Theβand Ep in hemodialysis patients is higher that those patients not receiving dialysis (P<0.05). Theβand Ep in peritoneal dialysis patients is lower that those patients not receiving dialysis (P<0.05). Theβ, Ep and PWVβin hemodialysis patients are higher than those in peritoneal patients (P<0.05). And AC is lower than control group (P<0.05). In uremia patients, Vascular stiffness was positively correlated with age, duration of dialysis, IMT, pulse pressure, Dd, and negatively correlated with△D(P<0.05). Conclusions Femoral artery endothelial function in uremia patients was damaged earlier than morphological changes. Vascular intimal injury resulting from hemodialysis is more serious, and vascular elasticity decrease is correlated with age, dialysis duration and pulse pressure. Echo-tracking technique could provide quantification evidences for early diagnosis of the femoral artery endothelial dysfunction in uremia patients. The echo-tracking technique has certain significance in early intervention and improvement the prognosis, however, the influence factor of the accuracy of measurement should be consider during the procedure.
2.Renal safety of adefovir dipivoxil for two-year treatment in Chinese patients with chronic hepatitis B

Objective To assess the renal safety of adefovir dipivoxil (ADV) in Chinese patients with chronic hepatitis B (CHB).Methods A retrospective study was performed on 1 013 CHB patients from Chinese ADV multicenter clinical trials (ADF30001 and ADF106632).All patients were administrated with ADV 10 mg daily.The serum creatinine and phosphorus levels were measured in different time pointsduring 104-week treatment.Nephrotoxicity was defined as an increase ≥44.2 μ mol/L from baseline in serum creatinine or a serum phosphorus value of < 0.4845 mmol/L on two consecutive occasions.Paired t test was used to analyze serum creatinine and phosphorous data before treatment and at different time points during treatment.Results At week 28,week 52,week 80 and week 104,the median levels of serum creatinine were 74.963,76.996,76.820 and 77.969 μmol/L,respectively,and there were no significant differences from baseline (t =0.91,0.23,0.59 and 0.97,P > 0.05).No patient experienced a ≥ 44.2 μmol/L increase from baseline.For serum phosphorus,the median levels at week 28,week 80 and week 104 were 1.098,1.088 and 1.048 mmol/L,and there were significant decreases from baseline (t =2.34,3.06 and 4.94,P <0.05 or < 0.01).The cumulative incidence of serum phosphorus abnormality was 0.8% (8/104).There were no confirmed serum phosphorous decreases to < 0.4845 mmol/L.Conclusion Two years treatment with ADV (10 mg/d) does not result in marked nephrotoxicity,indicating that ADV 10 mg daily is well tolerated by Chinese patients.

Objective: To study the effects of the total flavonoids of tartary buckwheat germ (TFTBG) on serum lipid and antioxidation in hypercholesterolemic rats. Method: According to body weight,60 Wistar rats were divided randomly into 6 groups: normal control group (NC), high-fat control group (HFC), Jiaogulan positive control group (JPC, Gynostemma pantaphyllum total glucoside tablet 0.032g/kg bw),TFTBG 1.0,0.5and TFTBG 0.2 g/kg bw (HD,MD,LD) group. Except NC group, all other groups were fed high-fat diet for hyperlipidemia model. NC group and HFC group were given distilled water 10 ml/kg bw. 4.ANALYSIS OF FATTY ACID AND UNSAPONIFIABLE MATTERS FROM TARTARY BUCKWHEAT OIL AND BUCKWHEAT OIL

Objective: To analyse the composition and content of the fatty acid and unsaponifiable matters in both tartary buckwheat seed oil and buckwheat seed oil. Methods: Fatty acids were obtained by the methods of KOH-EtOH(95%) saponification and treatment of inorganic acid. They were analyzed and determined by gas chromatography and GC/MS chromatography respectively. Results: 4-5 kinds of fatty acid were determined in both oils, and the content of unsaturated fatty acids in tatary buckwheat oil was 83.2% ( oleic acid 47.1%, linoleic acid 36.1%), buckwheat oil was 81.8% ( oleic acid 35.8% , linoleic acid 40.2% and linolenic acid 5.8%). The content of unsaponifiable matters in tartary buckwheat seed oil was 6.56%(?-sitosterol 54.37%), and in buckwheat seed oil was 21.90%(?-sitosterol 57.29%,?-tocopherol 1.41%). Conclusion: Both buckwheat oils were functional vegetable oils.

Objective:To investigate the effects of titanium surface modified by micro-arc oxidation (MAO)on the collagen secre-tion and the expression of osteogenesis-related genes of MC3T3-E1 cells.Methods:Pure titanium discs were divided into two groups:Titanium with modified surface by MAO(MAO group)and titanium with polished surface(PT group).Tissue culture polysty-rene plates (TC)were used as controls.Surface morphology of the samples was examined by field-emission scanning electron micros-copy(FESEM).Surface roughness of samples was measured by surface roughness meter.MC3T3-E1 cells were cultured on the sur-face of the samples and the collagen secretion on the samples was measured by sirius red-based colorimetric microassay after 12 days of culture.On day 16 of culture,the expression of osteogenesis-related genes was examined by RT-PCR.Results:A porous oxide layer was observed on the surface of MAO treated samples and the surface roughness was more than that of PT group(P<0.01 ). Cells in MAO group showed higher collagen secretion than in PT or TC groups(P<0.05).MAO treated surface induced higher OSX, COL-Iα1 and OPN expression than PT.Conclusion:MAO treated titanium surface may induce more collagen secretion of MC3T3-E1 cells than PT.
7.MHC antigen expression on the surface of bone marrow stromal stem cells after directional induction in vitro

BACKGROUND:Alogeneic bone marrow stromal stem cel transplantation for treatment of bone diseases is a hot topic. To seek effective methods for prevention of post-transplantation immune rejection is urgent to be solved.
OBJECTIVE:To explore the expression of MHC antigen on the surface of bone marrow stromal stem cels after osteogenic inductionin vitro.
METHODS:Bone marrow samples were extracted from rabbits to in vitro isolate and culture bone marrow stromal stem cels. Then, the cels were cultured in IMDM medium containing bone morphogenetic protein-2. Flow cytometry was used to analyze the expression of MHC antigen on the osteoblasts differentiated from bone marrow stromal stem cels.
RESULTS AND CONCLUSION:There was highly expressed MHC I antigen but no MHCII antigen on the osteoblasts differentiated from bone marrow stromal stem cels. After osteogenic induction, no immune rejection was found. These findings indicate that alogeneic or xenogeneic bone marrow stromal cel transplantation can be used in the treatment of bone defects.

9.Cerebral protection of dexmedetomidine in patients with heart valve replacement under CPB

Objective To investigate the cerebral protection of dexmedetomidine in patients with heart valve replacement under CPB.Methods Totally 80 cases of patients with heart valve replacement under CPB in Seventh people's hospital of Zhengzhou city from January 2015 to December 2015 were selected and divided into observation group and control group,40 cases in each group.Patients in observation group were treated with dexmedetomidine before anesthetic induction and during the operation,and patients in control group were treated with same dose of saline solution.The arterio-venous oxygen content difference (Da-jvO2),jugular venous oxygen saturation (SjvO2),cerebral oxygen extraction rate (CERO2),serum myelin basic protein (MBP),serum neuron-specific enolase (NSE),and S1003 protein content before anesthetic induction (T0) and when the ascending aorta open (T1),After 10 min of CPB (T2) and postoperative 6 h (T3) were compared between two groups,and occurring rates of MMSE and POCD were also compared.Results SjvO2 values of two groups at T1 were significant higher than T0 (P < 0.05),Da-jvO2,and CERO2 were significant lower than T0 (P < 0.05),SjvO2 values of observation group at T1 and T2 were significant higher than those of control group and Da-jvO2 and CERO2 were significant lower than those of control group (P < 0.05);At T2 and T3,MBP levels of observation group and control group were significant higher than T0 (P < 0.05),and those in observation group were significant lower than control group (P < 0.05);At T1,T2,and T3 of observation group and control group were significant higher than T0 (P <0.05),and NSE levels of observation group at T1,T2,T3 were significant lower than those of control group (P < 0.05),S-100β levels of observation group at T2 and T3 were significant lower than those of control group (P < 0.05).After 7 d,MMSE score of observation group was significant higher than that of control group and POCD occurring rate was lower (P < 0.05).Conclusion Dexmedetornidine can improve cerebral ischemia hypoxia state and reduce brain damage in patients with heart valve replacement under CPB.
10.Bone regeneration with bone marrow mesenchymal stem cells, platelet rich plasma and decellularized cartilage matrices complex

Objective:To study the bone regeneration capacity of the complex of bone marrow mesenchymal stem cells(BMSCs) and platelet rich plasma (PRP) with decellularized cartilage matrix (DCM).Methods:BMSCs were isolated from young rabbit and cultured;PRP were prepared from the fresh blood of rabbit and the DCM were come from the fresh ears of rabbits and processed into a mixture of BMSCs-PRP-DCM.The mixture was injected subcutaneously into nude mice,8 weeks after injection bone regeneration was examine by HE staining and Massons staining decellularization.Results:The BMSCs show good differentiation capacity in vitro and the cartilage fragments were well decellularized.Excellent endochondral ossification ability of the complex was observed by in vivo experiment.Conclusion:The BMSCs-PRP-DCM complex has good capacity of endochondral ossification.
