1.Protective effects of ulinastatin on islets isolated by automated method from canine pancreas

Objective To study the protective effects of ulinastatin (UTI) on canine islets isolated by automated method and to determine whether the addition of UTI during the in situ pancreatic perfusion process may improve islet recovery. Methods There was no significant difference in donor-related factors (age, gender, warm ischemia time, cold storage time, weight and pancreas weight) between the two experimental groups. Twenty donors were randomly divided into two groups. Pancreases were in situ perfused via abdominal aorta using cold hypertonic citric potassium solution (HC-A) 2 500 ml, group 1 (HC-A group, n =10). The pancreases were perfused with cold HC-A and UTI ( 10 000 U/kg), group 2 (HC-A+UTI group, n =10). Intraductal collagenase V (4℃) delivery with controlled perfusion was done, the pancreas was dissociated in system of Ricordi Chamber and their purified islets were separated with Ficoll density gradient centrifugation. The digestion time, islets remaining trapped in exocrine tissue, final islet purity, insulin and C-pep secretory activity, and islet recovery were observed. The purified islets were observed under light microscopy and electronic microscopy. Results UTI supplementation did not affect the digestion time, islets remaining trapped in exocrine tissue, final islet purify, insulin and C-pep secretory activity and their ultrastructure, and islet recovery was increased. In the HC-A+UTI group the yield of islets was [( 6.17 ? 2.86 )?10 4] IEQ, while in the HC-A group, that was [( 3.42 ? 1.47 )?10 4] IEQ ( P
2.Cerebral protection of dexmedetomidine in patients with heart valve replacement under CPB

Objective To investigate the cerebral protection of dexmedetomidine in patients with heart valve replacement under CPB.Methods Totally 80 cases of patients with heart valve replacement under CPB in Seventh people's hospital of Zhengzhou city from January 2015 to December 2015 were selected and divided into observation group and control group,40 cases in each group.Patients in observation group were treated with dexmedetomidine before anesthetic induction and during the operation,and patients in control group were treated with same dose of saline solution.The arterio-venous oxygen content difference (Da-jvO2),jugular venous oxygen saturation (SjvO2),cerebral oxygen extraction rate (CERO2),serum myelin basic protein (MBP),serum neuron-specific enolase (NSE),and S1003 protein content before anesthetic induction (T0) and when the ascending aorta open (T1),After 10 min of CPB (T2) and postoperative 6 h (T3) were compared between two groups,and occurring rates of MMSE and POCD were also compared.Results SjvO2 values of two groups at T1 were significant higher than T0 (P < 0.05),Da-jvO2,and CERO2 were significant lower than T0 (P < 0.05),SjvO2 values of observation group at T1 and T2 were significant higher than those of control group and Da-jvO2 and CERO2 were significant lower than those of control group (P < 0.05);At T2 and T3,MBP levels of observation group and control group were significant higher than T0 (P < 0.05),and those in observation group were significant lower than control group (P < 0.05);At T1,T2,and T3 of observation group and control group were significant higher than T0 (P <0.05),and NSE levels of observation group at T1,T2,T3 were significant lower than those of control group (P < 0.05),S-100β levels of observation group at T2 and T3 were significant lower than those of control group (P < 0.05).After 7 d,MMSE score of observation group was significant higher than that of control group and POCD occurring rate was lower (P < 0.05).Conclusion Dexmedetornidine can improve cerebral ischemia hypoxia state and reduce brain damage in patients with heart valve replacement under CPB.
3.Effects of irradiation on proliferation and differentiation of MC3T3-E1 osteoblastic cells in vitro.

OBJECTIVEThe aim of this study is to investigate the effects of irradiation on the proliferation and differentiation of MC3T3-E1 osteoblastic cells.
METHODSMC3T3-E1 cells were irradiated 24 h after initial seeding. Gamma-radiation was administered at 0, 4, and 8 Gy as single doses by using a 60Co source. Cell proliferation was assessed at days 1, 3, 5, and 7 post-irradiation by using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenylte-trazolium bromide assay. The collagen secretion of the cells was measured through sirius red staining at day 12 post-irradiation. The expressions of osteogenesis-related genes were assessed through real time fluorescence quantitative polymerase chain reaction at day 16 post-irradiation. The matrix mineralization caused by cells was evaluated through alizarin red staining at day 28 post-irradiation.
RESULTSThe cells exposed to 4 Gy or 8 Gy demonstrated significantly lower proliferation rates compared with the non-irradiated group. Doses of 4 Gy or more significantly inhibited the expressions of osteogenesis-related genes (Osterix and osteocalcin). Collagen secretion and cell mineralization were significantly reduced by the 8 Gy dose.
CONCLUSION60Co gamma-rays dose-dependently suppress the proliferation, collagen secretion, and mineralization of MC3T3-E1 cells. Furthermore, radiation seems to dose-dependently inhibit the expressions of osteogenesis-related genes of the cells.

4.Effects of aspirin and verapamil alone or in combination on mesenteric microcirculation of rats

AimTo study the effects of aspirin (Asp) and verapamil(Ver)alone or in combination on mesenteric microcirculation of rats. MethodsAcute microcirculation disturbance(AMD) was produced by highmolecular weight dextran(Mr 480, 000) 360 mg· kg-1 iv. Arteriol and venul blood flow velocity and diameter (ABFV, VBFV, AD, VD) and blood flow state(BFS) were observed by intravital microcirculation method. Results Asp 2.5, 5 mg · kg-i, Ver 0.3, 0.6 mg · kg-1,Asp+ Ver(1 + 0.15), (2. 5+ 0. 3) mg· kg-1 iv significant increase ABFV by 11.1%, 31.3%, 18.7%,19.5%, 26.5%, 37.3%, VBFV by 12.5%, 5.7%, 2.5%, 3.7%, 30%, 34.7%,AD by 4.3%, 17.9%, 31.5%, 35%, 20%, 38.1% and VDby 2.2%,4.2%, 25%, 31.5%,5.8%,23.5%espectively, and got a raise in the number of capillaries and a marked improvement of BFS. Asp + Ver(1+0.15, 2.5+0.3) mg · kg-1 iv could reverse AMD. Conclusion Asp is superior to Ver in the increase of BFV, but Ver is superior to Asp in expansion of blood vessel. Asp in combination with Ver produces marked synergistic action and protection againist AMD.
5.Probe into sleep quality in the patients with irritable bowel syndrome

Objective To investigate the sleep features in the patients with irritable bowel syndrome (IBS) and compare the sleep quality between those IBS patients who were with and without anxiety and depression.Methods Pittsburgh sleep quality index questionnaire (PSQI), self-rating anxiety scale (SAS) and self-rating depression scale (SDS) were measured in the 145 IBS patients and 59 regular physical examination volunteers.IBS patients were also divided into two subgroups-patients with or without anxiety and depression based on cutoff scores of SAS and SDS.Comparisons of sleep quality were made between subgroups, and between IBS patients and volunteer controls.Results Compared with the controls,the SAS raw score, SDS raw score and SAS positive incidence in IBS patients were shown statistically significant differences (P < 0.05 ), while the SDS positive incidence had no statistically significant difference(P > 0.05 ).PSQI total scores were significantly higher in the IBS patients without anxiety and depression (P < 0.05), 3 domains (sleep quality, sleep disturbances and daytime function disorder) were also found statistically significant differences ( P < 0.05 ), compared with the controls.The IBS patients with anxiety and depression were statistically significantly different from the controls ( P < 0.05 ) in 6 domains (sleep quality, sleep latency, sleep efficiency, sleep disturbances, sleep time and daytime function disorder) and significantly higher PSQI total scores( P < 0.05 ).Statistically significant differences (P <0.05) were also found in all 7 domains and with higher PSQI total scores in IBS patients with anxiety and depression, compared with IBS patients without anxiety and depression.Conclusions IBS patients were more likely to have sleep abnormality, mainly in sleep quality, sleep disturbances and daytime function disorder and PSQI total scores.The abnormalities of these factors were independent of emotional disorder.However, emotional disorder worsened the sleep disorder in IBS patients.
6.In vitro differentiation of islet-like cells from mouse bone-marrow stem cells on a PLGA scaffold

Objective To investigated the potential influences of poly(lactic-co-glycolicacid)(PLGA)scaffold as a platform on the differentiation of mouse bone-marrow stem cells to islet-like cells.Methods Mouse bonemarrow stem cells were grown and differentiated in culture with or without PLGA scaffold,and cell morphology and functions were compared within these groups.Results The PLGA scaffold showed fine biological compatibility.Differentiated islet-like cells were dithizone (DTZ) positive,insulin and C-peptide double positive,glucagon positive and somatostatin positive in both groups.Under electron microscope there were ultrastructures similar to that of islet β cells in cells of both groups.Cells with PLGA scaffold secreted more insulin under high level glucose stimulation (P<0.01).Conclusions PLGA scaffold was biologically compatible and improved function of the differentiated islet-like cells.

Objectives:To investigate the effect of TP 5 on the immune function and infection rate in patients undergoing cardiopulmonary bypass(CPB) operation. Methods:Ninety four patients were divided into two groups to observe the effect of TP 5 on the immune function and infection rate. Results:The immunological parameters in the TP 5 group was better than the control group, and postoperative infection was prevented. Conclusions:TP 5 can improve patients' cell mediated immunity and reduce infection rate.
10.Quality Control and Management of Pathological Tissue Dehydration

Objective To study full-automatic tissue hydroextractor that its factors of influence anhydration quality in the process of anhydration. Methods The quality of tissue hydroextractor was increased by various angles of reagent examination, correct clipping and standard management etc. Results The quality of hydroextrator and the high quality of tissue section was improved by replacing reagents timely, clipping tissue properly, canonical management and performing specification and maintenance of the machinery, and the efficiency and accuracy of pathological diagnosis was enhanced too. Conclusion In foundation of strictly quality control, the advanced machine integrates with the skill and sense of responsibility of the operators. The quality of tissue dehydration can be improved effectively.
