2.Effect of minimally invasive percutaneous nephrolithotomy titanium laser lithotripsy for upper ureteral calculi

Objective To analyze the clinical effect of minimally invasive percutaneous nephrolithotomy titanium laser lithotripsy(MPCNL)on upper ureteral calculi.Methods According to digital table,68 patients with upper ureteral calculi were randomly divided into the study group and control group,cach group in 34 cases.The study group received the MPCNL treatment,the control group received extracorporeal shock wave lithotripsy(ESWL)therapy. The therapeutic effects were compared.Results The success rate of gravel,stone clearance rates of the study group were 91 .1 8% and 91 .1 8%,which were higher than 70.59%,67.65% of the control group,there were significant differences between the two groups(χ2 =4.660,5.757,P =0.031 ,0.0164).Conclusion Percutaneous nerve titanium laser lithotripsy for upper ureteral stones can achieve good therapeutic effect.
3.Determination of Protocatechuic Acid and Protocatechuic Aldehyde in Fuxuekang Granules by RP-HPLC

OBJECTIVE: To establish a RP-HPLC method for the simultaneous determination of Protocatechuic acid and Protocatechuic aldehyde in Fuxuekang granules. METHODS: The chromatography was carried on Achirom Bond-1 C18 column(250 mm?4.6 mm,5 ?m). The mobile phase was composed of methanol - water (20∶80,pH=2.80 adjusted by glacial acetic acid) at a flow rate of 1.0 mL?min-1. The detection wavelengths were programmed at 256 nm for Protocatechuic acid and at 280 nm for Protocatechuic aldehyde. RESULTS: The linear ranges for Protocatechuic acid and Protocatechuic aldehyde were 5.49~73.22 ?g?mL-1(r=0.999 9) and 4.81~64.13 ?g?mL-1(r=0.999 9), respectively, and their average recoveries were 96.86%(RSD=2.52%,n=9) and 97.55%(RSD=3.82%,n=9) respectively. CONCLUSION: The method is simple, sensitive, and reproducible, and it is applicable for the quality control of Fuxuekang granules.
4.Construction of novel thioredoxin fusion protein expression system and the production of recombinant Lf-CATH2.

The objective of this study was to construct an improved thioredoxin fusion protein expression system, and express the cathelicidin-derived peptide, Lf-CATH2. The improved fusion vector Lf-CATH2-pET32α(-TS) was successfully constructed by firstly deleting the thrombin site and S tag from the pET-32α vector, then inserting the Lf-CATH2 plus a thrombin site instead. Afterwards, Lf-CATH2 was expressed in Escherichia coli as fusion protein. After the cleavage by thrombin, Lf-CATH2 was released and subsequently separated using affinity chromatography. The antimicrobial activity of purified Lf-CATH2 was also examined. The improved expression vector significantly increased enzyme cleavage efficiency by 37%, and Lf-CATH2 could be expressed in high yield and maintain the biological activity. This novel thioredoxin fusion protein expression system enables a quick production of high-yield bioactive cationic peptides like cathelicidins.

5.Histogenesis,differential diagnosis and prognosis of melanotic schowannoma

Purpose:To study the histogenesis,the differential diagnosis and the prognosis of melanotic schwannoma.Methods:The age of the patients was 4,17,28 years old, respectively.Three cases of melanotic schwannoma were examined using histologic (HE), immunohistochemical (ABC method) and electron microscopic techniques. Results: The diameter of tumor was 2,4,8, respectively. Histological examination showed pigmented spindle cells arranged in interlacing fascicles and occasional nuclear palisading. The pigmented polygonal cells were loosely arranged in sheets. After bleaching, the nuclei were oval and some showed slight pleomoprohism with prominent nucleoli, and nuclear mitoses were found. There was local invasion in two cases. Immunohistochemical stains for S 100 protein, HMB45, Vim, Leu 7, NSE were positive. Ultrastructrual examinations showed a lot of melanosomes in all stages of development and seldom basal lamina. Conclusions:①Melanotic schwannoma usually occurs in young adults. Most of the tumors are benign or only potentially malignant. Malignant MS is rare,and require follow up. ②MS originate from neural crest with a proliferation of the bipotential cell.
6.Expression of DLC1 mRNA and protein in breast carcinoma and non-cancerous breast tissues and its relationship with Ki-67

Purpose To investigate the expression of DLC1 and its relationship with Ki-67 in cancerous and non-cancerous tissues of the breast.Methods In situ hybridization and immunohistochemiscal EnVision method were used to detect the expression of DLC1 mRNA and protein and Ki-67 in 52 invasive breast ductal carcinomas and 42 non-cancerous mammary tissues, including 22 mammary fibroadenomas and 20 paracancerous tissues.Results The positive rates of DLC1 mRNA and protein expression in the breast carcinomas (50% and 57.7%) was significantly lower than that in the non-cancerous mammary tissues (90.5% and 92.9%) (χ~2=17.518 and 10.729,P<0.01).The expression of DLC1-mRNA was positively related to DLC1protein (r_s=0.379,P<0.01). The positive rate of Ki-67 expression was 61.5% in the breast carcinomas, but no expression was observed in the all non-cancerous tissues (χ~2=39.186,P<0.01).Correlation analysis showed that DLC1 expression was negatively correlated with Ki-67 expression (r_s=-0.507,P<0.01).Conclusions Lower or no expression of DLC1 mRNA and protein may play an important role in the pathogenesis and progression in breast carcinoma. DLC1 may inhibit the proliferation of the breast carcinoma cells,which indicates that it may act as a new molecular marker of breast carcinoma.Combining detection of DLC1 and Ki-67 may be useful parameters for evaluating the biological behaviors of breast carcinoma.
7.Efficacy and safety of CT-guided percutaneous catheter drainage in treatment of uniloculated deep neck abscesses

Twenty-three patients with uniloculated deep neck abscesses (UDNA),in whom the antibiotic therapy failed and CT-guided percutaneous catheter drainage (PCD) was performed from January 2005 to June 2015,were included in the study.Catheter placement was carried out using Trocar technique in all cases.Open surgical drainage was performed when PCD procedures failed.The abscess was completely drained and open surgical drainage was avoided in 19 cases (83%);the surgical drainage was performed because of muhiple internal septation in 3 (13%) case,and 1 (4%) case died from uremia.In this series the technical success rate and clinical success rate of PCD were 96% (22/23) and 83% (19/23),respectively.All patients were followed-up by CT scan.No other complications and no mortality occurred during the procedure,while postoperative pneumatosis developed in 1 case.CT-guided PDC is a safe and highly effective low-cost procedure for the treatment of patients with UDNA who failed medical therapy,it may be considered as an alternative to open surgery.
8.Retroperitoneal laparoscopic partial nephrectomy without blockage of renal pedicle for the treatment of uncomplex renal tumor

Objective To evaluate the efficacy and clinical significance of partial laparoscopic resection of renal pedicle without renal pedicle vascular resection in the treatment of low renal complex tumors.Methods Retrospective analysis of retroperitoneal laparoscopic renal artery clamping without partial nephrectomy in the treatment of the patients with low-grade complex renal tumors in 10 cases(observation group) and the clinical data of retroperitoneal laparoscopic renal pedicle occlusion of renal vascular resection in 10 patients (control group) were analyzed.The amount of bleeding,operation time,the average hospitalization time,drainage tube placement after operation,the hemoglobin (Hb),serum creatinine(Ccr) and glomerular filtration rate(GFR) changes were observed in the two groups.Results The operation time,bleeding volume,postoperative drainage tube placed time,postoperative hospitalization time of the observation group were (103.0 ± 42.7) min,(100.0 ± 70.4) mL,(3.5 ± 1.2) d,(5.0 ± 1.2) d,respectively,which of the control group were (129.2 ± 57.5) min,(89.2 ± 9.5) mL,(6.5 ± 9.7) d,(6.3 ± 3.4) d,respectively,there were statistically significant differences between the two groups (t =4.23,3.23,1.57,4.24,all P <0.05).Before operation,the Hb,Ccr and GFR between the two groups had no statistically significant differences(all P <0.05).After operation,the Hb,Ccr and GFR of the control group were (85.7 ± 18.2) g/L,(77.9 ± 22.1) μmol/L,(61.4 ± 50.9) mL · min-1 · 1.73 (m2)-1,respectively,which of the observation group were (95.6 ± 13.5) g/L,(70.2 ± 10.5) μmol/L,(85.5 ± 5.1) mL · min-1 · 1.73 (m2)-1,respectively,there were statistically significant differences between the two groups(t =9.34,7.44,7.34,all P <0.05).Conclusion Retroperitoneal laparoscopic renal partial nephrectomy without blocking renal pedicle vessels is a safe and effective method for the treatment of low complexity renal tumors.It has advantages of simple operation and is beneficial to the recovery of patients'function.
9.Exogenous carbon monoxide protects against the lung injury induced by ischemia-reperfusion of hind limbs in rats

AIM: To study the mechanism of protective effect of exogenous carbon monoxide (CO) in the lung injury induced by ischemia-reperfusion (IR) of hind limbs in rats. METHODS: Thirty-two SD rats were randomly divided into 4 groups: control, control+CO, IR and IR+CO. A rat model of ischemia in hind limbs and the reperfusion lung injury was made. The rats in IR+CO and control+CO groups were exposed to air containing 2.5?10~ -8 CO for 1 h before reperfusion or the corresponding control time point, while the other two groups were exposed to the routine air. The lung tissue structure, polymorphonuclear leukocyte (PMN) count, wet-to-dry weight ratio (W/D), malondialdehyde (MDA) content and the animal survival rate were observed. The carboxyhemoglobin (COHb) levels in artery blood were detected with CO-oximeter and the expression of intercellular adhesion molecule-1 (ICAM-1) in the lung was detected by Western blotting. RESULTS: Compared to control, the animal mortality, lung PMNs number, W/D , MDA content and ICAM-1 expression were all significantly increased in IR group. Compared with the IR group, the blood COHb level was significantly increased and the animal mortality, lung PMNs number, W/D, MDA content and ICAM-1 expression were all significantly decreased in IR+CO group. CONCLUSION: These data suggest that exogenous CO attenuate limb IR- induced lung injury by down-regulatiny ICAM-1 expression and suppressing PMN sequestration in the lung following limb IR in rats.
10.Change of c-fos in CCK-8 against the pulmonary artery smooth muscle cell injury induced by LPS

AIM: To observe the role of c-fos in the protection by cholecystokinin-octapeptide (CCK-8) against pulmonary artery smooth muscle cell (PASMC) injury induced by lipopolysaccharide (LPS). METHODS: The ultrastructure of PASMC was observed under transmission electron microscope (TEM). MDA content,LDH release and the rate of trypan blue in PASMC were measured,and immunocytochemistry technique was adopted to observe the c-fos protein expression. RESULTS: The TEM results showed significant PASMC structural injury in LPS group and alleviated structural changes in LPS+CCK-8 group. CCK-8 reduced the increase in the rate of trypan blue uptake,MDA content and LDH release in PASMC induced by LPS. LPS lightly increased c-fos protein expression,which was enhanced by CCK-8. CONCLUSION: CCK-8 attenuated the injury of PASMC induced by LPS,which may be concerned with the increase in c-fos protein expression.
